1Expression of rabbit defensin NP-1 gene in transgenic tobacco plants and its activity against bacterial wilt显示文摘A cDNAderived 200 bp fragment encoding a rabbit defensin NP1 mature peptide was ligated into the GUSless pBI121 to yield pBIC35NP1. Tobacco leaf discs were genetically transformed with an Agrobacterium tumefaciens strain harbouring pBIC35NP1. Northern blot analysis showed that the defensin NP1 expression cassette was normally transcribed in transgenic plants. 2Diagnostic value of whole-body MRI with diffusion-weighted sequence for detection of peritoneal metastases in colorectal malignancy显示文摘Objective:To assess the diagnostic accuracy of whole-body MRI using diffusion-weighted sequence(WB-DWI)to determine the peritoneal cancer index(PCI)in correlation with surgical and histopathological findings.Methods:Twenty-seven patients underwent preoperative WB-MRI,followed by cytoreductive surgery for primary tumors of the appendix(n=15),colorectum(n=12),and associated peritoneal disease.A total of 351 regions were retrospectively reviewed.The sensitivity,specificity,and accuracy were calculated at 13 anatomical sites.The WB-DWI PCI and PCI type were compared with surgical and histopathological findings.Results:No statistical difference was found between the WB-DWI PCI and surgical PCI(P=0.574).WB-DWI correctly predicted the PCI type in 24 of 27 patients with high accuracy(88.9%),including 10 of 10 patients with small-volume tumor,12 of 14 with moderate-volume tumor,and 2 of 3 with large-volume tumor.WB-DWI correctly depicted tumors in 163 of 203 regions,with 40false-negative and 23 false-positive regions.The overall sensitivity,specificity,and accuracy of WB-DWI for the detection of peritoneal tumors were 80.3%,84.5%,and 82.1%,respectively.For lesions<0.5 cm in diameter,WB-DWI demonstrated good sensitivity(69.4%).Conclusions:WB-DWI accurately predicted PCI before surgery in patients undergoing evaluation for cytoreductive surgery.Huan Zhang Weixing Dai Caixia Fu Xu Yan Alto Stemmer Tong Tong Guoxiang Cai 2018Cancer Biology & Medicine2018,15,2:3
3Closed form algorithm of double-satellite TDOA+AOA localization based on WGS-84 model显示文摘In this paper,we consider the double-satellite localization under the earth ellipsoid model of the Wideband Geodetic System(WGS-84)using the Time Difference of Arrival(TDOA)and the Angle-of-Arrival(AOA).Several closed-form solution algorithms via the pseudolinearization of the measurement equations are presented to efficiently estimate the location.These algorithms include the Weighted Least Squares(WLS),the Constrained Total Least Squares(CTLS),and the Taylor-Series Iteration(TSI).Performance comparison of the proposed methods with the Cramér-Rao Lower Bound(CRLB)in the simulation is shown to demonstrate that the proposed algorithms are feasible and have stable performance.Wanchun LI Ruibin CHEN Yuning GUO Caixia FU 2019Chinese Journal of Aeronautics2019,32,10:3

7Molecular insights of nanozymes from design to catalytic mechanism显示文摘Emerging as cost-effective potential alternatives to natural enzymes,nanozymes have attracted increasing interest in broad fields.To exploit the in-depth potential of nanozymes,rational structural engineering and explicit catalytic mechanisms at the molecular scale are required.Recently,impressive progress has been made in mimicking the characteristics of natural enzymes by constructing metal active sites,binding pockets,scaffolds,and delicate allosteric regulation.Ingenious in-depth studies have been conducted with advances in structural characterization and theoretical calculations,unveiling the“black box”of nanozymecatalytic mechanisms.This review introduces the state-of-art synthesis strategies by learning from the natural enzyme counterparts and summarizes the general overview of the nanozyme mechanism with a particular emphasis on the adsorbed intermediates and descriptors that predict the nanozyme activity.The emerging activity assessment methodology that illustrates the relationship between electrochemical oxygen reduction and enzymatic oxygen reduction is discussed with up-to-date advances.Future opportunities and challenges are presented in the end to spark more profound work and attract more researchers from various backgrounds to the flourishing field of nanozymes.Yuan Xu Zhixin Zhou Nankai Deng Kangchun Fu Caixia Zhu Qing Hong Yanfei Shen Songqin Liu Yuanjian Zhang 2023Science China Chemistry2023,66,5:1
