1The correlation between rat retinal nerve fiber layer thickness around optic disc by using optical coherence tomography and histological measurements显示文摘AIM:To explore the correlation between the retinal nerve fiber layer(RNFL) thickness by using optical coherence tomography(OCT) and by histological measurements in normal adult rats and optic nerve transected rats.·METHODS:The RNFL thickness of 36 rats was scanned in a circle 3.46mm far from the optic disc by OCT.The two experimental groups were the normal group(n =20 rats) and the optic nerve transected group(n =16 rats).The latter group included 4 groups(n =4 /group)surviving for 1 day,3,5 and 7 days.Then the RNFL thickness of the same retina area was also measured by NF-200 immunohistochemical staining method.Linear regression was used to analyze the correlation between the data obtained from these two methods.·RESULTS:The RNFL thickness of normal right eyes around optic disc by OCT was 72.35 ±5.71μm and that ofthe left eyes was 72.65 ±5.88μm(P =0.074).The RNFL thickness of the corresponding histological section by immunohistochemistry was 37.54 ±4.05μm(right eyes)and 37.38 ±4.23μm(left eyes)(P =0.059).There was a good correlation between the RNFL thickness measured by OCT and that measured by histology(R2=0.8131).After optic nerve transection,the trend of the RNFL thickness was thinner with the prolonged survival time.The correlation of the thickness detected by the above two methods was approximately(R2=0.8265).Value of the RNFL thickness in rats around optic disc measured by OCT was obviously higher than that measured by common histological measurement in normal adult rats and optic nerve transected rats.·CONCLUSION:The RNFL thickness measured by OCT has a strong correlation with that measured by histological method.Through OCT scanning,we found that the thickness of RNFL gradually becomes thinner in a time-dependent manner.Ju-Fang Huang Hui-Min Yu Lei Shang Ruo-Fei Ma Ngobe Akume Cynthia Yan-Qun Cao Jia Luo Le-Ping Zeng Dan Chen Kun Xiong 2013International Journal of Ophthalmology(English edition)2013,6,4:2

16A machine learning model for colorectal liver metastasis post-hepatectomy prognostications显示文摘Background:Currently,surgical resection is the mainstay for colorectal liver metastases(CRLM)management and the only potentially curative treatment modality.Prognostication tools can support patient selection for surgical resection to maximize therapeutic benefit.This study aimed to develop a survival prediction model using machine learning based on a multicenter patient sample in Hong Kong.Methods:Patients who underwent hepatectomy for CRLM between 1 January 2009 and 31 December 2018 in four hospitals in Hong Kong were included in the study.Survival analysis was performed using Cox proportional hazards(CPH).A stepwise selection on Cox multivariable models with Least Absolute Shrinkage and Selection Operator(LASSO)regression was applied to a multiply-imputed dataset to build a prediction model.The model was validated in the validation set,and its performance was compared with that of Fong Clinical Risk Score(CRS)using concordance index.Results:A total of 572 patients were included with a median follow-up of 3.6 years.The full models for overall survival(OS)and recurrence-free survival(RFS)consist of the same 8 established and novel variables,namely colorectal cancer nodal stage,CRLM neoadjuvant treatment,Charlson Comorbidity Score,pre-hepatectomy bilirubin and carcinoembryonic antigen(CEA)levels,CRLM largest tumor diameter,extrahepatic metastasis detected on positron emission-tomography(PET)-scan as well as KRAS status.Our CRLM Machine-learning Algorithm Prognostication model(CMAP)demonstrated better ability to predict OS(C-index=0.651),compared with the Fong CRS for 1-year(C-index=0.571)and 5-year OS(C-index=0.574).It also achieved a C-index of 0.651 for RFS.Conclusions:We present a promising machine learning algorithm to individualize prognostications for patients following resection of CRLM with good discriminative ability.Cynthia Sin Nga Lam Alina Ashok Bharwani Evelyn Hui Yi Chan Vernice Hui Yan Chan Howard Lai Ho Au Margaret Kay Ho Shireen Rashed Bernard Ming Hong Kwong Wentao Fang Ka Wing Ma Chung Mau Lo Tan To Cheung 2023Hepatobiliary Surgery and Nutrition2023,12,4:0
