| 1 | Cloning of a gene-edited macaque monkey by somatic cell nuclear transfer显示文摘Cloning of macaque monkeys by somatic cell nucleus transfer(SCNT) allows the generation of monkeys with uniform genetic backgrounds that are useful for the development of non-human primate models of human diseases. Here, we report the feasibility of this approach by SCNT of fibroblasts from a macaque monkey(Macaca fascicularis), in which a core circadian transcription factor BMAL1 was knocked out by clustered regularly interspaced short palindromic repeat/Cas9 gene editing(see accompanying paper).Out of 325 SCNT embryos transferred into 65 surrogate monkeys, we cloned five macaque monkeys with BMAL1 mutations in both alleles without mosaicism, with nuclear genes identical to that of the fibroblast donor monkey. Further peripheral blood m RNA analysis confirmed the complete absence of the wild-type BMAL1 transcript. | 2 | Numerical simulation and experimental investigation of natural convection heat transfer of molten salt around fine wire显示文摘In order to get the natural convection heat transfer mechanism of molten salt, the experimental investigation of natural convective heat transfer of LiNO3was studied after it was simulated by numerical calculation. Experiment was carried out on the nat-ural convection heat transfer of air and water around the fine wire using the method of Joule heating. The results showed that the natural convection heat transfer of air and water around the fine wire agreed well with Fand's correlation. Based on the aforementioned experiment, the natural convection heat transfer of molten salt LiNO3was studied by experiment and the same results were got. | 4 | Targeting and exploitation of tumor-associated neutrophils to enhance immunotherapy and drug delivery for cancer treatment显示文摘Neutrophils,the most abundant leukocytes in human blood,are essential fighter immune cells against microbial infection.Based on the finding that neutrophils can either restrict or promote cancer progression,tumor-associated neutrophils(TAN)are classified into anti-tumor N1 and pro-tumor N2 subsets.One of the major mechanisms underlying the tumor-promoting function of N2-TANs is suppression of adaptive immune cells,in particular,cytotoxic T lymphocytes.Currently,no established methodologies are available that can unequivocally distinguish immunosuppressive TANs and granulocytic/polymorphonuclear myeloid-derived suppressor cells(G/PMN-MDSC).In view of the critical role of PMN-MDSCs in immune evasion and resistance to cancer immunotherapy,as established from data obtained with diverse cancer models,therapeutic strategies targeting these cells have been actively developed to enhance the efficacy of immunotherapy.Here,we have reviewed the available literature on strategies targeting PMN-MDSCs and summarized the findings into four categories:(1)depletion of existing PMN-MDSCs,(2)blockade of the development of PMNMDSCs,(3)blockade of PMN-MDSC recruitment,(4)inhibition of immunosuppressive function.Owing to their high mobility to inflamed organs and ability to trespass the blood-brain barrier,neutrophils are outstanding candidate carriers in nanoparticle-based therapies.Another attractive application of neutrophils in cancer therapy is the use of neutrophil membrane-derived nanovesicles as a surrogate of extracellular vesicles for more efficient and scalable drug delivery.In the second part of the review,we have highlighted recent advances in the field of neutrophil-based cancer drug delivery.Overall,we believe that neutrophil-based therapeutics are a rapidly growing area of cancer therapy with significant potential benefits. | Yuting Zhang Guoqiang Liu Miaomiao Sun Xin Lu | 2020 | Cancer Biology & Medicine2020,17,1: | 9 |

| 10 | Generation of obese rat model by transcription activator-like effector nucleases targeting the leptin receptor gene显示文摘The laboratory rat is a valuable mammalian model organism for basic research and drug discovery. Here we demonstrate an efficient methodology by applying transcription activator-like effector nucleases(TALENs) technology to generate Leptin receptor(Lepr) knockout rats on the Sprague Dawley(SD) genetic background. Through direct injection of in vitro transcribed m RNA of TALEN pairs into SD rat zygotes, somatic mutations were induced in two of three resulting pups. One of the founders carrying bi-allelic mutation exhibited early onset of obesity and infertility. The other founder carried a chimeric mutation which was efficiently transmitted