| 1 | Over-expression of WOX1 Leads to Defects in Meristem Development and Polyamine Homeostasis in Arabidopsis显示文摘In plants,the meristem has to maintain a separate population of pluripotent cells that serve two main tasks,i.e.,self-maintenance and organ initiation,which are separated spatially in meristem.Prior to our study,WUS and WUS-like WOX genes had been reported as essential for the development of the SAM.In this study,the consequences of gain of WOX1 function are described.Here we report the identification of an Arabidopsis gain-of-function mutant wox1-D,in which the expression level of the WOX1(WUSCHEL HOMEOBOX 1) was elevated and subtle defects in meristem development were observed.The wox1-D mutant phenotype is dwarfed and slightly bushy,with a smaller shoot apex.The wox1-D mutant also produced small and dark green leaves,and exhibited a failure in anther dehiscence and male sterility.Molecular evidences showed that the transcription of the stem cell marker gene CLV3 was down-regulated in the meristem of wox1-D but accumulated in the other regions,i.e.,in the root-hypocotyl junction and at the sites for lateral root initiation.The fact that the organ size and cell size in leaves of wox1-D are smaller than those in wild type suggests that cell expansion is possibly affected in order to have partially retarded the development of lateral organs,possibly through alteration of CLV3 expression pattern in the meristem.An S-adenosylmethionine decarboxylase(SAMDC) protein,SAMDC1,was found able to interact with WOX1 by yeast two-hybrid and pull-down assays in vitro.HPLC analysis revealed a significant reduction of polyamine content in wox1-D.Our results suggest that WOX1 plays an important role in meristem development in Arabidopsis,possibly via regulation of SAMDC activity and polyamine homeostasis,and/or by regulating CLV3 expression. | Yanxia Zhang Renhong Wu Genji Qin Zhangliang Chen Hongya Gu Li-Jia Qu | 2011 | Journal of Integrative Plant Biology2011,53,6: | 23 |

| 3 | Evaluating and Improving Wind Forecasts over South China: The Role of Orographic Parameterization in the GRAPES Model显示文摘Unresolved small-scale orographic(SSO) drags are parameterized in a regional model based on the Global/Regional Assimilation and Prediction System for the Tropical Mesoscale Model(GRAPES TMM). The SSO drags are represented by adding a sink term in the momentum equations. The maximum height of the mountain within the grid box is adopted in the SSO parameterization(SSOP) scheme as compensation for the drag. The effects of the unresolved topography are parameterized as the feedbacks to the momentum tendencies on the first model level in planetary boundary layer(PBL)parameterization. The SSOP scheme has been implemented and coupled with the PBL parameterization scheme within the model physics package. A monthly simulation is designed to examine the performance of the SSOP scheme over the complex terrain areas located in the southwest of Guangdong. The verification results show that the surface wind speed bias has been much alleviated by adopting the SSOP scheme, in addition to reduction of the wind bias in the lower troposphere. | 7 | Satellite-observed trends in the Arctic sea ice concentration for the period 1979–2016显示文摘Arctic sea ice cover has decreased dramatically over the last three decades. This study quanti?es the sea ice concentration(SIC) trends in the Arctic Ocean over the period of 1979–2016 and analyzes their spatial and temporal variations. During each month the SIC trends are negative over the Arctic Ocean, wherein the largest(smallest) rate of decline found in September(March) is-0.48%/a(-0.10%/a).The summer(-0.42%/a) and autumn(-0.31%/a) seasons show faster decrease rates than those of winter(-0.12%/a) and spring(-0.20%/a) seasons. Regional variability is large in the annual SIC trend. The largest SIC trends are observed for the Kara(-0.60%/a) and Barents Seas(-0.54%/a), followed by the Chukchi Sea(-0.48%/a), East Siberian Sea(-0.43%/a), Laptev Sea(-0.38%/a), and Beaufort Sea(-0.36%/a). The annual SIC trend for the whole Arctic Ocean is-0.26%/a over the same period. Furthermore, the in?uences and feedbacks between the SIC and three climate indexes and three climatic parameters, including the Arctic Oscillation(AO), North Atlantic Oscillation(NAO), Dipole anomaly(DA), sea surface temperature(SST), surface air temperature(SAT), and surface wind(SW), are investigated. Statistically, sea ice provides memory for the Arctic climate system so that changes in SIC driven by the climate indices(AO, NAO and DA) can be felt during the ensuing