2Inositol Pyrophosphate lnsP8 Acts as an Intracellular Phosphate Signal in Arabidopsis显示文摘The maintenance of cellular phosphate(Pi)homeostasis is of great importance in living organisms.The SPX domain-containing protein 1(SPX1)proteins from both Arabidopsis and rice have been proposed to act as sensors of Pi status.The molecular signal indicating the cellular Pi status and regulating Pi homeostasis in plants,however,remains to be identified,as Pi itself does not bind to the SPX domain.Here,we report the identification of the inositol pyrophosphate lnsP8 as a signaling molecule that regulates Pi homeostasis in Arabidopsis.Polyacrylamide gel electrophoresis profiling of InsPs revealed that lnsP8 level positively cor­relates with cellular Pi concentration.We demonstrated that the homologs of diphosphoinositol pentaki-sphosphate kinase(PPIP5K),VIH1 and VIH2,function redundantly to synthesize lnsP8,and that the vih1 vih2 double mutant overaccumulates Pi.SPX1 directly interacts with PHR1,the central regulator of Pi star­vation responses,to inhibit its function under Pi-replete conditions.However,this interaction is compro­mised in the vih1 vih2 double mutant,resulting in the constitutive induction of Pi starvation-induced genes,indicating that plant cells cannot sense cellular Pi status without lnsP8.Furthermore,we showed that lnsP8 could directly bind to the SPX domain of SPX1 and is essential for the interaction between SPX1 and PHR1.Collectively,our study suggests that lnsP8 is the intracellular Pi signaling molecule serving as the ligand of SPX1 for controlling Pi homeostasis in plants.Jinsong Dong Guojie Ma Liqian Sui Mengwei Wei Viswanathan Satheesh Ruyue Zhang Shenghong Ge Jinkai Li Tong-En Zhang Christopher Wittwer Henning JJessen Huiming Zhang Guo-Yong An Dai-Yin Chao Dong Liu Mingguang Lei 2019Molecular Plant2019,12,11:8
3Meta-analysis of the effects of 1-methylcyclopropene(1-MCP)treatment on climacteric fruit ripening显示文摘1-Methylcyclopropene(1-MCP)is an inhibitor of ethylene perception that is widely used to maintain the quality of several climacteric fruits during storage.A large body of literature now exists on the effects of 1-MCP on climacteric fruit ripening for different species and environmental conditions,presenting an opportunity to use meta-analysis to systematically dissect these effects.We classified 44 ripening indicators of climacteric fruits into five categories:physiology and biochemistry,quality,enzyme activity,color,and volatiles.Meta-analysis showed that 1-MCP treatment reduced 20 of the 44 indicators by a minimum of 22%and increased 6 indicators by at least 20%.These effects were associated with positive effects on delaying ripening and maintaining quality.Of the seven moderating variables,species,1-MCP concentration,storage temperature and time had substantial impacts on the responses of fruit to 1-MCP treatment.Fruits from different species varied in their responses to 1-MCP,with the most pronounced responses observed in rosaceous fruits,especially apple,European pear fruits,and tropical fruits.The effect of gaseous 1-MCP was optimal at 1μl/l,with a treatment time of 12–24 h,when the storage temperature was 0℃for temperate fruits or 20℃for tropical fruits,and when the shelf temperature was 20℃,reflecting the majority of experimental approaches.These findings will help improve the efficacy of 1-MCP application during the storage of climacteric fruits,reduce fruit quality losses and increase commercial value.Jing Zhang Yuanchun Ma Chao Dong Leon A.Terry Christopher B.Watkins Zhifang Yu Zong-Ming(Max)Cheng 2020Horticulture Research2020,7,1:7

5A new single-cell protein from Clostridium autoethanogenum as a functional protein for largemouth bass(Micropterus salmoides)显示文摘Clostridium autoethanogenum protein(CAP)is a new single-cell protein source originating from inactivated bacteria.An in vitro digestion experiment and an 8-wk growth experiment were conducted to evaluate the molecular weight distribution of the CAP hydrolysate,and the effects of dietary CAP levels on the growth performance,plasma parameters,hepatic and intestinal health,and the diversity of gutadherent microbiota of largemouth bass(Micropterus salmoides).The fish(initial body weight of 47.99±0.01 g)were fed diets where CAP gradually replaced 0%(CAP0),12.5%(CAP12.5),25%(CAP25),37.5%(CAP37.5)and 50%(CAP50)of low-temperature steam dried anchovy fish meal(LTFM)in the diet.Results showed that the content of peptides below 1,000 Da in the CAP hydrolysate(0.56 mg/mL)was higher than that of the LTFM hydrolysate(0.48 mg/mL).Dietary CAP inclusion had no negative effect on growth performance,while whole-body lipid content significantly reduced in the CAP25 and CAP50 groups(P<0.05).The plasma alanine aminotransferase activities and triglyceride concentrations in the CAP inclusion groups were significantly lower than those in the CAP0 group(P<0.05).The plasma aspartate aminotransferase activity was significantly reduced in the CAP37.5 group(P<0.05).The richness and diversity of the gut-adhesive microbiota and the proportion of Clostridium sensu stricto 12 in the CAP50 group were significantly higher than those in the CAP0 group(P<0.05).Dietary CAP inclusion inhibited inflammatory responses by down-regulating the mRNA levels of interleukin 1b(IL1b),IL10 and transforming growth factor b1(P<0.05)in the liver.The mRNA levels of acetyl-CoA carboxylase 1 were significantly down-regulated in the CAP12.5,CAP25 and CAP37.5 groups(P<0.05),while that of fatty acid synthase was significantly down-regulated in the CAP50 group(P<0.05).These results demonstrate that dietary CAP inclusion could improve the hepatic and intestinal health of largemouth bass,and can be helpful to further develop CAP as a functional feed ingredient.Shifeng Ma Xiaofang Liang Pei Chen Jie Wang Xu Gu Yuchang Qin Christophe Blecker Min Xue 2022Animal Nutrition2022,,3:6

