2Molecular subtyping and genomic profiling expand precision medicine in refractory metastatic triple-negative breast cancer:the FUTURE trial显示文摘Triple-negative breast cancer(TNBC)is a highly heterogeneous disease,and molecular subtyping may result in improved diagnostic precision and targeted therapies.Our previous study classified TNBCs into four subtypes with putative therapeutic targets.Here,we conducted the FUTURE trial(ClinicalTrials.gov identifer:NCT03805399),a phaseⅠb/Ⅱsubtyping-based and genomic biomarker-guided umbrella trial,to evaluate the efficacy of these targets.Patients with refractory metastatic TNBC were enrolled and stratifed by TNBC subtypes and genomic biomarkers,and assigned to one of these seven arms:(A)pyrotinib with capecitabine,(B)androgen receptor inhibitor with CDK4/6 inhibitor,(C)anti PD-1 with nab-paclitaxel,(D)PARP inhibitor included,(E)and(F)anti-VEGFR included,or(G)mTOR inhibitor with nab paclitaxel.The primary end point was the objective response rate(ORR).We enrolled 69 refractory metastatic TNBC patients with a median of three previous lines of therapy(range,1-8).Objective response was achieved in 20(29.0%,95%confidence interval(CI):18.7%-41.2%)of the 69 intention-to-treat(ITT)patients.Our results showed that immunotherapy(arm C),in particular,achieved the highest ORR(52.6%,95%CI:28.9%-75.6%)in the ITT population.Arm E demonstrated favorable ORR(26.1%,95%C:10.2%-48.4%in the ITT population)but with more high grade(23)adverse events.Somatic mutations of TOP2A and CD8 immunohistochemical score may have the potential to predict immunotherapy response in the immunomodulatory subtype of TNBC.In conclusion,the phaseⅠb/ⅡFUTURE trial suggested a new concept for TNBC treatment,demonstrating the clinical beneft of subtyping-based targeted therapy for refractory metastatic TNBC.Yi-Zhou Jiang Yin Liu Yi Xiao Xin Hu Lin Jiang Wen-ia Zuo Ding Ma Jiahan Ding Xiaoyu Zhu Jianjun Zou Claire Verschraegen Daniel G.Stover Virginia Kaklamani Zhong-Hua Wang Zhi-Ming Shao 2021Cell Research2021,31,2:37
3China National Medical Products Administration approval summary:anlotinib for the treatment of advanced non-small cell lung cancer after two lines of chemotherapy显示文摘Background:On May 8,2018,the China National Medical Products Administration(NMPA)approved anlotinib,an orally administered anti-angiogenesis inhibitor,for the treatment of patients with advanced non-small cell lung can-cer(NSCLC)who have progressed after treatment with two or more lines of prior systemic chemotherapy.Main body of the abstract:China NMPA reviewed and inspected a regional double-blinded,placebo-controlled,Phase III trial comparing the overall survival(OS)of NSCLC patients between the anlotinib and placebo arms.A total of 437 patients were randomized(2:1)to receive either anlotinib(n=294)or placebo(n=143)once daily on a 2-week on and 1-week off schedule.Patients with epidermal growth factor receptor(EGFR)or activating anaplastic lymphoma kinase(ALK)genomic tumor aberrations should have disease progression on NMPA-approved therapy.Anlotinib is the first NMPA-approved drug for patients with advanced NSCLC who have progressed on at least two lines of prior systemic chemotherapies in China.The approval was based on a statistically and clinically significant improvement in median OS with anlotinib(9.46 months)compared with placebo[6.37 months;hazard ratio(HR])=0.70,95%confidence interval(CI)=0.55-0.89;two-sided log-rank P=0.002].The confirmed objective response rate(ORR)was 9.2%in the anlotinib arm and 0.7%in the placebo arm.The median duration of response(DoR)was 4.83 months,with a 95%CI of 3.31-6.97 months.The toxicity profile of anlotinib was consistent with that of known anti-angiogenesis inhibitors.Common adverse drug reactions(ADRs)in anlotinib-treated patients included hypertension(67.4%),hand-foot syndrome(43.9%),hemoptysis(14.0%),thyroid stimulating hormone(TSH)elevation(46.6%),and corrected QT interval(QTc)prolongation(26.2%).Short conclusion:Anlotinib demonstrated a clinically significant OS prolongation as a novel therapeutic option for advanced or metastatic NSCLC following at least two lines of chemotherapy.Ming Zhou Xiaoyuan Chen Hong Zhang Lin Xia Xin Tong Limin Zou Ruimin Hao Jianhong Pan Xiao Zhao Dongmei Chen Yuanyuan Song Yueli Qi Ling Tang Zhifang Liu Rong Gao Yuankai Shi Zhimin Yang 2019Cancer Communications2019,39,1:35