8New loci for refractive errors and ocular biometric parameters in young Chinese Han adults显示文摘Myopia has become a major public health issue with an increasing prevalence.There are still individuals who experience similar environmental risk factors and,yet,remain non-myopic.Thus,there might be genetic factors protecting people from myopia.Considering the opposite ocular characteristics of primary angle closure glaucoma(PACG)to myopia and possible common pathway between them,we propose that certain risk genes for PACG might act as a protective factor for myopia.In this study,2,678 young adults were genotyped for 37 targeted single nucleotide polymorphisms.Compared with emmetropia,rs1401999(allele C:OR=0.795,P=0.03;genotype in dominant model:OR=0.759,P=0.02)and rs1258267(allele A:OR=0.824,P=0.03;genotype in dominant model:OR=0.603,P=0.01)were associated with low to moderate myopia and high myopia,respectively.Genotype under recessive model of rs11024102 was correlated with myopia(OR=1.456,P=0.01),low to moderate myopia(OR=1.443,P=0.02)and high myopia(OR=1.453,P=0.02).However,these associations did not survive Bonferroni correction.Moreover,rs1401999,rs1258267,and rs11024102 showed associations with certain ocular biometric parameters in different groups.Our study suggests that ABCC5,CHAT and PLEKHA7 might be associated with refractive errors by contributing to the regulation of ocular biometry,in terms of uncorrected results and their biological functions.Yunyun Sun Zi-Bing Jin Shifei Wei Hongyan Jia Kai Cao Jianping Hu Caixia Lin Wenzai An Jiyuan Guo He LiJing Fu Shi-Ming Li Ningli Wang the Anyang University Students Eye Study Group the Anyang University Students Eye Study Group 2022Science China(Life Sciences)2022,65,10:1
9Changes in the Urban Surface Thermal Environment of a Chinese Coastal City Revealed by Downscaling MODIS LST with Random Forest Algorithm显示文摘Analysis of the formation and evolution of urban surface thermal environment is crucial for urban planning and improving the environment of a settlement.Qingdao was selected in this study as a typical coastal city undergoing rapid urbanization,and the spatiotemporal dynamic characteristics of its urban surface thermal environment from2010 to 2019 were evaluated.The random forest(RF)algorithm was adopted to obtain its land surface temperature(LST)map with 30-m resolution by downscaling the Moderate Resolution Imaging Spectroradiometer(MODIS)LST product;the remote sensing indices emphasizing different land cover types,LST calculated by the radiative transfer equation,and elevation were used as input variables in the algorithm.The heat island intensity(HII),urban heat island(UHI)volume,and UHI grade were used to analyze the spatiotemporal dynamic characteristics of the urban surface thermal environment in Qingdao.The results show an increasing trend in average HII between 1.1 and 2.52°C in the study area over the past 10 years.The northern city appeared to have the highest UHI volume,while change of the UHI volume in Huangdao District of southwestern Qingdao was the most significant.The areas with high HII have gradually expanded during the last 10 years,and the areas with a 10-yr persistently high HII are distributed mainly in old urban areas with high building density and a dense population.Different factors may influence UHI,such as artificial heat sources,surface heat sources,and hybrid heat sources.Finally,adjusting the urban structure,increasing the vegetated area,and changing building colors are proposed to mitigate UHI in the areas with continuously high HII.Nuo XU Fan DENG Bingqi LIU Caixia LI Hancong FU Huan YANG Jiahua ZHANG 2021Journal of Meteorological Research2021,35,5:1
10Electrolyte-dependent formation of solid electrolyte interphase and ion intercalation revealed by in situ surface characterizations显示文摘The formation of solid electrolyte interphase(SEI) and ion intercalation are two key processes in rechargeable batteries, which need to be explored under dynamic operating conditions. In this work, both planar and sandwich model lithium batteries consisting of Li metal | ionic liquid electrolyte | graphite electrode have been constructed and investigated by a series of in situ surface analysis platforms including atomic force microscopy, Raman and X-ray photoelectron spectroscopy. It is found that the choice of electrolyte, including the concentration and contents, has a profound effect on the SEI formation and evolution, and the subsequent ion intercalation. A smooth and compact SEI is preferably produced in highconcentration electrolytes, with FSI^(-) salt superior to TFSI^(-) salt, facilitating the lithiation/delithiation to achieve high capacity and excellent cycle stability, while suppressing the co-intercalation of electrolyte solvent ions. 13Exploration of Zinc(II)Complexes as Potent Inhibitors Against Protein Tyrosine Phosphatase IB显示文摘Although