1塔里木盆地库车坳陷烃源灶特征和天然气成藏过程显示文摘根据模拟实验数据,建立了库车坳陷典型源岩天然气产率和碳同位素的化学动力学参数。基于探井和人工井(13条地震剖面共230个人工井点)地质数据和今、古地温数据,采用BasinMod软件,分析了各井点成熟度和天然气产率随时间的变化规律,查明了三叠、侏罗系源岩的生烃历史和烃源灶的演化特征,指出,库车坳陷中生界源岩自新近纪以来快速埋藏和成熟,特别是5×106a以来,上新统库车组快速沉积,引起成熟度急剧增高,进入干气阶段,表现为5×106a以来气源灶极高的生气速率,这是库车坳陷形成高效天然气藏的重要原因。库车坳陷的前缘隆起带和克依构造带捕获了同一腐殖型气源灶(T-J)不同阶段生成的天然气;前缘隆起带的牙哈凝析气田主要捕获了生油窗阶段近源生成的天然气,表现为天然气甲烷碳同位素相对较轻;克拉2气藏主要是晚期(5×106a以来)捕获了镜质体反射率在1.0%~2.5%阶段生成的天然气,表现为天然气甲烷碳同位素相对较重。王飞宇 杜治利 张水昌 张宝民 陈建平 Bernhard Cramer 2009新疆石油地质2009,30,4:18
2煤系有机质多阶段成气的分子碳同位素表征及其对高过成熟干酪根生气潜力评价的启示显示文摘采用开放体系在线程序升温裂解-色谱-碳同位素比值质谱技术(PY/GC-IRMS)考察了煤系有机质热演化过程中甲烷瞬时生成速率与碳同位素组成特征,提出煤系有机质不同热演化进程甲烷生成的四个阶段:生油窗阶段、主生气阶段、晚期生气阶段(芳环侧链断裂)、开环-缩聚作用阶段。以一级反应动力学为基础,采用高斯分布算法,计算了各阶段甲烷产出的动力学参数。研究结果对煤成气资源量的正确评价,特别是近年来有机质高演化阶段成气潜力的评价具有重要指示意义。孙永革 杨中威 Bernhard CRAMER 2013地球化学2013,42,2:9
3To stay or to leave: Stem cells and progenitor cells navigating the S1P gradient显示文摘Most hematopoietic stem progenitor cells (HSPCs) reside in bone marrow (BM), but a small amount of HSPCs have been found to circulate between BM and tissues through blood and lymph. Several lines of evidence suggest that sphingosine-1-phosphate (S1P) gradient triggers HSPC egression to blood circulation after mobilization from BM stem cell niches. Stem cells also visit certain tissues. After a temporary 36 h short stay in local tissues, HSPCs go to lymph in response to S1P gradient between lymph and tissue and eventually enter the blood circulation. S1P also has a role in the guidance of the primitive HSPCs homing to BM in vivo, as S1P analogue FTY720 treatment can improve HSPC BM homing and engraftment. In stress conditions, various stem cells or progenitor cells can be attracted to local injured tissues and participate in local tissue cell differentiation and tissue rebuilding through modulation the expression level of S1P1, S1P2 or S1P3 receptors. 4Relative quantification of phosphoproteomic changes in grapevine (Vitis vinifera L.) leaves in response to abscisic acid显示文摘In a previous transcriptomic analysis,abscisic acid(ABA)was found to affect the abundance of a number of transcripts in leaves of Cabernet Sauvignon grapevines with roots that had been exposed to 10μM ABA for 2 h.Other work has indicated that ABA affects protein abundance and protein phosphorylation as well.In this study we investigated changes in protein abundance and phosphorylation of Cabernet Sauvignon grapevine leaves.Protein abundance was assessed by both label-free and isobaric-label quantitive proteomic methods.Each identified common proteins,but also additional proteins not found with the other method.Overall,several thousand proteins were identified and several hundred were quantified.In addition,hundreds of phosphoproteins were identified.Tens of proteins were found to be affected in the leaf after the roots had been exposed to ABA for 2 h,more than half of them were phosphorylated proteins.Many phosphosites were confirmed and several new ones were identified.ABA increased the abundance of some proteins,but the majority of the proteins had their protein abundance decreased.Many of these proteins were involved in growth and plant organ development,including proteins involved in protein synthesis,photosynthesis,sugar and amino-acid metabolism.This study provides new insights into how ABA regulates plant responses and acclimation to water deficits.Supakan Rattanakan Iniga George Paul A Haynes Grant R Cramer 2016Horticulture Research2016,3,1:3
5Cholangiocarcinoma, gone without the Wnt?显示文摘Cholangiocarcinoma(CCA) is a relatively rare malignancy of the intra- or extra-hepatic bile ducts that is classified according to its anatomical localization as intrahepatic, perihilar or distal. Overall, CCA has a dismal prognosis due to typical presentation at an advanced irresectable stage, lack of effective non-surgical treatments, and a high rate of disease recurrence. CCA frequently arises on a background of chronic liver inflammation and cholestasis. Chronic inflammation is accompanied by enhanced cell turnover with generation of additional inflammatory stimuli, and a microenvironment rich in pro-inflammatory mediators and proliferative factors that enable accumulation of mutations, transformation and expansion of mutated cells. A recent study by Boulter et al implicates the Wnt signaling cascade in cholangiocarcinogenesis. Wnt ligands Wnt7 B and Wnt10 A were found to be highly overexpressed in human CCA tissue. Wnt7 B protein was present throughout the tumor stroma, and often co-localized with a subset of CD68+ macrophages. To address in a direct manner