| 1 | A novel miR167a-OsARF6-OsAUX3 module regulates grain length and weight in rice显示文摘Grain size is one of the most important factors that control rice yield,as it is associated with grain weight(GW).To date,dozens of rice genes that regulate grain size have been isolated;however,the regulatory mechanism underlying GW control is not fully understood.Here,the quantitative trait locus qGL5 for grain length(GL)and GW was identified in recombinant inbred lines of 9311 and Nipponbare(NPB)and fine mapped to a candidate gene,OsAUX3.Sequence variations between 9311 and NPB in the OsAUX3 promoter and loss of function of OsAUX3 led to higher GL and GW.RNA sequencing,gene expression quantification,dual-luciferase reporter assays,chromatin immunoprecipitation-quantitative PCR,and yeast one-hybrid assays demonstrated that OsARF6 is an upstream transcription factor regulating the expression of OsAUX3.OsARF6 binds directly to the auxin response elements of the OsAUX3 promoter,covering a single-nucleotide polymorphism site between 9311 and NPB/Dongjin/Hwayoung,and thereby controls GL by altering longitudinal expansion and auxin distribution/content in glume cells.Furthermore,we showed that miR167a positively regulate GL and GW by directing OsARF6 mRNA silencing.Taken together,our study reveals that a novel miR167a-OsARF6-OsAUX3 module regulates GL and GW in rice,providing a potential target for the improvement of rice yield. | Jiyue Qiao Hongzhen Jiang Yuqing Lin Lianguang Shang Mei Wang Dongming Li Xiangdong Fu Markus Geisler Yanhua Qi Zhenyu Gao Qian Qian | 2021 | Molecular Plant2021,14,10: | 8 |
| 2 | Effects of Plant Growth-Promoting Rhizobacteria and N Source on Plant Growth and N and P Uptake by Tomato Grown on Calcareous Soils显示文摘Introducing specific microorganisms into the soil ecological system is an important strategy for improving nutrient use efficiency.Two pot experiments were conducted in the greenhouse from December 3, 2012 to January 25, 2013(Experiment 1) and March 11 to April 23, 2013(Experiment 2) to evaluate the effect of nitrogen(N) source and inoculation with plant growth-promoting rhizobacteria(PGPR) on plant growth and N and phosphorus(P) uptake in tomato(Lycopersicon esculentum Mill.) grown on calcareous soils from South Florida, USA.Treatments included urea, controlled release urea(a controlled release fertilizer, CRF) each at low and high N rates and with or without inoculation of PGPR.A mixture of PGPR strains Bacillus amyloliquefaciens IN937 a and Bacillus pumilus T4 was applied to the soil during growing periods of tomato.Treatments with PGPR inoculation increased plant height compared to treatments without PGPR in both experiments.Inoculation with PGPR increased shoot dry weight and shoot N uptake for the same N rate and N source.In both experiments, only at high N rate, CRF and urea treatments with PGPR had significantly(P < 0.05)greater shoot biomass than those without PGPR.Only at high N rate, CRF treatment with PGPR significantly increased shoot N uptake by 39.0% and 10.3% compared to that without PGPR in Experiments 1 and 2, respectively.Meanwhile, presence of PGPR in the soil increased shoot P uptake for all treatments in Experiment 1 and for most treatments in Experiment 2.In Experiment 1, only at low N rate, CRF treatment with PGPR significantly increased shoot P uptake compared with that without PGPR.In Experiment2, a significant increase in shoot P uptake by inoculation of PGPR was only observed in CRF treatment at high N rate.Results from this study indicate that inoculation with PGPR may increase plant growth and N and P uptake by tomato grown on calcareous soils.However, the effect of PGPR varied and was influenced by many factors such as N source, N rate, and soil fertility.Further investigations are warranted to confirm the effect of PGPR under different soil conditions. | Xiaohui FAN Shouan ZHANG Xiaodan MO Yuncong LI Yuqing FU Zhiguang LIU | 2017 | Pedosphere2017,27,6: | 8 |