protein tyrosine phosphatases(PTPs)do not contain any metals,their activities can be inhibited by some metal complexes.Here we investigated the inhibition of two zinc complexes with Schiff base ligands against PTPs activity to explore their effect on the cellular metabolism.It has been found that they are potent inhibitors against four recombinant PIPs5 including protein tyrosine phosphatase 1B(PTP1B),T cell protein tyrosine phosphatase(TCPTP),megakaryocyte protein tyrosine phosphatase 2(PTP-MEG2)S and Src-homology phosphatase 1(SHP-1),with exception of Src-homology phosphatase 2(SHP-2).Moreover,tlaey showed moderate selective inhibition against PTP1B with the IG50 values of 0.15 and 0.36μmol/L.Meanwhile,the complexes also inhibited cellular phosphatase activities efficiently.Comparing the inhibitory potency over PTPs mediated by the zinc ion,we found that zinc complexes might be easily developed into potent and selective inhibitors against certain PTP by rationally modifying the organic ligands moieties.LI Xinhua YUAN Caixia LU Liping ZHU Miaoli XING Shu FU Xueqi 2019Chemical Research in Chinese Universities2019,35,2:1
14Intercalation-etching of graphene on Pt(111) in H_2 and O_2 observed by in-situ low energy electron microscopy显示文摘Graphene layers are often exposed to gaseous environments in their synthesis and application processes, and interactions of graphene surfaces with molecules particularly H_2 and O_2 are of great importance in their physico-chemical properties. In this work, etching of graphene overlayers on Pt(111) in H_2 and O_2 atmospheres were investigated by in-situ low energy electron microscopy. Significant graphene etching was observed in 10^(-5) Torr H_2 above 1023 K, which occurs simultaneously at graphene island edges and interiors with a determined reaction barrier at 5.7 eV. The similar etching phenomena were found in 10.7 Torr O_2 above 973 K, while only island edges were reacted between 823 and 923 K. We suggest that etching of graphene edges is facilitated by Pt-aided hydrogenation or oxidation of edge carbon atoms while intercalation-etching is attributed to etching at the interiors at high temperatures. 15Visualizing surface-enriched Li storage with a nanopore-array model battery显示文摘The coupling of model batteries and surface-sensitive techniques provides an indispensable platform for interrogating the vital surface/interface processes in battery systems.Here,we report a sandwich-format nanopore-array model battery using an ultrathin graphite electrode and an anodized aluminum oxide(AAO)film.The porous framework of AAO regulates the contact pattern of the electrolyte with the graphite electrode from the inner side,while minimizing contamination on the outer surface.This model battery facilitates repetitive charge-discharge processes,where the graphite electrode is reversibly intercalated and deintercalated,and also allows for the in-situ characterizations of ion intercalation in the graphite electrode.The ion distribution profiles indicate that the intercalating Li ions accumulate in both the inner and outer surface regions of graphite,generating a high capacity of~455 mAh·g^(-1)(theory:372 mAh·g^(-1)).The surface enrichment presented herein provides new insights towards the mechanistic understanding of batteries and the rational design strategies.Shiwen Li Guohui Zhang Chao Wang Caixia Meng Xianjin Li Yanxiao Ning Qiang Fu 2023Nano Research2023,16,4:0
16Qu-2,a robust poplar suspension cell line for molecular biology显示文摘Populus spp.have long been used as model woody plant species for molecular biology research.However,tissues of poplar are often recalcitrant to experimental procedures for molecular studies.We generated a hormone autotrophic poplar suspension cell line from a hybrid of Populus alba×P.berolinensis‘Yinzhong’,named Qu-2.Qu-2 cells are suitable as a model biological system for studying woody plants.Qu-2 cells have many advantages over suspension cell lines derived so far from any other woody plants.Qu-2 cells are very easy to cultivate and can grow on several common plant culture media without the addition of any plant hormone.They show exceptionally high growth rates,reaching an approximately 150-fold increase in biomass after one week of culturing.Another important unique characteristic of Qu-2 cells is that they can be cryopreserved and readily reactivated.Qu-2 cells are suitable for molecular manipulations such as protoplast production,transient transformation,and RNA-seq analysis.Therefore,Qu-2 cells have the great potential to be an excellent model cell line in tree molecular biological research,ranging from physiology to gene function.The Qu-2 cells will be made available to the plant community for research.Caixia Liu Kailong Li Meng Wang Erqin Fan Chuanping Yang Junhui Wang Pengyue Fu Xiaolan Ge Heike W.Sederoff Ronald RSederoff Vincent LChiang Sui Wang Guanzheng Qu 2021Journal of Forestry Research2021,32,2:0
