| 1 | Baicalin induces ferroptosis in bladder cancer cells by downregulating FTH1显示文摘Ferroptosis is a non-apoptotic regulated cell death caused by iron accumulation and subsequent lipid peroxidation.Currently,the therapeutic role of ferroptosis on cancer is gaining increasing interest.Baicalin an active component in Scutellaria baicalensis Georgi with anticancer potential various cancer types;however,the effects of baicalein on bladder cancer and the underlying molecular mechanisms remain largely unknown.In the study,we investigated the effect of baicalin on bladder cancer cells5637 and KU-19-19.As a result,we show baicalin exerted its anticancer activity by inducing apoptosis and cell death in bladder cancer cells.Subsequently,we for the first time demonstrate baicalin-induced ferroptotic cell death in vitro and in vivo,accompanied by reactive oxygen species(ROS) accumulation and intracellular chelate iron enrichment.The ferroptosis inhibitor deferoxamine but not necrostatin-1,chloroquine(CQ),N-acetyl-L-cysteine,L-glutathione reduced,or carbobenzoxy-valyl-alanyl-aspartyl-[O-methyl]-fluoromethylketone(Z-VAD-FMK) rescued baicalin-induced cell death,indicating ferroptosis contributed to baicalin-induced cell death.Mechanistically,we show that ferritin heavy chain1(FTH1) was a key determinant for baicalin-induced ferroptosis.Overexpression of FTH1 abrogated the anticancer effects of baicalin in both 5637 and KU19-19 cells.Taken together,our data for the first time suggest that the natural product baicalin exerts its anticancer activity by inducing FTH1-dependent ferroptosis,which will hopefully provide a prospective compound for bladder cancer treatment. | Na Kong Xiaying Chen Jiao Feng Ting Duan Shuiping Liu Xueni Sun Peng Chen Ting Pan Lili Yan Ting Jin Yu Xiang Quan Gao Chengyong Wen Weirui Ma Wencheng Liu Mingming Zhang Zuyi Yang Wengang Wang Ruonan Zhang Bi Chen Tian Xie Xinbing Sui Wei Tao | 2021 | Acta Pharmaceutica Sinica B2021,11,12: | 23 |

| 3 | Chemical Mechanism of Flower Color Microvariation in Paeonia with Yellow Flowers显示文摘Paeonia species are important ornamental plants,including three types:tree peony,herbaceous peony,and Itoh hybrid peony.Paeonia with yellow color is desired and becoming popular.However,the chemical mechanism of the formation of yellow flowers in Paeonia is still unclear.In this study,14 representative samples were selected:four samples of three species of Paeonia,four herbaceous peony cultivars,three lutea hybrid tree peony cultivars,and three Itoh hybrid peony cultivars.The petal extracts of the samples were determined by HPLC-DAD and HPLCQ-TOF-MS/MS.In total,29 flavonoids were isolated and identified,including 28 flavonol glycosides and one chalcone derivative.There were significant differences in the composition and content of flavonoids in petals of different cultivars(species).The total content of flavonoids was between 19.430 and 143.043 mg·g^−1 DW.Flavonol glycosides were detected in all samples.Chalconaringenin 2′-O-glucoside was detected from one herbaceous peony cultivar‘Golden Wheel’lutea hybrid tree peony cultivars,and Itoh peony cultivars.The content of chalconaringenin 2′-O-glucoside in the samples was more than 43% of total flavonoids.The pigment type and content of lutea hybrid tree peonies and Itoh hybrid peonies are similar.Total flavonoids,especially quercetin 3-O-galloylglucoside and chalconaringenin 2′-O-glucoside contribute greatly to the formation of yellow flowers of Paeonia.The results of this study provide a comprehensive understanding of the chemical mechanism for yellow flower coloration of Paeonia,and lay the foundation for yellow flower breeding of Paeonia. | Yong Yang Bing Li Chengyong Feng Qian Wu Qianyu Wang Shanshan Li Xiaonan Yu Liangsheng Wang | 2020 | Horticultural Plant Journal2020,6,3: | 10 |

