2Proteins Related to the Biocontrol of Pythium Damping-off in Maize with Trichoderma harzianum Rifai显示文摘Induced resistance has been evidenced as one of mechanisms of Trichoderma to control plantdiseases, however, no study showed the change of host proteomics in Trichoderma-induced resistance ofmaize against damping-off caused by Pythium ultimum Trow. The mechanism of Trichoderma harzianumRifai for controlling maize seedling disease caused by Pythium ultimum Trow was investigated firstly byproteome technique and the result suggested that T. harzianum strain T22 was not only able to promoteseedling growth but also protein accumulation. One-dimensional electrophoresis assay showed that morebands appeared on the gel with T22 or T22 combined with P. ultimum (T22 + P. ultimum) treatment than withother treatments. Enzyme assay showed that two chitinases of the root sample were more activated in thetreatments with T22 than in the other treatments without T22. Proteins in the seedling roots from the varioustreatments were separated through protein extraction and 2-D electrophoresis technique. In the seedlingsproduced from the T22-treated seeds, there were 104 up-regulated proteins and 164 down-regulated pro-teins relative to the control, and 97 and 150, respectively, after treatment with T22 + P. ultimum; however,with P. ultimum alone the values were much lower than with the other two treatments. The correlationcoefficient values were 0.72, 0.51 and 0.49 for the comparison of protein spot distribution on gel amongcontrol with T22, P. ultimum and T22 + P. ultimum, respectively. So it seemed that P. ultimum infection wasmore effective than T22 in interfering with the host proteome profile. Furthermore, analysis with MALDI-TOF-MAS showed that some important proteins associated with defensive reactions were identified in T22or T22 + P. ultimum treatments, including endochitinase, pathogenesis-related protein PRMS (pathogenesis-related maize seed), GTP-binding protein, isoflavone reductase and other proteins related to respiration. Allthose proteins are probably part of the network of resistance or development-related proteins. 3Endoscopic ultrasound-guided vs endoscopic retrograde cholangiopancreatography biliary drainage for obstructed distal malignant biliary strictures: A systematic review and meta-analysis显示文摘BACKGROUND For palliation of malignant biliary obstruction(MBO), the gold-standard method of biliary drainage is endoscopic retrograde cholangiopancreatography(ERCP)with the placement of metallic stents. Endoscopic ultrasound(EUS)-guided drainage is an alternative that is typically reserved for cases of ERCP failure.Recently, however, there have been robust randomized clinical trials(RCTs)comparing EUS-guided drainage and ERCP as primary approaches to MBO.AIM To compare EUS guidance and ERCP in terms of their effectiveness and safety in palliative biliary drainage for MBO.METHODS This was a systematic review and meta-analysis, in which we searched the MEDLINE, Excerpta Medica, and Cochrane Central Register of Controlled Trials databases. Only RCTs comparing EUS and ERCP for primary drainage of MBO were eligible. All of the studies selected provided data regarding the rates of technical and clinical success, as well as the duration of the procedure, adverse events, and stent patency. We assessed the risk of biases using the Jadad score and the quality of evidence using the Grading of Recommendations Assessment,Development and Evaluation criteria.RESULTS The database searches yielded 5920 records, from which we selected 3 RCTs involving a total of 222 patients(112 submitted to EUS and 110 submitted to ERCP). In the EUS and ERCP groups, the rate of technical success was 91.96%n and 91.81%, respectively, with a risk difference(RD) of 0.00%(95%CI:-0.07, 0.07;P = 0.97; I^2 = 0%). The clinical success was 84.81% and 85.53% in the EUS and ERCP groups, respectively, with an RD of-0.01%(95%CI:-0.12, 0.10; P = 0.90; I^2 =0%). The mean difference(MD) for the duration of the procedure was-0.12%(95%CI:-8.20, 7.97; P = 0.98; I^2 = 84%). In the EUS and ERCP groups, there were14 and 25 adverse events, respectively, with an RD of-0.06%(95%CI:-0.23, 0.12; P= 0.54; I^2 = 77%). The MD for stent patency was 9.32%(95%CI:-4.53, 23.18; P =0.19; I^2 = 44%). The stent dysfunction rate was significantly lower in the EUS t group(MD =-0.22%; 95 CI:-0.35,-0.08; P = 0.001; I^2 = 0%).CONCLUSION EUS represents an interesting alternative to ERCP for MBO drainage,demonstrating lower stent dysfunction rates compared with ERCP. 4Changes in the sediment load of the Lancang-Mekong River over the period 1965-2003显示文摘Changes in the sediment load of a river can have important impacts on river channel evolution,nutrient fluxes,aquatic ecology and delta erosion and sedimentation,and the possibility of changes in the sediment load of the Lancang-Mekong River has attracted increasing concern in recent years.Existing studies present conflicting findings regarding the nature and magnitude of recent changes in the sediment load of the