| 2 | TRIM47 is a novel endothelial activation factor that aggravates lipopolysaccharide-induced acute lung injury in mice via K63-linked ubiquitination of TRAF2显示文摘Endothelial activation plays an essential role in the pathogenesis of sepsis-induced acute lung injury,however,the detailed regulatory mechanisms remain largely unknown.Here,we reported that TRIM47,an E3 ubiquitin ligase of the tripartite motifcontaining protein family,was highly expressed in vascular endothelial cells.TRIM47-deficient mice were effectively resistant to lipopolysaccharide(LPS)-induced acute lung injury and death by attenuating pulmonary inflammation.TRIM47 was upregulated during TNFα-induced endothelial activation in vitro.Knockdown of TRIM47 in endothelial cells inhibited the transcription of multiple pro-inflammatory cytokines,reduced monocyte adhesion and the expression of adhesion molecules,and suppressed the secretion of IL-1βand IL-6 in endothelial cells.By contrast,overexpression of TRIM47 promoted inflammatory response and monocyte adhesion upon TNFαstimulation.In addition,TRIM47 was able to activate the NF-κB and MAPK signaling pathways during endothelial activation.Furthermore,our experiments revealed that TRIM47 resulted in endothelial activation by promoting the K63-linked ubiquitination of TRAF2,a key component of the TNFαsignaling pathway.Taken together,our studies demonstrated that TRIM47 as a novel activator of endothelial cells,promoted LPS-induced pulmonary inflammation and acute lung injury through potentiating the K63-linked ubiquitination of TRAF2,which in turn activates NF-κB and MAPK signaling pathways to trigger an inflammatory response in endothelial cells. | Yisong Qian Ziwei Wang Hongru Lin Tianhua Lei Zhou Zhou Weilu Huang Xuehan Wu Li Zuo Jie Wu Yu Liu Ling-Fang Wang Xiao-Hui Guan Ke-Yu Deng Mingui Fu Hong-Bo Xin | 2022 | Signal Transduction and Targeted Therapy2022,7,6: | 2 |
| 3 | Surface sulfur vacancies enhanced electron transfer over Co-ZnS quantum dots for efficient degradation of plasticizer micropollutants by peroxymonosulfate activation显示文摘Peroxymonosulfate(PMS)activation in heterogeneous processes is a promising water treatment technology.Nevertheless,the high energy consumption and low efficiency during the reaction are ineluctable,due to electron cycling rate limitation.Herein,a new strategy is proposed based on a quantum dots(QDs)/PMS system.Co-ZnS QDs are synthesized by a water phase coprecipitation method.The inequivalent lattice-doping of Co for Zn leads to the generation of surface sulfur vacancies(SVs),which modulates the surface of the catalyst to form an electronic nonequilibrium surface.Astonishingly,the plasticizer micropollutants can be completely degraded within only tens of seconds in the Co-Zn S QDs/PMS system due to this type of surface modulation.The interfacial reaction mechanism is revealed that pollutants tend to be adsorbed on the cobalt metal sites as the electron donors,where the internal electrons of pollutants are captured by the metal species and transferred to the surface SVs.Meanwhile,PMS adsorbed on the SVs is reduced to radicals by capturing electrons,achieving effective electron recovery.Dissolved oxygen(DO)molecules are also easily attracted to catalyst defects and are reduced to O_(2)^(·-),further promoting the degradation of pollutants. | Yuting Gu Tingting Gao Fagen Zhang Chao Lu Wenrui Cao Ziwei Fu Chun Hu Lai Lyu | 2022 | Chinese Chemical Letters2022,33,8: | 2 |
| 4 | Study on the Resistance of Pseudoperonospora cubensis to Cymoxanil显示文摘[Objective] The paper was to clarify the sensitivity and resistance of Pseudoperonospora cubensis to cymoxanil.[Method] A total of 69 strains were collected from 13 major cucumber producing areas in eight provinces and cities of Guangdong, Hubei, Jiangsu, Shandong, Beijing, Liaoning, Jilin and Heilongjiang from 2012 to 2016, and their sensitivity and resistance to cymoxanil was determined using leaf disc floating method.[Result] The EC50 of cymoxanil to P. cubensis were 0.006 8-0.134 1 μg/mL and the average EC50 was 0.062 4 μg/mL. The sensitivity of P. cubensis to cymoxanil presented a single-peak curve, which was approximately normal distribution, and no strains with decreased sensitivity were found. The average EC50 (0.062 4 μg/mL) could be used as the sensitivity baseline of P. cubensis to cymoxanil. Among the 69 strains, 16 were low-resistance strains, accounting for 23.19% of the total strains;53 were sensitive strains, accounting for 76.81%, which were dominant populations;and no medium-resistance and high resistance strains were found. | 6 | Modeling colorectal tumorigenesis using the organoids derived from conditionally immortalized mouse intestinal crypt cells (ciMICs)显示文摘Intestinal cancers are developed from intestinal epithelial stem cells(ISCs)in intestinal crypts through a multi-step process involved in genetic mutations of oncogenes and tumor suppressor genes.ISCs play a key role in maintaining the homeostasis of gut epithelium.In 2009,Sato et al established