| 3 | Bioprospection of marine actinomycetes: recent advances,challenges and future perspectives显示文摘In exploring new sources for economically important products, marine environment draws particular attention because of its remarkable diversity and extreme conditions;it is known to produce metabolic products of great value. It represents untapped source for the discovery of novel secondary metabolites with varying potential such as antibiotic, anti-tumor, antifouling and cytotoxic properties. Marine actinomycetes distributed throughout the marine environment from shallow to deep sea sediments have proved to be a finest source for this discovery. Secondary metabolites derived from marine actinomycetes have proved their worth in industries based on the research on their properties and wide range applications. Spotlight of the review is range of marine based actinomycetes products and significant research in this field. This shows the capability of marine actinomycetes as bioactive metabolite producers. Additionally, the present review addresses some effective and novel approaches of procuring marine microbial compounds utilizing the latest screening strategies of drug discovery from which traditional resources such as marine actinobacteria has decreased due to declining yields. The aim is in the context of promoting fruitful and profitable results in the near future. The recent surfacing of new technologies for bioprospection of marine actinomycetes are very promising, resulting in high quality value added products, and will be de?ning a new era for bioactive compounds with medical and biotechnological applications. | Swati Sharma Abhay B. | 6 | Assessing the bird guild patterns in heterogeneous land use types around Jammu,Jammu and Kashmir,India显示文摘Land-use sprawl in the Himalayas has caused the conversion of natural habitat into human-modified habitats,thus degrading ecosystem health.Adaptation of birds to changing physical environment can be well understood by analyzing their habitat preferences,and foraging dynamics explored to a limited extent in the Himalayan region,as yet.To achieve a comprehensive understanding of avian guild structure,we used multivariate statistical techniques to classify bird species according to their similarities in foraging patterns and habitat preferences.Observations based on habitat and diet affinities accounted for rich avian diversity with a total of 208 bird species(about 15%of country’s avifauna)recorded from six different sites during 1 year survey.Unweighted pair-group average cluster analysis performed on the families revealed ten feeding and fifteen habitat guilds among 63 bird families observed.Subtropical forests harbored more species followed by urban forests and agricultural landscapes.Insectivorous and omnivorous outnumbered other feeding guilds in the study area.Bird assemblages were richer in protected areas and semidisturbed landscapes and did not show significant variation between the seasons.Results of the study revealed that different functional groups of birds behaved differently,primarily induced by choice of food.The site heterogeneity favored avifaunal persistence by providing favorable foraging,roosting,and nesting opportunities to birds.Composition of avian guilds indicated level of intactness and ecological integrity of ecosystems studied.This outcome thus sets the background for long-term analysis of bird-habitat relationship and their foraging dynamics.The study has the relevance for decision-makers to integrate avian guild structure as an essential ingredient in formulating conservation strategies. | Asha Sohil Neeraj Sharma | 2020 | Ecological Processes2020,9,1: | 0 |
| 7 | Homologous recombination mediates stable Fah gene integration and phenotypic correction in tyrosinaemia mouse-model显示文摘AIM To stably correct tyrosinaemia in proliferating livers of fumarylacetoacetate-hydrolase knockout(Fah-/-) mice by homologous-recombination-mediated targeted addition of the Fah gene.METHODS C57 BL/6 Fah?exon5 mice served as an animal model for human tyrosinaemia type 1 in our study. The vector was created by amplifying human Fah c DNA including the TTR promoter from a lentivirus plasmid as described. The Fah expression cassette was flanked by homologous arms(620 bp and 749 bp long) of the Rosa26 gene locus. Mice were injected with 2.1 × 108 VP of this vector(r AAV8-ROSA26.HAL-TTR.FahROSA26.HAR) via the tail vein. Mice in the control group were injected with 2.1 × 108 VP of a similar vector but missing the homologous arms(r AAV8-TTR.Fah). Primary hepatocytes from Fah-/-recipient mice, treated with our vectors, were isolated and 1 × 106 hepatocytes were transplanted into secondary Fah-/-recipient mice by injection into the spleen. Upon either vector application or hepatocyte transplantation NTBC treatment was stopped in recipient mice. RESULTS Here, we report successful HR-mediated genome editing by integration of a Fah gene expression cassette into the 'safe harbour locus' Rosa26 by recombinant AAV8. Both groups of mice showed long-term survival, weight gain and FAH positive clusters as determined by immunohistochemistry analysis of liver sections in the absence of NTBC treatment. In the group of C57 BL/6 Fah?exon5 mice, which have been transplanted with hepatocytes from a mouse injected with r AAV8-ROSA26.HAL-TTR.Fah-ROSA26.HAR 156 d before, 6 out of 6 mice showed long-term survival, weight gain and FAH positive clusters without need for NTBC treatment. In contrast only 1 out 5 mice, who received hepatocytes from r AAV8-TTR.Fah treated mice, survived and showed few and smaller FAH positive clusters. These results demonstrate that homologous recombinationmediated Fah gene transfer corrects the phenotype in a mouse model of human tyrosinaemia type 1(Fah-/-mice) and is long lasting in a proliferating state of the liver as shown by withdrawal of NTBC treatment and serial transplantation of isolated hepatocytes from primary Fah-/-recipient mice into secondary Fah-/-recipient mice. | 8 | Rice straw management through biofuel,biochar,mushroom cultivation,and paper production to overcome environmental pollution in North India显示文摘Rice is the prominent food grain required by more than half of the world's population to fulfill their nutritional demand.With the continuous growth in the population at the global level,rice production has also been elevated.However,high rice production also creates a new problem in waste management worldwide.Rice straw,generated after rice harvest,possesses meager nutritional value,due to which it is less preferred as fodder and burned in the field.Paddy burning is one of the major causes of air pollution,leading to lung,heart,eye,and skin-related diseases and even premature death.This stubble burning also decreases soil fertility.In this review article,we have discussed the various economic uses of paddy straw which will help to reduce air pollution through the decline in paddy straw burning.Biochar is produced from paddy straw,which can be mixed into the soil to restore fertility and reduce toxic metals'bioavailability.The generation of biofuels such as biobutanol,bioethanol,and biogas from rice straw with their mechanism of synthesis is also discussed in this article.Rice straw can also be utilized in the preparation of solid fuel.Along with this,mushroom cultivation in paddy straw houses is also described.Paddy straw can be used for the pulp and paper industries,which will help to reduce the tree dependence of these industries.Apart from this,a bibliometric analysis of the Scopus database on rice straw uses for the last 20 years was done,including a bibliographic keyword analysis to show published documents'trends.This review will give an elaborated overview of the alternative uses of rice straw with a quantitative analysis of air pollution caused by paddy straw burning.This review will also help to improve the current uses of paddy straw for industrial and commercial benefits to make it more economical. | Vikram Kumar Pooja Singh Jyoti Sharma Sakshi Saini Priyanka Sharma Asha Sharma | 2023 | Waste Disposal and Sustainable Energy2023,5,4: | 0 |
