1Triptolide (PG-490) induces apoptosis of dendritic cells through sequential p38 MAP kinase phosphorylation and caspase 3 activation显示文摘Dendritic cells (DCs) are the most potent antigen-presen ting cells that play crucial roles in the regulation of immune response. Triptol ide, an active component purified from the medicinal plant Tripterygium wilfor dii Hook F., has been demonstrated to act as a potent immunosuppressive drug c apab le of inhibiting T cell activation and proliferation. However, little is known a bout the effects of triptolide on DCs. The present study shows that triptolide d oes not affect phenotypic differentiation and LPS-induced maturation of murine DCs. But triptolide can dramatically reduce cell recovery by inducing apoptosis of DCs at concentration as low as 10 ng/ml, as demonstrated by phosphatidylserin e exposure, mitochondria potential decrease, and nuclear DNA condensation. Tript olide induces activation of p38 in DCs, which precedes the activation of caspase 3. SB203580, a specific kinase inhibitor for p38, can block the activation of caspase 3 and inhibit the resultant apoptosis of DCs. 2Identification of an HLA-A~* 0201-restricted CD8^+ T-cell epitope SSp-1 of SARS-CoV spike protein显示文摘A novel coronavirus, severe acute respiratory syndrome (SA RS)-associated coronavirus (SARS-CoV), has been identified as the causal agent of SARS. Spike (S) protein is a major structural glycoprotein of the SARS virus and a potential target for SARS-specific cell-mediated immune responses. A pa nel of S protein-derived peptides was tested for their binding affinity to HLA -A *0201 molecules. Peptides with high affinity for HLA-A *0201 were then as se ssed for their capacity to elicit specific immune responses mediated by cytotoxi c T lymphocytes (CTLs) both in vivo, in HLA-A2.1/K b transgenic mice, a nd in vitro, from peripheral blood lymphocytes (PBLs) harvested from healthy HLA-A 2.1 + donors. SARS-CoV protein-derived peptide-1 (SSp-1 RLNEVAKNL), induced pepti de-specific CTLs both in vivo (transgenic mice) and in vitro (human PBL s), which specifically released interferon-gamma (IFN-gamma) upon stimulation with SSp-1-pulsed autologous dendritic cells (DCs) or T2 cells. SSp-1-specif ic CTLs also lysed major histocompatibility complex (MHC)-matched tumor cell lines engineered to express S proteins. HLA-A *0201-SSp-1 tetramer staining re vealed the presence of significant populations of SSp-1-specific CTLs in SSp- 1-induced CD8 + T cells. 3MiR-122-5p suppresses the proliferation, migration, and invasion of gastric cancer cells by targeting LYN显示文摘Gastric cancer(GC)is one of malignant tumors with high mortality and morbidity in the world.MicroRNA-122(miR-122)acts as a tumor suppressor in a variety of cancers and has been found to be dominant in gastric adenocarcinoma.However,the specific biological function of miR-122-5p in GC is not completely clear.In this study,we found that miR-122-5p was low-expressed in GC tissues and cell lines by using qRT-PCR.Overexpression of miR-122-5p inhibited the proliferation,migration,and invasion of GC cells by using CCK-8 and transwell assays.On the contrary,downregulation of miR-122-5p promoted the proliferation,migration,and invasion of GC cells.In addition,we found that the expression of LYN,an Src family tyrosine kinase,was inversely correlated with miR-122-5p expression in GC tissues by using western blot analysis,immunohistochemistry,and qRT-PCR assays.Meanwhile,luciferase assay results indicated that LYN is a direct target of miR-122-5p in GC cells.Moreover,silencing LYN expression by its siRNA inhibited the proliferation,migration,and invasion of GC cells.Importantly,overexpression of LYN restored miR-122-5p-mediated inhibition of the proliferation,migration,and invasion of GC cells.Taken together,our results indicated miR-122-5p inhibited the proliferation,migration,and invasion by targeting LYN in GC.Lei Meng Zhangming Chen Zhe Jiang Ting Huang Jie Hu Panquan Luo Hongli Zhang Mengqi Huang Lei Huang Yu Chen Ming Lu A-man Xu Songcheng Ying 2020Acta Biochimica et Biophysica Sinica2020,52,1:12

