2Genomic structure of metabotropic glutamate receptor 7 and comparison of genomic structures of extracellular domains of mGluR family显示文摘Metabotropic glutamate receptor 7, coupled with a chemical neurotransmitter L-glutamate, plays an important role in the development of many psychiatric and neurological disorders. To study the biological and genetic mechanism of the mGluR7-related diseases, a physical map covering the full-length mGluR7 genomic sequence has been constructed through seed clone screening and fingerprinting database searching. These BAC clones in the physical map have been sequenced with shotgun strategy and assembled by Phred-Phrap-Consed software; the error rate of the final genoniic sequence is less than 0.01%. mGluR7 spans 880 kb genoniic region, the GC content and repeat content of mGluR7 genoniic sequence are 38% and 37.5% respectively. mGluR7 has a typical 'house-keeping' promoter and consists of 11 exons, with introns ranging from 6 kb to 285 kb. mGluR7a and mGluR7b are two known alternatively splicing variants. 3Functional characterization of the NF-kB binding site in the human NOD2 promoter显示文摘Nucleotide-binding and oligomerization domain 2(NOD2),a member of the NOD protein family,plays an important role in innate immunity.In response to pathogen attack,NOD2 stimulates cytokine and defensin production by activating nuclear factor(NF)-kB,a key transcription factor responsible for mediating downstream reactions.However,the mechanism linking NOD2 regulation and NF-kB activation is poorly understood.Using bioinformatics,we found a completely preserved canonical NF-kB binding site in the NOD2 core promoter(216 to 225 bp)in both humans and chimpanzees.The functional role of this NF-kB binding site was investigated using the enhanced green fluorescent protein(EGFP)reporter system,site-directed mutagenesis,the NF-kB activation inhibitor(JSH-23)and the chromatin immunoprecipitation(ChIP)assay.The results show that the NF-kB binding site is critical for regulation of the NOD2 gene.Either deletion of the NF-kB binding elements within the NOD2 promoter or treatment with an NF-kB activation inhibitor could lead to a significant loss of NOD2 promoter activity as detected by reporter gene assay.The canonical NF-kB binding site was bound by NF-kB as determined by the ChIP method.Based on these results,we suggest a positive feedback regulation between NF-kB and NOD2,which may represent an efficient mechanism in response to pathogen invasion.Chaofeng Hu Liping Sun Yiling Hu Daxiang Lu Huadong Wang Suisheng Tang 2010Cellular & Molecular Immunology2010,7,4:3

5Pervasive hybridization during evolutionary radiation of Rhododendron subgenus Hymenanthes in mountains of southwest China显示文摘Radiations are especially important for generating species biodiversity in mountainous ecosystems.The contribution of hybridization to such radiations has rarely been examined.Here,we use extensive genomic data to test whether hybridization was involved in evolutionary radiation within Rhododendron subgenus Hymenanthes,whose members show strong geographic isolation in the mountains of southwest China.We sequenced genomes for 143 species of this subgenus and 93 species of four other subgenera,and found that Hymenanthes was monophyletic and radiated during the late Oligocene to middle Miocene.Widespread hybridization events were inferred within and between the identified clades and subclades.This suggests that hybridization occurred both early and late during diversification of subgenus Hymenan thes,although the extent to which hybridization,speciation through mixing-isolation-mixing or hybrid speciation,accelerated the diversification needs further exploration.Cycles of isolation and contact in such and other montane ecosystems may have together promoted species radiation through hybridization between diverging populations and species.Similar radiation processes may apply to other montane floras in this region and elsewhere.Yazhen Ma Xingxing Mao Ji Wang Lei Zhang Yuanzhong Jiang Yuying Geng Tao Ma Liming Cai Shuangquan Huang Pete Hollingsworth Kangshan Mao Minghui Kang Yiling Li Wenlu Yang Haolin Wu Yang Chen Charles C.Davis Nawal Shrestha Richard H.Ree Zhenxiang Xi Quanjun Hu Richard I.Milne Jianquan Liu 2022National Science Review2022,9,12:1