| 5 | Efficacy and safety of darbepoetin alfa injection replacing epoetin alfa injection for the treatment of renal anemia in Chinese hemodialysis patients:A randomized,open-label,parallel-group,noninferiority phase III trial显示文摘Background:This study was to explore the clinical efficacy and safety of darbepoetin alfa injection replacing epoetin alfa injection(recombinant human erythropoietin injection,rHuEPO)for the treatment of anemia associated with chronic kidney failure in Chinese patients undergoing hemodialysis.Method:This study was a multicenter,randomized,open-label,intergroup parallel control phase III noninferiority trial from April 19,2013 to September 9,2014 at 25 sites.In this study,the members of the darbepoetin alfa group underwent intravenous administration once per week or once every two weeks.The members of the control drug epoetin alfa group underwent intravenous administration two or three times per week.All subjects underwent epoetin alfa administration during the 8-week baseline period.After that,subjects were randomly assigned to the darbepoetin alfa group or epoetin alfa group.The noninferiority in the changes of the average Hb concentrations from the baseline to the end of the evaluation period(noninferiority threshold:-1.0 g/dl)was tested between the two treatments.The time-dependent hemoglobin(Hb)concentration and the maintenance rate of the target Hb concentration(the proportion of subjects with Hb concentrations between 10.0 and 12.0 g/dl)were also evaluated.Iron metabolism,including changes in the serum iron,total iron-binding capacity,ferritin,transferrin saturation,and comparisons of the dose adjustments between the two groups during the treatment period were analyzed further.Adverse events(AEs)were also observed and compared,and the safety was analyzed between the two treatment groups.The conversion rate switching from epoetin alfa to darbepoetin alfa was also discussed.SAS?software version 9.2 was used to perform all statistical analyses.Descriptive statistics were used for all efficacy,safety,and demographic variable analyses,including for the primary efficacy indicators.Results:Four hundred and sixty-six patients were enrolled in this study,and ultimately 384 cases were analyzed for safety,including 267 cases in the darbepoetin alfa group and 117 cases in the epoetin alfa group.There were 211 cases in the per-protocol set,including 152 cases in the darbepoetin alfa group and 59 cases in the epoetin alfa group.The changes in the average Hb concentrations from the baseline to the end of the evaluation period were-0.07 and-0.15 g/dl in the darbepoetin alfa group and epoetin alfa group respectively.The difference between the two groups was 0.08 g/dl(95%confidence interval[CI]:-0.22 to 0.39),and the lower limit of the 95%CI was-0.22>-1.0 g/dl.The average Hb concentrations of the two groups were 10.88-11.43 g/dl(darbepoetin alfa)and 10.91-11.38 g/dl(epoetin alfa)during the study period of Weeks 0-28,with the maintenance rates of the target Hb concentration ranging within 71%-87%and 78%-95%in the darbepoetin alfa group and epoetin alfa group respectively.During the period of comparison between the two groups,the incidence of AEs in the darbepoetin alfa group was 61.42%,while in the epoetin alfa group it was 56.41%.All of the adverse events and reactions in the study were those commonly associated with hemodialysis.Conclusion:The overall efficacy and safety of darbepoetin alfa for the treatment of Chinese renal anemia patients undergoing hemodialysis are consistent with those of epoetin alfa. | Bicheng Liu Nan Chen Jinghong Zhao Aiping Yin Xiongfei Wu Changying Xing Gengru Jiang Junzhou Fu Mei Wang Rong Wang Jianying Niu Ping Fu Zhaohui Ni Fanfan Hou Jiuyang Zhao Jing Chen Yuqing Chen Wei Shi Jianghua Chen Wenge Li Gang Xu Ling Zhong Wenhu Liu Guohua Ding Yuichiro Kondo Changhe Yue Changlin Mei | 2022 | Chronic Diseases and Translational Medicine2022,8,2: | 1 |

