1Whole-genome sequencing of 508 patients identifies key molecular features associated with poor prognosis in esophageal squamous cell carcinoma显示文摘Esophageal squamous cell carcinoma(ESCC)is a poor-prognosis cancer type with limited understanding of its molecular etiology.Using 508 ESCC genomes,we identified five novel significantly mutated genes and uncovered mutational signature clusters associated with metastasis and patients’outcomes.Several functional assays implicated that NFE2L2 may act as a tumor suppressor in ESCC and that mutations in NFE2L2 probably impaired its tumor-suppressive function,or even conferred oncogenic activities.Additionally,we found that the NFE2L2 mutations were significantly associated with worse prognosis of ESCC.We also identified potential noncoding driver mutations including hotspot mutations in the promoter region of SLC35E2 that were correlated with worse survival.Approximately 5.9%and 15.2%of patients had high tumor mutation burden or actionable mutations,respectively,and may benefit from immunotherapy or targeted therapies.We found clinically relevant coding and noncoding genomic alterations and revealed three major subtypes that robustly predicted patients’outcomes.Collectively,we report the largest dataset of genomic profiling of ESCC useful for developing ESCC-specific biomarkers for diagnosis and treatment.Yongping Cui Hongyan Chen Ruibin Xi Heyang Cui Yahui Zhao Enwei Xu Ting Yan Xiaomei Lu Furong Huang Pengzhou Kong Yang Li Xiaolin Zhu Jiawei Wang Wenjie Zhu Jie Wang Yanchun Ma Yong Zhou Shiping Guo Ling Zhang Yiqian Liu Bin Wang Yanfeng Xi Ruifang Sun Xiao Yu Yuanfang Zhai Fang Wang Jian Yang Bin Yang Caixia Cheng Jing Liu Bin Song Hongyi Li Yi Wang Yingchun Zhang Xiaolong Cheng Qimin Zhan Yanhong Li Zhihua Liu-Show 2020Cell Research2020,30,10:19
2Roof pre-blasting to prevent support crushing and water inrush accidents显示文摘Support crushing and water inrush when mining under an unconsolidated confined aquifer in the Qidong Coal Mine was prevented by roof pre-blasting. The mechanism and applicable conditions for this method have been studied. The results show that when an overburden structure that may cause support crushing and a water inrush accident exists the weakening of the primary key stratum, which thereby reduces its weighting step, roof pre-blasting is both feasible and effective. If the position of the primary key stratum can be moved upward to exceed 10 times the mining height the possibility of support crushing and water inrush disaster caused by key stratum compound breakage will be lowered. The overburden structure of the number 7121 working face was considered during the design of a technical proposal involving roof pre-blasting. After comprehensively analyzing the applicability of roof pre-blasting the resulting design prevented support crushing and water inrush disasters from happening at the number 7121 working face and laid a solid foundation for mining safely.Wang Xiaozhen 2012International Journal of Mining Science and Technology2012,22,3:10
3A Model Output Machine Learning Method for Grid Temperature Forecasts in the Beijing Area显示文摘In this paper, the model output machine learning (MOML) method is proposed for simulating weather consultation, which can improve the forecast results of numerical weather prediction (NWP). During weather consultation, the forecasters obtain the final results by combining the observations with the NWP results and giving opinions based on their experience. It is obvious that using a suitable post-processing algorithm for simulating weather consultation is an interesting and important topic. MOML is a post-processing method based on machine learning, which matches NWP forecasts against observations through a regression function. By adopting different feature engineering of datasets and training periods, the observational and model data can be processed into the corresponding training set and test set. The MOML regression function uses an existing machine learning algorithm with the processed