| 1 | Nia1 and Nia2 are Involved in Exogenous Salicylic Acid-induced Nitric Oxide Generation and Stomatal Closure in Arabidopsis显示文摘Phytohormone salicylic acid (SA) plays important roles in plant responses to environmental stress. However, knowledge about the molecular mechanisms for SA affecting the stomatal movements is limited. In this paper, we demonstrated that exogenous SA significantly induced stomatal closure and nitric oxide (NO) generation in Arabidopsis guard cells based on genetic and physiological data. These effects were significantly inhibited by the NO scavenger c-PTIO, NO synthase (NOS) inhibitor L-NAME or nitrate reductase suppressor tungstate respectively, implying that NOS and nitrate reductase (NR) participate in SA-evoked stomatal closing. Furthermore, the effects of SA promotion of stomatal closure and NO synthesis are significantly suppressed in NR single mutants of nia1, nia2 or double mutant nia1/nia2, compared with the wild type plants. This suggests that both Nia1 and Nia2 are involved in SA-stimulated stomatal closure. | 5 | Influence of Ganoderma lucidum spores on the levels of neuropeptide Y and somatostatin in brains of seizure rats显示文摘BACKGROUND: Previous research has revealed that somatostatin can induce epilepsy, and that the levels of neuropeptide Y may increase and become more active in brain areas with epileptic seizures. OBJECTIVE: To observe the effect of Ganoderma lucidum spore powder on the neuropeptide Y and somatostatin content in the cerebral cortex and hippocampal regions of seizure rats induced by pentylenetetrazol (PTZ). Furthermore, to verify any effect of Ganoderma lucidum spore powder on inhibition of epileptic seizures. DESIGN, TIME AND SETTING: A randomized group animal study was performed in August 2007 in the School of Basic Medical Sciences, Jiamusi University (Jiamusi, Heilongjiang, China). MATERIALS: Thirty healthy, male, Wistar rats, aged 12 weeks and weighing 180–220 g, were taken as the experimental animals. PTZ (Sigma Company, United States) was used to induce epilepsy. Ganoderma lucidum spores (Leyss, ex Fr variety) were purchased from Jiamusi City Wild Growing Case of the Ganoderma Lucidum (China). Rabbit anti-somatostatin antibodies and secondary antibodies were purchased from Wuhan Boster Company (China). Neuropeptide Y radioimmunoassay kit was purchased from Beijing Furui Biotechnology Company (China). METHODS: Thirty rats were randomly divided into three groups: a control group, an epilepsy model group and a Ganoderma lucidum spore-treated group. Each group contained 10 animals. Rats in the epilepsy model group were treated with intraperitoneal injections of PTZ and gastric perfusion of physiologic saline. In the Ganoderma lucidum spore-treated group, intraperitoneal injection of PTZ and gastric perfusion of Ganoderma lucidum spore powder were administered. The blank control group was only administered with the physiological saline by intraperitoneal injection and gastric perfusion. MAIN OUTCOME MEASURES: Immunohistochemical staining and radioimmunoassay methods were used to observe the changes of somatostatin and neuropeptide Y content in brain tissue of epileptic rats, as well as the morphology of neurons. RESULTS: All 30 rats were involved in the result analysis, without any loss. The number of somatostatin immunoreacted positive cells in the cerebral cortex and hippocampus was significantly increased in the epilepsy model group compared to the blank control group (P < 0.01). The number of somatostatin immunoreacted positive cells in cerebral cortex and hippocampus was significantly decreased in the Ganoderma lucidum spore-treated group compared to the epilepsy model group (P < 0.01). The contents of neuropeptide Y in cerebral cortex and hippocampus were significantly increased in the epilepsy model group compared to the blank control group (P < 0.01). The contents of neuropeptide Y in the cerebral cortex and hippocampus were significantly decreased in the Ganoderma lucidum spore-treated group compared to the epilepsy model group (P < 0.05). The epilepsy