1Inhibition of SARS-CoV-2 (previously 2019-nCoV) infection by a highly potent pan-coronavirus fusion inhibitor targeting its spike protein that harbors a high capacity to mediate membrane fusion显示文摘The recent outbreak of coronavirus disease(COVID-19)caused by SARS-CoV-2 infection in Wuhan,China has posed a serious threat to global public health.To develop specific anti-coronavirus therapeutics and prophylactics,the molecular mechanism that underlies viral infection must first be defined.Therefore,we herein established a SARS-CoV-2 spike(S)protein-mediated cell-cell fusion assay and found that SARS-CoV-2 showed a superior plasma membrane fusion capacity compared to that of SARS-CoV.We solved the X-ray crystal structure of six-helical bundle(6-HB)core of the HR1 and HR2 domains in the SARS-CoV-2 S protein S2 subunit revealing that several mutated amino acid residues in the HR1 domain may be associated with enhanced interactions with the HR2 domain.We previously developed a pan-coronavirus fusion inhibitor,EK1,which targeted the HR!domain and could inhibit infection by divergent human coronaviruses tested,including SARS-CoV and MERS-CoV.Here we generated a series of lipopeptides derived from EK1 and found that EK1C4 was the most potent fusion inhibitor against SARS-CoV-2 S protein-mediated membrane fusion and pseudovirus infection with IC50s of 1.3 and 15.8 nM,about 241-and 149-fold more potent than the original EK1 peptide,respectively.EK1C4 was also highly effective against membrane fusion and infection of other human coronavirus pseudoviruses tested,including SARS-CoV and MERS-CoV,as well as SARSr-CoVs,and potently inhibited the replication of 5 live human coronaviruses examined,including SARS-CoV-2.Intranasal application of EK1C4 before or after challenge with HCoV-OC43 protected mice from infection,suggesting that EK1C4 could be used for prevention and treatment of infection by the currently circulating SARS-CoV-2 and other emerging SARSr-CoVs.Shuai Xia Meiqin Liu Chao Wang Wei Xu Qiaoshuai Lan Siliang Feng Feifei Qi Linlin Bao Lanying Du Shuwen Liu Chuan Qin Fei Sun Zhengli Shi Yun Zhu Shibo Jiang Lu Lu 2020Cell Research2020,30,4:81

4Positive role of nitrite as electron acceptor on anoxic denitrifying phosphorus removal process显示文摘Literatures revealed that the electron acceptor-nitrite could be inhibitory or toxic in the denitrifying phosphorus removal process. Batch test experiments were used to investigate the inhibitory effect during the anoxic condition. The inoculated activated sludge was taken from a continuous double-sludge denitrifying phosphorus and nitrogen removal system. Nitrite was added at the anoxic stage. One time injection and sequencing batch injection were carried on in the denitrifying dephosphorus procedure. The results indicated that the nitrite concentration higher than 30 mg/L would inhibit the anoxic phosphate uptake severely,and the threshold inhibitory concentration was dependent on the characteristics of the activated sludge and the operating conditions; instead,lower than the inhibitory concentration would not be detrimental to anoxic phosphorus uptake,and it could act as good electron acceptor for the anoxic phosphate accumulated. Positive effects performed during the denitrifying biological dephosphorus all the time. 7Highly active OMS-2 for catalytic ozone decomposition under humid conditions显示文摘Four kinds of cryptomelane-type octahedral molecular sieve(OMS)-2-X(the X represents the molar ratio of KMnO4/MnAc2) were prepared as catalytic materials for ozone decomposition through a one-step hydrothermal reaction of KMnO4 and MnAc2, by changing their molar ratios. These samples were characterized by N2 adsorption–desorption, X-ray di raction(XRD), transmission electron microscopy(TEM), scanning electron microscopy(SEM), temperature programmed reduction by H2(H2-TPR) and X-ray photoelectron spectroscopy(XPS). Among them, the OMS-2-0.7 sample showed the best O3 conversion of 92% under high relative humidity(RH) of 90% and gas hourly space velocity of 585,000 h-1. This was accordingly thought as a possible way for purifying ozone-containing waste gases under high RH atmospheres. The e ciency of ozone decomposition of the prepared OMS-2-X sample was found to be related to specific surface area, particle size, surface oxygen vacancies, and Mn3+ cation amounts. 