dataset to revise the output of NWP models combined with the observations, so as to improve the results of weather forecasts. To test the new approach for grid temperature forecasts, the 2-m surface air temperature in the Beijing area from the ECMWF model is used. MOML with different feature engineering is compared against the ECMWF model and modified model output statistics (MOS) method. MOML shows a better numerical performance than the ECMWF model and MOS, especially for winter. The results of MOML with a linear algorithm, running training period, and dataset using spatial interpolation ideas, are better than others when the forecast time is within a few days. 4Multiple Dolomitization and Fluid Flow Events in the Precambrian Dengying Formation of Sichuan Basin, Southwestern China显示文摘The Precambrian Dengying Formation is a set of large-scale, extensively dolomitized, carbonate reservoirs occurring within the Sichuan Basin. Petrographic and geochemical studies reveal dolomitization was a direct result of precipitation by chemically distinct fluids occurring at different times and at different intensities. Based on this evidence, dolomitization and multiple fluid flow events are analyzed, and three types of fluid evolution models are proposed. Results of analysis show that Precambrian Dengying Formation carbonates were deposited in a restricted peritidal environment(630–542 Ma). A high temperature and high Mg^(2+) concentration seawater was a direct result of dolomitization for the micrite matrix, and for fibrous aragonite in primary pores. Geochemical evidence shows low δ^(18)O values of micritic dolomite varying from-1.29‰ to-4.52‰ PDB, abundant light rare earth elements(REEs), and low dolomite order degrees. Microbes and meteoric water significantly altered dolomite original chemical signatures, resulting in algal micritic dolomite and the fine-grained, granular, dolosparite dolomite having very negative δ^(18)O values. Finely crystalline cement dolomite(536.3–280 Ma) and coarsely crystalline cement dolomite have a higher crystallization degree and higher order degree. The diagenetic sequence and fluid inclusion evidence imply a linear correlation between their burial depth and homogenization temperatures, which closely resemble the temperature of generated hydrocarbon. Compared with finely crystalline dolomite, precipitation of coarsely crystalline dolomite was more affected by restricted basinal fluids. In addition, there is a trend toward a more negative δ^(18)O value, higher salinity, higher Fe and Mn concentrations, REE-rich. Two periods of hydrothermal fluids are identified, as the exceptionally high temperatures as opposed to the temperatures of burial history, in addition to the presence of high salinity fluid inclusions. The early hydrothermal fluid flow event was characterized by hot magnesium-and silicon-rich fluids, as demonstrated by the recrystallized matrix dolomite that is intimately associated with flint, opal, and microcrystalline quartz in intergranular or intercrystalline pores. This event was likely the result of a seafloor hydrothermal chimney eruption during Episode I of the Tongwan Movement(536.3±5.5 Ma). In contrast, later hydrothermal fluids, which caused precipitation of saddle dolomite, were characterized by high salinity(15–16.05 wt% NaCl equivalent) and homogenization temperatures(250 to 265°C), δ^(18)O values that were more enriched, and REE signatures. Geochemical data and the paragenetic sequence indicate that this hydrothermal fluid was related to extensive Permian large igneous province activity(360–280 Ma). 5Heteroatom-doped porous carbon from methyl orange dye wastewater for oxygen reduction显示文摘Banana peel-derived porous carbon(BPPC) was prepared from banana peel and used as an adsorbent for methyl orange(MO) wastewater removal. BPPC-MO50 is a N,S-doped BPPC obtained via secondary carbonization. The BPPC-MO50 exhibited a high specific surface area of1774.3 m^2/g. Heteroatom-doped porous carbon(PC) was successfully synthesized from the BPPC absorbed MO at high temperature and used for oxygen reduction. The BPPC-MO50 displayed the highest ORR onset potential among all carbon-based electrocatalysts, i.e., 0.93 V vs.reversible hydrogen electrode(RHE). This is the first report to describe porous carbon-activated materials from agriculture and forestry waste that is used for adsorption of dyes from wastewater via an enhanced heteroatom(N,S) content. 