17MutL homolog 1 methylation and microsatellite instability in sporadic colorectal tumors among Filipinos显示文摘BACKGROUND Colorectal cancer(CRC)ranks third in terms of incidence and second in mortality worldwide.In CRC,the silencing of mismatch repair genes,including the mutL homolog 1(hMLH1)has been linked to microsatellite instability(MSI),the lengthening or shortening of microsatellite repeats.Very limited data have been presented so far on the link of hMLH1 methylation and MSI in Southeast Asia populations with sporadic CRC,and on its clinical significance.AIM To investigate the significance of the MSI status and hMLH1 methylation in CRC Filipino patients.METHODS Fifty-four sporadic CRC patients with complete clinical data were included in this study.Genomic DNA from CRC tumor biopsies and their normal tissue counterparts were profiled for MSI by high resolution melting(HRM)analysis using the Bethesda Panel of Markers(BAT25,BAT26,D2S123,D5S346,and D17S250).hMLH1 methylation screening was performed using bisulfite conversion and methylation specific polymerase chain reaction.Statistical analysis was conducted to calculate their associations to clinicopathological characteristics and survival relevance(Kaplan-Meier curves and the log-rank test).RESULTS hMLH1 methylation was observed in 9%and 35%of CRC and normal samples,respectively.Higher incidence of consistently methylated hMLH1 found in both normal and CRC was noticed for relation to location of tumor(P<0.05).As for MSI status,D2S123 the most common unstable microsatellite and MSI-high(MSIH)was the most common MSI profile,counted for 46%and 50%of normal and CRC tissues,respectively.The presence of MSI-low(MSI-L)and microsatellite stable(MSS)was 43%and 11%for normal,and 31%and 19%for CRC samples.The mean month of patients’survival was shorter in patients whose normal and tumor tissues had methylated compared to those with unmethylated hMLH1 and with MSI-H compared to those with MSI-L/MSS(P<0.05).This was supported by significant difference in Kaplan-Meier with log-rank analysis.This data indicated that hMLH1 methylation and high MSI status have prognostic value.CONCLUSION This study showed the clinical significance of hMLH1 methylation and MSI status in sporadic CRC Filipino patients,especially in the normal part of the tumor.Loraine Kay D Cabral Cynthia A Mapua Filipinas F Natividad Caecilia H C Sukowati Edgardo R Cortez Ma Luisa D Enriquez 2021World Journal of Gastrointestinal Oncology2021,13,12:0
18Mass transport in a highly immiscible alloy on extended shear deformation显示文摘Forced mixing to a single-phase or supersaturated solid solution(SSS)and its prerequisite microstructure evolution in immiscible systems has been a focus of research for fundamental science and practical applications.Controlling the formation of SSS by shear deformation could enable a material design beyond conventional equilibrium microstructure in immiscible systems.Here,a highly immiscible Cu-50 at.%Cr binary alloy(mixing enthalpy of∼20 kJ mol^(−1))was employed to investigate the microstructure evolution and localized tendencies of SSS during severe shear deformation.Our results demonstrate the dislocation mediated microstructural refinement process in each phase of the binary alloy and the mechanisms associated with localized solute supersaturation as a function of shear strain.Pronounced grain refinement in the softer Cu phase occurs owing to the strain localization driving the preferential dynamic recrystallization.The grain refinement of the Cr phase,however,is enabled by the progressive evolution of grain lamination,splitting,and fragmentation as a function of shear strain.The solute supersaturation is found to be strongly dependent on the local environments that affect the dislocation activity,including the level of microstructure refinement,the interfacial orientation relationship,the mechanical incompatibility,and the localized preferential phase oxidation.Ab initio simulations confirm that it is more favorable to oxidize Cr than Cu at incoherent Cu/Cr interfaces,limiting the mass transport on an incoherent boundary.Our results unveil the mechanism underpinning the non-equilibrium mass transport in immiscible systems upon severe deformation that can be applied to produce immiscible alloys with superior mechanical properties.Miao Song Jia Liu Xiaolong Ma Qin Pang Matthew J.Olszta Joshua Silverstein Madhusudhan R.Pallaka Peter V.Sushko Suveen N.Mathaudhu Cynthia Powell Arun Devaraj Bharat Gwalani 2023Journal of Materials Science & Technology2023,,3:0
