1Component analysis of Chinese medicine and advances in fuming-washing therapy for knee osteoarthritis via unsupervised data mining methods显示文摘OBJECTIVE: To analyze the component law of Chinese medicines in fuming-washing therapy for knee osteoarthritis(KOA), and develop new fuming-washing prescriptions for KOA through unsupervised data mining methods.METHODS: Chinese medicine recipes for fuming-washing therapy for KOA were collected and recorded in a database. The correlation coefficient among herbs, core combinations of herbs, andnew prescriptions were analyzed using modified mutual information, complex system entropy cluster, and unsupervised hierarchical clustering, respectively.RESULTS: Based on analysis of 345 Chinese medicine recipes for fuming-washing therapy, 68 herbs occurred frequently, 33 herb pairs occurred frequently, and 12 core combinations were found.Five new fuming-washing recipes for KOA were developed.CONCLUSION: Chinese medicines for fuming-washing therapy of KOA mainly consist of wind-dampness-dispelling and cold-dispersing herbs, blood-activating and stasis-resolving herbs,and wind-dampness-dispelling and heat-clearing herbs. The treatment of fuming-washing therapy for KOA also includes dispelling wind-dampness and dispersing cold, activating blood and resolving stasis, and dispelling wind-dampness and clearing heat. Zhenzhutougucao(Herba Speranskiae Tuberculatae), Honghua(Flos Carthami), Niuxi(Radix Achyranthis Bidentatae), Shenjincao(Herba Lycopodii Japonici), Weilingxian(Radix et Rhizoma Clematidis Chinensis), Chuanwu(Radix Aconiti), Haitongpi(Cortex Erythrinae Variegatae), Ruxiang(Olibanum),Danggui(Radix Angelicae Sinensis), Caowu(Radix Aconiti Kusnezoffii), Moyao(Myrrha), and Aiye(Folium Artemisiae Argyi) are the main herbs used in the fuming-washing treatment for KOA.Jun Liu Jianke Pan Yanping Wang Dingkun Lin Dan Shen Hongjun Yang Xiang Li Minghui Luo Xuewei Cao 2013Journal of Traditional Chinese Medicine2013,33,5:32
2Flow Dynamics of a Spiral-groove Dry-gas Seal显示文摘The dry-gas seal has been widely used in different industries.With increased spin speed of the rotator shaft,turbulence occurs in the gas film between the stator and rotor seal faces.For the micro-scale flow in the gas film and grooves,turbulence can change the pressure distribution of the gas film.Hence,the seal performance is influenced.However,turbulence effects and methods for their evaluation are not considered in the existing industrial designs of dry-gas seal.The present paper numerically obtains the turbulent flow fields of a spiral-groove dry-gas seal to analyze turbulence effects on seal performance.The direct numerical simulation(DNS) and Reynolds-averaged Navier-Stokes(RANS) methods are utilized to predict the velocity field properties in the grooves and gas film.The key performance parameter,open force,is obtained by integrating the pressure distribution,and the obtained result is in good agreement with the experimental data of other researchers.Very large velocity gradients are found in the sealing gas film because of the geometrical effects of the grooves.Considering turbulence effects,the calculation results show that both the gas film pressure and open force decrease.The RANS method underestimates the performance,compared with the DNS.The solution of the conventional Reynolds lubrication equation without turbulence effects suffers from significant calculation errors and a small application scope.The present study helps elucidate the physical mechanism of the hydrodynamic effects of grooves for improving and optimizing the industrial design or seal face pattern of a dry-gas seal.WANG Bing ZHANG Huiqiang CAO Hongjun 2013Chinese Journal of Mechanical Engineering2013,26,1:18
3Intrastriatal glial cell line-derived neurotrophic factors for protecting dopaminergic neurons in the substantia nigra of mice with Parkinson disease显示文摘BACKGROUND:Substantia nigra is deep in position and limited in range,the glial cell line-derived neurotrophic factor(GDNF)injection directly into substantia nigra has relatively greater damages with higher difficulty.GDNF injection into striatum,the target area of dopaminergic neuron,may protect the dopaminergic neurons in the compact part of substantia nigra through retrograde transport.OBJECTIVE:To investigate the protective effect of intrastriatal GDNF on dopaminergic neurons in the substantia nigra of mice with Parkinson disease(PD),and analyze the action pathway.DESIGN:A controlled observation.SETTING:Neurobiological Laboratory of Xuzhou Medical College.MATERIALS:Twenty-four male Kunming mice of 7-8 weeks old were used.GDNF,1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine(MPTP)were purchased from Sigma Company(USA);LEICAQWin image processing and analytical system.METHODS:The experiments were carried out in the Neurobiological Laboratory of Xuzhou Medical College from September 2005 to October 2006.The PD models were established in adult KunMing mice by intraperitoneal injection of MPTP.The model mice were were randomly divided into four groups with 6 mice in each group:GDNF 4-day group,phosphate buffer solution(PSB)4-day group,GDNF 6-day group and PSB 6-day group.Mice in the GDNF 4 and 6-day groups were administrated with 1 μL GDNF solution(20 μg/L,dispensed with 0.01 mol/L PBS)injected into right striatum at 4 and 6 days after model establishment.Mice in the PSB 4 and 6-day groups were administrated with 0.01 mol/L PBS of the same volume to the same injection at corresponding time points.