| 1 | CPSF30-L-mediated recognition of mRNA m^(6)A modification controls alternative polyadenylation of nitrate signaling-related gene transcripts in Arabidopsis显示文摘N6-methyladenosine(m^(6)A),a ubiquitous internal modification of eukaryotic mRNAs,plays a vital role in almost every aspect of mRNA metabolism.However,there is little evidence documenting the role of m^(6)A in regulating alternative polyadenylation(APA)in plants.APA is controlled by a large protein-RNA complex with many components,including CLEAVAGE AND POLYADENYLATION SPECIFICITY FACTOR30(CPSF30).In Arabidopsis,CPSF30 has two isoforms and the longer isoform(CPSF30-L)contains a YT512-B Homology(YTH)domain,which is unique to plants.In this study,we showed that CPSF30-L YTH domain binds to m^(6)A in v itro.In the cpsf30-2 mutant,the transcripts of many genes including several important nitrate signaling-related genes had shifts in polyadenylation sites that were correlated with m^(6)A peaks,indicating that these gene transcripts carrying m^(6)A tend to be regulated by APA.Wild-type CPSF30-L could rescue the defects in APA and nitrate metabolism in cpsf30-2,but m^(6)A-binding-defective mutants of CPSF30-L could not.Taken together,our results demonstrated that m^(6)A modification regulates APA in Arabidops is and revealed that the m^(6)A reader CPSF30-L affects nitrate signaling by controlling APA,shedding new light on the roles of the m^(6)A modification during RNA 3-end processing in nitrate metabolism. | Yiteng Hou Jing Sun Baixing Wu Yangyang Gao Hongbo Nie Zhentian Nie Shuxuan Quan Yong Wang Xiaofeng Cao Sisi Li | 2021 | Molecular Plant2021,14,4: | 12 |
| 2 | Magnetic field data processing methods of the China Seismo-Electromagnetic Satellite显示文摘The High Precision Magnetometer(HPM) on board the China Seismo-Electromagnetic Satellite(CSES) allows highly accurate measurement of the geomagnetic field; it includes FGM(Fluxgate Magnetometer) and CDSM(Coupled Dark State Magnetometer)probes. This article introduces the main processing method, algorithm, and processing procedure of the HPM data. First, the FGM and CDSM probes are calibrated according to ground sensor data. Then the FGM linear parameters can be corrected in orbit, by applying the absolute vector magnetic field correction algorithm from CDSM data. At the same time, the magnetic interference of the satellite is eliminated according to ground-satellite magnetic test results. Finally, according to the characteristics of the magnetic field direction in the low latitude region, the transformation matrix between FGM probe and star sensor is calibrated in orbit to determine the correct direction of the magnetic field. | 4 | Dust levitation and transport over the surface of the Moon显示文摘Exposed to space plasma and solar radiation, electrostatic potential may build up in the lunar regolith, leading to a wealth of dust phenomena, including levitation, oscillation, and transport over the surface. Based on plasma sheath theory,the global near-surface plasma environment is modeled, and the dynamics of charged dust are investigated. Results show that sub-micron sized dust particles can be levitated by the electric field over the surface, forming a dust belt that changes in position and thickness depending on the solar zenith angle. On the dayside of the Moon, stably levitated particles are about ten times smaller, and collect in a thinner belt closer to the surface than do those on the nightside. Although the size and charge of stably levitated dust particles are dependent on ambient plasma conditions, initial charge and velocity, which are closely related to the dynamics of dust particles including charging, oscillation, and damping, will determine whether, or not, a particle can attain stable levitation. Horizontal electrostatic dust transport near to the terminator region may lead to net deposition of dust from the dark into the sunlit hemisphere. | 5 | In vitro comparison of the hemocompatibility of diamond-like carbon and carbon nitride coatings with different atomic percentages of N显示文摘Carbon nitride (CNx) and diamond-like carbon (DLC) coatings were prepared by dc magnetron sputtering at room temperature. Different partial pressures of N2 were used to synthesize