6Rapid, automated, and reliable antimicrobial susceptibility test from positive blood culture by CAST‐R显示文摘Antimicrobial susceptibility tests(ASTs)are pivotal in combating multidrug resistant pathogens,yet they can be time‐consuming,labor‐intensive,and unstable.Using the AST of tigecycline for sepsis as the main model,here we establish an automated system of Clinical Antimicrobials Susceptibility Test Ramanometry(CAST‐R),based on D2O‐probed Raman microspectroscopy.Featuring a liquid robot for sample pretreatment and a machine learning‐based control scheme for data acquisition and quality control,the 3‐h,automated CAST‐R process accelerates AST by>10‐fold,processes 96 paralleled antibiotic‐exposure reactions,and produces high‐quality Raman spectra.The Expedited Minimal Inhibitory Concentration via Metabolic Activity is proposed as a quantitative and broadly applicable parameter for metabolism‐based AST,which shows 99%essential agreement and 93%categorical agreement with the broth microdilution method(BMD)when tested on 100 Acinetobacter baumannii isolates.Further tests on 26 clinically positive blood samples for eight antimicrobials,including tigecycline,meropenem,ceftazidime,ampicillin/sulbactam,oxacillin,clindamycin,vancomycin,and levofloxacin reveal 93%categorical agreement with BMD‐based results.The automation,speed,reliability,and general applicability of CAST‐R suggest its potential utility for guiding the clinical administration of antimicrobials.Pengfei Zhu Lihui Ren Ying Zhu Jing Dai Huijie Liu Yuli Mao Yuandong Li Yuehui He Xiaoshan Zheng Rongze Chen Xiaoting Fu Lili Zhang Lijun Sun Yuanqi Zhu Yuetong Ji Bo Ma Yingchun Xu Jian Xu Qiwen Yang 2022mLife2022,1,3:3
7Cobalt Sulfide Confined in N-Doped Porous Branched Carbon Nanotubes for Lithium-Ion Batteries显示文摘Lithium-ion batteries(LIBs) are considered new generation of large-scale energy-storage devices.However,LIBs suffer from a lack of desirable anode materials with excellent specific capacity and cycling stability.In this work,we design a novel hierarchical structure constructed by encapsulating cobalt sulfide nanowires within nitrogen-doped porous branched carbon nanotubes(NBNTs)for LIBs.The unique hierarchical Co9S8@NBNT electrode displayed a reversible specific capacity of 1310 mAhg-1 at a current density of 0.1 Ag-1,and was able to maintain a stable reversible discharge capacity of 1109 mAhg-1 at a current density of 0.5 Ag-1 with coulombic efficiency reaching almost 100% for 200 cycles.The excellent rate and cycling capabilities can be ascribed to the hierarchical porosity of the one-dimensional Co9S8@NBNT internetworks,the incorporation of nitrogen doping,and the carbon nanotube confinement of the active cobalt sulfide nanowires offering a proximate electron pathway for the isolated nanoparticles and shielding of the cobalt sulfide nanowires from pulverization over long cycling periods.Yongsheng Zhou Yingchun Zhu Bingshe Xu Xueji Zhang Khalid A.Al.Ghanim Shahid Mahboob 2019Nano-Micro Letters2019,11,2:3
8Reactive oxygen species in paraventricular nucleus involved in cardiac sympathetic afferent reflex in rats显示文摘Objective: The present study was designed to determine if reactive oxygen species (ROS) in the paraventricular nucleus (PVN) were involved in modulating cardiac sympathetic afferent reflex (CSAR) in anesthetized rats. Methods: Malondialdehyde (MDA), the end product of lipid peroxidation, in the PVN, was determined by thiobarbituric acid (TBA) spectrometric method. Renal sympathetic nerve activity (RSNA) and arterial pressure were recorded in sinoaortic-denervated and cervical-vagotomized rats. The CSAR was evaluated by the response of the RSNA evoked by epicardial application of bradykinin (BK, 0.4 μg). Results: The MDA in the PVN was significantly increased after epicardial application of BK compared with control (2.0±0.3 vs 0.8±0.1 nmol/mg protein, P<0.01). Microinjection of a superoxide anion scavenger, tiron (20 nmol) into the PVN significantly inhibited the CSAR evoked by BK (12.3±1.9 vs 4.2±1.2%, P<0.01) and decreased MDA level (1.9±0.3 vs 0.6±0.1 nmol/mg protein, P<0.01) compared with control. 