5Voltage-gated sodium channel Nav1.5 promotes proliferation, migration and invasion of oral squamous cell carcinoma显示文摘The protein voltage-gated sodium channel Nav1.5 is highly upregulated in various types of cancer and, in general, promotes cancer cell invasiveness and metastatic progression. A previous study found that Nav1.5 was highly expressed in poorly differentiated oral squamous cell carci noma (OSCC). However, whether Nav1.5 enhances invasiveness and metastasis of OSCC are still unknown. In this study, we found that Nav1.5 was highly expressed in OSCC cell lines compared with normal oral keratinocyte HOK cell line by using western blot analysis. CCK-8 assay results revealed that downregulation of Nav1.5 expression by its specific siRNA reduced proliferation of OSCC HSC-3 cells. Moreover, transwell assay results showed Nav1.5 knockdown significantly inhibited migration and invasion of HSC-3 cells. Meanwhile, qRT-PCR and western blot analysis results showed that epidermal growth factor (EGF) induced Nav1.5 expression in a time-and dosedependent manner. In addition, EGF promoted proliferation, migration and invasion of HSC-3 cells. Importantly, the Nav1.5 inhibitor tetrodotoxin significantly inhibited the proliferation of HSC-3 cells and impeded the migration and invasion of HSC-3 cells. Furthermore, it was found that siRNAmediated knockdown of Nav1.5 also lessened the proliferation of HSC-3 cells and blocked the migration and invasion of HSC-3 cells. 6Tunable seesaw-like 3D capacitive sensor for force and acceleration sensing显示文摘To address the resource-competing issue between high sensitivity and wide working range for a stand-alone sensor,development of capacitive sensors with an adjustable gap between two electrodes has been of growing interest.While several approaches have been developed to fabricate tunable capacitive sensors,it remains challenging to achieve,simultaneously,a broad range of tunable sensitivity and working range in a single device.In this work,a 3D capacitive sensor with a seesaw-like shape is designed and fabricated by the controlled compressive buckling assembly,which leverages the mechanically tunable configuration to achieve high-precision force sensing(resolution~5.22 nN)and unprecedented adjustment range(by~33 times)of sensitivity.The mechanical tests under different loading conditions demonstrate the stability and reliability of capacitive sensors.Incorporation of an asymmetric seesaw-like structure design in the capacitive sensor allows the acceleration measurement with a tunable sensitivity.These results suggest simple and low-cost routes to high-performance,tunable 3D capacitive sensors,with diverse potential applications in wearable electronics and biomedical devices.Jilong Ye Fan Zhang Zhangming Shen Shunze Cao Tianqi Jin Xiaogang Guo Zhihong Li Li Lin Yihui Zhang 2021npj Flexible Electronics2021,5,1:2
7Optimal Weight and Parameter Estimation of Multi-structure and Unequal-Precision Data Fusion显示文摘Measured data fusion process is an effective way to improve the data process precision. In this paper,the fusion weight is firstly introduced, and then we study the optimal weight and parameter estimation using multistructure and unequal-precision data fusion. For the linear regression model, it is theoretically proved that the optimal weight is only related to the data measure precision,which is consistent with the classical Gauss-Markov theorem. 8A 0.02% THD and 80 dB PSRR filterless class D amplifier with direct lithium battery hookup in mobile application显示文摘This paper presents a fully integrated CMOS filterless class D amplifier that can directly hook up lithium battery in mobile application The proposed amplifier embodies a 2-order feedback path architecture instead of direct feedback of output to input of the integrator to decrease the high frequency intermodulation distortion associated with direct feedback and eliminate the integrator input common mode disturbance from the output in ternary modulation. The prototype class D amplifier realized in 0.35 μm digital technology achieves a THD+N of 0.02% when delivering 400 m W to an 8 Ω load from V_(DD) =3.6 V. The PSRR of the prototype class D amplifier is 80 dB at217 Hz. Furthermore a filterless method that can eliminate the external LC filter is employed which offers great advantages of saving PCB space and lowering system cost. In addition the prototype class D amplifier can operate in large voltage range with V_(DD)range from 2.5 to 4.2V in mobile application. 