| 1 | Chromosome analysis of esophageal squamous cell carcinoma cell line KYSE 410-4 by repetitive multicolor fluorescence in situ hybridization显示文摘Chromosome aberrations are distinctive features of human malignant tumors. Analysis of chromosomal changes can illuminate the molecular mechanisms underlying the development and progression of cancer. To establish the technique of multicolor fluorescence in situ hybridization (M-FISH) for identifying chromosome aberrations in esophageal carcinoma cell line KYSE 410-4, four pools of 6-color whole-chromosome painting probes have been designed and hybridized on the same metaphase spread by four rounds of repetitive FISH. Repetitive 6-color M-FISH was successfully established and the cytogenetic abnormalities in KYSE 410-4 cells were characterized. Chromosome gains occurred at 2q, 3, 8, 17p, and X. An isochromosome 3q was visualized in the cell line, which might be one intermedi- ate mechanism leading to 3p losses and/or 3q gains. Furthermore, 16 structural arrangements were detected, including four derivative chromosomes. The rearrangement of the centromeric regions accounted for approximately 44% of all rearrangements. The results added a more complete and accurate information of the genetic alterations to the classical cytogenetic description of KYSE 410-4 and provided a detailed cytogenetic background data for appropriate use of the cell line. | 3 | Flavone from Zhongjiefeng(Herba Sarcandrae Glabrae) inhibits platelet apoptosis in immune-induced bone marrow failure through mitochondrial pathway显示文摘OBjECTIVE: To investigate the effect of Flavone from Zhongjiefeng(Herba Sarcandrae Glabrae) on the platelet number in immune-induced bone marrow failure(BMF) and its mechanism of mitochondrial apoptotic pathway.METHODS: Immune-induced BMF model, established in mice, was randomly divided into four groups: normal control group without BMF, BMF control group, cyclosporine(CSA) group and flavone group(n = 10 in each group). Mice were given0.027 g/kg cyclosporine or 0.2 g/kg flavone lavage daily in either the cyclosporine or flavone group respectively. Platelet count, mitochondrial transmembrane potential(ΔΨm), cytochrome C(Cyt C), phosphatidylserine(PS), changes of calcium ion(Ca^(2+)),and protein expression of mitochondrial apoptotic pathway including B-cell lymphoma-2(bcl-2) Homologous Antagonist-Killer Protein(Bak), bcl-2-associated X protein(Bax), caspase-3, caspase-8, and caspase-9 were examined and compared.RESULTS: Compared with the normal control group, the BMF group had significantly lower levels of platelet count, ΔΨm, and expressions of caspase family proteins as well as higher levels of Cyt C, PS,Ca^(2+), and expressions of Bak and Bax(all P < 0.05).Compared with the BMF group, the CSA and flavone groups had significantly higher ΔΨm and expressions of caspase family proteins(all P < 0.05)whereas the levels of Cyt C, PS, Ca^(2+), and expressions of Bak and Bax were reduced(all P < 0.05).More importantly, the flavone group had higher levels of Cyt C, Ca^(2+)and expressions of Bak and Bax compared with the CSA group(all P < 0.05), while the levels of PS and caspase family proteins were reduced(all P < 0.05).CONCLUSION: Flavone from Zhongjiefeng(Herba Sarcandrae Glabrae) significantly increases the platelet number and prevents its apoptosis through mitochondrial pathway. | Jiang Yiling Zheng Qin Zhang Aiping Cui Lele Xia Lemin Luo Meihong | 2017 | Journal of Traditional Chinese Medicine2017,37,5: | 2 |

| 5 | Regulatory variation within 3’UTR of STAT5A correlates with sudden cardiac death in Chinese populations显示文摘Definitive diagnosis to sudden cardiac death(SCD)is often challenging since the postmortem examination on SCD victims could hardly demonstrate an adequate cause of death.It is therefore important to uncover the inherited risk component to SCD.Signal transducer and activators of transcription 5 A(STAT5A)is a member of the STAT family and a transcription factor that is activated by many cell ligands and associated with various cardiovascular processes.In this study,we performed a systematic variant screening on the STAT5A to filter potential functional genetic variations.Based on the screening results,an insertion/deletion polymorphism(rs3833144)in 3’UTR of STAT5A was selected as the candidate variant.A total of 159 SCD cases and 668 SCD matched