| 2 | Rapid reconstitution of NK1 cells after allogeneic transplantation is associated with a reduced incidence of graft-versus-host disease显示文摘The balance between immunostimulation and immunoregulation in T cell immunity is achieved by maintaining specific ratios of Th1, Th2, Th3 and Tr1 cells. Here, we investigate levels of type 1(IFN-gamma; NK1), type 2(IL-13; NK2), type 3(TGF-beta;NK3) and regulatory(IL-10; NKr) cytokines in peripheral blood to assess the cytokine profiles of natural killer(NK) cells following human allogeneic hematopoietic stem cell transplantation(allo-HSCT). NK2 and NK3 cell expansion was observed after allo-HSCT; levels of NKr cells reached donor levels at day 15, though levels of NK1 cells were consistently lower than donor levels until day 60 after allo-HSCT. Multivariate analysis showed that a higher level of NK1 cells by day 15 was associated with a lower overall risk of acute graft-versus-host disease(GVHD)(HR 0.157, P=0.010) as well as II-IV acute GVHD(HR0.260, P=0.059). Furthermore, higher levels of NK1 cells by day 15 were correlated with lower rates of cytomegalovirus(CMV)reactivation(HR 0.040, 0.005–0.348, P=0.003). | 3 | Platelet transfusion refractoriness after T-cell-replete haploidentical transplantation is associated with inferior clinical outcomes显示文摘Haploidentical stem cell transplantation (haplo-SCT) has been an alternative source of bone marrow for patients without human leukocyte antigen (HLA)-matched donors. The aim of this study was to investigate the relationships between platelet transfusion refractoriness (PTR) and clinical outcomes in the setting of haplo-SCT. Between May 2012 and March 2014, 345 patients who underwent unmanipulated haplo-SCT were retrospectively enrolled. PTR occurred in 20.6% of all patients. Patients in the PTR group experienced higher transplant-related mortality (TRM, 43.7% vs. 13.5%, P<0.001), lower overall survival (OS, 47.9%vs. 76.3%, P<0.001) and lower leukemia-free survival (LFS, 47.9% vs. 72.3%, P<0.001) compared to patients in the non-PTR group. The multivariate analysis showed that PTR was associated with TRM (P=0.002), LFS (P<0.001), and OS (P<0.001).The cumulative incidences of PTR in patients receiving >12 platelet (PLT) transfusions (third quartile of PLT transfusions) were higher than in patients receiving either >6 (second quartile) or >3 (first quartile) PLT transfusions (56.1% vs. 41.6% vs. 28.2%,respectively; P<0.001). The multivariate analysis also showed that PTR was associated with the number of PLT transfusions(P<0.001). | 4 | Improved clinical outcomes of rhG-CSF-mobilized blood and marrow haploidentical transplantation compared to propensity score-matched rhG-CSF-primed peripheral blood stem cell haploidentical transplantation:a multicenter study显示文摘The effects of haploidentical rhG-CSF-mobilized blood and marrow transplantation(HBMT) on hematological malignances are well established. Previous prospective single-center studies have demonstrated better survival after HBMT versus haploidentical rhG-CSF-mobilized peripheral blood stem cell transplantation(HPBSCT) for acute leukemia(AL) not in remission(NR) or in more than the second complete remission(>CR2). To test the hypothesis that HBMT is still superior to HPBSCT for patients with AL, multiple myeloma(MM), or non-Hodgkin lymphoma(NHL) in CR1/CR2 and for patients with chronic myeloid leukemia in the first and second chronic phase lacking a matched donor, we designed a propensity score method-based multicenter study.Hematopoietic recovery, acute graft-versus-host disease(aGVHD), and chronic GVHD were comparable between the HBMT group(n=168) and the HPBSCT group(n=42). No significant differences were found in non-relapse mortality rate(20.17%±3.58%and 27.24%±7.16%, P=0.18) or relapse rate(19.96%±3.72% and 28.49%±8.25%, P=0.32) between the HBMT group and the HPBSCT group. HBMT recipients had better overall survival(65.0%±4.2% and 54.2%±8.3%, P=0.037) and disease-free survival(59.9%±4.6% and 44.3%±8.7%, P=0.051). Multivariate analysis showed that HPBSCT was associated with poorer DFS(HR(95%CI), 1.639(0.995–2.699), P=0.052). | 5 | Response of Pinus taiwanensis growth to climate changes at its southern limit of Daiyun Mountain, China's Mainland Fujian Province显示文摘The Pinus taiwanensis trees of a Chinese endemic species form pure forests at infertile sites in humid subtropical China, which can aid soil reservation and ecological resiliency at such sites. Herein, we used dendrochronological methods to investigate the growth patterns and their relationship with climate by analyzing 158 cores of 79 P. taiwanensis trees at 4 sites in their southernmost distribution in China's Mainland at Daiyun Mountain in Fujian Province. Tree growths at sites with favorable conditions