② On the 12th day after model establishment,the midbrain tissue section of each mice was divided into 3 areas from rostral to caudal sides.The positive neurons of tyroxine hydroxylase(TH)and calcium binding protein(CB)with obvious nucleolus and clear outline were randomly selected for the measurement,and the number of positive neurons in unit area was counted.MAIN OUTCOME MEASURES:Number of positive neurons of TH and CB in midbrain substantia nigra of mice in each group.RESULTS:All the 24 mice were involved in the analysis of results.The numbers of TH+ and CB+ neurons in the GDNF 4-day group(54.33±6.92,46.33±5.54)were obviously more than those in the PBS 4-day group(27.67±5.01,21.50±5.96,P < 0.01).The numbers of TH+ and CB+ neurons in the GDNF 6-day group(75.67±5.39,69.67±8.69)were obviously more than those in the PBS 6-day group(27.17±4.50,21.33±5.72,P < 0.01)and those in the GDNF 4-day group(P < 0.01).CONCLUSION:Intrastriatal GDNF can protect dopaminergic neurons in substantia nigra of PD mice,and it may be related to the increase of CB expression.Chenghua Xiao Yanqiang Wang Hongmei Liu Hongjun Wang Junping Cao Dianshuai Gao 2007Neural Regeneration Research2007,2,4:4
4Pathological features of COVID-19-associated lung injury:a preliminary proteomics report based on clinical samples显示文摘The COVID-19 pandemic has emerged as a global health emergency due to its association with severe pneumonia and relative high mortality.However,the molecular characteristics and pathological features underlying COVID-19 pneumonia remain largely unknown.To characterize molecular mechanisms underlying COVID-19 pathogenesis in the lung tissue using a proteomic approach,fresh lung tissues were obtained from newly deceased patients with COVID-19 pneumonia.After virus inactivation,a quantitative proteomic approach combined with bioinformatics analysis was used to detect proteomic changes in the SARS-CoV-2-infected lung tissues.We identified significant differentially expressed proteins involved in a variety of fundamental biological processes including cellular metabolism,blood coagulation,immune response,angiogenesis,and cell microenvironment regulation.Several inflammatory factors were upregulated,which was possibly caused by the activation of NF-κB signaling.Extensive dysregulation of the lung proteome in response to SARS-CoV-2 infection was discovered.Our results systematically outlined the molecular pathological features in terms of the lung response to SARS-CoV-2 infection,and provided the scientific basis for the therapeutic target that is urgently needed to control the COVID-19 pandemic.Ling Leng Ruiyuan Cao Jie Ma Danlei Mou Yunping Zhu Wei Li Luye Lv Dunqin Gao Shikun Zhang Feng Gong Lei Zhao Bintao Qiu Haiping Xiang Zhongjie Hu Yingmei Feng Yan Dai Jiang Zhao Zhihong Wu Hongjun Li Wu Zhong 2020Signal Transduction and Targeted Therapy2020,5,1:4

15Arrhythmogenic properties of dismantling cadherin-mediated adhesion in murine hearts显示文摘Objective:To evaluate the arrhythmogenic effects of dismantling cadherin-mediated adhesion by recombinant mouse aminopeptidase N(rmAPN)in murine hearts.Methods:rmAPN was incubated with cultured neonatal rat cardiomyocytes as well as being infused in adult mice.The cell-cell connections were immunolabelled and ob- served by laser confocal microscopy.Disruption of the N-terminal of N-cadherin(N-cad)was detected by western blot and quantitative immunofluorescence.The risk of inducible ventricular tachyarrhythmia was evaluated in mice by an electrophysiological study.Results:Disrupted cell-cell contact was observed in cultured neonatal rat cardiomyocytes in response to 30-40 ng/μL rmAPN.Loss of the N-terminal in N-cad and altered distribution of connexin 43(Cx43)were observed in hearts from rmAPN-infused mice.In addition,a reduction of phosphorylated Cx43 was also detected concomitant with redistribution of Cx43.Electrophysiological studies of rmAPN-infused mice showed prolonged QRS duration and increased inducibility of ventricular tachycardias.Conclusion: Disruption of N-cad by rmAPN contributes to gap junction remodeling and may elicit arrhythmogenic effects.The disorder of adherent junctions by proteolytic enzymes may play an important role in arrhythmogenic mechanisms in correlated diseases.Hongjun Zhu Hegui Wang Xiwen Zhang Xiaofeng Hou Kejiang Cao Jiangang Zou 2010The Journal of Biomedical Research2010,24,4:1