7Effects of grazing intensity on seed production of Caragana stenophylla along a climatic aridity gradient in the Inner Mongolia Steppe,China显示文摘As the increases of climatic aridity and grazing intensity,shrubs play an increasingly important role in grassland ecosystem in arid and semi-arid regions,and its abundance also generally increases.However,the effects of climatic aridity and grazing intensity on sexual reproduction of shrubs in grassland remain largely unclear.In order to understand the effects of grazing intensity and climatic drought stress,and their interaction on seed production of shrub species,we examined the seed number,seed weight and seed yield of Caragana stenophylla under three grazing intensities(fenced,mild grazing and severe grazing) across a climatic aridity gradient(semi-arid,arid,very arid and intensively arid zones) in the Inner Mongolia Steppe,northern China during 2012–2013.The seed number,seed weight and seed yield gradually increased from the semi-arid to the very arid zones,but decreased from the very arid to the intensively arid zones in fenced plots.The seed number and seed yield decreased from the semi-arid to the intensively arid zones in mild and severe grazing treatment plots,therefore,grazing enhanced the suppression effect of climatic aridity on seed production of C.stenophylla.The seed number and seed yield gradually decreased as grazing intensity increased.The seed weight was highest in severe grazing plots,followed by the mild grazing plots and then the fenced plots.Precipitation varied interannually during the study period.We observed that the seed number,seed weight and seed yield were lower in the low precipitation year(2013) than in the high precipitation year(2012).As climatic drought stress increased,the negative effects of grazing on seed production of C.stenophylla also gradually increased.Our results indicated that climatic drought stress may contribute to the encroachment of C.stenophylla shrub in arid zones by promoting its seed production.However,grazing had negative effects on sexual reproduction of C.stenophylla,and the combined effects of drought stress and grazing seriously suppressed sexual reproduction of C.stenophylla in the intensively arid zone.XIE Lina CHEN Weizhong Christopher A GABLER HAN Lei GUO Hongyu CHEN Qing MA Chengcang GU Song 2016Journal of Arid Land2016,8,6:5
8Varicella zoster meningitis complicating combined anti-tumor necrosis factor and corticosteroid therapy in Crohn's disease显示文摘Opportunistic viral infections are a well-recognized complication of anti-tumor necrosis factor (TNF) therapy for inflammatory bowel disease (IBD). Cases of severe or atypical varicella zoster virus infection, both primary and latent reactivation, have been described in association with immunosuppression of Crohn's disease (CD) patients. However, central nervous system varicella zoster virus infections have been rarely described, and there are no previous reports of varicella zoster virus meningitis associated with anti-TNF therapy among the CD population. Here, we present the case of a 40-year-old male with severe ileocecal-CD who developed a reactivation of dermatomal herpes zoster after treatment with prednisone and adalimumab. The reactivation presented as debilitating varicella zoster virus meningitis, which was not completely resolved despite aggressive antiviral therapy with prolonged intravenous acyclovir and subsequent oral valacyclovir. This is the first reported case of opportunistic central nervous system varicella zoster infection complicating anti-TNF therapy in the CD population. 9Amnion epithelial cells——a novel therapy for ischemic stroke?显示文摘Stroke is a leading cause of death and disability and new therapies are desperately needed. Given the complex nature of ischemic brain injury, it has been postulated that cell-based therapies may be useful. However, cell resources, invasive extraction procedures, immunological rejection, tumorigenesis and ethical challenges make it unlikely that many stem cell types could serve as a practical source for therapy. By contrast, these issues do not pertain to human amnion epithelial cells(h AECs), which are placenta-derived stem cells. We recently assessed the effects of systemically delivered hAECs on stroke outcome using four animal models of stroke. We demonstrated that when injected intravenously after ischemia onset, hAECs migrate preferentially to the spleen and injured brain to limit apoptosis and inflammation, and attenuate early brain infiltration of immune cells, progression of infarction and systemic immunosuppression and to ultimately ameliorate functional deficits. When administration of hAECs is delayed by 1-3 days poststroke, long-term functional recovery can still be enhanced in young and aged mice of either sex. Moreover, our proof-of-principle findings suggest that h AECs are effective at limiting post-stroke infarct development in non-human primates. 11Extracts from cotton over the whole growing season induce Orobanche cumana(sunflower broomrape) germination with significant cultivar interactions显示文摘Five cotton cultivars and their parents were tested for induction of germination of Orobanche cumana Wallr.(sunflower broomrape) seeds in pot and field experiments. Germination rates induced by cotton root extracts were the highest followed by stem extracts then leaf extracts. Cotton seedlings at the six-leaf stage induced higher germination than seedling at the two-and four-leaf stage, in all five cotton cultivars and their parents. In the field, the highest concentration of cotton root extracts gave the highest germination rate of O. cumana, and the lowest concentration of cotton root extracts gave the lowest germination rate. Methanol extracts of cotton rhizosphere soil gave the highest germination of O. cumana. In general,the root, stem and leaf extracts were more active at the sixleaf stage than other seedling stages. In conclusion,extracts of cotton rhizosphere soil and tissues have high activity in the seedling stage.
