1China National Medical Products Administration approval summary:anlotinib for the treatment of advanced non-small cell lung cancer after two lines of chemotherapy显示文摘Background:On May 8,2018,the China National Medical Products Administration(NMPA)approved anlotinib,an orally administered anti-angiogenesis inhibitor,for the treatment of patients with advanced non-small cell lung can-cer(NSCLC)who have progressed after treatment with two or more lines of prior systemic chemotherapy.Main body of the abstract:China NMPA reviewed and inspected a regional double-blinded,placebo-controlled,Phase III trial comparing the overall survival(OS)of NSCLC patients between the anlotinib and placebo arms.A total of 437 patients were randomized(2:1)to receive either anlotinib(n=294)or placebo(n=143)once daily on a 2-week on and 1-week off schedule.Patients with epidermal growth factor receptor(EGFR)or activating anaplastic lymphoma kinase(ALK)genomic tumor aberrations should have disease progression on NMPA-approved therapy.Anlotinib is the first NMPA-approved drug for patients with advanced NSCLC who have progressed on at least two lines of prior systemic chemotherapies in China.The approval was based on a statistically and clinically significant improvement in median OS with anlotinib(9.46 months)compared with placebo[6.37 months;hazard ratio(HR])=0.70,95%confidence interval(CI)=0.55-0.89;two-sided log-rank P=0.002].The confirmed objective response rate(ORR)was 9.2%in the anlotinib arm and 0.7%in the placebo arm.The median duration of response(DoR)was 4.83 months,with a 95%CI of 3.31-6.97 months.The toxicity profile of anlotinib was consistent with that of known anti-angiogenesis inhibitors.Common adverse drug reactions(ADRs)in anlotinib-treated patients included hypertension(67.4%),hand-foot syndrome(43.9%),hemoptysis(14.0%),thyroid stimulating hormone(TSH)elevation(46.6%),and corrected QT interval(QTc)prolongation(26.2%).Short conclusion:Anlotinib demonstrated a clinically significant OS prolongation as a novel therapeutic option for advanced or metastatic NSCLC following at least two lines of chemotherapy.Ming Zhou Xiaoyuan Chen Hong Zhang Lin Xia Xin Tong Limin Zou Ruimin Hao Jianhong Pan Xiao Zhao Dongmei Chen Yuanyuan Song Yueli Qi Ling Tang Zhifang Liu Rong Gao Yuankai Shi Zhimin Yang 2019Cancer Communications2019,39,1:35

3Location-aided and secure routing protocol for heterogeneous multi-hop wireless networks显示文摘Most of the existing routing protocols for heterogeneous multi-hop wireless networks(HWMNs) use on-demand routing protocols owing to their suitability for dynamic topology environments, but it causes wastes of network resources with large number of data packets being broadcast. Furthermore, some nodes in heterogeneous wireless multi-hop networksmay be malicious or selfish nodes which can easily lead to link attacks. A novel routing protocol called trust-based secure routing protocol with auxiliary of nodes' location information(TSRAL) is proposed for the establishment of a secure routing protocol with a lower overhead for HWMNs which combines the location information and trust value of nodes to select the next forwarding nodes. The destination node selects an optimal path from multiple paths according to the total trust value and the hop number of routes. 4Disruption of the bHLH transcription factor Abnormal Tapetum 1 causes male sterility in watermelon显示文摘Although male sterility has been identified as a useful trait for hybrid vigor utilization and hybrid seed production,its underlying molecular mechanisms in Cucurbitaceae species are still largely unclear.Here,a spontaneous male-sterile watermelon mutant,Se18,was reported to have abnormal tapetum development,which resulted in completely aborted pollen grains.Map-based cloning demonstrated that the causal gene Citrullus lanatus Abnormal Tapetum 1(ClATM1)encodes a basic helix-loop-helix(bHLH)transcription factor with a 10-bp deletion and produces a truncated protein without the bHLH interaction and functional(BIF)domain in Se18 plants.qRT–PCR and RNA in situ hybridization showed that ClATM1 is specifically expressed in the tapetum layer and in microsporocytes during stages 6–8a of anther development.The genetic function of ClATM1 in regulating anther development was verified by CRISPR/Cas9-mediated mutagenesis.Moreover,ClATM1 was significantly downregulated in the Se18 mutant,displaying a clear dose effect at the transcriptional level.Subsequent dual-luciferase reporter,β-glucuronidase(GUS)activity,and yeast one-hybrid assays indicated that ClATM1 could activate its own transcriptional expression through promoter binding.Collectively,ClATM1 is the first male sterility gene cloned from watermelon,and its self-regulatory activity provides new insights into the molecular mechanism underlying anther development in plants.Ruimin Zhang Jingjing Chang Jiayue Li Guangpu Lan Changqing Xuan Hao Li Jianxiang Ma Yong Zhang Jianqiang Yang Shujuan Tian Li Yuan Xian Zhang Chunhua Wei 2021Horticulture Research2021,8,1:3