healthy controls was enrolled to perform a case-control study and evaluate the association between rs3833144 and SCD susceptibility in Chinese populations.Logistic regression analysis showed that the deletion allele of rs3833144 had significantly increased the SCD risk(odds ratio(OR)=1.54;95%confidence interval(CI)=1.18-2.01;P=0.000955).Further genotype-expression eQTL analysis showed that samples with deletion allele appeared to lower expression of STAT5A,and in silico prediction suggested the local 3 D structure changes of STAT5A mRNA caused by the variant.On the other hand,the bioinformatic analysis presented that promoters of RARA and PTGES3L-AARSD1 could interact with rs3833144,and eQTL analysis showed the higher expression of both genes in samples with deletion allele.Dual-luciferase activity assays also suggested the significant regulatory role of rs3833144 in gene transcription.Our current data thus suggested a possible involvement of rs3833144 to SCD predisposition in Chinese populations and rs3833144 with potential function roles may become a candidate marker for SCD diagnosis and prevention. | Huan Yu Yadong Guo Zhenzhen Yang Qing Zhang Jiabin Xu Qi Yang Yiling Qu Rui Tan Lijuan Li Yan He Chengtao Li Suhua Zhang Bin Luo Yuzhen Gao | 2022 | Forensic Sciences Research2022,7,4: | 0 |
| 6 | Enhancing effect of choline chloride-based deep eutectic solvents with polyols on the aqueous solubility of curcumin–insight from experiment and theoretical calculation显示文摘The development of green solvents for enhancing aqueous solubility of drug curcumin remains a challenge. This study explores the enhancing effect of deep eutectic solvents(DESs) on the aqueous solubility of curcumin(CUR) via experiment and theoretical calculation. Choline chloride-based DESs with polyols 1,2-propanediol(1,2-PDO), 1,3-propanediol, ethylene glycol, and glycerol as hydrogen bond donors were prepared and used as co-solvents. The CUR aqueous solubility increased with increasing the DESs content at temperature of 303.15-318.15 K, especially in aqueous ChCl/1,2-PDO(mole ratio 1:4) solutions. The positive apparent molar volume values and reduced density gradient analysis confirmed the existence of strong interactions between CUR and solvent. The van der Waals interactions and hydrogen bonding coexisted in DESs monomer retained the stability of DESs structure after introducing CUR. Moreover,the lower interaction energy of DESs…CUR system than that of the counterpart DESs further proved the strong interaction between CUR and DESs. | 8 | Maize-soybean relay cropping increases soybean yield synergistically by extending the post-anthesis leaf stay-green period and accelerating grain filling显示文摘Relay cropping of Poaceae and Fabaceae promotes high yield and land-use efficiency by allowing a double harvest.However,it is difficult to increase yield synergistically because of the reduced photosynthetic abilities of legume leaves under the shade of graminoids.Leaf photosynthetic capacity in relay cropping systems is associated with ecological niche differentiation and photosynthetic compensation after restoration of normal light.We conducted a field experiment in southwest China in 2020–2021 to evaluate the effects of three cropping patterns:maize–soybean relay cropping(IMS),monoculture maize(MM),and monoculture soybean(SS),and N application levels:no N application(NN:0 kg N ha^(−1)),reduced N(RN:180 kg N ha^(−1)),and conventional N(CN:240 kg N ha^(−1)).Compared to monocropping,relay cropping increased the stay-green traits of maize and soybean by 13%and 89%,respectively.Relay cropping prolonged the leaf stay-green duration in the maize and soybean lag phase by almost 4 and 8 days,respectively.Relay cropping maize(IM)increased the leaf area index(LAI)by 79.4%to 88.5%under NN and 55.5%to 148%under RN.Relay cropping soybean(IS)increased the LAI from 115%to 437%at days 40 to 50 after anthesis.IM increased yield by 65.6%.IS increased yield by 9.7%.HI and system yield were at their highest values under RN.In the relay cropping system,reduced N application extended green leaf duration,increased photosynthesis inside the canopy at multiple levels,ultimately increases soybean yield synergistically. | Yiling Li Ping Chen Zhidan Fu Kai Luo Ping Lin Chao Gao Shanshan Liu Tian Pu Taiwen Yong Wenyu Yang | 2023 | The Crop Journal2023,11,6: | 0 |