9Seismic behavior of breakwaters on complex ground by numerical tests:Liquefaction and post liquefaction ground settlements显示文摘A large number of breakwaters have been constructed along coasts to protect humans and infrastructures from tsunamis.There is a risk that foundation soils of these structures may liquefy,or partially liquefy during the earthquake preceding a tsunami,which would greatly reduce the structures’capacity to resist the tsunami.It is necessary to consider not only the soil’s liquefaction behavior due to earthquake motions but also its post-liquefaction behavior because this behavior will affect the breakwater’s capacity to resist an incoming tsunami.In this study,numerical tests based on a sophisticated constitutive model and a soil-water coupled finite element method are used to predict the mechanical behavior of breakwaters and the surrounding soils.Two real breakwaters subjected to two different seismic excitations are examined through numerical simulation.The simulation results show that,earthquakes affect not only the immediate behavior of breakwaters and the surrounding soils but also their long-term settlements due to post-earthquake consolidation.A soil profile with thick clayey layers beneath liquefied soil is more vulnerable to tsunami than a soil profile with only sandy layers.Therefore,quantitatively evaluating the seismic behavior of breakwaters and surrounding soils is important for the design of breakwater structures to resist tsunamis.Gu Linlin Zhang Feng Bao Xiaohua Shi Zhenming Ye Guanlin Ling Xianzhang 2018Earthquake Engineering and Engineering Vibration2018,17,2:3
10Sensitivity of SARS-CoV-2 to different temperatures显示文摘This study was designed to investigate the sensitivity of SARS-CoV-2 to different temperatures,to provide basic data and a scientific basis for the control of COVID-19 epidemic.The virus was dispersed in 1 mL basal DMEM medium at a final concentration of 103.2 TCID 50/mL and then incubated at 4,22,30,35,37,38,39 and 40°C for up to 5 days.The infectivity of residual virus was titrated using the Vero E6 cell line.The results showed that the virus remained viable for 5 days at 4°C,and for 1 day only at 22 and 30°C.We found that the infectivity of the virus was completely lost after less than 12 hours at 37,38 and 39°C,while at 40°C,the inactivation time of the virus was rapidly reduced to 6 hours.We show that SARS-CoV-2 is sensitive to heat,is more stable at lower temperatures than higher temperature,remains viable for longer at lower temperatures,and loses viability rapidly at higher temperatures.Qi Lv Mingya Liu Feifei Qi Shuran Gong Shasha Zhou Shisheng Zhan Linlin Bao 2020Animal Models and Experimental Medicine2020,3,4:3
11Integrated histopathological,lipidomic,and metabolomic profiles reveal mink is a useful animal model to mimic the pathogenicity of severe COVID-19 patients显示文摘Severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)is transmitted on mink farms between minks and humans in many countries.However,the systemic pathological features of SARS-CoV-2-infected minks are mostly unknown.Here,we demonstrated that minks were largely permissive to SARS-CoV-2,characterized by severe and diffuse alveolar damage,and lasted at least 14 days post inoculation(dpi).We first reported that infected minks displayed multiple organ-system lesions accompanied by an increased inflammatory response and widespread viral distribution in the cardiovascular,hepatobiliary,urinary,endocrine,digestive,and immune systems.The viral protein partially co-localized with activated Mac-2+macrophages throughout the body.Moreover,we first found that the alterations in lipids and metabolites were correlated with the histological lesions in infected minks,especially at 6 dpi,and were similar to that of patients with severe and fatal COVID-19.Particularly,altered metabolic pathways,abnormal digestion,and absorption of vitamins,lipids,cholesterol,steroids,amino acids,and proteins,consistent with hepatic dysfunction,highlight metabolic and immune dysregulation.Enriched kynurenine in infected minks contributed to significant activation of the kynurenine pathway and was related to macrophage activation.Melatonin,which has significant anti-inflammatory and immunomodulating effects,was significantly downregulated at 6 dpi and displayed potential as a targeted medicine.Our data first illustrate systematic analyses of infected minks to recapitulate those observations in severe and fetal COVID-19 patients,delineating a useful animal model to mimic SARS-CoV-2-induced systematic and severe pathophysiological features and provide a reliable tool for the development of effective and targeted treatment strategies,vaccine research,and potential biomarkers.Zhiqi Song Linlin Bao Wei Deng Jiangning Liu Erjun Ren Qi Lv Mingya Liu Feifei Qi Ting Chen Ran Deng Fengdi Li Yunpeng Liu Qiang Wei Hong Gao Pin Yu Yunlin Han Wenjie Zhao Junjun Zheng Xujian Liang Fuhe Yang Chuan Qin 2022Signal Transduction and Targeted Therapy2022,7,2:2