7Gefitinib and fostamatinib target EGFR and SYK to attenuate silicosis:a multi-omics study with drug exploration显示文摘Silicosis is the most prevalent and fatal occupational disease with no effective therapeutics,and currently used drugs cannot reverse the disease progress.Worse still,there are still challenges to be addressed to fully decipher the intricated pathogenesis.Thus,specifying the essential mechanisms and targets in silicosis progression then exploring anti-silicosis pharmacuticals are desperately needed.In this work,multi-omics atlas was constructed to depict the pivotal abnormalities of silicosis and develop targeted agents.By utilizing an unbiased and time-resolved analysis of the transcriptome,proteome and phosphoproteome of a silicosis mouse model,we have verified the significant differences in transcript,protein,kinase activity and signaling pathway level during silicosis progression,in which the importance of essential biological processes such as macrophage activation,chemotaxis,immune cell recruitment and chronic inflammation were emphasized.Notably,the phosphorylation of EGFR(p-EGFR)and SYK(pSYK)were identified as potential therapeutic targets in the progression of silicosis.To inhibit and validate these targets,we tested fostamatinib(targeting SYK)and Gefitinib(targeting EGFR),and both drugs effectively ameliorated pulmonary dysfunction and inhibited the progression of inflammation and fibrosis.Overall,our drug discovery with multi-omics approach provides novel and viable therapeutic strategies for the treatment of silicosis.Mingyao Wang Zhe Zhang Jiangfeng Liu Meiyue Song Tiantian Zhang Yiling Chen Huiyuan Hu Peiran Yang Bolun Li Xiaomin Song Junling Pang Yanjiang Xing Zhujie Cao Wenjun Guo Hao Yang Jing Wang Juntao Yang Chen Wang 2022Signal Transduction and Targeted Therapy2022,7,6:1

13Peridynamic Meso-scale Modeling for Degradation in Transverse Mechanical Properties of Composites with Micro-void Defects显示文摘The effect of micro-void on transverse stiffness and strength for fiber-reinforced composites subjected to load perpendicular to fiber is studied using the state-based peridynamic(PD)theory.The heterogeneous microstructure with micro-voids are discretized with irregular and non-uniform grids on mesoscale.The PD representative volume element with randomly distributed and sized micro-voids has been established with periodic boundary conditions.A parametric study was performed to evaluate the effects of void fraction and void size on transverse properties.It was found that the transverse modulus decreases with the void fraction in the range of 0.5–2%.A smaller void has a greater impact on the transverse modulus than a larger void under the same void content.The crack growth path for the multi-fiber model subjected to transverse tension was analyzed.The PD predictions show that cracks initiate at the interface between fiber and matrix,then deflect to the micro-voids nearby.The PD predictions match well with the analytical and experimental observations available in the literature.Jiayue Wang Yin Yu Zhongcheng Mu Yile Hu 2022Acta Mechanica Solida Sinica2022,35,5:0
14Clinical evaluation of 4D dynamic dose for thoracic tumor stereotactic body radiation therapy with variable parameters显示文摘Objective:To evaluate the dose uncertainty in stereotactic body radiation therapy induced by respiratory motion using a 4D dynamic dose(4DDD)reconstruction method.Methods:A retrospective analysis was conducted on five lung cancer patients who received static intensitymodulated radiation therapy.The 4DDD was constructed using beam delivery log files,four-dimensional computed tomography(4DCT)scans,and treatment plans.To evaluate the impact of respiratory motion,4DDD calculations were performed with 10 starting phases for each field.A total of 270 field doses were simulated and calculated.The differences between the cumulative volume histogram in whole-course treatment and the field doses'gamma passing rate(GPR)were compared.The correlations between plan complexity metrics and the dose deviation caused by respiratory motion were evaluated independently.The phase distributions of 398 subfields were calculated and evaluated for the influence of dose rate and breathing frequency.Results:The GPRs of all fields were different among various starting phases,with the highest range from 62.20% to 76.87% for 2 mm/3%GPR.The deviation of mean point dose was(5.42±5.21)%,and the deviation in the mean dose and D98% within the internal gross tumor volume were(0.97±0.71)% and(0.77±0.53)%,respectively.There was a significant correlation between the beam aperture-to-volume(BA2V)ratio and the average 2 mm/2%GPR(R?0.601,P<0.01).Lower dose rates led to a more homogeneous distribution of phases among subfields(t?44.100,P<0.001).Conclusions:Different beam starting phases had a limited impact on the overall treatment evaluation.However,the respiratory motion could be observed to induce dose deviations using the 4DDD reconstruction model,particularly for fields with small BA2V.Yiling Zeng Yu Chang Sheng Zhang Jun Han Hongyuan Liu Feng Xiao Bo Pang Bin Hu Junping Cheng Hong Quan Zhiyong Yang 2023Radiation Medicine and Protection2023,4,3:0
