1miR-25 modulates triacylglycerol and lipid accumulation in goat mammary epithelial cells by repressing PGC-1beta显示文摘Background: The goat(Caprahircus) is one of the most important livestock animals. Goat milk fat is an important component in the nutritional quality of goat milk. Growing evidence points to the critical roles of microRNAs(miRNAs) in lipid metabolism.Results: Using a highly sensitive method of S-poly(T) plus for miRNAs detection, we analyze the expression patterns of 715 miRNAs in goat mammary gland tissues at different stages of lactation. We observed that miR-25 expression had an inverse relationship with milk production. Overexpression of miR-25 significantly repressed triacylglycerol synthesis and lipid droplet accumulation. To explore the regulatory mechanism of miR-25 in milk lipid metabolism,we analyzed its putative target genes with bioinformatics analysis followed by 3′-UTR assays. Peroxisome proliferative activated receptor gamma coactivator 1 beta(PGC-1 beta), a key regulator of lipogenics was identified as a direct target of miR-25 with three specific sites within its 3′-UTR. 2Evaluation of dolomite reservoirs in the Longwangmiao Formation,Lower Cambrian in Northern Sichuan basin,China显示文摘With the extended petroleum exploration of the carbonate reservoir in the Longwangmiao Formation,Lower Cambrian in Sichuan basin,looking for a new favorable exploration area is the primary task in current exploration.This paper focusing on the dolomite reservoir in the Longwangmiao Formation in the Northern Sichuan basin,will evaluate the reservoir conditions,including reservoir lithology,reservoir spaces,physical properties and other basic characteristics.Combined with sedimentary environment study and diagenesis analysis,the genetic mechanism of Longwangmiao reservoir in study area will be discussed.It indicates that the reservoir rocks of Longwangmiao Formation in the study area are composed of dolarenite and crystalline dolomite,and the main reservoir spaces are intergranular dissolved pores,intragranular dissolved pores and intercrystalline dissolved pores.However,most of dissolved pores have been filled with bitumen,which lead to the reservoir properties very poor,both the porosity and permeability are worse than that of central Sichuan basin,so the reservoir in the Longwangmiao Formation in the Northern Sichuan basin is low-porosity and low-permeability reservoir.During the Early Cambrian Longwangmiao period,the study was near the old-land,and the energy of sea water was not enough to deposit many grain shoals,which cause the sediments are small with lots of quartz and muds.Moreover,because lacking of large scale karstification in the late diagenesis,there isn't much dissolved pores and vugs develop in the Longwangmiao Formation.Some Intergranular dissolved pores and intercrystalline dissolved pores formed by the early atmosphere water dissolved exist as some reservoir spaces,but most of them have been filled with bitumen,which was residual when the ancient oil reservoir transformed.Above factors cause the Longwangmiao Formation in the Northern Sichuan basin become not a very favorable reservoir in Sichuan basin.Xuefei Yang Yaping Wang Xingzhi Wang Deming Zeng Longbo Xu Zisang Huang 2017Petroleum2017,3,4:3

4An improved Tet-on system in micro RNA overexpression and CRISPR/Cas9-mediated gene editing显示文摘Background: Tetracycline(Tet)-regulated expression system has become a widely applied tool to control gene activity. This study aimed to improve the Tet-on system with superior regulatory characteristics.Results: By comprehensively comparing factors of transactivators, Tet-responsive elements(TREs), orientations of induced expression cassette, and promoters controlling the transactivator, we developed an optimal Tet-on system with enhanced inducible efficiency and lower leakiness. With the system, we successfully performed effective inducible and reversible expression of micro RNA, and presented a more precise and easily reproducible fine-tuning for confirming the target of a mi RNA. 9Resource prediction and assessment based on 3D/4D big data modeling and deep integration in key ore districts of North China显示文摘The North China district has been subjected to significant research with regard to the ore-forming dynamics,processes,and quantitative forecasting of gold deposits;it accounts for the highest number of gold reserves and annual products in China.Based on the top-level design of geoscience theory and the method adopted by the National Key R&D Project(deep process and