1Transplantation of engineered exosomes derived from bone marrow mesenchymal stromal cells ameliorate diabetic peripheral neuropathy under electrical stimulation显示文摘Diabetic peripheral neuropathy(DPN)is a long-term complication associated with nerve dysfunction and uncontrolled hyperglycemia.In spite of new drug discoveries,development of effective therapy is much needed to cure DPN.Here,we have developed a combinatorial approach to provide biochemical and electrical cues,considered to be important for nerve regeneration.Exosomes derived from bone marrow mesenchymal stromal cells(BMSCs)were fused with polypyrrole nanoparticles(PpyNps)containing liposomes to deliver both the cues in a single delivery vehicle.We developed DPN rat model and injected intramuscularly the fused exosomal system to understand its long-term therapeutic effect.We found that the fused system along with electrical stimulation normalized the nerve conduction velocity(57.60±0.45 m/s)and compound muscle action potential(16.96±0.73 mV)similar to healthy control(58.53±1.10 m/s;18.19±1.45 mV).Gastrocnemius muscle morphology,muscle mass,and integrity were recovered after treatment.Interestingly,we also observed paracrine effect of delivered exosomes in controlling hyperglycemia and loss in body weight and also showed attenuation of damage to the tissues such as the pancreas,kidney,and liver.This work provides a promising effective treatment and also contribute cutting edge therapeutic approach for the treatment of DPN.Anamika Singh Alok Raghav Parvaiz Ahmad Shiekh Ashok Kumar 2021Bioactive Materials2021,6,8:4
2Efficient Microorganism Compost Benefits Plant Growth and Improves Soil Health in Calendula and Marigold显示文摘Organic farming needs to be promoted as a means of sustaining soil health and reducing the cost of farming, especially for small farmers.The present study deals with the effect of organic compost, prepared using Efficient Microorganism(EM) consortium and applied along with full or half of the recommended dose of chemical fertilizers, on the growth of Calendula and Marigold plants, soil physico-chemical parameters and soil enzyme activities. Soil enzyme activities were improved with the increase in the rate of EM compost application in both Calendula and Marigold. Carotenoid pigment increased by 46.11% and 12.19% with application of EM compost over the control in Calendula and Marigold flowers respectively. Soil humus, available nitrogen and organic carbon content also increased due to the supplementation of EM compost resulting in better soil fertility. For Calendula, treatment T5(Half dose NPK + EM compost 20 000 kg·hm^(-2)) was found to be the most promising in terms of acid phosphatase(82.63 μg p-Nitrophenyl Phosphate·g^(-1)·h^(-1)), dehydrogenase(10.46 μg Triphenyl Formazan·g^(-1)·d^(-1)) and β-glucosidase(0.30 IU·g^(-1)) activities. 3A secondary bile acid from microbiota metabolism attenuates ileitis and bile acid reduction in subclinical necrotic enteritis in chickens显示文摘Background:Clostridium perfringens-induced chicken necrotic enteritis(NE)is responsible for substantial economic losses worldwide annually.Recently,as a result of antibiotic growth promoter prohibition,the prevalence of NE in chickens has reemerged.This study was aimed to reduce NE through titrating dietary deoxycholic acid(DCA)as an effective antimicrobial alternative.Materials and methods:Day-old broiler chicks were assigned to six groups and fed diets supplemented with 0(basal diet),0.8,1.0 and 1.5 g/kg(on top of basal diet)DCA.The birds were challenged with Eimeria maxima(20,000 oocysts/bird)at d 18 and C.perfringens(109 CFU/bird per day)at d 23,24,and 25 to induce NE.The birds were sacrificed at d 26 when ileal tissue and digesta were collected for analyzing histopathology,mRNA accumulation and C.perfringens colonization by real-time PCR,targeted metabolomics of bile acids,fluorescence in situ hybridization(FISH),or terminal deoxynucleotidyl transferase dUTP nick end labeling(TUNEL)assay.Results:At the cellular level,birds infected with E.maxima and C.perfringens developed subclinical NE and showed shortening villi,crypt hyperplasia and immune cell infiltration in ileum.Dietary DCA alleviated the NE-induced ileal inflammation in a dose-dependent manner compared