| 1 | Dental stem cell and dental tissue regeneration显示文摘The teeth are highly differentiated chewing organs formed by the development of tooth germ tissue located in the jaw and consist of the enamel, dentin, cementum, pulp, and periodontal tissue. Moreover, the teeth have a complicated regulatory mechanism, special histologic origin, diverse structure, and important function in mastication, articulation, and aesthetics. These characteristics, to a certain extent, greatly complicate the research in tooth regeneration. Recently, new ideas for tooth and tissue regeneration have begun to appear with rapid developments in the theories and technologies in tissue engineering. Numerous types of stem cells have been isolated from dental tissue, such as dental pulp stem cells (DPSCs), stem cells isolated from human pulp of exfoliated deciduous teeth (SHED), periodontal ligament stem cells (PDLSCs), stem cells from apical papilla (SCAPs), and dental follicle cells (DFCs). All these cells can regenerate the tissue of tooth. | 2 | Factors Associated with Thrombolysis Outcome in Ischemic Stroke Patients with Atrial Fibrillation显示文摘The outcome of early intravenous thrombolysis for ischemic stroke in patients with atrial fibrillation(AF)is worse than that without thrombosis. How to increase the efficacy of intravenous thrombolysis for AF-related ischemic stroke remains largely unknown. In this study, we investigated factors that influence the effect of intravenous thrombolysis in these patients. Our results showed that thrombolysis was independently associated with a favorable outcome(P \ 0.001) and did not influence the mortality of AF-related ischemic stroke, although it increased the risk of hemorrhage within 24 h after treatment. Risk factors for a poor outcome at admission were:heart failure(P = 0.045); high systolic pressure(P = 0.039); high blood glucose(P = 0.030); and a high National Institutes of Health Stroke Scale(NIHSS) score(P \ 0.001). Moreover, high systolic pressure at admission(P = 0.007), high blood glucose(P = 0.027), and a high NIHSS score(P \ 0.001) were independent risk factors for mortality at 3 months. Besides thrombolysis, a high NIHSS score(P = 0.006) and warfarin taken within 48 h before stroke onset(P = 0.032) were also independent risk factors for symptomatic hemorrhage within 24 h after treatment. | 5 | Observation and modeling on irregular purple soil water infiltration process显示文摘The infiltration process is a critical link between surface water and groundwater. In this research, a specific device to observe infiltration processes in homogeneous and heterogeneous soils with triangular and inverted triangular profiles was designed, and the Green-Ampt model was employed for the process simulation. The results indicate that(1) the wetting front in coarse texture soils transports faster than in fine texture soils;(2) for the homogeneous case, the wetting front in triangularshaped soils transports faster than the inverted triangular type, but the triangular-shaped soils show a lower infiltration rate;(3) in the initial step, the wetting front in triangular-shaped soils shows higher transport speed, but depicts lower speed with increase in the time;(4) both the wetting front and infiltration rate show a significant exponential relation with the time. | 6 | Ionomycin ameliorates hypophosphatasia via rescuing alkaline phosphatase deficiency-mediated L-type Ca^(2+) channel internalization in mesenchymal stem cells显示文摘The loss-of-function mutations in the ALPL result in hypophosphatasia(HPP), an inborn metabolic disorder that causes skeletal mineralization defects. In adults, the main clinical features are early loss of primary or secondary teeth, osteoporosis, bone pain,chondrocalcinosis, and fractures. However, guidelines for the treatment of adults with HPP are not available. Here, we show that ALPL deficiency caused a reduction in intracellular Ca2+ influx, resulting in an osteoporotic phenotype due to downregulated osteogenic differentiation and upregulated adipogenic differentiation in both human and mouse bone marrow mesenchymal stem cells(BMSCs). Increasing the intracellular level of calcium in BMSCs by ionomycin treatment rescued the osteoporotic phenotype in alpl+/- mice and BMSC-specific(Prrx1-alpl-/-) conditional alpl knockout mice. Mechanistically, ALPL was found to be required for the maintenance of intracellular Ca2+ influx, which it achieves