a three-dimensional culture system,which mimicked the niche microenvironment by employing the niche factors,and successfully grew crypt ISCs into organoids or Mini-guts in vitro.Since then,the intestinal organoid technology has been used to delineate cellular signaling in ISC biology.However,the cultured organoids consist of heterogeneous cell populations,and it was technically challenging to introduce genomic changes into three-dimensional organoids.Thus,there was a technical necessity to develop a twodimensional ISC culture system for effective genomic manipulations.In this study,we established a conditionally immortalized mouse intestinal crypt(ciMIC)cell line by using a piggyBac transposon-based SV40 T antigen expression system.We showed that the ciMICs maintained long-term proliferative activity under two-dimensional niche factor-containing culture condition,retained the biological characteristics of intestinal epithelial stem cells,and could form intestinal organoids in three-dimensional culture.While in vivo cell implantation tests indicated that the ciMICs were non-tumorigenic,the ciMICs overexpressing oncogenic b-catenin and/or KRAS exhibited high proliferative activity and developed intestinal adenoma-like pathological features in vivo.Collectively,these findings strongly suggested that the engineered ciMICs should be used as a valuable tool cell line to dissect the genetic and/or epigenetic underpinnings of intestinal tumorigenesis. | Xiaoxing Wu Zhaoxia Li Hongyu Zhang Fang He Min Qiao Huaxiu Luo Jing Zhang Meng Zhang Yukun Mao William Wagstaff Yongtao Zhang Changchun Niu Xia Zhao Hao Wang Linjuan Huang Deyao Shi Qing Liu Na Ni Kai Fu Rex C.Haydon Russell R.Reid Hue H.Luu Tong-Chuan He Ziwei Wang Houjie Liang Bing-Qiang Zhang Ning Wang | 2021 | Genes & Diseases2021,8,6: | 1 |
| 7 | Geochronology and Petrogenesis of Granitoid Intrusions in the Feidong District, Southern Tan-Lu Fault Zone, China显示文摘The Feidong district is located in the southern segment of the Tan-Lu fault zone that separates the South China Block(NCB)from the North China Craton(NCC).We report zircon U-Pb geochronology and Hf isotope data,as well as whole-rock geochemistry for Xishanyi granodiorite and Jianshan granite in the Feidong district.Zircon U-Pb dating results show that the emplacement ages of the Xishanyi and Jianshan intrusions are 124±3 Ma and 130±1 Ma respectively,coeval with magmatic events linked to large-scale lithospheric thinning in eastern China.The whole-rock geochemistry of the Xishanyi and Jianshan intrusions demonstrate that they are peraluminous,high potassium calc-alkaline I-type granites with adakitic characteristics.Both intrusions underwent weak crustal assimilation during emplacement.The in situ zirconε(Hf)(t)values of the Xishanyi granodiorites range from-26.4 to-21.8,with T(DM2)model ages of 2552 to 2841 Ma.The in situ zirconε(Hf)(t)values of the Jianshan granite are from-27.5 to-23.0 with T(DM2)model ages of 2632 to 2904 Ma.The peak age of inherited zircon grains from the Xishanyi granodiorite and the Jianshan granite were^2.07 Ga and^1.94 Ga,respectively.After compared with the regional magmatism,we suggest that both the Xishanyi and Jianshan granitoid intrusions were derived from partial melting of the NCC lower crust. | FU Xiang ZHANG Dayu YAO Ziwei REN Kangda DENG Yufeng LI Jiahao JIANG Ren YUAN Feng | 2020 | Acta Geologica Sinica(English Edition)2020,94,6: | 1 |

| 9 | Low-Energy Driven Ring-Opening Behavior of Benzocyclobutene Derivatives显示文摘It's urgent to develop benzocyclobutene(BCB)-based polymers with low curing temperature for temperature-sensitive applications such as liquid crystal displays(LCDs)and flexible electronics.Herein,the effect of substituents on the ring-opening behavior of BCB derivatives was investigated.The ring-opening activation energy barriers(ΔGA)of BCB derivatives with one or two substituents on the four-membered alkyl ring were systematically calculated using the B3LYP function.Both mono-and di-substituted BCBs adopted the conrotatory ring-opening process,obeying the Woodward-Hoffmann's Rules upon heating.The mono-/di-substituted BCBs exhibited 8.2%—69%lowerΔGA compared with BCB,attributed to the electronic effects of the substituents.Disubstituted BCBs with both electron-donating and electron-withdrawing groups,e.g.,1-NH_(2)-8-NO_(2)-BCB,demonstrated the lowestΔGA.In addition,BCB derivatives with amide/ester/acyloxy group modified on C1 position were synthesized as model molecules,and their ring-opening temperature can be decreased by 20℃ compared to the unsubstituted one,also consistent with our calculation results.This work combined theoretical calculation methods with experimental results to provide valuable insights into the design and synthesis of BCB derivatives and next-generation BCB functional packaging materials with low ring-opening temperature. | Ziwei Yuan Quan Sun Jinchong Xiao Pingxia Zhang Konstantin S.Levchenko Dmitry Y.Demin Wenxin Fu | 2023 | Chinese Journal of Chemistry2023,41,23: | 0 |