metallogenic mechanism of North China Craton(NCC)metallogenic system),this paper systematically collects and constructs the geoscience data(district,camp,and deposit scales)in four key gold districts of North China(Jiaojia-Sanshandao,Southern Zhaoping,Wulong,and Qingchengzi).The settings associated with the geological dynamics of gold deposits were quantitatively and synthetically analyzed,namely:NCC destruction,metallogenic events,genetic models,and exploration models.Three-dimensional(3D)and four-dimensional(4D)geological modeling was performed using the big data on the districts,while the district-scale 3D exploration criteria were integrated to construct a quantitative exploration model.Among them,FLAC3D modelling and the Geo Cube software(version 3.0)were used to implement the numerical simulation of the 3D geological models and the constraints of the fluid saturation parameters of the Jiaojia fault to reconstruct the 4D fault structure models of the Jiaojia fault(with a depth of 5000 m).Using Geo Cube3.0,multiple integration modules(general weights of evidence(Wof E),Boost Wof E,Fuzzy Wof E,Logistic Regression,Information Entropy,and Random Forest)and exploration criteria were integrated,while the C-V fractal classification of A,B and C targets in four districts was carried out.The research results are summarized in the following four areas:(1)Four gold districts in the study area have more than three targets(the depth is 3000 m),and the class A,B and C targets exhibit a good spatial correlation with gold bodies that are controlled by mining engineering at depths greater than 1000 m.(2)The Boost Wof E method was used to identify the target optimization in 3D spaces(at depths of 3000–5000 m)of the Jiaojia-Sanshandao,Southern Zhaoping,and Wulong districts.(3)The general Wof E method is based on the Bayesian theory in 3D space and provides robust integration and target optimization that are suitable for the Jiaojia-Sanshandao and Southern Zhaoping districts in the Jiaodong area;it can also be applied to the Wulong district in the Liaodong area using a quantitative genetic model and an exploration model.Random forest is a multi-objective integration and target optimization method for 3D spaces,and it is suitable for the complex exploration model in the Qingchengzi district of the Liaodong area.The genetic model and exploration criteria associated with the exploration model of the Qingchengzi district were constrained by the common characteristics of the gold fault structure,magmatic rock emplacement in North China,and the strata fold and interlayer detachment structure.(4)Based on the gold reserves and the 3D block unit model of the Sanshandao gold deposit in the Jiaojia-Sanshandao district,the gold contents of the 3D block units in class A and B targets of the ore concentration were estimated to be 65.5%and 25.1%,respectively.The total Au resources of the optimized targets below a depth of 3000 m were 3908 t(including 1700 t reserves),and the total Au resources of the targets at depths from 3000 to 5000 m were 936 t.The study shows that the deep gold deposits in the four gold districts of North China exhibit a strong'transport-deposition'spatial correlation with potential targets.These'transport-deposition'spatial models represent the tectonic-magmatic-hydrothermal activities of the metallogenic system associated with the NCC destruction events and indicate the Au enrichment zones.Gongwen WANG Zhiqiang ZHANG Ruixi LI Junjian LI Deming SHA Qingdong ZENG Zhenshan PANG Dapeng LI Leilei HUANG 2021Science China Earth Sciences2021,64,9:1
10Pore-fillings of dolomite reservoirs in Sinian Dengying Formation in Sichuan basin显示文摘The Sinian Dengying Formation in the Sichuan basin is an important potential reservoir for oil and gas exploration in the periphery of the Sichuan basin.Its reservoirs were well developed.And many kinds of fillings which lead to the decrease of pores can be seen in reservoir pores and caves.In this paper,the pore fillings of the Dengying Formation reservoir in Sichuan basin are the object of study.The types and characteristics of the fillings are studied in detail by means of rock slices analysis,cathode luminescence,trace elements and inclusion temperature measurement.The study shows that the fillings include dolomite fillings and siliceous quartz fillings and bitumen fillings.Among them,dolomite fillings can be divided into botryoidal dolomite fillings,foliated dolomite fillings,finely-medium crystalline dolomite fillings and coarse crystalline dolomite fillings.50%–80%of