1Cell–cell contact with proinflammatory macrophages enhances the immunotherapeutic effect of mesenchymal stem cells in two abortion models显示文摘Mesenchymal stem cells(MSCs),which are pluripotent cells with immunomodulatory properties,have been considered good candidates for the therapy of several immune disorders,such as inflammatory bowel diseases,concanavalin A-induced liver injury,and graft-versus-host disease.The embryo is a natural allograft to the maternal immune system.A successful pregnancy depends on the timely extinction of the inflammatory response induced by embryo implantation,followed by the switch to a tolerant immune microenvironment in both the uterus and the system.Excessive infiltration of immune cells and serious inflammatory responses are triggers for embryo rejection,which results in miscarriage.Here,we demonstrated that adoptive transfer of MSCs could prevent fetal loss in a lipopolysaccharide(LPS)-induced abortion model and immune response-mediated spontaneous abortion model.The immunosuppressive MSCs alleviated excessive inflammation by inhibiting CD4+T cell proliferation and promoting the decidual macrophage switch to M2 in a tumor necrosis factor-stimulated gene-6(TSG-6)-dependent manner.Cell-tocell contact with proinflammatory macrophages increased the TSG-6 production by the MSCs,thereby enhancing the suppressive regulation of T cells and macrophages.Moreover,proinflammatory macrophages in contact with the MSCs upregulated the expression of CD200 on the stem cells and facilitated the reprogramming of macrophages towards an anti-inflammatory skew through the interaction of CD200 with CD200R on proinflammatory macrophages.Therefore,the results demonstrate that a TSG-6-mediated paracrine effect,reinforced by cell-to-cell contact between MSCs and proinflammatory macrophages,is involved in the mechanism of MSC-mediated abortion relief through the induction of immune tolerance.Our study also indicates the potential application of MSCs in clinical recurrent miscarriages.Yanhong Li Di Zhang Ling Xu Lin Dong Ji Zheng Yikong Lin Jiefang Huang Yanyun Zhang Yu Tao Xingxing Zang Dajin Li Meirong Du 2019Cellular & Molecular Immunology2019,16,12:23
2Safety and efficacy of ciprofol vs.propofol for sedation in intensive care unit patients with mechanical ventilation:a multi-center,open label,randomized,phase 2 trial显示文摘Background:Ciprofol(HSK3486;Haisco Pharmaceutical Group Co.,Ltd.,Chengdu,China),developed as a novel 2,6-disubstituted phenol derivative showed similar tolerability and efficacy characteristics as propofol when applicated as continuous intravenous infusion for 12 h maintenance sedation in a previous phase 1 trial.The phase 2 trial was designed to investigate the safety,efficacy,and pharmacokinetic characteristics of ciprofol for sedation of patients undergoing mechanical ventilation.Methods:In this multicenter,open label,randomized,propofol positive-controlled,phase 2 trial,39 Chinese intensive care unit patients receiving mechanical ventilation were enrolled and randomly assigned to a ciprofol or propofol group in a 2:1 ratio.The ciprofol infusion was started with a loading infusion of 0.1-0.2 mg/kg for 0.5-5.0 min,followed by an initial maintenance infusion rate of 0.30 mg·kg^(-1)·h^(-1),which could be adjusted to an infusion rate of 0.06 to 0.80 mg·kg^(-1)·h^(-1),whereas for propofol the loading infusion dose was 0.5-1.0 mg/kg for 0.5-5.0 min,followed by an initial maintenance infusion rate of 1.50 mg·kg^(-1)·h^(-1),which could be adjusted to 0.30-4.00 mg·kg^(-1)·h^(-1)to achieve-2 to+1 Richmond Agitation-Sedation Scale sedation within 6-24 h of drug administration.Results:Of the 39 enrolled patients,36 completed the trial.The median(min,max)of the average time to sedation compliance values for ciprofol and propofol were 60.0(52.6,60.0)min and 60.0(55.2,60.0)min,with median difference of 0.00(95%confidence interval:0.00,0.00).In total,29(74.4%)patients comprising 18(69.2%)in the ciprofol and 11(84.6%)in the propofol group experienced 86 treatment emergent adverse events(TEAEs),the majority being of severity grade 1 or 2.Drug-and sedation-related TEAEs were hypotension(7.7%vs.23.1%,P=0.310)and sinus bradycardia(3.8%vs.7.7%,P=1.000)in the ciprofol and propofol groups,respectively.The plasma concentration-time curves for ciprofol and propofol were similar.Conclusions:ciprofol is comparable to propofol with good tolerance and efficacy for sedation of Chinese intensive care unit patients undergoing mechanical ventilation in the present study setting.Trial registration:ClinicalTrials.gov,NCT04147416.Yongjun Liu Xiangyou Yu Duming Zhu Jun Zeng Qinhan Lin Bin Zang Chuanxi Chen Ning Liu Xiao Liu Wei Gao Xiangdong Guan 2022Chinese Medical Journal2022,,9:19