5Magma mixing of granites at Lianghe:In-situ zircon analysis for trace elements, U-Pb ages and Hf isotopes显示文摘In this paper, we report an integrated study of trace element, U-Pb age and Hf isotopic composition of zircons from alkali feldspar granites, granodiorites and diorite enclaves in a recently discovered ring complex at Lianghe in western Yunnan, Chi na. The granitoids showed identical U-Pb ages of 127, 115 and 122 Ma, from felsic to mafic, but had different zircon trace el ements and Hf isotopic compositions. Trace element content decreased with a gradual increase in εHf(t) values of ?9.1 to ?5.4, ?4.5 to 0, and 3.6 to 6.2, respectively. Results indicate that changes in zircon trace elements generally correlate with changes in Hf isotope signatures within single samples and among various granitoids. These relationships reflect the mixing of felsic and mafic magmas. Evidence indicates that depleted mantle-derived mafic magma underplating caused ancient crustal melting, and then formed large-scale granites in Lianghe during the Early Cretaceous. 7GSDMB promotes non-canonical pyroptosis by enhancing caspase-4 activity显示文摘Gasdermin B (GSDMB) has been reported to be associated with immune diseases in humans, but the detailed molecular mechanisms remain unsolved. The N-terminus of GSDMB by itself, unlike other gasdermin family proteins, does not induce cell death. Here, we show that GSDMB is highly expressed in the leukocytes of septic shock patients, which is associated with increased release of the gasdermin D (GSDMD) N-terminus. GSDMB expression and the accumulation of the N-terminal fragment of GSDMD are induced by the activation of the non-canonical pyroptosis pathway in a human monocyte cell line. The downregulation of GSDMB alleviates the cleavage of GSDMD and cell death. Consistently, the overexpression of GSDMB promotes GSDMD cleavage, accompanied by increased LDH release. We further found that GSDMB promotes caspase-4 activity, which is required for the cleavage of GSDMD in non-canonical pyroptosis, by directly binding to the CARD domain of caspase-4. 8Mitochondrial ROS promote macrophage pyroptosis by inducing GSDMD oxidation显示文摘Disrupted mitochondrial membrane potential (MMP) and reactive oxygen species (ROS) generation are often associated with macrophage pyroptosis. It remains unclear how these forms of mitochondrial dysfunction relate to inflammasome activation and gasdermin-D (Gsdmd) cleavage, two central steps of the pyroptotic process. Here, we also found MMP collapse and ROS generation induced by Nlrp3 inflammasome activation as previous studies reported. The elimination of ROS alleviated the cleavage of Gsdmd, suggesting that Gsdmd cleavage occurs downstream of ROS release. Consistent with this result, hydrogen peroxide treatment augmented the cleavage of Gsdmd by caspase-1. Indeed, four amino acid residues of Gsdmd were oxidized under oxidative stress in macrophages. The efficiency of Gsdmd cleavage by inflammatory caspase-1 was dramatically reduced when oxidative modification was blocked by mutation of these amino acid residues. 