| 1 | Multi-Stage Metamorphism of the UHP Pelitic Gneiss from the Southern Altyn Tagh HP/UHP Belt,Western China:Petrological and Geochronological Evidence显示文摘The kyanite-bearing garnet pelitic gneiss from the Jianggalesavi area in southern Altyn Tagh high pressure/ultra-high pressure belt was proved to have been experienced UHP metamorphism (>12 GPa) by the discovery of kyanite and spinel exsolution microstructure in quartz (precursor stishovite). In this study, three stages of retrograded metamorphism (M2-M4) after the UHP metamorphism (Ml) were identified for the UHP pelitic gneiss. The HP granulite-facies stage (M2) was characterized by the mineral assemblage of garnet+kyanite+K?feldspar+nitile+qiiartz±ilinenite, recording the P-T condition of >1.12 GPa and ?850-930℃. The granulite-facies stage (M3) was represented by the mineral assemblage of garnet rim+K-feldspar+sillimanite (SillJ+biotite (Bti)+plagioclase (Pli)+ilmenite+quartz, and confined under P-T conditions of 0.5-0.8 GPa and^770-795℃. The late cooling stage M4 was accompanied by the appearance of fine-grained Pl2, Sill2 and Bt2 in the matrix, and the P-T conditions were 0.4-0.6 GPa and <675℃. A clockvvised P-T path was obtained for the pelitic gneiss in the P-T pseudosection, which showed a deep subduction/collision processes with subsequent exhumation and cooling. Com? bined with the corresponding multistage metamorphic assemblages, the age dating results implied that the zircons from the gneiss have integrated the recording peak metamorphic (Ml,484±3 Ma) and retrograded metamorphic ages (M2 to M3, 450±2 Ma). There was about 32 Ma interval during the first exhumation from the upper mantle depth (>350 km) to the lower crust depth (-40-20 km), resulting in an average exhumation rate of 9.11-9.70 mm/yr. In the southern Altyn Tagh region, the HP and UHP rocks from different areas had identical peak metamorphic ages. Therefore, contemporary UHP and HP rocks with different metamorphic evolutions were recognized coexisting in the same orogenic belt, which can be interpreted by the model of subduction channel. | 3 | Characterization and Expression Analysis of Protein Kinase C Gene from Dunaliella salina显示文摘Protein kinase C (PKC) has a crucial role in signal transduction for a variety of biologically active substances which activate cellular functions and proliferation. We previously isolated the full-length PKC gene from Dunaliella salina (DsPKC) using rapid amplification of cDNA ends (RACE) and RT-PCR methods. And we submitted the mRNA sequence of DsPKC gene to NCBI (Genbank No. JN625213). In the present paper, the DsPKC gene open reading frame obtained by PCR was cloned into pGS-21a vector and transformed into Escherichia coli to generate the fusion protein. Bioinformatics analysis revealed that DsPKC gene was a member of serine/threonine kinase with two conserved domains and highly conserved motifs. The DsPKC was highly expressed upon induction with isopropyl-β-d-thiogalactoside (IPTG) at a final concentration of 0.2 mmol L 1 at 37℃. Under salt stress, the fu- sion protein Green Fluorescent Protein (GFP)-DsPKC was transferred from the cytoplasm to the cell membrane. The expression pat- tern of DsPKC gene was analyzed using real-time quantitative PCR, and indicated that DsPKC gene was up-regulated by 3.0 mol L 1 NaCl at 12 h, which was significantly higher than in control values (P < 0.05). | 5 | Expression, purification, and subcellular localization of phospholipase C in Dunaliella salina显示文摘Plants possess effective mechanisms to respond quickly to the external environment. Rapid activation of phosphatidylinositol-specific phospholipase C (PLC) enzymes occurs after a stimulus. The PLC in Dunaliella salina plays important roles in growth and stress responses. However, the molecular basis of PLC action in D. salina remains little understood. To gain insight into the potential biological functions of this enzyme, we cloned a phospholipase C gene from D. salina in a previous study, named DsPLC (GenBank No. KF573428). Here, we present the prokaryotic expression, purification, and characterization of the DsPLC gene. The entire coding region of DsPLC was inserted into an expression vector pET32a, and the DsPLC gene