Lancang-Meking River and the authors have attempted to assemble the most reliable data on annual sediment loads for the period 1965-2003,to assess recent trends in the sediment load of the river.The changes in annual sediment load at 7 stations on the river are analyzed.Important sediment contributing areas are found in the reaches between Gajiu and Yunjinghong,Chiang Saen and Luang Prabang and downstream of Nong Khai.The sediment load increased at Gajiu,Yunjinghong and Chiang Saen over the period 1985-1992 because of serious soil erosion caused by the expansion of cultivation,the replacement of natural forest by plantations and land disturbance associated with hydropower dam construction.A marked reduction in sediment load occurred at Gajiu after the impoundment of the Manwan Hydropower dam on the Lancang River,but this reduction was not evident downstream at Yunjinghong and the stations further downstream.Significant increases in sediment load appeared at Mukdahan and Khong Chiam.These contrasting patterns of change reflect the influence of sediment contributions from the intervening catchment areas and channel systems as well as storage and remobilization of sediment from the channel system and the impact of hydraulic works such as irrigation systems.The long term mean annual sediment load of the Mekong River at its mouth is estimated to be ca.145×106ta-1,which is lower than previously reported values and it seems likely that this will be reduced in the foreseeable future.LIU Cheng HE Yun WALLING Des E WANG JianJun 2013Science China(Technological Sciences)2013,56,4:6
5The maternally expressed polycomb group gene OsEMF2a is essential for endosperm cellularization and imprinting in rice显示文摘Cellularization is a key event in endosperm development.Polycomb group(PcG)genes,such as Fertilization-Independent Seed 2(FIS2),are vital for the syncytium-to-cellularization transition in Arabidopsis plants.In this study,we found that OsEMF2a,a rice homolog of the Arabidopsis PcG gene Embryonic Flower2(EMF2),plays a role similar to that of FIS2 in regard to seed development,although there is limited sequence similarity between the genes.Delayed cellularization was observed in osemf2a,associated with an unusual activation of type I MADS-box genes.The cell cycle was persistently activated in osemf2a caryopses,which was likely caused by cytokinin overproduction.However,the overaccumulation of auxin was not found to be associated with the delayed cellularization.As OsEMF2a is a maternally expressed gene in the endosperm,a paternally inherited functional allele was unable to recover the maternal defects of OsEMF2a.Many imprinted rice genes were deregulated in the defective hybrid seeds of osemf2a(♀)/9311(♂)(m9).The paternal expression bias of some paternally expressed genes was disrupted in m9 due to either the activation of maternal alleles or the repression of paternal alleles.These findings suggest that OsEMF2a-PRC2-mediated H3K27me3 is necessary for endosperm cellularization and genomic imprinting in rice.Xiaojun Cheng Meiyao Pan Zhiguo E Yong Zhou Baixiao Niu Chen Chen 2021Plant Communications2021,2,1:5
6The m^(6)A methylome of SARS-CoV-2 in host cells显示文摘The newly identified Severe Acute Respiratory Syndrome Coronavirus 2(SARS-CoV-2)has resulted in a global health emergency because of its rapid spread and high mortality.The molecular mechanism of interaction between host and viral genomic RNA is yet unclear.We demonstrate herein that SARS-CoV-2 genomic RNA,as well as the negative-sense RNA,is dynamically N6-methyladenosine(m^(6)A)-modified in human and monkey cells.Combined RIP-seq and miCLIP analyses identified a total of 8 m^(6)A sites at single-base resolution in the genome.Especially,epidemic strains with mutations at these identified m^(6)A sites have emerged worldwide,and formed a unique cluster in the US as indicated by phylogenetic analysis.Further functional experiments showed that m^(6)A methylation negatively regulates SARS-CoV-2 infection.SARS-CoV-2 infection also triggered a global increase in host m^(6)A methylome,exhibiting altered localization and motifs of m^(6)A methylation in mRNAs.Altogether,our results identify m^(6)A as a dynamic epitranscriptomic mark mediating the virus–host interaction.Jun’e Liu Yan-Peng Xu Kai Li Qing Ye Hang-Yu Zhou Hanxiao Sun Xiaoyu Li Liu Yu Yong-Qiang Deng Rui-Ting Li Meng-Li Cheng Bo He Jia Zhou Xiao-Feng Li Aiping Wu Chengqi Yi Cheng-Feng Qin 2021Cell Research2021,31,4:5