9Reservoir architecture patterns of sandy gravel braided distributary channel A case study of Triassic Upper Karamay Formation,Xinjiang oilfield显示文摘The purpose of this study was to discuss shape,scale and superimposed types of sandy gravel bodies in sandy-gravel braided distributary channel.Lithofacies analysis,hierarchy bounding surface analysis and subsurface dense well pattern combining with outcrops method were used to examine reservoir architecture patterns of sandy gravel braided distributary channel based on cores,well logging,and outcrops data,and the reservoir architecture patterns of sandy gravel braided distributary channels in different grades have been established.The study shows:(1)The main reservoir architecture elements for sandy gravel braided channel delta are distributary channel and overbank sand,while reservoir flow barrier elements are interchannel and lacustrine mudstone.(2)The compound sand bodies in the sandy gravel braided delta distributary channel take on three shapes:sheet-like distributary channel sand body,interweave strip distributary channel sand body,single strip distributary channel sand body.(3)Identification marks of single distributary channel include:elevation of sand body top,lateral overlaying,“thick-thin-thick”feature of sand bodies,interchannel mudstone and overbank sand between distributary channels and the differences in well log curve shape of sand bodies.(4)Nine lithofacies types were distinguished in distributary channel unit interior,different channel units have different lithofacies association sequence.Senlin Yin Zhangming Hu Xiang Ren Xiaojun Wu Yanhui Chen Lijun Zheng Zhibin Jiang 2016Petroleum2016,2,2:2
10An inexact multi-objective programming approach for strategic environmental assessment on regional development plan显示文摘This paper presents the development of an inexact multi-objective programming (IMOP) model and its application to the strategic environmental assessment (SEA) for the regional development plan for the Hunnan New Zone (HNZ) in Shenyang City, China. Inexact programming and multi-objective programming methods are employed to effectively account for extensive uncertainties in the study system and to reflect various interests from different stakeholders, respectively. In the case study, balancing-economy-and-environment scenario and focusing-industry-development scenario are analyzed by the interactive solution process for addressing the preferences from local authorities and compromises among different objectives. Through interpreting the model solutions under both scenarios, analysis of industrial structure, waste water treatment plant (WWTP) expansion, water consumption and pollution generation and treatment are undertaken for providing a solid base to justify and evaluate the HNZ regional development plan. 11Imperfection sensitivity of mechanical properties in soft network materials with horseshoe microstructures显示文摘Developments of soft network materials with rationally distributed wavy microstructures have enabled many promising applications in bio-integrated electronic devices,due to their abilities to reproduce precisely nonlinear mechanical properties of human tissues/organs.In practical applications,the soft network materials usually serve as the encapsulation layer and/or substrate of bio-integrated electronic devices,where deterministic holes can be utilized to accommodate hard chips,thereby increasing the filling ratio of the device system.Therefore,it is essential to understand how the hole-type imperfection affects the stretchability of soft network materialswith various geometric constructions.Thiswork presents a systematic investigation of the imperfection sensitivity of mechanical properties in soft network materials consisting of horseshoe microstructures,through combined computational and experimental studies.A factor of imperfection insensitivity of stretchability is introduced to quantify the influence of hole imperfections,as compared to the case of perfect soft network materials.Such factor is shown to have different dependences on the arc angle and normalized width of horseshoe microstructures for triangular network materials.The soft triangular and Kagome network materials,especially with the arc angle in the range of(30?,60?),are found to be much more imperfection insensitive than corresponding traditional lattice materials with straight microstructures.Differently,the soft honeycomb network materials are not as imperfection insensitive as traditional honeycomb lattice materials.Jianxing Liu Xinyuan Zhu Zhangming Shen Yihui Zhang 2021Acta Mechanica Sinica2021,37,7:1
