3Rapid isolation and identification of Mycobacterium tuberculosis from pulmonary tuberculosis patients显示文摘Objective:The Study aimed to develop a rapid inexpensive and simple method for preliminary isolation and detection of M.tuberculosis from clinical specimens.Also to probe the impact of deteriorating health measures on the pulmonary tuberculosis control program in Basra city.Methods:A simple monophasic-diphasic setup (MDCS) was used for the isolation and preliminary identification of M.tuberculosis.This setup consists of a slanted Lowenstein-Jensen medium,the bottom of it is covered with 1 mL of tuberculosis broth thus establishing a diphasic solid liquid environment at the bottom of the screw capped test tube and above them a monophasic one.Results:During 7 months period from the beginning of December 2003 to the end of June 2004,1 295 cases were managed in Tuberculosis and Chest Diseases Clinic in Basra city.Only 348 cases were diagnosed as active pulmonary tuberculosis.They consist of 232 new cases and 116 previously registered ones.The MDCS method showed higher recovery of isolation rate of M.tuberculosis(92.3%) than Lowenstein-Jensen,and pyruvate methods with 78.1%and 51.9%respectively.Also this method revealed more rapid appearance of results of 12 days than Lowenstein-Jensen,and pyruvate media with 22 days and 23 days respectively by the traditional culturing methods.Concurrently the study revealed that drug resistance against one or more anti tuberculosis drugs was 23.1%for new cases and 70.8%for the old ones.Multi drug resistance accounts for 20%of total isolates from old cases.This was based on using the critical concentration and 1%proportional procedures.Tuberculosis was more common among males than females with 69.4%and 30.6%respectively. A significant correlation(P<0.05) was found between the productive age group(25-54) and incidence of tuberculosis. Tuberculosis was higher among married(83.8%) than unmarried peoples(22%).Children,husbands and wives showed higher infection rates than others indicating kinship influence.Failure of treatment was highly affected by improper use of anti tuberculosis drugs.Conclusion:The MDCS method exhibited several advantages over other culturing techniques exemplified by elimination of transport medium,establishment of simultaneous but two separate environments to accomplish both the detection and preliminary identification of M.tuberculosis,providing flexibility in the kinds of media used in the liquid and solid phases and it is inexpensive.Al-Sulami AA Ali ZM Al-Maleky DA 2009Asian Pacific Journal of Tropical Medicine2009,2,3:3
4Fungal diversity notes 367-490:taxonomic and phylogenetic contributions to fungal taxa显示文摘This is a continuity of a series of taxonomic papers where materials are examined,described and novel combinations are proposed where necessary to improve our traditional species concepts and provide updates on their classification.In addition to extensive morphological descriptions and appropriate asexual and sexual connections,DNA sequence data are also analysed from concatenated datasets(rDNA,TEF-a,RBP2 and b-Tubulin)to infer phylogenetic relationships and substantiate systematic position of taxa within appropriate ranks.Wherever new species or combinations are being proposed,we apply an integrative approach(morphological and molecular data as well as ecological features wherever applicable).Notes on 125 fungal taxa are compiled in this paper,including eight new genera,101 new species,two new combinations,one neotype,four reference specimens,new host or distribution records for eight species and one alternative morphs.The new genera introduced in this paper are Alloarthopyrenia,Arundellina,Camarosporioides,Neomassaria,Neomassarina,Neotruncatella,Paracapsulospora and 5Toxicity assessment of polyethylene microplastics in combination with a mix of emerging pollutants on Physalaemus cuvieri tadpoles显示文摘Studies in recent years have shown that aquatic pollution by microplastics(MPs)can be considered to pose additional stress to amphibian populations.However,our knowledge of how MPs affect amphibians is very rudimentary,and even more limited is our understanding of their effects in combination with other emerging pollutants.Thus,we aimed to evaluate the possible toxicity of polyethylene MPs(PE-MPs)(alone or in combination with a mix of pollutants)on the health of Physalaemus cuvieri tadpoles.After 30 days of exposure,multiple biomarkers were measured,including morphological,biometric,and developmental indices,behavioral parameters,mutagenicity,cytotoxicity,antioxidant and cholinesterase responses,as well as the uptake and accumulation of PE-MPs in animals.Based on the results,there was no significant change in any of the parameters measured in tadpoles exposed to treatments,but induced stress was observed in tadpoles exposed to PE-MPs combined with themixture of pollutants,reflecting significant changes in physiological and biochemical responses.Through principal component analysis(PCA)and integrated biomarker response(IBR)assessment,effects induced by pollutants in each test group were distinguished,confirming that the exposure of P.cuvieri tadpoles to the PE-MPs in combination with a mix of emerging pollutants induces an enhanced stress response,although the uptake and accumulation of PE-MPs in these animalswas reduced.Thus,our study provides newinsight into the danger to amphibians of MPs coexisting with other pollutants in aquatic environments.Amanda Pereira da Costa Araújo Thiarlen Marinho da Luz Mohamed Ahmed Ibrahim Ahmed Mir Mohammad Ali Md.Mostafizur Rahman Bojan Nataraj Daniela de Melo e Silva DamiàBarceló Guilherme Malafaia 2023Journal of Environmental Sciences2023,,5:2