10Simulation and Experiment Based on FSMLC Method with EUPI Hysteresis Compensation for a Piezo-Driven Micro Position Stage显示文摘Micro/nano positioning technologies have been attractive for decades in industrial and scientific applications fields. The actuators have inherent hysteresis that can cause system unexpected behave in some extend. In this research, the authors used extented unparallel Prandtl-Ishlinshii(EUPI)models to represent the input-output relationship of a piezo-driven micro position stage. Integral inverse(I-I) compensator is used for compensating the hysteresis characteristics of the micro positioning stage and compared with direct inverse(D-I) compensator and inverse model(I-M) compensator. However,the accuracy and the robustness of the I-I compensator are worse when there is noisy in the system,a novel sliding-mode-like-control with EUPI(SMLC-EUPI) method was proposed and analyzed by different trajectory tracking experiments in Matlab environment. Though the above strategies can alleviate most deviation, the adjustment of the SMLC’s parameters is very complex. So the fuzzy method is used to adjust these parameters and be verified by trajectory tracking experiments. 13Immunostimulant nanomodulator boosts antitumor immune response in triple negative breast cancer by synergism of vessel normalization and photothermal therapy显示文摘Tumor vascular dysfunction and immune suppression predict poor outcomes of tumor therapy.Combination of photothermal therapy(PTT)and vessel normalization with tumor immunotherapy is promising to augment antitumor benefit.Herein,we develop a potential immunostimulatory nanomodulator for treatment of triple-negative breast cancer(TNBC)treatment via synergism of PTT,vessel normalization,and priming of tumoral suppressive immune microenvironment by blocking transforming growth factor-β(TGF-β)pathway.The nanomodulator,namely Vac@Apt@BPs,is developed by conjugation of TGF-βinhibitor Vactosertib(Vac)and nucleolin-recognizing aptamer(Apt)on the surface of black phosphorus nanoparticles(BPs).Vac@Apt@BPs show good accumulation in TNBC via aptamer-induced active targeting of TNBC.Via the blockade of TGF-βsignaling,Vac@Apt@BPs effectively inhibit the formation of tumor neovascular,and normalize the vessels to recover vascular integrity and alleviate the hypoxia stress.Together with the tumor eradication and immunogenic cell death via PTT,robust immune response was boosted by promoted maturation of dendritic cells,suppression of regulatory T cells,and stimulation of effective T cells.This synergistic therapeutic strategy potentially suppresses the growth of TNBC in mice.Hao Jiang Yilang He Jia Zhao Ruimin Chang Hailun He Tan Li Xingyu Zhang Bo Shu Wenxuan Zhang Huaiyu Wang Jing Liu Shubing Zhang Yuetao Zhao 2023Nano Research2023,16,8:0
14MiR-16-5p plays an inhibitory role in human non-small cell lung cancer through Fermitin family member 2显示文摘Increasing evidence indicates that aberrant expressions of some microRNAs are associated with cancer progression.However,the roles and biological mechanisms of miRNA-16-5p in human non-small cell lung cancer(NSCLC)are not to be well studied.Here,we validated that the expression of miR-16-5p was decreased significantly in NSCLC samples and cell lines.The correlation between the clinicopathological features of NSCLC and the miR-16-5p expression showed that the expression of miR-16-5p in non-small cell lung cancer was linked with the advanced TNM stage,positive lymph node metastasis,with short overall survival(OS).Also,a negative correlation between miR-16-5p and Fermitin family member 2(FERMT2)was observed,implying there may be a potential link about their regulation.The hypothesis was further confirmed by in-silico analysis and dual-luciferase reporter assay.Moreover,we demonstrated that the transfections of miR-16-5p mimics could alter some biological characteristics of NSCLC cells remarkably accomplished by the expression variance of FERMT2 in vitro and in vivo assays.Summarily,this study demonstrated that miR-16-5p,as a tumor suppression factor in NSCLC by targeting FERMT2,could serve as one promising biomarker in the prediction for NSCLC patients.JUNQI GUO YUN YANG WEI ZHAO ZHONGHAI YAN XIA YANG YUNFEI YAN RUIMIN HAO JINXIA HU FEI JIAO 2021BIOCELL2021,45,3:0