generally showed an age-related growth trend, which decreased from approximately 5 mm to 0.5 mm in about a century. Trees in moderately stressed environments established high growth in their juvenile periods but were highly susceptible to environmental stresses such as a sharp growth decline in the 1990 s. The temperature in February and the moisture in July are the major limiting factors for most of the tree growths, except for a few extremely stressed P. taiwanensis trees. The growth of the dwarf P. taiwanensis trees, with all ring diameters of the first 20 years less than 4 mm, is mainly limited by the dry climate in May. | 9 | The in situ observation of modelled sea ice drift characteristics in the Bohai Sea显示文摘Sea ice drift is mainly controlled by ocean currents, local wind, and internal ice stress. Information on sea ice motion, especially in situ synchronous observation of an ice velocity, a current velocity, and a wind speed, is of great significance to identify ice drift characteristics. A sea ice substitute, the so-called 'modelled ice', which is made by polypropylene material with a density similar to Bohai Sea ice, is used to complete a free drift experiment in the open sea. The trajectories of isolated modelled ice, currents and wind in the Bohai Sea during non-frozen and frozen periods are obtained. The results show that the currents play a major role while the wind plays a minor role in the free drift of isolated modelled ice when the wind is mild in the Bohai Sea. The modelled ice drift is significantly affected by the ocean current and wind based on the ice–current–wind relationship established by a multiple linear regression. The modelled ice velocity calculated by the multiple linear regression is close to that of the in situ observation, the magnitude of the error between the calculated and observed ice velocities is less than12.05%, and the velocity direction error is less than 6.21°. Thus, the ice velocity can be estimated based on the observed current velocity and wind speed when the in situ observed ice velocity is missing. And the modelled ice of same thickness with a smaller density is more sensitive to the current velocity and the wind speed changes. | 12 | Fabrication and characterization of a PAM modified PHBV/BG scaffold显示文摘In order to improve the bioactivity and mechanical strength of the scaffold used in bone repair simultaneously, a novel porous PAM-poly (β-hydroxybutyrate-co-β-hydroxyvalerate) (PHBV)/bioactive glass (BG) scaffold was prepared by photo-initiated polymerization. PAM was used to improve the hydrophilicity of PHBV matrix while the BG particles were added to increase the bioactivity and strength of the matrix synchronously. The grafted amide group and Si-O moieties from acrylamide and the added BG were confirmed by Fourier Transform Infrared Spectrometry (FTIR). The micromorphology of the scaffolds before and after grafting was observed by scanning electron microscopy (SEM). The resulting images demonstrate that the PAM-PHBV/BG scaffold has a well connected pore structure and appropriate pore size which may be convenient for cells to grow and discharge metabolites. The specific gravity method was used to evaluate the pore property of the scaffold and the result shows that the scaffold has an average porosity up to 82.0%. Mercury intrusion porosimetry (MIP) indicated that the pores of PAM-PHBV/BG scaffold were mainly distributed between 75 and 150 μm. The compressive strength test was adopted to evaluate the mechanical property of the scaffold. The result shows that the PAM-PHBV/BG scaffold has a relatively high compressive strength (0.91 MPa) when compared with the pure PHBV scaffold. Besides, the properties of the pure PHBV scaffold, PHBV/BG scaffold were also evaluated. For the purpose of monitoring and investigating molecular events in living cells at real-time, the surface plasmon resonance based cytosensor (SBCS) for cell cultur-ing and signal monitoring was established, and on the basis of it, the corresponding technology was also established by monitoring and analyzing SPR re-sponses induced in rat C6 glioma cells by phorbol 12-myristate 13-acetate (PMA). The SPR signals induced by PMA in living C6 cells were significantly different from those groups without cells. These re-sponses were strongly dependent on and saturable to the concentrations of PMA, and could be sup-pressed by the specific and potent PKC inhibitors, which indicated that the measured signal could be the reflection of the redistribution of intracellular components near the cell membrane triggered by the activation of PKC. This research provides a quantita-tive and non-invasive technique to study the spa-tio-temporal characteristics of the cellular effects in living cells at real-time.