was successfully expressed in Escherichia coli. The DsPLC protein was purified and identified using a polyclonal antibody and western blotting. Expressing DsPLC fused with a green fluorescent protein (GFP) in onion showed that DsPLC-GFP was localized to the intracellular membrane. Quantitative real-time PCR analysis revealed that the relative expression of the DsPLC gene was induced significantly by 3.0-mol/L NaCl at 4 h. Our results support the importance of PLC enzymes in plant defense signaling. | 6 | Cloning of Ds MAPK Gene and Construction of Antisense Expression Vector显示文摘In order to investigate the role of MAPK gene in adaptation of Dunaliella salina to hypersaline environment,the Ds MAPK gene of D. salina was amplified by PCR. After inverted insertion of the open reading frame of Ds MAPK gene into downstream sequence of 35 S promoter of plant expression vector,an antisense expression vector of Ds MAPK gene was successfully constructed and introduced into D. salina cells by LiA c/PEG-mediated method. The expression of Ds MAPK gene in transgenic D. salina was analyzed by semi-quantitative RT-PCR method. The results showed that the expression of Ds MAPK gene in D. salina was significantly inhibited at the transcriptional level. | 9 | Subcellular Localization of Small GTP-binding Protein DsRab in Dunaliella salina显示文摘With the total RNA of Dunaliella salina as a template,the cD NA sequence of D. salina small GTP-binding protein gene was amplified by RT-PCR technique,and cloned onto pM Dl8-T simple vector,the recon was subjected to PCR detection and restriction endonuclease analysis,and the total sequence of DNA was determined. The results showed that the cloned fragment was 612 bp,and shared 100% homology with reported D. salina DsRab gene( GenB ank: JN989548). The target gene fragment was inserted downstream of pM DCG 35 S promoter,constructing subcellular localization recombinant vector pM DCG-DsRab. The successfully constructed subcellular localization recombinant vector pM DCG-DsRab was transformed into Agrobacterium tumefaciens LBA4404,and positive single clones were screened and used for transinfection of onion epidemical cells by Agrobacterium-mediated method,and the instant expression of DsRab was observed under fluorescence microscope. The results showed that the fusion protein GFP-DsRab was successfully expressed in onion epidemical cells,and mainly distributed on cytomembrane. | 12 | Auricularia polytricha and Flammulina velutipes ameliorate inflammation and modulate the gut microbiota via regulation of NF-κB and Keap1/Nrf2 signaling pathways on DSS-induced inflammatory bowel disease显示文摘Flammulina velutipes and Auricularia polytricha are two popular dietary mushrooms,especially in the Northeast of China.They have revealed many pharmacological effects due to their bioactive compounds,but their possible mechanism of action involved in inflammation remains poorly known.Therefore,Inflammatory bowel disease(IBD)which is a chronic disease and progressive inflammation in the gastrointestinal tract were used to evaluate their anti-inflammatory and modulatory properties on the intestinal microbiota via the signaling pathways involved by oral administration of F.velutipes(FVE)and A.polytricha(APE).Mice were daily received 3%DSS in the drinking water followed by the daily administration of FVE and APE.The food consumption,weight variation and DAI score were assessed during the experiment and inflammatory parameters,mRNA and protein expressions to determine the possible signaling pathways involved,and the gut microbiota from the cecum by high-throughput sequencing.Data showed that APE and FVE improved clinical symptoms,inflammatory cytokines and oxidative markers.They also attenuated the mRNA and proteins expressions in colon tissue.Moreover,APE and FVE regulated DSS-induced intestinal dysbiosis and stimulated the SCFAs production.Overall,APE and FVE could improve DSS-induced IBD in mice suggesting that they represent novels therapeutic drugs and could be used as prebiotic to modulate the gut microbiota and preserve intestinal integrity. | Ivan Stève Nguepi Tsopmejio Miao Ding Jiali Wei Cong Zhao Yu Jiang Yuting Li Hui Song | 2022 | Food Bioscience2022,47,3: | 0 |