9Metabolic analyses reveal different mechanisms of leaf color change in two purple-leaf tea plant (Camellia sinensis L.) cultivars显示文摘Purple-leaf tea plants,as anthocyanin-rich cultivars,are valuable materials for manufacturing teas with unique colors or flavors.In this study,a new purple-leaf cultivar“Zixin”(“ZX”)was examined,and its biochemical variation and mechanism of leaf color change were elucidated.The metabolomes of leaves of“ZX”at completely purple,intermediately purple,and completely green stages were analyzed using ultra-performance liquid chromatography quadrupole time of flight mass spectrometry(UPLC-QTOF-MS).Metabolites in the flavonoid biosynthetic pathway remained at high levels in purple leaves,whereas intermediates of porphyrin and chlorophyll metabolism and carotenoid biosynthesis exhibited high levels in green leaves.In addition,fatty acid metabolism was more active in purple leaves,and steroids maintained higher levels in green leaves.Saponin,alcohol,organic acid,and terpenoid-related metabolites also changed significantly during the leaf color change process.Furthermore,the substance changes between“ZX”and“Zijuan”(a thoroughly studied purple-leaf cultivar)were also compared.The leaf color change in“Zijuan”was mainly caused by a decrease in flavonoids/anthocyanins.However,a decrease in flavonoids/anthocyanins,an enhancement of porphyrin,chlorophyll metabolism,carotenoid biosynthesis,and steroids,and a decrease in fatty acids synergistically caused the leaf color change in“ZX”.These findings will facilitate comprehensive research on the regulatory mechanisms of leaf color change in purple-leaf tea cultivars.Jiazhi Shen Zhongwei Zou Xuzhou Zhang Lin Zhou Yuhua Wang Wanping Fang Xujun Zhu 2018Horticulture Research2018,5,1:22
10LARGE-EDDY SIMULATION OF FLOW AROUND CYLINDER ARRAYS AT A SUBCRITICAL REYNOLDS NUMBER显示文摘The complex three-dimensional turbulent flows around a cylinder array with four cylinders in an in-line square configuration at a subcritical Reynolds number of 1.5×104 with the spacing ratio at L/D=1.5 and 3.5 were investigated using the Large Eddy Simulation (LES). The full field vorticity and velocity distributions as well as turbulent quantities were calculated in detail and the near wake structures were presented. The results show that the bi-stable flow nature was observed at L/D=1.5 and distinct vortex shedding of the upstream cylinders occurred at L/ D=3.5 at Re =1.5×104. The techniques of Laser Doppler Anemometry (LDA) and Digital Particle Image Velocimetry (DPIV) are also employed to validate the present LES method. The results show that the numerical predictions are in excellent agreement with the experimental measurements. 12A method to estimate concentrations of surface-level particulate matter using satellite-based aerosol optical thickness显示文摘We propose a new method to estimate surface-level particulate matter(PM)concentrations by using satellite-retrieved Aerosol Optical Thickness(AOT).This method considers the distribution and variation of Planetary Boundary Layer(PBL)height and relative humidity(RH)at the regional scale.The method estimates surface-level particulate matter concentrations using the data simulated by an atmospheric boundary layer model RAMS and satellite-retrieved AOT.By incorporation MODIS AOT,PBL height and RH simulated by RAMS,this method is applied to estimate the surface-level PM 2.5 concentrations in North China region.The result is evaluated by using 16 ground-based observations deployed in the research region,and the result shows a good agreement between estimated PM 2.5 concentrations and observations,and the coefficient of determination R2 is 0.61 between the estimated PM 2.5 concentrations and the observations.In addition,surface-level PM 2.5 concentrations are also estimated by using MODIS AOT,ground-based LIDAR observations and RH measurements.A comparison between the two estimated PM 2.5 concentrations shows that the new method proposed in this paper is better than the traditional method.The coefficient of determination R2 is improved from 0.32 to 0.62.TAO JinHua ZHANG MeiGen CHEN LiangFu WANG ZiFeng SU Lin GE Cui HAN Xiao ZOU MingMin 2013Science China Earth Sciences2013,56,8:20