whether Wnt signaling is engaged in development of CCA, Boulter et al explored the Wnt signaling pathway in an experimental model that recapitulates the multi-stage progression of human CCA.Wnt ligands found to be elevated in human CCA were also upregulated during the course of CCA development following thioacetamide treatment. Wnt10 a increased during the(pre-cancerous) regenerative phase, while Wnt7 b induction paralleled tumor growth. Along with upregulation of target genes, the findings demonstrate that the canonical Wnt pathway is progressively activated during cholangio-carcinogenesis. Macrophage depletion,eliminating a major source of Wnt7 b, prevented activation of the canonical Wnt cascade, and resulted in reduced number and volume of tumors in this model. Moreover,specific inhibitors of the canonical Wnt pathway(ICG-001 and C-59) caused reduction of tumor area and number,in xenograft and thioacetamide models of CCA. The aggregated findings show that experimental, and presumably human CCA, is a Wnt-driven tumor. 6A rapid dehydration leaf assay reveals stomatal response differences in grapevine genotypes显示文摘A simple and reliable way of phenotyping plant responses to dehydration was developed.Fully-developed leaves were detached and placed in a closed plastic box containing a salt solution to control the atmospheric water potential in the container.Three hours of dehydration(weight loss of the leaf)was optimal for measuring changes in stomatal response to dehydration.Application of the plant hormone abscisic acid(ABA)prior to leaf detachment decreased the amount of water loss,indicating that the assay was able to detect differences based on a stomatal response to dehydration.Five different Vitis genotypes(V.riparia,V.champinii,V.vinifera cv.Shiraz,V.vinifera cv.Grenache and V.vinifera cv.Cabernet Sauvignon)with known differences in drought tolerance were screened for their dehydration response and the results obtained corresponded to previous reports of stomatal responses in the vineyard.Significant differences in stomatal density along with differences in the amount and rate of water lost indicate differences in dehydration sensitivity among the genotypes screened.Differences in stomatal response to ABA were also detected.Shiraz had the lowest stomatal density and the highest ABA sensitivity among the genotypes screened,yet Shiraz lost the most amount of water,indicating that it was the least sensitive to dehydration.Despite having the highest stomatal density and intermediate stomatal sensitivity to ABA,V.riparia lost the smallest amount of water,indicating that it was the most sensitive to dehydration.The assay presented here represents a simple and reliable phenotyping method for plant responses to leaf dehydration.Daniel W Hopper Ryan Ghan Grant R Cramer 2014Horticulture Research2014,1,1:2

10Preliminary Estimation of Paleoproductivity via TOC and Habitat Types:Which Method Is More Reliable?—A Case Study on the Ordovician-Silurian Transitional Black Shales of the Upper Yangtze Platform, South China显示文摘New total organic carbon (TOC) data from the two Ordovician-Silurian transitional graptolite-bearing black shale intervals, the Wufeng (五峰) Formation and the Longmaxi (龙马溪) Formation in Central Guizhou (贵州) and West Hubei (湖北), respectively, as well as previously reported TOC data from the same intervals in other places on the Yangtze platform of South China, have been used to produce an initial estimate of the primary paleoproductivity via a conventional inverse method (i.e., Rpp-inverse). The values of the Rpp-inverse are estimated to be 32 (43-21) gC/(m2·a) (Wufeng Formation) and 21 (27-16) gC/(m2·a) (Longmaxi Formation). Also, simultaneously, the habitat types (i.e., HT; cf., BA: benthic assemblage) and their temporal and spatial changes have been documented from the same succession, and an initial estimate of the primary paleoproductivity has been produced using a forward method (i.e., Rpp-forward). Being bounded mainly by the peritidal to inner-shelf environment shelly-facies or mixed-facies successions with BA1 to BA3 faunas both at the top and the base, which indicates the habitat types from HT II1 to HT III2, the biohabitat type of the two graptolite-bearing black shale intervals can be limited to HT III to HT IV, corresponding to the inner shelf to the outer shelf, with depths from roughly 60 m to 200-300 m. Based on the current data from the South China Sea and the southern part of the East China Sea, values of Rpp-forward should be about 100 to 400 gC/(m2·a).