CNx to evaluate the relationship between the atomic percentage of nitrogen and hemocompatibility. Auger electron spectroscopy and atomic force microscopy indicated atomic percentages of N of 0.12 and 0.22 and that the CNx coatings were smooth. An in vitro study of the hemocompatibility of the coatings revealed that both CNx coatings had better anticoagulant properties and lower platelet adhesion than DLC. Compared with CN0.12, the CN0.22 coating showed longer dynamic clotting time (about 42 min), static clotting time (23.6 min) and recalcification time (45.6 s), as well as lower platelet adhesion (102 cells μm 2), aggregation, and activation. | 6 | A correlative denoising autoencoder to model social influence for top-N recommender system显示文摘In recent years,there are numerous works been proposed to leverage the techniques of deep learning to improve social-aware recommendation performance.In most cases,it requires a larger number of data to train a robust deep learning model,which contains a lot of parameters to fit training data.However,both data of user ratings and social networks are facing critical sparse problem,which makes it not easy to train a robust deep neural network model.Towards this problem,we propose a novel correlative denoising autoencoder(CoDAE)method by taking correlations between users with multiple roles into account to learn robust representations from sparse inputs of ratings and social networks for recommendation.We develop the CoDAE model by utilizing three separated autoencoders to learn user features with roles of rater,truster and trustee,respectively.Especially,on account of that each input unit of user vectors with roles of truster and trustee is corresponding to a particular user,we propose to utilize shared parameters to learn common information of the units that corresponding to same users.Moreover,we propose a related regularization term to learn correlations between user features that learnt by the three subnetworks of CoDAE model.We further conduct a series of experiments to evaluate the proposed method on two public datasets for Top-N recommendation task.The experimental results demonstrate that the proposed model outperforms state-of-the-art algorithms on rank-sensitive metrics of MAP and NDCG. | Yiteng PAN Fazhi HE Haiping YU | 2020 | Frontiers of Computer Science2020,14,3: | 3 |

| 10 | Therapeutic silencing miR-146b-5p improves cardiac remodeling in a porcine model of myocardial infarction by modulating the wound reparative phenotype显示文摘Fibrotic remodeling is an adverse consequence of immune response-driven phenotypic modulation of cardiac cells following myocardial infarction(Ml).MicroRNA-146b(miR-146b)is an active regulator of immunomodulation,but its function in the cardiac inflammatory cascade and its clinical implication in fibrotic remodeling following Ml remain largely unknown.Herein,miR-146b-5p was found to be upregulated in the infarcted myocardium of mice and the serum of myocardial ischemia patients.Gain-and loss-of-function experiments demonstrated that miR-146b-5p was a hypoxia-induced regulator that governed the pro-fibrotic phenotype transition of cardiac cells.Overexpression of miR-146b-5p activated fibroblast proliferation,migration,and fibroblast-to-myofibroblast transition,impaired endothelial cell function and stress survival,and disturbed macrophage paracrine signaling.Interestingly,the opposite effects were observed when miR-146b-5p expression was inhibited.Luciferase assays and rescue studies demonstrated that the miR-146b-5p target genes mediating the above phenotypic modulations included interleukin 1 receptor associated kinase 1(IRAKI)and carcinoembryonic antigen related cell adhesion molecule 1(CEACAM1).Local delivery of a miR-146b-5p antagomir significantly reduced fibrosis and cell death,and upregulated capillary and reparative macrophages in the infarcted myocardium to restore cardiac remodeling and function in both mouse and porcine Ml models.Local inhibition of miR-146b-5p may represent a novel therapeutic approach to treat cardiac fibrotic remodeling and dysfunction following Ml. | Yiteng Liao Hao Li Hao Cao Yun Dong Lei Gao Zhongmin Liu Junbo Ge Hongming Zhu | 2021 | Protein & Cell2021,12,3: | 1 |