| 5 | Dynamics and Small Signal Stability Analysis of PMSG-based Wind Farm with an MMC-HVDC System显示文摘The permanent magnet synchronous generator (PMSG)-based wind farm with a modular multilevel converter (MMC) based HVDC system exhibits various oscillations and can experience dynamic instability due to the interactions between different controllers of the wind farm and MMC stations, which have not been properly examined in the existing literatures. This paper presents a dynamic modeling approach for small signal stability analysis of PMSG-based wind farms with a MMC- HVDC system. The small signal model of the study system is validated by the comprehensive electromagnetic transient (EMT) simulations in PSCAD/EMTDC. Then the eigenvalue approach and participation factors analysis are utilized to comprehensively evaluate the impact of different controllers, system’s parameters and the circulating current suppressing controller (CCSC) on the small signal stability of the entire system. From eigenvalue analysis, it is revealed that as the output active power of the wind farm increases within the rated range, the overall system will exhibit a sub-synchronous oscillation (SSO) instability mode, an extremely weak damping mode, and a low frequency oscillation instability mode. From participation factors analysis, it is observed that the SSO mode and weak damping mode are primarily related to the internal dynamics of the MMC, which can be suppressed or improved by CCSC. It is determined that the low frequency oscillation mode is primarily caused by the interactions between the phase locked loop (PLL) control of the wind farm and the voltage and frequency (V-F) control of the MMC station. | 6 | Extracts from Huangqi(Radix Astragali Mongoliciplus) and Ezhu(Rhizoma Curcumae Phaeocaulis) inhibit Lewis lung carcinoma cell growth in a xenograft mouse model by impairing mitogen-activated protein kinase signaling, vascular endothelial growth factor prod显示文摘OBJECTIVE: To study the anti-tumor effects of the extracts from Huangqi(Radix Astragali Mongolici)and Ezhu(Rhizoma Curcumae Phaeocaulis) on the growth of Lewis lung carcinoma(LLC) in a xenograft mouse model and to investigate the possible underlying mechanism.METHODS: LLC tumor-bearing C57 BL/6 mice were treated with normal saline, cisplatin(2 mg/kg intraperitoneally every other day), or Huangqi(Radix Astragali Mongolici) and Ezhu(Rhizoma Curcumae Phaeocaulis)(1∶1, 2∶1, or 3∶1 ratio;5, 8, or 11 g/kg crude drug intragastrically every day) for 15 d.Body weights and tumor volumes were measured every other day. Tumors were excised on day 15 and analyzed. Tumor microvessel density(MVD)was assessed by immunohistochemical staining of CD34;and expression of vascular endothelial cell growth factor(VEGF), the mitogen-activated protein kinases p38 mitogen-activated protein kinase(MAPK), extracellular signal-regulated kinases 1 and 2(ERK1/2), and Jun N-terminal kinase(JNK)and their phosphorylated forms were assessed by Western blotting.RESULTS: Treatment with cisplatin caused a significant loss of body weight compared with controls,whereas Huangqi(Radix Astragali Mongolici) and Ezhu(Rhizoma Curcumae Phaeocaulis) extract combinations had no effect. Extracts from Huangqi(Radix Astragali Mongolici) and Ezhu(Rhizoma Curcumae Phaeocaulis) significantly decreased tumor weight and tumor MVD compared with controls,and at the 3∶1 treatment group had similar efficacy to cisplatin in reducing MVD. Tumors from Huangqi(Radix Astragali Mongolici) and Ezhu(Rhizoma Curcumae Phaeocaulis) treatments also showed decreased p38 MAPK, p-p38 MAPK, ERK1/2, p-ERK1/2,JNK, and p-JNK expression compared with the control group(all P < 0.01). | 8 | Radial Grip Rigidity of the Matching of Lengthened Shrink-fit Holder and Cutter in High-speed Milling显示文摘Though the lengthened shrink-fit holder (LSFH) is widely applied in high speed milling of the parts characterized by deep cavities at present,its design and selection mainly depends on the experience and lacks a correct theoretical guidance.In this paper,attention is focus on the radial grip rigidity of the matching of LSFH and cutter in high speed milling.Based on the experiment modal analysis (EMA) technique,an accurate finite element model of the matching of LSFH and cutter is established firstly.Subsequently,the influence of different interference,grip length and spindle speed on the grip rigidity of LSFH are analyzed.The analysis results show that there is a reasonable interference and grip length between the LSFH and cutter so that to have a steepless grip and have a good radial grip rigidity and at the same time to avoid the strength of LSFH to exceed it’s yield limit which will reduce the precision and service life of LSFH,besides when spindle speed reach a extension the weakening influence of the centrifugal force on the radial grip rigidity of the matching of LSFH and cutter should been taken into account.Finally,the finite element analysis results are verified based on the construction of measurement method of the grip rigidity and the results fit very well.The studies provide a theoretical basis for the design,selection and the serialization and standardization of the matching of LSFH and cutter. | ZHOU Houming WANG Chengyong DENG Jianxin PENG Ruitao | 2012 | Chinese Journal of Mechanical Engineering2012,25,1: | 5 |