9Genome-wide identification and expression analysis of NIN-like Protein(NLP)genes reveals their potential roles in the response to nitrate signaling in watermelon显示文摘To balance the relationship between high yield and low nitrogen supply,the nitrogen utilization efficiency of watermelon needs to be improved urgently.Nodule inception-like Protein(NLP)transcription factors play a key node role in nitrate response and growth and development of plant,however,comprehensive analysis of the NLP gene family in watermelon is unclear.This study explored the functional classification,evolutionary characteristics,and expression profile of the ClNLP gene family.Three ClNLPs were categorized into three groups according to their gene structure and phylogeny.All of them contained the conserved RWP-RK and PB1 domains.Evolutionary analysis of ClNLPs revealed that ClNLP1 and ClNLP3 underwent strong purified selection.In addition,cis-acting elements related to plant hormones and abiotic stresses were present in the ClNLP promoter.According to tissue-specific analysis ClNLP was widely expressed in roots,stems,leaves,flowers and fruits,and ClNLP1 was significantly induced in the roots of different nitrogen utilization varieties under different nitrate nitrogen supply.The SRTING functional protein association network suggested that ClNLP1 is associated with most genes,such as NRT1.1,NRT2.1,NIA1,and NIR1,and the dual-luciferase reporter assay found that ClNLP1 positively regulates the expression of ClNRT2.1.We speculated that ClNLP1 might play a central role in regulating the response of watermelon to nitrate nitrogen.Gaopeng Yuan Dexi Sun Yifan Wang Guolin An Weihua Li Wenjing Si Junpu Liu Yingchun Zhu 2022Horticultural Plant Journal2022,8,5:2
10Selection of nitrite-degrading and biogenic amine-degrading strains and its involved genes显示文摘Objectives:Accumulation of nitrite and biogenic amines(BAs)in fermented meat products is a matter of public health concern.The study aimed to screen nitrite-degrading and BA-degrading strains from sour porridges and sausages and bacon products in China.Materials and Methods:After screening out 12 strains,the degradation of nitrite,the degradation of BAs,the activities of nitrite-reducing enzymes,and the detection of genes involved in the BAs were assessed by spectrophotometry method with hydrochloric acid naphthalene ethylenediamine,high-performance liquid chromatography,GENMED kit,and polymerase chain reaction,respectively.Results:Pediococcus pentosaceus labelled M SZ12 and M GC 2,Lactobacillus plantarum labelled M SZ22,and Staphylococcus xylosus labelled Y CC 3 were selected.The activity of nitrite-reducing enzyme in M SZ22 was 2.663 units/mg.The degradation rate of total BAs of M SZ22 was 93.24%.The degradation rates of nitrite and BAs of M SZ12 were 86.49%and 37.87%,respectively.The activity of nitrite-reducing enzyme in M SZ12 was up to 1.962 units/mg.M GC 2 showed higher degradation rates of nitrite(89.19%)and Y CC 3 showed higher degradation rates of BAs(36.16%).The genes encoding the multicopper oxidases(suf I/D2EK17)were detected in the four strains,which also did not contain BAs(histidine decarboxylase(hdc),tyrosine decarboxylase(tdc),ornithine decarboxylase(odc),lysine decarboxylase(ldc))formation encoding genes.Conclusion:These four strains(M SZ12,M GC 2,M SZ22,and Y CC 3)are promising candidates to use as starter cultures for nitrite and BAs in fermented sausages.Yuxin Li Zhihui Yu Yingchun Zhu Zhixiang Cao 2020Food Quality and Safety2020,4,4:2