4Protective Effect of Hydroxysafflor Yellow A on Bleomycin-Induced Pulmonary Inflammation and Fibrosis in Rats显示文摘Objective: To observe the effect of hydroxysafflor yellow A(HSYA), an active ingredient of a traditional Chinese herbal medicine Carthamus tinctorius L., on lung inflammation and pulmonary fibrosis induced by bleomycin(BLM) in rats. Methods: Animals were divided into 6 groups including normal group, model group, three HSYA groups and dexamethasone(DXM) group. Three doses of HSYA(35.6, 53.3, and 80.0 mg·kg^(–1)·day^(–1)) were intraperitoneally(i.p.) injected in rats for 3 weeks after BLM administration and DXM was used as the positive control(n=8 or 12). Arterial blood gas was assayed and morphological changes were observed. Lung mRNA expressions of tumor necrosis factor(TNF)-α, interleukin(IL)-1β, IL-6 and some cytokines in lung tissue were detected by real-time polymerase chain reaction. Nuclear factor-κB p65 or α-smooth muscle actin(α-SMA) protein distribution in rat lung tissue was observed by immunohistochemistry. Results: On the 7 th day after BLM administration, lung tissue showed serious inflammation. Treatment with HSYA or DXM ameliorated lung inflammation. After treatment with HSYA or DXM, oxygen partial pressure(PaO_2) increased(HSYA 80.0 mg·kg^(–1), P<0.01) and CO_2 partial pressure(PaCO_2) decreased(HSYA 53.3, 80.0 mg·kg^(–1), P<0.05). Moreover, the mRNA expression of TNF-α, IL-1β, and IL-6; and the number of NF-κB p65 positive cells was lower in HSYA 53.3 and 80.0 mg·kg^(–1) groups than those in the model group(all P<0.05). Twenty-one days after BLM administration, HSYA or DXM treatment ameliorated fibrosis, increased PaO_2(HSYA 53.3, 80.0 mg·kg^(–1), P<0.01), and decreased PaCO_2(53.3 and 80.0 mg·kg^(–1), P<0.05). Further, the mRNA expression of TGF-β1, α-SMA, and collagen Ⅰ as well as the number of α-SMA positive cells increased in the model group and HSYA can attenuate these changes(53.3, 80.0 mg·kg^(–1), P<0.05). Hematoxylin and eosin and Masson's trichrome staining indicated that the fibrosis and collagen deposition were ameliorated in HSYA groups(53.3, 80.0 mg·kg^(–1), P<0.05). 6BubR1 phosphorylates CENP-E as a switch enabling the transition from lateral association to end-on capture of spindle microtubules显示文摘Error-free mitosis depends on accurate chromosome attachment to spindle microtubules, powered congression of those chromosomes, their segregation in anaphase, and assembly of a spindle midzone at mitotic exit. The centromere-associated kinesin motor CENP-E, whose binding partner is BubR1, has been implicated in congression of misaligned chromosomes and the transition from lateral kinetochore-microtubule association to end-on capture. Although previously proposed to be a pseudokinase, here we report the structure of the kinase domain of Drosophila melanogaster BubR1, revealing its folding into a conformation predicted to be catalytically active. BubR1 is shown to be a bona fide kinase whose phosphorylation of CENP-E switches it from a laterally attached microtubule motor to a plus-end microtubule tip tracker. Computational modeling is used to identify bubristatin as a selective BubR1 kinase antagonist that targets the αN1 helix of N-terminal extension and αC helix of the BubR1 kinase domain. Inhibition of CENP-E phosphorylation is shown to prevent proper microtubule capture at kinetochores and, surprisingly, proper assembly of the central spindle at mitotic exit. 8Robust superhydrophobic polyurethane sponge functionalized with perfluorinated graphene oxide for efficient immiscible oil/water mixture, stable emulsion separation and crude oil dehydration显示文摘In recent years, graphene oxide(GO), prepared by the modified Hummers’ method, and its derivatives have become a focus of research owing to their outstanding physical and chemical properties and low cost. Drawing inspiration from the mussel protein,a facile and environmentally-friendly method was employed to fabricate superhydrophobic/superoleophilic reduced graphene oxide(rGO) derivative. The preparation comprises two steps: coating GO nanosheets with polydopamine(PDA) and subsequent reaction with 1H,1H,2H,2H-perfluorodecanethiol. Due to the excellent adhesive ability of PDA, the resulting f PDA modified rGO nanosheets(rGO-f PDA) were firmly immobilized onto polyurethane(PU) sponge skeleton by a simple drop-coating method. The as-prepared rGO-f PDA functionalized sponge exhibited superhydrophobic behavior with a water contact angle of 162°±2°, high organic adsorption capacity, recyclability and stable oil/water separation behavior under different acidic/alkaline conditions. 