17Revealing the effect of electrolyte coordination structures on the intercalation chemistry of batteries显示文摘In-depth understanding of the electrolyte-dependent intercalation chemistry in batteries through direct operando/in situ characterizations is crucial for the development of the high-performance batteries.Herein,taking the Al/graphite battery as a model system,the effect of electrolyte coordination structure on the intercalation processes has been investigated over the batteries with either 1-hexyl-3-methylimidazolium chloride(HMICl)-AlCl_(3) or 1-ethyl-3-methylimidazolium chloride(EMICl)-AlCl_(3) ionic liquid electrolyte using operando X-ray photoelectron spectroscopy(XPS)and X-ray diffraction.With a weaker anion-cation interaction in HMI-based electrolyte,the XPS-derived atomic ratio between cointercalated N and intercalated Al is 0.9,which is lower than 1.6 for EMI-based electrolyte.Attributed to the additional de-solvation process,the batteries with the HMI-based electrolyte show a lower ionic diffusion rate,capacity,and cycling performance,which agree with the operando characterization results.Our findings highlight the critical role of the electrolyte coordination structure on the(co-)intercalation chemistry.Chao Wang Xianjin Li Guiming Zhong Caixia Meng Shiwen Li Guohui Zhang Yanxiao Ning Xianfeng Li Qiang Fu 2023Journal of Energy Chemistry2023,,6:0
18Genomic Perspectives on the Emerging SARS-CoV-2 Omicron Variant显示文摘A new variant of concern for SARS-CoV-2,Omicron(B.1.1.529),was designated by the World Health Organization on November 26,2021.This study analyzed the viral genome sequencing data of 108 samples collected from patients infected with Omicron.First,we found that the enrichment efficiency of viral nucleic acids was reduced due to mutations in the region where the primers anneal to.Second,the Omicron variant possesses an excessive number of mutations compared to other variants circulating at the same time(median:62 vs.45),especially in the Spike gene.Mutations in the Spike gene confer alterations in 32 amino acid residues,more than those observed in other SARS-CoV-2 variants.Moreover,a large number of nonsynonymous mutations occur in the codons for the amino acid residues located on the surface of the Spike protein,which could potentially affect the replication,infectivity,and antigenicity of SARS-CoV-2.Third,there are 53 mutations between the Omicron variant and its closest sequences available in public databases.Many of these mutations were rarely observed in public databases and had a low mutation rate.In addition,the linkage disequilibrium between these mutations was low,with a limited number of mutations concurrently observed in the same genome,suggesting that the Omicron variant would be in a different evolutionary branch from the currently prevalent variants.To improve our ability to detect and track the source of new variants rapidly,it is imperative to further strengthen genomic surveillance and data sharing globally in a timely manner.Wentai Ma Jing Yang Haoyi Fu Chao Su Caixia Yu Qihui Wang Ana Tereza Ribeiro de Vasconcelos Georgii A.Bazykin Yiming Bao Mingkun Li 2022Genomics, Proteomics & Bioinformatics2022,20,1:0

20Step-confined thin film growth via near-surface atom migration显示文摘Understanding of thin film growth mechanism is crucial for tailoring film growth behaviors,which in turn determine physicochemical properties of the resulting films.Here,vapor-growth of tungsten carbide overlayers on W(110)surface is investigated by real time low energy electron microscopy.The surface growth is strongly confined by surface steps,which is in contrast with overlayer growth crossing steps in a so-called carpet-like growth mode for example in graphene growth on metal surfaces.Density functional theory calculations indicate that the step-confined growth is caused by the strong interaction of the forming carbide overlayer with the substrate blocking cross-step growth of the film.Furthermore,the tungsten carbide growth within each terrace is facilitated by the supply of carbon atoms from near-surface regions at high temperatures.These findings suggest the critical role of near-surface atom diffusion and step confinement effects in the thin film growth,which may be active in many film growth systems.Caixia Meng Junfeng Gao Rongtan Li Yanxiao Ning Yuan Chang Rentao Mu Qiang Fu Xinhe Bao 2020Nano Research2020,13,6:0