to the progenies. Through phenotyping of the resulting three lines of rats bearing distinct mutations in the Lepr locus, we found that the strains with a frame-shifted or premature stop codon mutation led to obesity and metabolic disorders. However, no obvious defect was observed in a strain with an in-frame 57 bp deletion in the extracellular domain of Lepr. This suggests the deleted amino acids do not significantly affect Lepr structure and function. This is the first report of generating the Lepr mutant obese rat model in SD strain through a reverse genetic approach. | 15 | Surface sulfur vacancies enhanced electron transfer over Co-ZnS quantum dots for efficient degradation of plasticizer micropollutants by peroxymonosulfate activation显示文摘Peroxymonosulfate(PMS)activation in heterogeneous processes is a promising water treatment technology.Nevertheless,the high energy consumption and low efficiency during the reaction are ineluctable,due to electron cycling rate limitation.Herein,a new strategy is proposed based on a quantum dots(QDs)/PMS system.Co-ZnS QDs are synthesized by a water phase coprecipitation method.The inequivalent lattice-doping of Co for Zn leads to the generation of surface sulfur vacancies(SVs),which modulates the surface of the catalyst to form an electronic nonequilibrium surface.Astonishingly,the plasticizer micropollutants can be completely degraded within only tens of seconds in the Co-Zn S QDs/PMS system due to this type of surface modulation.The interfacial reaction mechanism is revealed that pollutants tend to be adsorbed on the cobalt metal sites as the electron donors,where the internal electrons of pollutants are captured by the metal species and transferred to the surface SVs.Meanwhile,PMS adsorbed on the SVs is reduced to radicals by capturing electrons,achieving effective electron recovery.Dissolved oxygen(DO)molecules are also easily attracted to catalyst defects and are reduced to O_(2)^(·-),further promoting the degradation of pollutants. | Yuting Gu Tingting Gao Fagen Zhang Chao Lu Wenrui Cao Ziwei Fu Chun Hu Lai Lyu | 2022 | Chinese Chemical Letters2022,33,8: | 2 |

| 17 | SERS study on the synergistic effects of electric field enhancement and charge transfer in an Ag2S quantum dots/plasmonic bowtie nanoantenna composite system显示文摘Localized surface plasmon resonance(LSPR)of nanostructures and the interfacial charge transfer(CT)of semiconductor materials play essential roles in the study of optical and photoelectronic properties.In this paper,a composite substrate of Ag2S quantum dots(QDs)coated plasmonic Au bowtie nanoantenna(BNA)arrays with a metalinsulator-metal(MIM)configuration was built to study the synergistic effect of LSPR and interfacial CT using surface-enhanced Raman scattering(SERS)in the near-infrared(NIR)region.The Au BNA array structure with a large enhancement of the localized electric field(E-field)strongly enhanced the Raman signal of adsorbed p-aminothiophenol(PATP)probe molecules.Meanwhile,the broad enhanced spectral region was achieved owing to the coupling of LSPR The as-prepared Au BNA array structure facilitated enhancements of the excitation as well as the emission of Raman signal simultaneously,which was established by finite-difference time-domain simulation.Moreover,Ag2S semiconductor QDs were introduced into the BNA/PATP system to further enhance Raman signals,which benefited from the interfacial CT resonance in the BNA/Ag2S-QDs/PATP system.As a result,the Raman signals of PATP in the BNA/Ag2S-QDs/PATP system were strongly enhanced under 785 nm laser excitation due to the synergistic effect of E-field enhancement and interfacial CT.Furthermore,the SERS polarization dependence effeas of the BNA/Ag2S-QDs/PATP system were also investigated.The SERS spectra indicated that the polarization dependence of the substrate increased with decreasing polarization angles(θpola)of excitation from p-polarized(θpola=90°)excitation to s-polarized(θpola=0°)excitation.This study provides a strategy using the synergistic effect of interfacial CT and E-field enhancement for SERS applications and provides a guidance for the development of SERS study on semiconductor QD-based plasmonic substrates,and can be farther extended to other material-nanostructure systems for various optoelectronic and sensing applications. | BIN WANG CHEN ZHAO HUANYU LU TINGTING ZOU SUBHASH C.SINGH ZHI YU CHAONAN YAO XIN ZHENG JUN XING YUTING ZOU CUNZHU TONG WEILI YU BO ZHAO CHUNLEI GUO | 2020 | Photonics Research2020,8,4: | 1 |