| 7 | Influences of yellow and green lights on the visual response of western flower thrips and field verification显示文摘This study aimed to determine the effects of spectral light characteristics on the visual response of the western flower thrips,the strengthening mechanism of thrips response behavior regulated by light,and thrips response characteristics to contrast light.Light with combined and single wavelength were tested by using a self-made behavior response device for thrips.Light sources for trapping thrips were made to verify the trapping effect on thrips in a greenhouse,and the reasons for changes in thrips behavior were analyzed to characterize the mechanism of their phototactic response.The results showed that the light mode(single,contrast,combined light)affected the thrips visual response and approach response,whereas in contrast light,the effects were optimal.Combination light inhibited the thrips visual response,and when the illumination increased,the thrips visual response to single and combination light intensified,and the thrips approach sensitivity to green light increased in contrast and combination light.However,the light mode did not affect the thrips visual response and sensitivity to spectral light characteristics.The degree of thrips visual response to yellow light was stronger than that to green light,while the degree of thrips visual response to green light was stronger than that to yellow light,indicating that the photo-induced mechanism of the thrips visual response differed from that of the thrips approach response.Moreover,in the greenhouse,the trapping effect of different light sources on thrips was positively correlated with temperature.The trapping effect of green light was optimal,followed by a yellow light source,while the difference of light intensity(illumination,illumination energy)and its photo-thermal intensity between yellow and green light was the reason for the differences in the degree of visual trends and the trapping effects of thrips.However,the sensitivity of thrips responding to different light depended on the difference in the heterogeneous stimulation intensity of different spectral light.Thus,light brightness and photo-thermal effects were the causes of thrips visual responses,while bio-photoelectric reaction effects caused thrips to produce a visual response and affected the degree of the thrips visual response.The results reveal the underlying causes of pest control by light,and provide a theoretical basis for the research and development of pest induction equipment and light arrangements. | Qihang Liu Mingqian Zhao Jin Miao Guangchun Fu Yuqing Wu | 2022 | International Journal of Agricultural and Biological Engineering2022,15,4: | 1 |
| 8 | Darbepoetin alfa injection versus epoetin alfa injection for treating anemia of Chinese hemodialysis patients with chronic kidney failure:A randomized,open-label,parallel-group,non-inferiority Phase III trail显示文摘Background:Erythropoietin is a glycoprotein that mainly regulates erythropoiesis.In patients with chronic renal failure with anemia,darbepoetin alfa can stimulate erythropoiesis,correct anemia,and maintain hemoglobin levels.This study was designed to demonstrate the efficacy and safety of darbepoetin alfa injections as being not inferior to epoetin alfa injections(Recombinant Human Erythropoietin injection,rHuEPO)when maintaining hemoglobin(Hb)levels within the target range(10.0-12.0 g/dL)for the treatment of renal anemia.Methods:Ninety-five patients were enrolled in this study from April 15,2013 to April 10,2014 at 25 sites.In this study,patients(n=95)aged 18-70 years were randomized into a once per week intravenous darbepoetin alfa group(n=56)and a twice or three times per week intravenous epoetin alfa group(n=39)for 28 weeks,who had anemia with hemoglobin levels between 6 g/dL and 10 g/dL due to chronic kidney disease(CKD)and were undergoing hemodialysis or hemofiltration with ESA-naive(erythropoiesis stimulating agent-naive).The primary efficacy profile was the mean Hb level(the non-inferiority margin was-1.0 g/dL,week 21-28);the secondary efficacy profiles were the Hb increase rate(week 0-4),the target Hb achievement cumulative rate and time,the change trends of the Hb levels,and the target Hb maintenance ratio.Adverse events(AEs)were observed and compared,and the efficacy and safety were analyzed between the two treatment groups.Additionally,the frequencies of dose adjustments between the darbepoetin alfa and epoetin alfa groups were compared