| 10 | Effect of optimal combination of Huangqi(Radix Astragali Mongolici) and Ezhu(Rhizoma Curcumae Phaeocaulis) on proliferation and apoptosis of A549 lung cancer cells显示文摘OBJECTIVE: To investigate the effect of optimal combination(E) of Huangqi(Radix Astragali Mongolici) and Ezhu(Rhizoma Curcumae Phaeocaulis) on proliferation and apoptosis of A549 lung cancer cells and the possible mechanism underpinning the action.METHODS: A uniform design method was used to optimize the E of Huangqi(Radix Astragali Mongolici) and Ezhu(Rhizoma Curcumae Phaeocaulis) in A549 lung cancer cells. MTS assay was applied to analyze the effect of the component formula ofHuangqi(Radix Astragali Mongolici) and Ezhu(Rhizoma Curcumae Phaeocaulis) on A549 cells viability in various uniform design groups. A549 cells with exponential growth in routine culture were exposed to Co Cl_2(200 μmol/L) to mimic hypoxic conditions. Group 0 was treated with RPMI-1640, the group Co Cl_2 was treated with Co Cl_2(200 μmol/L),the group DDP + Co Cl_2 was treated with 4 mg/L Cisplatin injection(DDP) + Co Cl_2(200 μmol/L), and the drug group was treated with various dose of E(0.5 E, 1 E, 2 E) + Co Cl_2(200 μmol/L). All groups were cultured for 24 h. Cell apoptosis was measured by Annexin V-FITC/propidium iodide double staining and flow cytometry. Western blot assay and quantitative real-time polymerase chain reaction(q RT-PCR) were employed to detect the protein and m RNA expression of B-celllymphoma-2(Bcl-2),Bcl-2-associated X protein(Bax) and cysteinyl aspartate specific proteinase-3(caspase-3).RESULTS: The E obtained by the uniform design was comprise of 200 mg/L Astragalus polysaccharide(X1) and 32 mg/L Curcumin(X3). Group DDP+Co Cl_2, group 1 E + Co Cl_2 and group 2 E + Co Cl_2 promoted the apoptosis of A549 cells(P < 0.05). Group1 E + Co Cl_2 and group 2 E + Co Cl_2 had no statistically significant differences compared with the group DDP + Co Cl_2(P > 0.05). Compared with group 0, various doses of E + Co Cl_2 could up-regulate the expression of Bax and caspase-3 and down-regulate the expression of Bcl-2 at protein and m RNA levels(P < 0.05).CONCLUSION: Astragalus polysaccharide and Curcumin was the optimal combination of Huangqi(Radix Astragali Mongolici) and Ezhu(Rhizoma Cur-cumae Phaeocaulis). E promoted the apoptosis of A549 cells. | 11 | Drilling Force and Temperature of Bone under Dry and Physiological Drilling Conditions显示文摘Many researches on drilling force and temperature have been done with the aim to reduce the labour intensiveness of surgery, avoid unnecessary damage and improve drilling quality. However, there has not been a systematic study of mid- and high-speed drilling under dry and physiological conditions(injection of saline). Furthermore, there is no consensus on optimal drilling parameters. To study these parameters under dry and physiological drilling conditions, pig humerus bones are drilled with medical twist drills operated using a wide range of drilling speeds and feed rates. Drilling force and temperature are measured using a YDZ-II01 W dynamometer and a NEC TVS-500 EX thermal infrared imager, respectively, to evaluate internal bone damage. To evaluate drilling quality, bone debris and hole morphology are observed by SEM(scanning electron microscopy). Changes in drilling force and temperature give similar results during drilling such that the value of each parameter peaks just before the drill penetrates through the osteon of the compact bone into the trabeculae of the spongy bone. Drilling temperatures under physiological conditions are much lower than those observed under dry conditions, while a larger drilling force occurs under physiological conditions than dry conditions. Drilling speed and feed rate have a significant influence on drilling force, temperature, bone debris and hole morphology. The investigation of the effect of drilling force and temperature on internal bone damage reveals that a drilling speed of 4500 r/min and a feed rate of 50 mm/min are recommended for bone drilling under physiological