to NE control birds.Consistent with the increased histopathological scores,subclinical NE birds suffered body weight gain reduction compared to the uninfected birds,an effect attenuated with increased doses of dietary DCA.At the molecular level,the highest dose of DCA at 1.5 g/kg reduced C.perfringens luminal colonization compared to NE birds using PCR and FISH.Furthermore,the dietary DCA reduced subclinical NE-induced intestinal inflammatory gene expression and cell apoptosis using PCR and TUNEL assays.Upon further examining ileal bile acid pool through targeted metabolomics,subclinical NE reduced the total bile acid level in ileal digesta compared to uninfected birds.Notably,dietary DCA increased total bile acid and DCA levels in a dose-dependent manner compared to NE birds.Conclusion:These results indicate that DCA attenuates NE-induced intestinal inflammation and bile acid reduction and could be an effective antimicrobial alternative against the intestinal disease.Mohit Bansal Ying Fu Bilal Alrubaye Mussie Abraha Ayidh Almansour Anamika Gupta Rohana Liyanage Hong Wang Billy Hargis Xiaolun Sun 2020Journal of Animal Science and Biotechnology2020,11,3:3
4Enhanced Lead Uptake by an Association of Plant and Earthworm Bioaugmented with Bacteria显示文摘Lead(Pb) is recorded as the second most hazardous pollutant of the environment.Previous cases of Pb bioremediation has been reported using single biosystem,but very few reports are available in biological approaches using multi-biosystems to achieve an enhanced bioremoval of Pb.The present study evaluated the capacity of a unique association of Pennisetum purpureum,a hyperaccumulator plant,and Lumbricus terrestris(earthworm)bioaugmented with a Pb-resistant bacterium,obtained from an industrially contaminated site and identified as isolate VITMVCJ1 Klebsiella variicola,to bioremediate Pb.The Pb-resistant gene was amplified in the bacterial isolate VITMVCJ1.The study was conducted for 60 d.Results verified that the bioaugmentation process enhanced1)root and shoot length of the plants,2)chlorophyll content of the plants,and 3)biofilm-producing ability of the microbes from the rhizosphere region of the plants.The total phenolic and flavonoid contents were found to be lower in the plants in the bioaugmented setup.The study also observed a reduction in the toxic effects of Pb on earthworm and plant.The earthworm was used to assess the Pb-induced stress syndrome after exposure to sublethal concentrations of Pb in the soil.A reduction in the content of malondialdehyde,a potential biomarker,on exposure to Pb demonstrated the role of the inoculum to alleviate heavy metal-induced stress in earthworms.All three symbionts accumulated Pb;Pb was accumulated mainly in the root of the plant,and poorly in the shoot of the plant and body mass of the earthworm.The bioaugmentation system exhibited stable and excellent uptake of Pb from the contaminated soils.The results of the present study suggest the positive effect of the synergistic association of the plant and earthworm with appropriate microbes for the bioremoval of Pb.anamika das jabez w.osborne 2018Pedosphere2018,28,2:3
5Comparative response of seedlings of selected native dry tropical and alien invasive species to CO_(2) enrichment显示文摘Global climate change and ongoing plant invasion are the two prominent ecological issues threatening biodiversity world wide.Among invasive species,Lantana camara and Hyptis suaveolens are the two most important invaders in the dry deciduous forest in India.We monitored the growth of these two invasive species and seedlings of four native dry deciduous species(Acacia catechu,Bauhinia variegata,Dalbergia latifolia and Tectona grandis)under ambient(375–395μmol mol^(-1))and elevated CO_(2)(700–750μmol mol^(-1))to study the differential growth response of invasive and native seedlings.Methods Seedlings of all the species were exposed to ambient and elevated CO_(2).After 60 days of exposure,seedlings were harvested and all the growth-related parameters like plant height;biomass of root,stem and leaves;total seedling biomass;R/S ratio;allocation parameters;net assimilation rate(NAR)and relative growth rate(RGR)were determined.Important Findings Biomass,RGR and NAR of all the species increased under elevated CO_(2)but the increase was higher in invasive species and they formed larger seedlings than natives.Therefore under the CO_(2)-enriched future atmosphere,competitive hierarchies could change and may interfere with the species composition of the invaded area.Purnima Raizada Anamika Singh A.S.Raghubanshi 2009Journal of Plant Ecology2009,2,2:2