17Sequentially immune-induced antibodies could cross-neutralize SARS-CoV-2 variants显示文摘Background:The Omicron(B.1.1.529)SARS-COV-2 variant has raised serious concerns because of its unprecedented rapid rate of spreading and the fact that there are 36 mutations in the spike protein.Since the vaccine-induced neutralizing antibody targets are the spike protein,this may lead to the possibility of vaccine-induced hu-moral immunity escape.Methods:We measured the neutralizing activity in vitro for Omicron and compared this with wild type(WH-09)and Delta variants in human and monkey sera from different types of immunity.The monkey sera samples were collected at 1 and 3 months post three-dose inactivated(PiCoVacc)and recombinant protein(ZF2001)vaccination.Human sera were collected from 1 month post three-dose inactivated vaccination.Results:In inactivated vaccine sera,at 1/3 months post three-dose,geometric mean titers(GMTs)of neutralization antibody(NAb)against the Omicron variant were 4.9/5.2-fold lower than those of the wild type.In recombinant protein vaccine sera,GMTs of NAb against Omicron were 15.7/8.9-fold lower than those of the wild type.In human sera,at 1 month post three-dose inactivated vaccination,GMTs of NAb against Omicron were 3.1-fold lower than those of the wild type.Conclusion:This study demonstrated that despite a reduction in neutralization titers,cross-neutralizing activity against Omicron and Delta variants was still observed after three doses of inactivated and recombinant protein vaccination.Qi Lv Shasha Zhou Feifei Qi Yaqing Zhang Fengdi Li Mingya Liu Linlin Bao 2022Animal Models and Experimental Medicine2022,5,1:1

19A broadly neutralizing human monoclonal antibody against the hemagglutinin of avian influenza virus H7N9显示文摘Background:The new emerging avian influenza A H7N9 virus,causing severe human infection with a mortality rate of around 41%.This study aims to provide a novel treatment option for the prevention and control of H7N9.Methods:H7 hemagglutinin(HA)-specific B cells were isolated from peripheral blood plasma cells of the patients previously infected by H7N9 in Jiangsu Province,China.The human monoclonal antibodies(mAbs)were generated by amplification and cloning of these HA-specific B cells.First,all human mAbs were screened for binding activity by enzyme-linked immunosorbent assay.Then,those mAbs,exhibiting potent affinity to recognize H7 HAs were further evaluated by hemagglutination-inhibiting(HAI)and microneutralizationin vitro assays.Finally,the lead mAb candidate was selected and tested against the lethal challenge of the H7N9 virus using murine models.Results:The mAb 6-137 was able to recognize a panel of H7 HAs with high affinity but not HA of other subtypes,including H1N1 and H3N2.The mAb 6-137 can efficiently inhibit the HA activity in the inactivated H7N9 virus and neutralize 100 tissue culture infectious dose 50(TCID_(50))of H7N9 virus(influenza A/Nanjing/1/2013)invitro,with neutralizing activity as low as 78 ng/mL.In addition,the mAb 6-137 protected the mice against the lethal challenge of H7N9 prophylactically and therapeutically.Conclusion:The mAb 6-137 could be an effective antibody as a prophylactic or therapeutic biological treatment for the H7N9 exposure or infection.Jingxin Li Li Zhang Linlin Bao Yuxiao Wang Lin Qiu Jialei Hu Rong Tang Huiyan Yu Jun Shan Yan Li Chuan Qin Fengcai Zhu 2022Chinese Medical Journal2022,,7:1
20Population genetic structure in wild and aquaculture populations of Hemibarbus maculates inferred from microsatellites markers显示文摘The objective of this study was to investigate 4 aquaculture populations Shanghai(SH),Hangzhou(HZ),Kaihua(KH)and Xianju(XJ)and one wild population Yingshan(YS)of spotted barbell(Hemibarbus maculates)to assess their genetic diversity level and investigate the genetic structure of the populations.The dendrogram and STRUCTURE revealed that the populations XJ,KH,and HZ jointly formed one cluster,to which the populations SH and YS were sequentially adhered.The genetic diversity of the cultured populations maintained better,possible due to favourable hatchery conditions that decreased the effect of environmental selection present in wild populations.The results of the present study will contribute to the management of spotted barbell genetic resources,but also demonstrates how the genetic diversity of freshwater species is vulnerable to human activity.Linlin Li Hungdu Lin Wenqiao Tang Dong Liu Baolong Bao Jinquan Yang 2017Aquaculture and Fisheries2017,2,2:1