8A Novel Model of Predicting Archie's Cementation Factor from Nuclear Magnetic Resonance(NMR) Logs in Low Permeability Reservoirs显示文摘The resistivity experimental measurements of core samples drilled from low permeability reservoirs of Ordos Basin,Northwest China,illustrate that the cementation factors are not agminate,but vary from 1.335 to 1.749.This leads to a challenge for the estimation of water and hydrocarbon saturation.Based on the analysis of Purcell equation and assumption that rock resistivity is determined by the parallel connection of numerous capillary resistances,a theoretical expression of cementation factor in terms of porosity and permeability is established.Then,cementation factor can be calculated if the parameters of porosity and permeability are determined.In the field application,porosity can be easily obtained by conventional logs.However,it is a tough challenge to estimate permeability due to the strong heterogeneity of low permeability reservoirs.Thus,the Schlumberger Doll Research(SDR)model derived from NMR logs has been proposed to estimate permeability.Based on the analysis of the theoretical expressions of cementation factor and SDR model,a novel cementation factor prediction model,which is relevant to porosity and logarithmic mean of NMR T2spectrum(T2lm),is derived.The advantage of this model is that all the input information can be acquired from NMR logs accurately.In order to confirm the credibility of the novel model,the resistivity and corresponding laboratory NMR measurements of 27 core samples are conducted.The credibility of the model is confirmed by comparing the predicted cementation factors with the core analyzed results.The absolute errors for all core samples are lower than 0.071.Once this model is extended to field application,the accuracy of water and hydrocarbon saturation estimation will be significantly improved.Liang Wang Zhiqiang Mao Yujiang Shi Qin'e Tao Yumei Cheng Yong Song 2014Journal of Earth Science2014,25,1:4
9Synthesis and biological evaluation of 8-substituted and deglucuronidated scutellarin and baicalin analogues as antioxidant responsive element activators显示文摘Flavonoids are important bioactive dietary compounds, which are proved to be antioxidant responsive element (ARE) activators to defend against electrophilic toxicants and oxidative stress. The activators induce ARE gene transcription through Nrf2 factor, a major transcriptional stimulator of cytoprotective genes, relieved from a Keap1 complex. In this report, based on the structures of two flavonoids, scutellarin and baicalin, which are extracted from two common Chinese plants Dengzhanhua (Erigeron breviscapus (vant) Hand-Mazz) and Huangqin (Scutellaria baicalensis Georigi), respectively, we synthesized several 8-substituted and deglucuronidated analogues and identified the ARE activation effects of these flavonoids. We found that the Baicalin, deglucurnonidated Baicalin and diaza cyclopenta derivative were more active. Their dose-dependent upregulation activities of ARE and NQO1 and induction effects of Nrf2 were testified. 10Clinical observation on pericardial effusion in patients with lung cancer treated by intrapericardial catheterization and infusion of highly agglutinative staphylococcin and cisplatin显示文摘Objective: To evaluate the therapeutic efficacy of injecting highly agglutinative staphylococcin (HASL) and cisplatin into pericardial cavity of lung cancer patients with pericardial effusion. Methods: 81 patients were randomized into two groups: 45 in the experimental group (HASL and Cisplatin) and 36 in the control group (Cisplatin). At first pericardial effusion was drained out from a intrapericardial catheter and then different drugs were infused, respectively. 24 h after perfusion the draining continued again until drainage quantity was less than 30 mL every day. The draining lasted 10–15 days. Results: The response rate was 91.1% for the experimental group and 80.6% for the control group. There was no significant difference between the two groups (P>0.05). The complete remission was 77.8% for the experimental group and 52.8% for the control group, which was statistically significant difference (P<0.05). The adverse effects were myelosuppression and nausea and vomiting, which were 35.6% and 40.0% in the experimental group and 72.2% and 66.7% in the control group, respectively (P<0.01, P<0.05). 17Increased expression of chondroitin sulphate proteoglycans in rat hepatocellular carcinoma tissues显示文摘AIM:To investigate the expression of chondroitin sulphate proteoglycans(CSPGs)in rat liver tissues of hepatocellular carcinoma(HCC).METHODS:Thirty male Sprague Dawley rats were randomly divided into two groups:control group(n=10) and HCC model group(n=20).Rats in the HCC model groups were intragastrically administrated with 0.2%(w/v)N-diethylnitrosamine(DEN)every 5 d for 16 wk,whereas 0.9%(w/v)normal saline was administered to rats in the control group.After 16 wk from the initiation of experiment,all rats were killed and livers were collected and fixed in 4%(w/v)paraformaldehyde.All tissues were embedded in paraffin and sectioned.Histological staining(hematoxylin and eosin and Toluidine blue)was performed to demonstrate the onset of HCC and the content of sulphated glycosaminoglycan(sGAG).Immunohistochemical staining was performed to investigate the expression of chondroitin sulphate(CS)/dermatan sulphate(DS)-GAG,heparan sulphate(HS)-GAG,keratan sulphate(KS)-GAG in liver tissues.Furthermore,expression and distribution of CSPG family members,including aggrecan,versican,biglycan and decorin in liver tissues,were also immunohistochemically