seasons. Positive SST trends can cause greater SIC reductions, which is observed in the Greenland and Barents Seas during the autumn and winter. In contrast, the removal of sea ice(i.e., loss of the insulating layer) likely contributes to a colder sea surface(i.e., decreased SST), as is observed in northern Barents Sea. Decreasing SIC trends can lead to an in-phase enhancement of SAT, while SAT variations seem to have a lagged in?uence on SIC trends. | 10 | Changes in Physicochemical,Cooking and Sensory Characteristics of Rice Shifted from Low-temperature Storage显示文摘The physicochemical, cooking, and sensory characteristics of stored rice were measured in order to investigate the quality changes in it after low-temperature storage. The quality of the stored rice was compared to that of the rice stored at ambient temperature(20℃, 30%–70% RH) at a 25 d interval during 200 d of storage. The rice was stored in a temperature controller at 4℃ for 3 months and later under a simulated condition similar to that of the main rice consumption areas in China(35℃, 80% RH and 30℃, 70% RH). The results showed that the fatty acid value, b value, and moisture content of rice stored at 35℃, 80% RH and 30℃, 70% RH had increased significantly, whereas its L value had decreased as compared to the rice stored at ambient temperature. Higher temperature storage caused a greater water uptake, whereas the dry mass in the residual cooking water notably reduced under the storage at 35℃ as compared to that at 20℃. Hardness increased and adhesiveness reduced under the storage at 35℃ as compared to that at 20℃. | 14 | Fabrication of aluminum matrix composite reinforced with carbon nanotubes显示文摘1.0wt.% carbon nanotube (CNT) reinforced 2024Al matrix composite was fabricated by cold isostatic press and subsequent hot extrusion techniques. The mechanical properties of the composite were measured by a tensile test. Mean-while,the fracture surfaces were examined using field emission scanning electron microscopy. The experimental results show that CNTs are dispersed homogeneously in the composite and that the interfaces of the Al matrix and the CNT bond well. Although the tensile strength and the Young’s modulus of the composite are enhanced markedly,the elongation does not decrease when compared with the matrix material fabricated under the same process. | 15 | Arctic sea ice volume export through the Fram Strait from combined satellite and model data:1979–2012显示文摘By combing satellite-derived ice motion and concentration with ice thickness fields from a popular model PIOMAS we obtain the estimates of ice volume flux passing the Fram Strait over the 1979–2012 period. Since current satellite and field observations for sea ice thickness are limited in time and space, the use of PIOMAS is expected to fill the gap by providing temporally continued ice thickness fields. Calculated monthly volume flux exhibits a prominent annual cycle with the peak record in March(roughly 145 km3/month) and the trough in August(10 km^3/month). Annual ice volume flux(1 132 km^3) is primarily attributable to winter(October through May) outflow(approximately 92%). Uncertainty in annual ice volume export is estimated to be 55 km^3(or 5.7%). Our results also verified the extremely large volume flux appearing between late 1980 s and mid-1990 s. Nevertheless, no clear trend was found in our volume flux results. Ice motion is the primary factor in the determination of behavior of volume flux. Ice thickness presented a general decline trend may partly enhance or weaken the volume flux trend. Ice concentration exerted the least influences on modulating trends and variability in volume flux. Moreover, the linkage between winter ice volume flux and three established Arctic atmospheric schemes were examined. | 18 | A model for nanogrinding based on direct evidence of ground chips of silicon wafers显示文摘Nanometer chips were directly fabricated using face nanogrinding carried out by ultrafine diamond grits at room temperature. Direct evidence for ground nanometer chips is cuboid, and the average ratio of width to thickness is 1.49. Chips of 9.0 nm in thickness, 13.3 nm in width, and 16.0 in diagonal were achieved and confirmed using transmission electron microscopy. Based on the nanometer chips observed, a model was proposed according to the mass conservation and fundamental mechanism of face grinding. The surface roughness and thickness of damaged layers measured experimentally are in good agreement with the prediction of the developed model. | 19 | Application of regional citrate anticoagulation in patients at