| 1 | Phosphatase activity and culture conditions of the yeast Candida mycoderma sp. and analysis of organic phosphorus hydrolysis ability显示文摘Orthophosphate is an essential but limiting macronutrient for plant growth. About 67%cropland in China lacks sufficient phosphorus, especially that with red soil. Extensive soil phosphorus reserves exist in the form of organic phosphorus, which is unavailable for root uptake unless hydrolyzed by secretory acid phosphatases. Thus, many microorganisms with the ability to produce phosphatase have been exploited. In this work, the activity of an extracellular acid phosphatase and yeast biomass from Candida mycoderma was measured under different culture conditions, such as p H, temperature, and carbon source. A maximal phosphatase activity of 8.47 × 105± 0.11 × 105U/g was achieved by C. Mycoderma in 36 hr under the optimal conditions. The extracellular acid phosphatase has high activity over a wide p H tolerance range from 2.5 to 5.0(optimum p H 3.5). The effects of different phosphorus compounds on the acid phosphatase production were also studied. The presence of phytin, lecithin or calcium phosphate reduced the phosphatase activity and biomass yield significantly. In addition, the p H of the culture medium was reduced significantly by lecithin. The efficiency of the strain in releasing orthophosphate from organic phosphorus was studied in red soil(used in planting trees) and rice soil(originating as red soil). The available phosphorus content was increased by 230% after inoculating20 days in rice soil and decreased by 50% after inoculating 10 days in red soil. | 4 | RBM4 dictates ESCC cell fate switch from cellular senescence to glutamine-addiction survival through inhibiting LKB1-AMPK-axis显示文摘Cellular senescence provides a protective barrier against tumorigenesis in precancerous or normal tissues upon distinct stressors.However,the detailed mechanisms by which tumor cells evade premature senescence to malignant progression remain largely elusive.Here we reported that RBM4 adversely impacted cellular senescence to favor glutamine-dependent survival of esophageal squamous cell carcinoma(ESCC)cells by dictating the activity of LKB1,a critical governor of cancer metabolism.The level of RBM4 was specifically elevated in ESCC compared to normal tissues,and RBM4 overexpression promoted the malignant phenotype.RBM4 contributed to overcome H-RAS-or doxorubicin-induced senescence,while its depletion caused P27-dependent senescence and proliferation arrest by activating LKB1-AMPK-mTOR cascade.Mechanistically,RBM4 competitively bound LKB1 to disrupt the LKB1/STRAD/MO25 heterotrimeric complex,subsequently recruiting the E3 ligase TRIM26 to LKB1,promoting LKB1 ubiquitination and degradation in nucleus.Therefore,such molecular process leads to bypassing senescence and sustaining cell proliferation through the activation of glutamine metabolism.Clinically,the ESCC patients with high RBM4 and low LKB1 have significantly worse overall survival than those with low RBM4 and high LKB1.The RBM4 high/LKB1 low expression confers increased sensitivity of ESCC cells to glutaminase inhibitor CB-839,providing a novel insight into mechanisms underlying the glutamine-dependency to improve the efficacy of glutamine inhibitors in ESCC therapeutics. | Lei Chen Wenjing Zhang Dan Chen Quan Yang Siwen Sun Zhenwei Dai Zhengzheng Li Xuemei Liang Chaoqun Chen Yuexia Jiao Lili Zhi Lianmei Zhao Jinrui Zhang Xuefeng Liu Jinyao Zhao Man Li Yang Wang Yangfan Qi | 2023 | Signal Transduction and Targeted Therapy2023,8,5: | 0 |
| 5 | Surface Photovoltage Studies of Moisture Sensitive Characteristic of Copper Tetrasulfonato-Phthalocyanine显示文摘The surface photovoltaic characteristic of copper tetrasulfonato- phthalocyanine (CuTsPc) in water vapor was studied by surface photovoltage spectroscopy (SPS). It was found that the adsorption of water vapor caused some marvelous changes of the photovoltaic response, that is, the SPS signal diminished rapidly and even reversed; the bands were widened and displayed a bathochromic shift. The dependence of surface photovoltage on the vapor pressure indicates that the reversed signal reaches to a maximum when the vapor pressure is 4.7×102 Pa. The time response velocity, reversibility, selectivity and reproducibili-ty were examined as well. All results obtained show that CuTsPc is of great significance in the manufacture of moisture sensitive devices.