the pores are filled by the dolomite fillings with botryoidal structure,and the pores can be partially full filled;0%–20%of pores are filled by foliated dolomite fillings;20–80%of the pores are filled by finely-medium dolomite fillings;20–80%of the pores are filled by coarse crystalline dolomite fillings.1%–5%of secondary pores are often filled by siliceous quartzs,and the pores can be partially full filled.Bitumen fillings often occur after the above-mentioned fillings,and the residual pores are often semi-filled by bitumens,which makes the original pores and throat smaller and reduces the effective porosity and permeability of the reservoir.So,it is a destructive diagenesis.The pore fillings make the secondary porosity in the reservoir significantly lower,which is not conducive to the formation and preservation of the Dengying Formation reservoir.Chao Chen Xuefei Yang Xingzhi Wang Deming Zeng Mingyou Feng Lin Xie Xiang Li 2020Petroleum2020,6,1:1

12Reservoir characteristics and genetic differences between the second and fourth members of Sinian Dengying Formation in northern Sichuan Basin and its surrounding areas显示文摘This study investigated the characteristics and genesis of reservoirs in the 2^(nd) and 4^(th) members of Sinian Dengying Formation in northern Sichuan and its surrounding areas, on the basis of outcrop, drilling cores and thin section observation and geochemical analysis. The reservoirs of 2^(nd) member are distributed in the middle part of the stratum. The reservoir quality is controlled by supergene karst and the distribution of mound-shoal complex. The bedded elongated isolated algal framework solution-cave and the residual 'grape-lace' cave, which are partially filled with multi-stage dolosparite, constituted the main reservoir space of the 2^(nd) member. There is no asphalt distribution in the pores. The pore connectivity is poor, and the porosity and permeability of the reservoir is relatively low. The reservoirs of 4^(th) member were distributed in the upper and top part of the stratum. The reservoir quality is controlled by burial dissolution and the distribution of mound-shoal complex. The bedded algal framework solution-pores or caves, intercrystalline pores and intercrystalline dissolved pores constituted the main reservoir space of the 4^(th) member. It's partially filled with asphalt and quartz, without any dolosparite fillings in the pores and caves. The pore connectivity is good. Most of the 4^(th) member reservoirs had medium-low porosity and low permeability, and, locally, medium-high porosity and medium permeability. Affected by the development of mound-shoal complex and heterogeneous dissolution, the platform margin along Ningqiang, Guangyuan, Jiange and Langzhong is the most favorable region for reservoir development. 18Versatile SrFeO_(x) for memristive neurons and synapses显示文摘Spiking neural network(SNN)consisting of memristor-based artificial neurons and synapses has emerged as a compact and energy-efficient hardware solution for spatiotemporal information processing.However,it is challenging to develop memristive neurons and synapses based on the same material system because the required resistive switching(RS)characteristics are different.Here,it is shown that SrFeO_(x)(SFO),an intriguing material system exhibiting topotactic phase transformation between insulating brownmillerite(BM)SrFeO_(2).5 phase and conductive perovskite(PV)SrFeO_(3) phase,can be engineered into both neuronal and synaptic devices.Using a BM-SFO single layer as the RS medium,the Au/BM-SFO/SrRuO_(3)(SRO)memristor exhibits nonvolatile RS behavior originating from the formation/rupture of PV-SFO filaments in the BM-SFO matrix.By contrast,using a PV-SFO(matrix)/BM-SFO(interfacial layer)bilayer as the RS medium,the Au/PV-SFO/BM-SFO/SRO memristor exhibits volatile RS behavior originating from the interfacial BM-PV phase transformation.Synaptic and neuronal characteristics are further demonstrated in the Au/BM-SFO/SRO and Au/PV-SFO/BM-SFO/SRO memristors,respectively.Using the SFO-based synapses and neurons,fully memristive SNNs are constructed by simulation,which show good performance on unsupervised image recognition.Our study suggests that SFO is a versatile material platform on which both neuronal and synaptic devices can be developed for constructing fully memristive SNNs.Kaihui Chen Zhen Fan Jingjing Rao Wenjie Li Deming Wang Changjian Li Gaokuo Zhong Ruiqiang Tao Guo Tian Minghui Qin Min Zeng Xubing Lu Guofu Zhou Xingsen Gao Jun-Ming Liu 2022Journal of Materiomics2022,8,5:0