determined.RESULTS:After 16 wk administration of DEN,malignant nodules were observed on the surface of livers from the HCC model group,and their hepatic lobule structures appeared largely disrupted under microscope.Toluidine blue staining demonstrated that there was an significant increase in sGAG content in HCC tissues when compared with that in the normal liver tissues from the control group[0.37±0.05 integrated optical density per stained area(IOD/area)and 0.21± 0.01 IOD/area,P<0.05].Immunohistochemical studies demonstrated that this increased sGAG in HCC tissues was induced by an elevated expression of CS/DS(0.28±0.02 IOD/area and 0.18±0.02 IOD/area,P< 0.05)and HS(0.30±0.03 IOD/area and 0.17±0.02 IOD/area,P<0.01)but not KS GAGs in HCC tissues.Further studies thereby were performed to investigate the expression and distribution of several CSPG components in HCC tissues,including aggrecan,versican,biglycan and decorin.Interestingly,there was a distinct distribution pattern for these CSPG components between HCC tissues and the normal tissues.Positive staining of aggrecan,biglycan and decorin was localized in hepatic membrane and/or pericellular matrix in normal liver tissues;however,their expression was mainly observed in the cytoplasm,cell membranes in hepatoma cells and/or pericellular matrix within HCC tissues.Semi-quantitative analysis indicated that there was a higher level of expression of aggrecan(0.43± 0.01 and 0.35±0.03,P<0.05),biglycan(0.32±0.01 and 0.25±0.01,P<0.001)and decorin(0.29±0.01 and 0.26±0.01,P<0.05)in HCC tissues compared with that in the normal liver tissues.Very weak versican positive staining was observed in hepatocytes near central vein in normal liver tissues;however there was an intensive versican distribution in fibrosis septa between the hepatoma nodules.Semi-quantitative analysis indicated that the positive rate of versican in hepatoma tissues from the HCC model group was much higher than that in the control group(33.61%and 21.28%,P <0.05).There was no positive staining in lumican and keratocan,two major KSPGs,in either normal or HCC liver tissues.CONCLUSION:CSPGs play important roles in the onset and progression of HCC,and may provide potential therapeutic targets and clinical biomarkers for this prevalent tumor in humans.Xiao-Li Jia Si-Yuan Li Shuang-Suo Dang Yan-An Cheng Xin Zhang Wen-Jun Wang Clare E Hughes Bruce Caterson 2012World Journal of Gastroenterology2012,18,30:2
18Rapid phenotypic evolution with shallow genomic differentiation during early stages of high elevation adaptation in Eurasian Tree Sparrows显示文摘Known as the‘third polar region’,the Qinghai-Tibet Plateau represents one of the harshest highland environments in the world and yet a number of organisms thrive there.Previous studies of birds,animals and humans have focused on well-differentiated populations in later stages of phenotypic divergence.The adaptive processes during the initial phase of highland adaptation remain poorly understood.We studied a human commensal,the Eurasian Tree Sparrow,which has followed human agriculture to the Qinghai-Tibet Plateau.Despite strong phenotypic differentiation at multiple levels,in particular in muscle-related phenotypes,highland and lowland populations show shallow genomic divergence and the colonization event occurred within the past few thousand years.In a one-month acclimation experiment investigating phenotypic plasticity,we exposed adult lowland tree sparrows to a hypoxic environment and did not observe muscle changes.Through population genetic analyses,we identified a signature of polygenic adaptation,whereby shifts in allele frequencies are spread across multiple loci,many of which are associated with muscle-related processes.Our results reveal a case of positive selection in which polygenic adaptation appears to drive rapid phenotypic evolution,shedding light on early stages of adaptive evolution to a novel environment.Yanhua Qu Chunhai Chen Ying Xiong Huishang She Yong E Zhang Yalin Cheng Shane DuBay Dongming Li Per G P Ericson Yan Hao Hongyuan Wang Hongfeng Zhao Gang Song Hailin Zhang Ting Yang Chi Zhang Liping Liang Tianyu Wu Jinyang Zhao Qiang Gao Weiwei Zhai Fumin Lei 2020National Science Review2020,7,1:2

20Experimental study by online measurement of the precipitation of nickel hydroxide: Effects of operating conditions显示文摘The objective of this work is using the online measurement method to study the process of precipitation of nickel hydroxide in a single-feed semi-batch stirred reactor with an internal diameter of D = 240 mm. The effects of impeller speed, impeller type, impeller diameter and feed location on the mean particle size d43 and particle size distribution(PSD) were investigated. d43 and PSD were measured online using a Malvern Insitec Liquid Process Sizer every 20 s. It was found that d43 varied between 13 μm and 26 μm under different operating conditions,and it decreased with increasing impeller diameter. When feeding at the off-bottom distance of D/2 under lower impeller speeds, d43 was significantly smaller than that at D/3.