by regulating L-type Ca2+ channel trafficking via binding to the α2δsubunits to regulate the internalization of the L-type Ca2+ channel. Decreased Ca2+ flux inactivates the Akt/GSK3β/β-catenin signaling pathway, which regulates lineage differentiation of BMSCs. This study identifies a previously unknown role of the ectoenzyme ALPL in the maintenance of calcium channel trafficking to regulate stem cell lineage differentiation and bone homeostasis. | 9 | Characterization and Expression Analysis of a Complement Component Gene in Sea Cucumber(Apostichopus japonicus)显示文摘The complement system plays a crucial role in the innate immune system of animals. It can be activated by distinct yet overlapping classical, alternative and lectin pathways. In the alternative pathway, complement factor B(Bf) serves as the catalytic subunit of complement component 3(C3) convertase, which plays the central role among three activation pathways. In this study, the Bf gene in sea cucumber(Apostichopus japonicus), termed Aj Bf, was obtained by rapid amplification of c DNA ends(RACE). The full-length c DNA of Aj Bf was 3231 bp in length barring the poly(A) tail. It contained an open reading frame(ORF) of 2742 bp encoding 913 amino acids, a 105 bp 5'-UTR(5'-terminal untranslated region) and a 384 bp 3'-UTR. Aj Bf was a mosaic protein with six CCP(complement control protein) domains, a VWA(von Willebrand factor A) domain, and a serine protease domain. The deduced molecular weight of Aj Bf protein was 101 k Da. Quantitative real time PCR(q RT-PCR) analysis indicated that the expression level of Aj Bf in A. japonicus was obviously higher at larval stage than that at embryonic stage. Expression detection in different tissues showed that Aj Bf expressed higher in coelomocytes than in other four tissues. In addation, Aj Bf expression in different tissues was induced significantly after LPS or Poly I:C challenge. | 12 | Identification of banana fusarium wilt using supervised classification algorithms with UAV-based multi-spectral imagery显示文摘The disease of banana Fusarium wilt currently threatens banana production areas all over the world.Rapid and large-area monitoring of Fusarium wilt disease is very important for the disease treatment and crop planting adjustments.The objective of this study was to evaluate the performance of supervised classification algorithms such as support vector machine(SVM),random forest(RF),and artificial neural network(ANN)algorithms to identify locations that were infested or not infested with Fusarium wilt.An unmanned aerial vehicle(UAV)equipped with a five-band multi-spectral sensor(blue,green,red,red-edge and near-infrared bands)was used to capture the multi-spectral imagery.A total of 139 ground sample-sites were surveyed to assess the occurrence of banana Fusarium wilt.The results showed that the SVM,RF,and ANN algorithms exhibited good performance for identifying and mapping banana Fusarium wilt disease in UAV-based multi-spectral imagery.The overall accuracies of the SVM,RF,and ANN were 91.4%,90.0%,and 91.1%,respectively for the pixel-based approach.The RF algorithm required significantly less training time than the SVM and ANN algorithms.The maps generated by the SVM,RF,and ANN algorithms showed the areas of occurrence of Fusarium wilt disease were in the range of 5.21-5.75 hm2,accounting for 36.3%-40.1%of the total planting area of bananas in the study area.The results also showed that the inclusion of the red-edge band resulted in an increase in the overall accuracy of 2.9%-3.0%.A simulation of the resolutions of satellite-based imagery(i.e.,0.5 m,1 m,2 m,and 5 m resolutions)showed that imagery with a spatial resolution higher than 2 m resulted in good identification accuracy of Fusarium wilt.The results of this study demonstrate that the RF classifier is well suited for the identification and mapping of banana Fusarium wilt disease from UAV-based remote sensing imagery.The results provide guidance for disease treatment and crop planting adjustments. | Huichun Ye Wenjiang Huang Shanyu Huang Bei Cui Yingying Dong Anting Guo Yu Ren Yu Jin | 2020 | International Journal of Agricultural and Biological Engineering2020,13,3: | 3 |