conditions. Drilling quality peaks under these optimal parameter conditions. | 13 | Comparison of developmental toxicity of different surface modified CdSe/ZnS QDs in zebrafish embryos显示文摘Quantum dots(QDs)are new types of nanomaterials.Few studies have focused on the effect of different surface modified QDs on embryonic development.Herein,we compared the in vivo toxicity of Cd Se/Zn S QDs with carboxyl(-COOH)and amino(-NH 2)modification using zebrafish embryos.After exposure,the two Cd Se/Zn S QDs decreased the survival rate,hatching rate,and embryo movement of zebrafish.Moreover,we found QDs attached to the embryo membrane before hatching and the eyes,yolk and heart after hatching.The attached amount of carboxyl QDs was more.Consistently,the Cd content in embryos and larvae was higher in carboxyl QD-treatment.We further observed that the two QDs caused zebrafish pericardial edema and cardiac dysfunction.In line with it,both carboxyl and amino QDs upregulated the transcription levels of cardiac development-related genes,and the levels were higher in carboxyl QD-treated groups.Furthermore,the chelator of Cd^2+diethylene triamine pentacetate acid could partially rescued the developmental toxicity caused by the two types of QDs suggesting that both the nature of QDs and the release of Cd^2+contribute to the developmental toxicity.In conclusion,the two Cd Se/ZnS QDs have developmental toxicity and affect the cardiac development,and the carboxyl QDs is more toxic possibly due to the higher affinity and more release to embryos and larvae.Our study provides new knowledge that the surface functional modification of QDs is critical on the development on aquatic species,which is beneficial to develop and applicate QDs more safely and environmentfriendly. | Naying Zheng Jinhui Yan Wang Qian Chao Song Zhenghong Zuo Chengyong He | 2021 | Journal of Environmental Sciences2021,33,2: | 3 |

| 16 | CO_2 sensing properties and mechanism of PrFeO_3 and NdFeO_3 thick film sensor显示文摘The CO_2 sensing of PrFeO_3 and NdFeO_3 sensors were investigated. Experimental results show that the resistances for PrFeO_3 and NdFeO_3 in CO_2 gas are larger than those in air and the responses for PrFeO_3and NdFeO_3 sensors increase with an increase in room-temperature relative humidity. When exposed to1000 ppm CO_2, the response of PrFeO_3 thick film based on nano-powders annealed at 700℃can reach8.44 at 160℃for the background of wet air with 58%of room-temperature relative humidity (RH),which is much larger than the corresponding value (3.03) in wet air with 25%RH. The sensing response S of NdFeO_3 thick-film sensor based on nano-powders annealed at 600℃to 3000 ppm CO_2 at the operating temperature 200℃can reach 2.36 for the background of wet air with 72%RH, which is larger than the corresponding value (1.83) in the air with 25%RH. | 18 | Efficacy of Liangxue Guyuan decoction(凉血固元汤)on radiation-induced intestinal injury in rats via the toll-like receptor 4/myeloid differentiation primary response 88/nuclear factor-kappa B pathway显示文摘OBJECTIVE:To evaluate the efficacy of Liangxue Guyuan decoction(凉血固元汤,LGD)on radiation-induced intestinal injury in rats,and the possible underlying mechanism of action.METHODS:A total of 255 male Sprague-Dawley rats were used.15 rats were assigned to the control group and the remaining 240 rats were exposed to a^(60)Co source at a dose of 11 Gy.Irradiated rats were randomly divided into model,dexamethasone(DXM),low-dose LGD(LGDl),and high-dose LGD(LGDh)groups and treated for 11 d.The survival rate,weight of body,intestinal pathology and the expression of toll-like receptor 4(TLR4),myeloid differentiation primary response 88(MyD88),and nuclear`factor-kappa B(NF-κB)were recorded.RESULTS:Radiation reduced the survival rate and weight of rats,destroyed the intestinal structure,induced an inflammatory reaction,and increased both protein and mRNA expression of TLR4,MyD88,and NF-κB in ileum.However,LGDh increased the survival rate,inhibited weight loss,alleviated inflammation and improve the expression of TLR4 pathway.CONCLUSIONS:LGD increased the survival rate and inhibit weight loss of irradiated rats,and reduced inflammation and intestinal injury.The underlying mechanism may involve regulation of the TLR4/MyD88/NF-κB pathway. | WANG Yuguo DOU Yongqi FENG Jian XU Chengyong WANG Qian | 2021 | Journal of Traditional Chinese Medicine2021,41,2: | 2 |