during the treatment period.SAS?software version 9.2 was used to perform all statistical analyses.Descriptive statistics were used for all efficacy,safety,and demographic variable analyses,including for the primary efficacy indicators.Results:The mean Hb level was 11.3 g/dL in the darbepoetin alfa group and 10.7 g/dL in the epoetin alfa group,respectively;the difference of the lower limits of the 95%confidence intervals(CI)between the two groups was 0.1 g/dL(>-1.0 g/dL),and non-inferiority was proven;the Hb levels started to increase in the first four weeks at a similar increase rate;no obvious differences were observed between the groups in the target Hb achievement cumulative rates,and the Hb levels as well as the target Hb level maintenance rate changed over time.The incidence of AEs was 62.5%in the darbepoetin alfa group and 76.9%in the epoetin alfa group.All the adverse events observed in the study were those commonly associated with hemodialysis.Conclusion:Darbepoetin alfa intravenously once per week can effectively increase Hb levels and maintain the target Hb levels well,which makes it not inferior to epoetin alfa intravenously twice or three times per week.Darbepoetin alfa shows an efficacy and safety comparable to epoetin alfa for the treatment of renal anemia. | Nan Chen Changying Xing Jianying Niu Bicheng Liu Junzhou Fu Jiuyang Zhao Zhaohui Ni Mei Wang Wenhu Liu Jinghong Zhao Ling Zhong Xiongfei Wu Wenge Li Yuqing Chen Wei Shi Jianghua Chen Aiping Yin Ping Fu Rong Wang Gengru Jiang Fanfan Hou Guohua Ding Jing Chen Gang Xu Yuichiro Kondo Yuliang Su Changlin Mei | 2022 | Chronic Diseases and Translational Medicine2022,8,1: | 1 |

| 16 | Method for the extraction of circulating nucleic acids based on MOF reveals cell-free RNA signatures in liver cancer显示文摘Cell-free RNA(cf RNA) allows assessment of health, status, and phenotype of a variety of human organs and is a potential biomarker to non-invasively diagnose numerous diseases. Nevertheless, there is a lack of highly efficient and bias-free cf RNA isolation technologies due to the low abundance and instability of cf RNA.Here, we developed a reproducible and high-efficiency isolation technology for different types of cell-free nucleic acids(containing cf RNA and viral RNA) in serum/plasma based on the inclusion of nucleic acids by metal-organic framework(MOF) materials, which greatly improved the isolation efficiency and was able to preserve RNA integrity compared with the most widely used research kit method. Importantly, the quality of cf RNA extracted by the MOF method is about 10-fold that of the kit method, and the MOF method isolates more than three times as many different RNA types as the kit method. The whole transcriptome mapping characteristics of cf RNA in serum from patients with liver cancer was described and a cf RNA signature with six cf RNAs was identified to diagnose liver cancer with high diagnostic efficiency(area under curve = 0.905 in the independent validation cohort) using this MOF method. | 20 | Increased expression of osteoprotegerin in vascular smooth muscle cells from spontaneously hypertensive rats显示文摘Background Osteoprotegerin (OPG) is a secreted protein of the tumor necrosis factor receptor family, which regulates bone mass by inhibiting osteoclast differentiation and activation. Although OPG is expressed ubiquitously and abundantly in many tissues and cell types including vascular cells, the role of OPG in other tissues is unknown.Our previous studies demonstrated that OPG was highly expressed in vascular smooth muscle cells (VSMC) and upregulated during vascular lesion formation. Methods and Results We documented, by Northern blot analysis,that the expression of OPG was more prevalent in the aorta and cultured VSMC from spontaneously hypertensive rats (SI-IR) compared to Wistar-Kyoto rats (WKY). In addition, we found that the expression of Angiotensin II (Ang II)type I receptor (AT1R) in SHR VSMC was at significantly increased levels than in WKY VSMC. Furthermore, Ang II potently induced the expression of OPG in VSMC in a time- and dose-dependent manner through the AT1R signaling pathway.
