1Mechanism of metastasis by membrane type 1-matrix metalloproteinase in hepatocellular carcinoma显示文摘AIM: To investigate the precise role of membrane type1-matrix metalloproteinase (MT1-MMP) in hepatocellular carcinoma (HCC) metastasis.METHODS: Human HCC cells Hep3B with overexpression of MT1-MMP were established by stable transfection, and compared with control cells carrying the empty vector.Cells were examined in vivo for their differences in the metastatic ability of athymic nude mice, and analyzed in vitro for their differences in invasion ability by invasion chamber coated with Matrigel, adhesion towards collagen I and migration through culture chamber. Cell proliferation and apoptosis in adherent and suspension status were evaluated by MTT and flow cytometry analysis.RESULTS: We found that overexpression of MT1-MMP could increase intrahepatic metastasis in nude mice with orthotopic implantation of HCC cells (incidence of 100%[MT1-MMP transfectants] vs 40% [vector control transfectants], P<0.05). MT1-MMP could also enhance cell invasion through Matrigel (107.7 vs 39.3 cells/field,P<0.001), adhesion towards matrix (0.30 vs 0.12absorbance unit at 540 nm, P<0.001), cell migration(89.3 vs 39.0 cells/field, P<0.001), and cell proliferation(24.3 vs 40.5 h/doubling, P<0.001). We also observed that MT1-MMP supported cell survival (71.4% vs 23.9%,P<0.001) with reduced apoptosis (43.7% vs51.0%, P<0.05)in an attachment-free environment.CONCLUSION: MT1-MMP overexpression could enhance metastasis. 2Prevention and management of hepatitis B virus reactivation in patients with hematological malignancies treated with anticancer therapy显示文摘Hepatitis due to hepatitis B virus(HBV) reactivation can be severe and potentially fatal, but is preventable. HBV reactivation is most commonly reported in patients receiving cancer chemotherapy, especially rituximabcontaining therapy for hematological malignancies and those receiving stem cell transplantation. All patients with hematological malignancies receiving anticancer therapy should be screened for active or resolved HBV infection by blood tests for hepatitis B surface antigen(HBs Ag) and antibody to hepatitis B core antigen(antiHBc). Patients found to be positive for HBs Ag should be given prophylactic antiviral therapy to prevent HBV reactivation. For patients with resolved HBV infection, no standard strategy has yet been established to prevent HBV reactivation. There are usually two options. One is pre-emptive therapy guided by serial HBV DNA monitoring, whereby antiviral therapy is given as soon as HBV DNA becomes detectable. However, there is little evidence regarding the optimal interval and period of monitoring. An alternative approach is prophylactic antiviral therapy, especially for patients receiving highrisk therapy such as rituximab, newer generation of anti-CD20 monoclonal antibody, obinutuzumab or hematopoietic stem cell transplantation. This strategy may effectively prevent HBV reactivation and avoid the inconvenience of repeated HBV DNA monitoring. Entecavir or tenofovir are preferred over lamivudine as prophylactic therapy. Although there is no well-defined guideline on the optimal duration of prophylactic therapy, there is growing evidence to recommend continuing prophylactic antiviral therapy for at least 12 mo after cessation of chemotherapy, and even longer for those who receive rituximab or who had high serum HBV DNA levels before the start of immunosuppressive therapy. Many novel agents have recently become available for the treatment of hematological malignancies, and these agents may be associated with HBV reactivation. Although there is currently limited evidence to guide the optimal preventive measures, we recommend antiviral prophylaxis in HBs Ag-positive patients receiving novel treatments, especially the Bruton tyrosine kinase inhibitors and the phosphatidylinositol 3-kinase inhibitors, which are B-cell receptor signaling modulators and reduce proliferation of malignant B-cells. 