12Anatomical variation of infra-pyloric artery origination: A prospective multicenter observational study (IPA-Origin)显示文摘Objective: Infra-pyloric artery(IPA) is an important anatomical landmark in treatment of gastric cancer and is the key vessel for pylorus-preserving gastrectomy and subgroup of infra-pyloric lymph nodes. However, its anatomical variation is not thoroughly understood. Our study aimed to clarify the origination of the IPA.Methods: We did this prospective, multicenter, open-label, observational study at gastric surgery departments of34 hospitals in China. Gastric cancer patients aged 18 years or older and scheduled to undergo elective total or distal gastrectomy were assigned. During the surgery, IPA dissecting and exposing the origination point with photographs or video clips were required. The primary outcome was the origination of the IPA. Analysis of variance, χ~2 tests and Fisher's tests were used to analyze the differences between groups. The study is registered at Clinicaltrials.gov(No. NCT03071237).Results: Between May 8 and July 31, 2017, 429 patients were assigned for the study, and 419(97.7%) patients had the IPA dissected and recorded through photograph or video and were included in the primary outcome analysis. The median age was 62 years old, and 73.7% were male. Among the patients, 78.5% received laparoscopic surgery. Single IPA origination was identified in 398(95.0%) patients, including gastroduodenal artery(GDA) in154(36.8%) patients, anterior superior pancreaticoduodenal artery(ASPDA) in 130(31.0%) patients, and right gastroepiploic artery(RGEA) in 114(27.2%) patients. Fifteen(3.6%) patients were identified with multiple IPA and 6(1.4%) patients were identified as IPA absence. The differences in the distribution of surgical approach(P=0.003) and geographic area(P=0.030) were statistically significant. 13Rapid and sensitive liquid chromatography–tandem mass spectrometric method for the quantitative determination of potentially harmful substance 5,5′-oxydimethylenebis (2-furfural) in traditional Chinese medicine injections显示文摘With the rapid development and wide application of traditional Chinese medicine injection(TCMI), a number of adverse events of some TCMIs have incessantly been reported and have drawn broad attention in recent years. Establishing effective and practical analytical methods for safety evaluation and quality control of TCMI can help to improve the safety of TCMIs in clinical applications. In this study, a sensitive and rapid high-performance liquid chromatography–tandem mass spectrometry(HPLC–MS/MS)method has been developed and validated for the quantitative determination of potentially harmful substance5,5′-oxydimethylenebis(2-furfural, OMBF) in TCMI samples. Chromatographic separation was performed on a C18 reversed-phase column(150 mm × 2.1 mm, 5 μm) by gradient elution, using methanol–water containing 0.1% formic acid as mobile phase at the flow rate of 0.3 m L/min. MS/MS detection was performed on a triple quadrupole mass spectrometer with positive electrospray ionization in the multiple reaction-monitoring mode. The method was sensitive with a limit of quantification of 0.3 ng/m L and linear over the range of 0.3–30 ng/m L(r = 0.9998). Intra-and inter-day precision for analyte was o9.52% RSD withrecoveries in the range 88.0–109.67% at three concentration levels. The validated method was successfully applied to quantitatively determine the compound OMBF in TCMIs and glucose injections. 14CZ-7, a new derivative of Claulansine F, ameliorates 2V0-induced vascular dementia in rats through a Nrf2-mediated antioxidant responses显示文摘Vascular dementia (VD) results from accumulated damage in the vascular system, which is characterized by progressive impairments in memory and cognition and is second only to Alzheimer’s disease (AD) in prevalence among all types of dementia. In contrast to AD, there is no FDA-approved treatment for VD owing to its multiple etiologies. In this study, we investigated whether CZ-7, a new derivative of Claulansine F (Clau F) with verified neuroprotective activity in vitro, could ameliorate the cognitive impairment of rats with permanent occlusion of bilateral common carotid arteries (2VO) and its potential mechanisms of action. The 2VO rats were orally administered CZ-7 (10, 20, 40?mg/kg) from day 27 to day 53 post-surgery. Morris water maze tests conducted at day 48–51 revealed that CZ-7 administration significantly reduced the escape latency in 2VO rats. After the rats were sacrificed on day 53, morphological studies using Nissl and terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) staining showed that administration of CZ-7 markedly attenuated the pathological changes in CA1–CA3 area of the hippocampus, including neuronal cell loss, nuclear shrinkage, and dark staining of neurons, and significantly decreased the chronic cerebral hypoperfusion-induced cell loss. Klüver–Barrera staining study revealed that CZ-7 administration significantly improved the white matter lesions. 