14Adenovirus-mediated wild-type p53 gene transfer in combination with bronchial arterial infusion for treatment of advanced non-small-cell lung cancer,one year follow-up显示文摘Objective:In the present study,we have examined the safety and efficacy of recombinant adenovirus encoding human p53 tumor suppressor gene(rAd-p53) injection in patients with advanced non-small-cell lung cancer(NSCLC) in the combination with the therapy of bronchial arterial infusion(BAI).Methods:A total of 58 patients with advanced NSCLC were enrolled in a non-randomized,two-armed clinical trial.Of which,19 received a combination treatment of BAI and rAd-p53(the combo group),while the remaining 39 were treated with only BAI(the control group).Patients were followed up for 12 months,with safety and local response evaluated by the National Cancer Institute's Common Toxicity Criteria and response evaluation criteria in solid tumor(RECIST),respectively.Time to progression(TTP) and survival rates were also analyzed by Kaplan-Meier method.Results:In the combo group,19 patients received a total of 49 injections of rAd-p53 and 46 times of BAI,respectively,while 39 patients in the control group received a total of 113 times of BAI.The combination treatment was found to have less adverse events such as anorexia,nausea and emesis,pain,and leucopenia(P<0.05) but more arthralgia,fever,influenza-like symptom,and myalgia(P<0.05),compared with the control group.The overall response rates(complete response(CR)+partial response(PR)) were 47.3% and 38.4% for the combo group and the control group,respectively(P>0.05).Patients in the combo group had a longer TTP than those in the control group(a median 7.75 vs 5.5 months,P=0.018).However,the combination treatment did not lead to better survival,with survival rates at 3,6,and 12 months in the combo group being 94.74%,89.47%,and 52.63%,respectively,compared with 92.31%,69.23%,and 38.83% in the control group(P=0.224).Conclusion:Our results show that the combination of rAd-p53 and BAI was well tolerated in patients with NSCLC and may have improved the quality of life and delayed the disease progression.A further study to better determine the efficacy of this combination therapy is warranted.Yong-song GUAN Yuan LIU Qing ZOU Qing HE Zi LA Lin YANG Ying HU 2009Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2009,10,5:20
15Histological Subtypes of Lung Cancer in Chinese Males from 2000 to 2012显示文摘Objective To characterize the histological and epidemiological features of male lung cancer patients in China. Methods The demographic and histological information about male lung cancer patients identified from 2000-01-01 to 2012-12-31,was collected from the Cancer Hospital of the Chinese Academy of Medical Sciences. Relative frequencies(RF) were estimated for major histological subtypes and compared according to the years of diagnosis and birth. Results The RF of adenocarcinoma(ADC) increased from 21.96% to 43.36% and the RF of squamous cell carcinoma(SCC) decreased from 39.11% to 32.23% from 2000 to 2012 in the 15 427 male lung cancer patients included in this study(Z=17.909,P<0.0001; Z=-6.117,P<0.0001). The RF of ADC increased from 28.72% in 2000-2004,36.88% in 2005-2008 to 48.61% in 2009-2012 in patients born after 1960. The age-adjusted RF of ADC in 2007-2012 increased consistently in all the investigated areas. Conclusion The increased RF of ADC in male lung cancer patients highlights the need for further investigation of the etiologic factors of these tumors. 16Minimally invasive surgery alone compared with intensity-modulated radiotherapy for primary stage I nasopharyngeal carcinoma显示文摘Background:The National Comprehensive Cancer Network guidelines recommend intensity-modulated radiotherapy(IMRT)as the primary curative treatment for newly diagnosed nasopharyngeal carcinoma(NPC),but the radiation-related complications and relatively high medical costs remain a consequential burden for the patients.Endoscopic nasopharyngectomy(ENPG)was successfully applied in recurrent NPC with radiation free and relatively low medical costs.In this study,we examined whether ENPG could be an effective treatment for localized stage I NPC.Methods:Ten newly diagnosed localized stage I NPC patients voluntarily received ENPG alone from June 2007 to September 2017 