high risk of bleeding during intermittent hemodialysis:a prospective multicenter randomized controlled trial显示文摘Objective:Safe and effective anticoagulation is essential for hemodialysis patients who are at high risk of bleeding.The purpose of this trial is to evaluate the effectiveness and safety of two-stage regional citrate anticoagulation(RCA)combined with sequential anticoagulation and standard calcium-containing dialysate in intermittent hemodialysis(IHD)treatment.Methods:Patients at high risk of bleeding who underwent IHD from September 2019 to May 2021 were prospectively enrolled in 13 blood purification centers of nephrology departments,and were randomly divided into RCA group and saline flushing group.In the RCA group,0.04 g/mL sodium citrate was infused from the start of the dialysis line during blood draining and at the venous expansion chamber.The sodium citrate was stopped after 3 h of dialysis,which was changed to sequential dialysis without anticoagulant.The hazard ratios for coagulation were according to baseline.Results:A total of 159 patients and 208 sessions were enrolled,including RCA group(80 patients,110 sessions)and saline flushing group(79 patients,98 sessions).The incidence of severe coagulation events of extracorporeal circulation in the RCA group was significantly lower than that in the saline flushing group(3.64%vs.20.41%,P<0.001).The survival time of the filter pipeline in the RCA group was significantly longer than that in the saline flushing group((238.34±9.33)min vs.(221.73±34.10)min,P<0.001).The urea clearance index(Kt/V)in the RCA group was similar to that in the saline flushing group with no statistically significant difference(1.12±0.34 vs.1.08±0.34,P=0.41).Conclusions:Compared with saline flushing,the two-stage RCA combined with a sequential anticoagulation strategy significantly reduced extracorporeal circulation clotting events and prolonged the dialysis time without serious adverse events. | Xiaoyan TANG Dezheng CHEN Ling ZHANG Ping FU Yanxia CHEN Zhou XIAO Xiangcheng XIAO Weisheng PENG Li CHENG Yanmin ZHANG Hongbo LI Kehui LI Bizhen GOU Xin WU Qian YU Lijun JIAN Zaizhi ZHU Yu WEN Cheng LIU Hen XUE Hongyu ZHANG Xin HE Bin YAN Liping ZHONG Bin HUANG Mingying MAO | 2022 | Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2022,23,11: | 4 |
| 20 | Impact of diabetes on promoting the growth of breast cancer显示文摘Background:Type Ⅱ diabetes mellitus(DM2)is a significant risk factor for cancers,including breast cancer.However,a proper diabetic breast cancer mouse model is notwell-established for treatment strategy design.Additionally,the precise diabetic signaling pathways that regulate cancer growth remain unresolved.In the present study,we established a suitable mouse model and demonstrated the pathogenic role of diabetes on breast cancer progression.Methods:We successfully generated a transgenic mouse model of human epidermal growth factor receptor 2 positive(Her2^(+) or ERBB2)breast cancer with DM2 by crossing leptin receptor mutant(Lepr^(db/+))mice with (MMTV-ErbB2/neu)mice.Themousemodelswere administrated with antidiabetic drugs to assess the impacts of controlling DM2 in affecting tumor growth.Magnetic resonance spectroscopic imaging was employed to analyze the tumor metabolism.Results:Treatment with metformin/rosiglitazone in MMTV-ErbB2/Lepr^(db/db) mousemodel reduced serum insulin levels,prolonged overall survival,decreased cumulative tumor incidence,and inhibited tumor progression.Anti-insulin resistance medications also inhibited glycolytic metabolism in tumors in vivo as indicated by the reduced metabolic flux of hyperpolarized ^(13)C pyruvate-to-lactate reaction.The tumor cells from MMTV-ErbB2/Lepr^(db/db) transgenic mice treated with metformin had reprogrammed metabolism by reducing levels of both oxygen consumption and lactate production.Metformin decreased the expression of Myc and pyruvate kinase isozyme 2(PKM2),leading to metabolism reprogramming.Moreover,metformin attenuated the mTOR/AKT signaling pathway and altered adipokine profiles.Conclusions:MMTV-ErbB2/Lepr^(db/db) mouse model was able to recapitulate diabetic HER2^(+) human breast cancer.Additionally,our results defined the signaling pathways deregulated in HER2^(+) breast cancer under diabetic condition,which can be intervened by anti-insulin resistance therapy. | Ping-Chieh Chou Hyun Ho Choi Yizhi Huang Enrique Fuentes-Mattei Guermarie Velazquez-Torres Fanmao Zhang Liem Phan Jaehyuk Lee Yanxia Shi James A.Bankson Yun Wu Huamin Wang Ruiying Zhao Sai-Ching Jim Yeung Mong-Hong Lee | 2021 | Cancer Communications2021,41,5: | 4 |