| 15 | Quantification and prediction of enteric methane emissions from Chinese lactating Holstein dairy cows fed diets with different dietary neutral detergent fiber/non-fibrous carbohydrate(NDF/NFC)ratios显示文摘Methane(CH_(4))emissions from ruminant production are a significant source of anthropogenic greenhouse gas production,but few studies have examined the enteric CH_(4)emissions of lactating dairy cows under different feeding regimes in China.This study aimed to investigate the influence of different dietary neutral detergent fiber/non-fibrous carbohydrate(NDF/NFC)ratios on production performance,nutrient digestibility,and CH_(4)emissions for Holstein dairy cows at various stages of lactation.It evaluated the performance of CH_(4)prediction equations developed using local dietary and milk production variables compared to previously published prediction equations developed in other production regimes.For this purpose,36 lactating cows were assigned to one of three treatments with differing dietary NDF/NFC ratios:low(NDF/NFC=1.19),medium(NDF/NFC=1.54),and high(NDF/NFC=1.68).A modified acid-insoluble ash method was used to determine nutrient digestibility,while the sulfur hexafluoride technique was used to measure enteric CH4 emissions.The results showed that the dry matter(DM)intake of cows at the early,middle,and late stages of lactation decreased significantly(P<0.01)from 20.9 to 15.4 kg d^(–1),15.3 to 11.6 kg d^(–1),and 16.4 to 15.0 kg d^(–1),respectively,as dietary NDF/NFC ratios increased.Across all three treatments,DM and gross energy(GE)digestibility values were the highest(P<0.05)for cows at the middle and late lactation stages.Daily CH_(4)emissions increased linearly(P<0.05),from 325.2 to 391.9 kg d^(–1),261.0 to 399.8 kg d^(–1),and 241.8 to 390.6 kg d^(–1),respectively,as dietary NDF/NFC ratios increased during the early,middle,and late stages of lactation.CH_(4)emissions expressed per unit of metabolic body weight,DM intake,NDF intake,or fat-corrected milk yield increased with increasing dietary NDF/NFC ratios.In addition,CH_(4)emissions expressed per unit of GE intake increased significantly(P<0.05),from 4.87 to 8.12%,5.16 to 9.25%,and 5.06 to 8.17%respectively,as dietary NDF/NFC ratios increased during the early,middle,and late lactation stages.The modelling results showed that the equation using DM intake as the single variable yielded a greater R^(2)than equations using other dietary or milk production variables.When data obtained from each lactation stage were combined,DM intake remained a better predictor of CH_(4)emissions(R^(2)=0.786,P=0.026)than any other variables tested.Compared to the prediction equations developed herein,previously published equations had a greater root mean square prediction error,reflecting their inability to predict CH_(4)emissions for Chinese Holstein dairy cows accurately.The quantification of CH_(4)production by lactating dairy cows under Chinese production systems and the development of associated prediction equations will help establish regional or national CH_(4)inventories and improve mitigation approaches to dairy production. | DONG Li-feng JIA Peng LI Bin-chang WANG Bei YANG Chun-lei LIU Zhi-hao DIAO Qi-yu | 2022 | Journal of Integrative Agriculture2022,21,3: | 2 |

| 18 | Going below and beyond the surface: Microneedle structure, materials, drugs, fabrication, and applications for wound healing and tissue regeneration显示文摘Microneedle, as a novel drug delivery system, has attracted widespread attention due to its non-invasiveness, painless and simple administration, controllable drug delivery, and diverse cargo loading capacity. Although microneedles are initially designed to penetrate stratum corneum of skin for transdermal drug delivery, they, recently, have been used to promote wound healing and regeneration of diverse tissues and organs and the results are promising. Despite there are reviews about microneedles, few of them focus on wound healing and tissue regeneration. Here, we review the recent advances of microneedles in this field. We first give an overview of microneedle system in terms of its potential cargos (e.g., small molecules, macromolecules, nucleic acids, nanoparticles, extracellular vesicle, cells), structural designs (e.g., multidrug structures, adhesive structures), material selection, and drug release mechanisms. Then we briefly summarize different microneedle fabrication methods, including their advantages and limitations. We finally summarize the recent progress of microneedle-assisted wound healing and tissue regeneration (e.g., skin, cardiac, bone, tendon, ocular, vascular, oral, hair, spinal cord, and uterine tissues).