7Fractal analysis of retinal vasculature in normal subjects on ultra-wide field fluorescein angiography显示文摘AIM: To evaluate the fractal feature of the retinal vasculature of normal eyes on a stereographic projected and montaged ultra-wide field(UWF) fluorescein angiography(FA).METHODS: Prospective, observational, cross-sectional study. Totally 59 eyes of 31 normal subjects were imaged using the Optos 200 Tx. Images obtained at different gaze angles stereographically projected and montaged. The early-phase UWF FA frames were processed to segment the retinal vasculature and the results were exported as binary masks. The fractal dimension(FD) was calculated using the box-counting method.RESULTS: The global FD for the entire retina was 1.6±0.04, with no difference between males and females(1.59±0.04 vs 1.61±0.04, P=0.084) or between right and left eyes(1.6±0.04 vs 1.6±0.05, P=0.61). FD was nonuniformly distributed among four quadrants(P<0.001) and decreased as the distance from the fovea increased(P<0.001). A negative association was observed between FD and age(R=-0.37, P=0.006), and this relationship was observed in the posterior and mid-peripheral retina(P<0.05) but absent in far-periphery(P>0.05).CONCLUSION: Fractal geometry is non-uniformly distributed across the retina in normal eyes and decreases from the fovea to the far-periphery. 8Census of Kuiper Comets-the TAOS Project显示文摘Cometary nuclei, the remnant icy bodies of planet formation, located at the outer edge of the solar system, are relatively poorly studied because of their small sizes and thus faintness. Thus far more than a hundred large members (size about 100 km) of this cometary population in the Kuiper belt have been identified. Smaller bodies (10 km) are much too faint to image directly and their number so far can only be extrapolated on theoretical grounds. The Taiwan-America Occultation Survey (TAOS) will conduct a census of the number of Kuiper belt objects down to a few km size by monitoring chance stellar occultations by these cometary nuclei. We will set up an array of small (50cm), wide-field (f/1 9) telescopes, each equipped with a 2K squared CCD camera, along a 7 km baseline in central Taiwan at an elevation above ~ 3000 m. The robot telescopes will operate in a coincidence mode, so the sequence and timing of any candidate occultation event can be recorded and distinguished against a false detection. The Lawrence Livermore National Laboratory, the National Central University, and the Academia Sinica, will each contribute one telescope system. Researchers from an international variety of institutes are also participating in the project. Our experiment provides the only means to study the cometary population in the small-sized end of the distrbution. 10Reconstruction of lateral root formation through single-cell RNA sequencing reveals order of tissue in itiation显示文摘Postembryonic organogenesis is critical for plant development.Underground,lateral roots(LRs)form the bulk of mature root systems,yet the ontogeny of the LR primordium(LRP)is not clear.In this study,we performed the single-cell RNA sequencing through the first four stages of LR formation in Arabidopsis.Our analysis led to a model in which a single group of precursor cells,with a cell identity different from their pericycle origins,rapidly reprograms and splits into a mixed ground tissue/stem cell niche fate and a vascular precursor fate.The ground tissue and stem cell niche fates soon separate and a subset of more specialized vascular cells form sucrose transporting phloem cells that appear to connect to the primary root.We did not detect cells resembling epidermis or root cap,suggesting that outer tissues may form later,preceding LR emergence.At this stage,some remaining initial precursor cells form the primordium flanks,while the rest create a reservoir of pluripotent cells that are able to replace the LR if damaged.Laser ablation of the central and lateral LRP regions showed that remaining cells restart the sequence of tissue initiation to form a LR.Collectively,our study reveals an ontological hierarchy for LR formation with an early and sequential split of main root tissues and stem cells.Laura Serrano-Ron Pablo Perez-Garcia Alvaro Sanchez-Corrionero Inmaculada Gude Javier Cabrera Pui-Leng Ip Kenneth D.Birnbaum Miguel A.Moreno-Risueno 2021Molecular Plant2021,14,8:2

18Testing heteroscedasticity by wavelets in a nonparametric regression model显示文摘In the nonparametric regression models, a homoscedastic structure is usually assumed. However, the homoscedasticity cannot be guaranteed a priori. Hence, testing the heteroscedasticity is needed. In this paper we propose a consistent nonparametric test for heteroscedasticity, based on wavelets. The empirical wavelet coefficients of the conditional variance in a regression model are defined first. Then they are shown to be asymptotically normal, based on which a test statistic for the heteroscedasticity is constructed by using Fan's wavelet thresholding idea.