seizures in the Ganoderma lucidum spore-treated group were obviously reduced compared to the epilepsy model group. | 7 | Artesunate and Dihydroartemisinin Inhibit Rabies Virus Replication显示文摘Rabies is caused by infection of rabies virus(RABV)and remains a serious threat to the global public health.Except for the requirement for cold chain and high cost of human rabies immune globulin,no small molecule drugs are currently available for clinical treatment of rabies.So,it is of great importance to identify novel compounds that can effectively inhibit RABV infection.Artesunate(ART)and dihydroartemisinin(DHA),two derivatives of artemisinin,are widely used for treatment of malaria in adults and children,showing high safety.In this study,we found that both ART and DHA were able to inhibit RABV replication in host cells at a low concentration(0.1μmol/L).The antiviral effects of ART and DHA were independent of viral strains and cell lines.Pre-treatment with ART or DHA for 2 h in vitro did not affect the viral replication in host cells,implying that ART and DHA neither reduced the viability of RABV directly nor inhibited the binding and entrance of the virus to host cells.Further studies revealed that ART and DHA inhibited RABV genomic RNA synthesis and viral gene transcription.Treatment with ART or DHA(5 mg/kg)by intramuscular injection improved,to some extent,the survival rate of RABV-challenged mice.Combination treatment with derivatives of artemisinin and mannitol significantly improved the survival rate of RABV-challenged mice.The results suggest that ART and DHA have a great potential to be explored as new anti-rabies agents for treatment of rabies. | Jun Luo Yue Zhang Yang Wang Qing Liu Jiesen Li Hongling He Yongwen Luo Shile Huang Xiaofeng Guo | 2021 | Virologica Sinica2021,36,4: | 2 |
| 8 | Effects of Ganoderma lucidum spore powder on astrocyte expression and glutamine synthetase activity in the hippocampal region of epileptic rats显示文摘BACKGROUND: Recent studies have demonstrated that astrocyte dysfunction plays a central role in inhibiting epileptic seizures and that regulation of astrocyte function may be a new target for treatment of epilepsy. OBJECTIVE: To observe the effects of Ganoderma lucidum spore powder (GLSP) on astrocyte morphology and glutamine synthetase (GS) activity in the hippocampal region of epileptic rats. DESIGN, TIME AND SETTING: A randomized, controlled animal experiment was performed at the Function Laboratory, College of Basic Medicine, Jiamusi University between October and December 2006. MATERIALS: A total of 30 Sprague Dawley (SD) rats were randomized to three groups (n = 10): control, model, and GLSP. GLSP was sourced from Jiamusi Wild Ganoderma Lucidum Planting Base and prepared to 30 g/L with physiological saline before use. Pentylenetetrazol (PTZ) (10 g/L) was provided by Sigma Company, USA. METHODS: The control group received intraperitoneal (i.p.) and intragastric (i.g.) physiological saline. Following epilepsy induction by i.p. administration of PTZ (35 mg/kg), rats from the model and GLSP groups were ig injected with physiological saline and GLSP (300 mg/kg), respectively. Each compound was administered once per day, for a total of 28 successive days. Epileptic seizure convulsions were graded 0–5. A higher grade indicated more severe epilepsy. Only those rats showing stage 2 or higher convulsions at least 5 times successively were included in further experiments. MAIN OUTCOME MEASURES: Immediately after injection, seizure activity was monitored for 30 minutes to determine the latent period and seizure duration; simultaneously, astrocyte numbers and GS activity in the hippocampal region of rats with epilepsy were detected by immunohistochemistry. RESULTS: All 30 rats were included in the final analysis. On day 28, following PTZ administration epileptic seizures were not found in the control group. In the GLSP group, rats exhibited rhythmic head nodding or facial spasms, and the latent period was significantly longer than that of the model group (P < 0.05). In the model group, the majority