8-OHdG and reactive oxygen species (ROS) immunofluorescent analyses showed that CZ-7 administration significantly decreased oxidative stress in CA1–CA3 area of the hippocampus. Finally, we found that the CZ-7-improved oxidative stress might be mediated via the Nrf2 pathway, evidenced by the double immunofluorescent staining of Nrf2 and the elevation of expression levels of oxidative stress proteins HO-1 and NQO1. 17Predictors of short-term outcomes related to central subfield foveal thickness after intravitreal bevacizumab for macular edema due to central retinal vein occlusion显示文摘AIM:To investigate the predictive factors for shortterm effects of intravitreal bevacizumab injections on central subfield foveal thickness(CSFT)in patients with macular edema(ME)secondary to central retinal vein occlusion(CRVO).·METHODS:This was a retrospective study in 60 eyes treated with intravitreal bevacizumab injections for ME due to CRVO.Follow-up was three months.The Early Treatment Diabetic Retinopathy Study(ETDRS)score and CSFT measured by spectral-domain optical coherence tomography(SD-OCT)were used to observe the changes in best-corrected visual acuity(BCVA).Baseline BCVA,CSFT,age,CRVO duration and the presence of cystoid macular edema(CME)or subretinal fluid(SRF)were analyzed as potential predictive factors of the effects of intravitreal bevacizumab injections.·RESULTS:BCVA improved from 0.9 log MAR at baseline to 0.6 log MAR at 3mo,which was associated with a significant reduction in CSFT from 721μm to 392μm 3mo after injection.About 50%of CME cases and more than90%of SRF cases responded to treatment with a complete resolution at 3mo.Age(P=0.036)and low baseline CSFT(P=0.037)were associated with a good 3-month prognosis.Patients>60 years old achieved better CME resolution(P=0.031)and lower CSFT at 3mo(305μm vs 474μm,P=0.003).·CONCLUSION:Intravitreal bevacizumab significantly improved visual acuity and CSFT in patients with CRVO after 3mo.Older age and lower baseline CSFT were good predictors of short-term CSFT outcomes.The retinalthickness response to bevacizumab might depend on the resolution of CME rather than SRF.Mei-Zi Wang Kang Feng Yao Lu Fang Qian Xin-Rong Lu Si-Wen Zang Lin Zhao 2016International Journal of Ophthalmology(English edition)2016,9,1:3

19Dynamics of algae growth and nutrients in experimental enclosures culturing bighead carp and common carp: Phosphorus dynamics显示文摘This is the third paper of the series about 'Dynamics of algae growth and nutrients in experimental enclosures culturing bighead carp and common carp'.In this paper,phosphorus dynamics were investigated under the condition of culturing bighead carp and common carp with added fish food(nitrogen dynamics is discussed in the second paper because their behaviors are so different from each other).Nearly fifty days' observation results indicated that the reservoir water was typical of 'phosphorus limited' water,and soluble reactive phosphorus(SRP) was the main constituent of measured total phosphorus(TP).The presence of fish food resulted in significantly higher SRP,dissolved total phosphorus(DTP) and TP concentrations in contrast with the reservoir water.Moreover,continuous supply of fish food led to the decline of total nitrogen to total phosphorus(TN:TP) from more than 100 to less than5.Variations in the ratio of TN to TP favored the growth of blue-green algae.Fish species affected phosphorus concentrations,and culturing bait-eating common carp contributed more to reducing the SRP,DTP and TP concentrations than culturing planktivorous bighead carp.0.5%,4.1%and 3.1%TP can be removed in enclosures with culturing bighead carp,common carp and mixed bighead carp and common carp,respectively.Abundant phosphorus in the fish culturing activities may be present as the uneaten food,algae cells,and within the water column and sediment,which should be taken into serious consideration for the target of future water eutrophication prevention and safety of the drinking water supply.Suiliang Huang Min Wu Changjuan Zang Shenglan Du Joseph Domagalski Magdalena Gajewska Feng Gao Chao Lin Yong Guo Baoyan Liu Shaoming Wang Yang Luo Adam Szymkiewicz Romuald Szymkiewicz 2016International Journal of Sediment Research2016,31,2:3
20Laser test of the prototype of CEE time projection chamber显示文摘A prototype thick-GEM-based cooling storage ring external-target experiment(CEE) time projection chamber(TPC) is constructed and tested with the pulsed ultraviolet laser beams. The results indicate that the prototype TPC has a good performance in three-dimensional track resolution. In X direction the position resolution is about 0.2 mm, and in Y direction the position resolution is about 0.5 mm.