15Transcription-wide impact by RESCUE-induced off-target single-nucleotide variants in mammalian cells显示文摘RNA base editing is a promising tool in precise molecular therapy. Currently, there are two widely used RNA base editors, REPAIRand RESCUE. REPAIR only facilitates A-to-I conversions, while RESCUE performs both A-to-I and C-to-U conversions. Thus, RESCUEcan generate twice the number of mutations compared to REPAIR. However, transcription-wide impact due to RESCUE-induced offtarget single-nucleotide variants (SNVs) is not fully appreciated. Therefore, to determine the off-target effects of RESCUE-mediatedediting, we employed transcription-wide sequencing on cells edited by RESCUE. The SNVs showed different off-target effects onmRNA, circRNA, lncRNA, and miRNA expression patterns and their interacting networks. 16Care Bundles:Enhanced Recovery After Delivery显示文摘The care of obstetrics patients has improved dramatically over the past few decades.Unfortunately,rates of cesarean section remain high,and decreasing this rate requires close care and follow-up in the prenatal outpatient setting.Counseling regarding cesarean delivery and vaginal delivery is imperative.Opportunities still exist in helping patients objectively weigh the decision for cesarean delivery versus vaginal delivery.Additional developments have occurred in the intrapartum and the postpartum setting,with an aim to improve patient and neonatal outcomes.Changes have been implemented for patients undergoing cesarean delivery including preoperative and intraoperative treatment of pain,nausea,and vomiting,as well as postoperative care bundles that improve patient outcomes.Innovations have also occurred in the care of postpartum patients after vaginal delivery,again with improvements in patient outcomes.This article summarizes the current evidence,provides care recommendations,and identifies the next steps in improving obstetrics care.Ling-Qun-Hu Plato J.Lysandrou Rebecca Minehart Jing-Ping Wang Yun Xia Yiling Hu Brett Worly 2023Maternal-Fetal Medicine2023,5,3:0
17Adjuvant chemotherapy versus adjuvant concurrent chemoradiotherapy after radical surgery for early-stage cervical cancer:a randomized,non-inferiority,multicenter trial显示文摘We conducted a prospective study to assess the non-inferiority of adjuvant chemotherapy alone versus adjuvant concurrent chemoradiotherapy (CCRT) as an alternative strategy for patients with early-stage (FIGO 2009 stage IB–IIA) cervical cancer having risk factors after surgery. The condition was assessed in terms of prognosis, adverse effects, and quality of life. This randomized trial involved nine centers across China. Eligible patients were randomized to receive adjuvant chemotherapy or CCRT after surgery. The primary end-point was progression-free survival (PFS). From December 2012 to December 2014, 337 patients were subjected to randomization. Final analysis included 329 patients, including 165 in the adjuvant chemotherapy group and 164 in the adjuvant CCRT group. The median follow-up was 72.1 months. The three-year PFS rates were both 91.9%, and the five-year OS was 90.6% versus 90.0% in adjuvant chemotherapy and CCRT groups, respectively. No significant differences were observed in the PFS or OS between groups. The adjusted HR for PFS was 0.854 (95% confidence interval 0.415–1.757;P = 0.667) favoring adjuvant chemotherapy, excluding the predefined non-inferiority boundary of 1.9. The chemotherapy group showed a tendency toward good quality of life. In comparison with post-operative adjuvant CCRT, adjuvant chemotherapy treatment showed non-inferior efficacy in patients with early-stage cervical cancer having pathological risk factors. 18Enhanced energy-storage performance in BNT-based lead-free dielectric ceramics via introducing SrTi_(0.875)Nb_(0.1)O_(3)显示文摘Environmentally friendly lead-free ceramics capacitors,with outstanding power density,rapid charging/discharging rate,and superior stability,have been receiving increasing attention of late for their ability to meet the critical requirements of pulsed power devices in low-consumption systems.However,the relatively low energy storage capability must be urgently overcome.Herein,this work reports on leadfree SrTi_(0.875)Nb_(0.1)O_(3)(STN)replacement of(Bi_(0.47)La_(0.03)Na_(0.5))_(0.94)Ba_(0.06)TiO_(3)(BLNBT)ferroelectric ceramics with excellent energy storage performance.Improving relaxor behaviour and breakdown strength(Eb),decreasing grain size,and mitigating large polarization difference are conductive to the enhancement of comprehensive energy storage performances.The phase-field simulation methods are further analysized evolution process of electrical tree in the experimental breakdown.In particular,the 0.70BLNBT-0.30STN ceramic exhibit a large discharged energy density of 4.2 J/cm^(3) with an efficiency of 89.3%at room temperature under electric field of 380 kV/cm.Additionally,for practical applications,the BLNBT-based ceramics achieve a high power density(~62.3 MW/cm^(3))and fast discharged time(~148.8 ns)over broad temperature range(20-200℃).Therefore,this work can provide a simple and effective guideline paradigm for acquiring high-performance dielectric materials in low-consumption systems operating in a wide range of temperatures and long-term operations.Lukang Wu Luomen Tang Yizan Zhai Yiling Zhang Jianjian Sun Di Hu Zhongbin Pan Zhen Su Yang Zhang Jinjun Liu 2022Journal of Materiomics2022,8,3:0