of rats exhibited myoclonus or generalized convulsions, and epileptic seizure duration was slightly (but not significantly) longer than that in the GLSP group (P > 0.05). Within 2–3 minutes of PTZ injection, the model group exhibited grade 4 or 5 epileptic seizures, and the GLSP group mostly showed grade 2 or 3, and occasionally grade 4 or 5, epileptic seizures. All rats recovered within 30 minutes. The model group exhibited significantly increased astrocytes (P < 0.05), with thicker cellular processes and a higher number of cellular processes, and significantly decreased GS activity (P < 0.05) than the control group. The astrocyte count was significantly decreased but GS activity was significantly increased in the GLSP group when compared with the model group (P < 0.05); however, astrocyte appearance was similar in both groups (P < 0.05). | 14 | ReishiMax inhibits mTORC1/2 by activating AMPK and inhibiting IGFR/PI3K/Rheb in tumor cells显示文摘Ganoderma lucidum(G.lucidum)extracts,as dietary supplements,have been found to exert potent anticancer activity,which is attributed to the presence of polysaccharides and triterpenes.However,the molecular mechanism underlying the anticancer action of G.lucidum extracts remains to be investigated.Here,we show that ReishiMax GLp,containing G.lucidum polysaccharides and triterpenes(GLPT),inhibited cell proliferation and induced cell death in human lung cancer cells(A549 and A427)and simultaneously suppressed the signaling pathways of mammalian target of rapamycin complexes 1 and 2(mTORC1 and mTORC2),respectively.Mechanistically,GLPT downregulated the phosphorylation and protein levels of insulin-like growth factor 1 receptor(IGFR)and phosphoinositide 3-kinase(PI3K)as well as the protein level of RAS homolog enriched in brain(Rheb).In addition,GLPT also activated the AMP-activated protein kinase(AMPK)network.This was evidenced by observations that GLPT increased the phosphorylation of AMPKα(T172)and its substrates tuberous sclerosis complex 2(TSC2,S1387)and regulatory-associated protein of mTOR(raptor,S792).Ectopic expression of dominant-negative AMPKαpartially mitigated the inhibitory effect of GLPT on mTORC1,indicating that GLPT inhibits mTORC1 partly by activating AMPK.The results suggest that G.lucidum extracts exert anticancer action at least partly by suppressing mTORC1/2 signaling via activation of AMPK and inhibition of IGFR/PI3K/Rheb in tumor cells. | Didem Sohretoglu Chao Zhang Jun Luo Shile Huang | 2019 | Signal Transduction and Targeted Therapy2019,4,1: | 1 |

| 18 | Reductive degradation of chloramphenicol by Geobacter metallireducens显示文摘Geobacter metallireducens is known to be capable of removing nitroaromatic compounds via an oxidation mode. However, little attention has been paid to investigate the reductive removal of chlorinated nitroaromatic compounds by G. metallireducens. In this study, G. metallireducens was used to reduce chloramphenicol(CAP), a typical chlorinated nitroaromatic antibiotic. Cyclic voltammograms and chronoamperometry highlighted a higher peak current for CAP reduction by G. metallireducens compared to the control without bacteria. G. metallireducens efficiently reduced CAP(20 mg/L) with acetate as the sole electron donor, and the removal efficiency reached(97.6±4.9)% within 6 d. Aromatic amine(AMCl2), AMCl(dechlorinated AMCl2) and AM(dechlorinated AMCl) were identified as reduction products by liquid chromatography-mass spectrometry. However, the removal efficiency declined to(25.0±3.6)% when the CAP dosage increased to 80 mg/L. Transcriptomic analysis indicated the significant upregulation of genes related to electron transfer, such as pilus assembly protein gene(2.8 folds), NADH-quinone oxidoreductase subunit K2 gene(4.5 folds) and many c-type cytochrome genes such as cytochrome c biogenesis protein Res B(Gmet 2901, 4.6 folds), cytochrome c(Gmet 0335, 4.4 folds) and cytochrome c7(Gmet 2902, 3.4 folds). Furthermore, a gene related to chlorinated contaminant removal(Gmet 1046, 5.4 folds) was also upregulated, possibly resulting in enhanced CAP reduction.