in Sun Yat-sen University Cancer Center.Simultaneously,the data of 329 stage I NPC patients treated with IMRT were collected and used as a reference cohort.The survival outcomes,quality of life(QOL),and medical costs between two groups were compared.Results:After a median follow-up of 59.0 months(95%CI 53.4-64.6),no death,locoregional recurrence,or distant metastasis was observed in the 10 patients treated with ENPG.The 5-year overall survival,local relapse-free survival,regional relapse-free survival,and distant metastasis-free survival among the ENPG-treated patients was similar to that among the IMRT-treated patients(100%vs.99.1%,100%vs.97.7%,100%vs.99.0%,100%vs.97.4%,respectively,P>0.05).In addition,compared with IMRT,ENPG was associated with decreased total medical costs($4090.42±1502.65 vs.$12620.88±4242.65,P<0.001)and improved QOL scores including dry mouth(3.3±10.5 vs.34.4±25.8,P<0.001)and sticky saliva(3.3±10.5 vs.32.6±23.3,P<0.001).Conclusions:ENPG alone was associated with promising long-term survival outcomes,low medical costs,and satisfactory QOL and might therefore be an alternative strategy for treating newly diagnosed localized stage I NPC patients who refused radiotherapy.However,the application of ENPG should be prudent,and prospective clinical tri-als were needed to further verify the results.You-Ping Liu Xing Lv Xiong Zou Yi-Jun Hua Rui You Qi Yang Le Xia Shao-Yan Guo Wen Hu Meng-Xia Zhang Si-Yuan Chen Mei Lin Yu-Long Xie Li-Zhi Liu Rui Sun Pei-Yu Huang Wei Fan Xiang Guo Ming-Huang Hong Ming-Yuan Chen 2019Cancer Communications2019,39,1:17
17Polyamine Accumulation in Transgenic Tomato Enhances the Tolerance to High Temperature Stress显示文摘Polyamines play an important role in plant response to abiotic stress.S-adenosyl-l-methionine decarboxylase(SAMDC) is one of the key regulatory enzymes in the biosynthesis of polyamines.In order to better understand the effect of regulation of polyamine biosynthesis on the tolerance of high-temperature stress in tomato,SAMDC cDNA isolated from Saccharomyces cerevisiae was introduced into tomato genome by means of Agrobacterium tumefaciens through leaf disc transformation.Transgene and expression was confirmed by Southern and Northern blot analyses,respectively.Transgenic plants expressing yeast SAMDC produced 1.7-to 2.4-fold higher levels of spermidine and spermine than wild-type plants under high temperature stress,and enhanced antioxidant enzyme activity and the protection of membrane lipid peroxidation was also observed.This subsequently improved the efficiency of CO2 assimilation and protected the plants from high temperature stress,which indicated that the transgenic tomato presented an enhanced tolerance to high temperature stress(38℃) compared with wild-type plants.Our results demonstrated clearly that increasing polyamine biosynthesis in plants may be a means of creating high temperature-tolerant germplasm.Lin Cheng Yijing Zou Shuli Ding Jiajing Zhang Xiaolin Yu Jiashu Cao Gang Lu 2009Journal of Integrative Plant Biology2009,51,5:16

20FORCE REDUCTION OF FLOW AROUND A SINUSOIDAL WAVY CYLINDER显示文摘A large eddy simulation of cross-flow around a sinusoidal wavy cylinder at Re = 3000 was performed and the load cell measurement was introduced for the validation test. The mean flow field and the near wake flow structures were presented and compared with those for a circular cylinder at the same Reynolds number. The mean drag coefficient for the wavy cylinder is smaller than that for a corresponding circular cylinder due to the formation of a longer wake vortex generated by the wavy cylinder. The fluctuating lift coefficient of the wavy cylinder is also greatly reduced. This kind of wavy surface leads to the formation of 3-D free shear layers which are more stable than purely 2-D free shear layers. Such free shear layers only roll up into mature vortices at further downstream position and significantly modify the near wake structures and the pressure distributions around the wavy cylinder.