7Identification of groundwater potential in hard rock aquifer systems using Remote Sensing, GIS and Magnetic Survey in Veppanthattai, Perambalur, Tamilnadu显示文摘Water is an essential natural resource without which life wouldn’t exist.The study aims to identify groundwater potential areas in Vepapanthattai taluk of Perambalur district,Tamil Nadu,India,using analytic hierarchy process(AHP)model.Remote sensing and magnetic parameters have been used to determine the evaluation indicators for groundwater occurrence under the ArcGIS environment.Groundwater occurrence is linked to structural porosity and permeability over the predominantly hard rock terrain,making magnetic data more relevant for locating groundwater potential zones in the research area.NE-SW and NW-SE trending magnetic breaks derived from reduction to pole map are found to be more significant for groundwater exploration.The lineaments rose diagram indicates the general trend of the fracture to be in the NE-SW direction.Assigned normalised criteria weights acquired using the AHP model was used to reclassify the thematic layers.As a result,the taluk’s low,moderate,and high potential zones cover 25.08%,25.68%and 49.24%of the study area,respectively.The high potential zones exhibit characteristics favourable for groundwater infiltration and storage,with factors as gentle slope of<3°,high lineament densities,magnetic breaks,magnetic low zones as indicative of dykes and cracks,lithology as colluvial deposits and land surface with dense vegetation.The depth of the fracture zones was estimated using power spectrum and Euler Deconvolution method.The groundwater potential mapping results were validated using groundwater level data measured from the wells,which indicated that the groundwater potential zoning results are consistent with the data derived from the real world.Muthamilselvan A Anamika Sekar Emmanuel Ignatius 2022Journal of Groundwater Science and Engineering2022,10,4:1

12Restoration of dysregulated CC chemokine signaling for monocyte/macrophage chemotaxis in head and neck squamous cell carcinoma patients by neem leaf glycoprotein maximizes tumor cell cytotoxicity显示文摘Previous studies have shown that the CC chemokine receptor CCR5 is downregulated on monocyte/macrophage(MO/Mw)surfaces in head and neck squamous cell carcinoma(HNSCC)patients(stage IIIB).Ligands(RANTES,MIP-1a and MIP-1b)of this chemokine receptor were also secreted in lesser quantity from MO/Mw of HNSCC patients in comparison with healthy individuals.In an aim to restore this dysregulated receptor–ligand signaling,we have used neem leaf glycoprotein(NLGP),a novel immunomodulator reported from our laboratory.NLGP upregulated CCR5 expression,as evidenced from studies on MO/Mw of peripheral blood from HNSCC patients as well as healthy individuals.Expression of RANTES,MIP-1a and MIP-1b was also upregulated following NLGP treatment of these cells in vitro.Interestingly,NLGP has little effect on the expression of CCR5 and the ligand RANTES in oral cancer cells.This restored CCR5 receptor–ligand signaling seen in MO/Mw was reflected in improved CCR5-dependent,p38 mitogen-activated protein kinase(MAPK)-mediated migration of MO/Mw after NLGP treatment to a standard chemoattractant.NLGP also induces better antigen presentation and simultaneous costimulation to effector T cells by MO/Mw by upregulating human leucocyte antigen(HLA)-ABC,CD80 and CD86.In addition,NLGP-treated MO/Mw-primed T cells can effectively lyse tumor cells in vitro.The effects of NLGP on monocyte migration and T cell-mediated oral tumor cell killing were further demonstrated in transwell assays with or without CCR5 neutralization.These results suggest a new approach in cancer immunotherapy by modulating dysregulated CCR5 signals from MO/Mw.Krishnendu Chakraborty Anamika Bose Tathagata Chakraborty Koustav Sarkar Shyamal Goswami Smarajit Pal Rathindranath Baral 2010Cellular & Molecular Immunology2010,7,5:1
