1Therapeutic potential of traditional Chinese medicine for the treatment of NAFLD:A promising drug Potentilla discolor Bunge显示文摘Nonalcoholic fatty liver disease(NAFLD)is characterized by excessive accumulation of hepatic lipids and metabolic stress-induced liver injury.There are currently no approved effective pharmacological treatments for NAFLD.Traditional Chinese medicine(TCM)has been used for centuries to treat patients with chronic liver diseases without clear disease types and mechanisms.More recently,TCM has been shown to have unique advantages in the treatment of NAFLD.We performed a systematic review of the medical literature published over the last two decades and found that many TCM formulas have been reported to be beneficial for the treatment of metabolic dysfunctions,including Potentilla discolor Bunge(PDB).PDB has a variety of active compounds,including flavonoids,terpenoids,organic acids,steroids and tannins.Many compounds have been shown to exhibit a series of beneficial effects for the treatment of NAFLD,including anti-oxidative and anti-inflammatory functions,improvement of lipid metabolism and reversal of insulin resistance.In this review,we summarize potential therapeutic effects of TCM formulas for the treatment of NAFLD,focusing on the medicinal properties of natural active compounds from PDB and their underlying mechanisms.We point out that PDB can be classified as a novel candidate for the treatment and prevention of NAFLD.Longshan Ji Qian Li Yong He Xin Zhang Zhenhua Zhou Yating Gao Miao Fang Zhuo Yu Robim M.Rodrigues Yueqiu Gao Man Li 2022Acta Pharmaceutica Sinica B2022,12,9:4

3Electrically driven single-photon sources显示文摘Single-photon sources are building blocks for photonic quantum information processes. Of the many single-photon generation schemes, electrically driven single-photon sources have the advantages of realizing monolithic integration of quantum light sources and detectors without optical filtering, thus greatly simplify the integrated quantum photonic circuits. Here, we review recent advances on electrically driven single-photon sources based on solid-state quantum emitters, such as semiconductor epitaxial quantum dots, colloidal quantum dots, carbon nanotubes, molecules, and defect states in diamond, SiC and layered semiconductors. In particular, the merits and drawbacks of each system are discussed. 10Sequentially releasing self-healing hydrogel fabricated with TGFβ3-microspheres and bFGF to facilitate rat alveolar bone defect repair显示文摘Resorption and loss of alveolar bone leads to oral dysfunction and loss of natural or implant teeth. Biomimetic delivery of growth factors based on stem cell recruitment and osteogenic differentiation, as the key steps in natural alveolar bone regenerative process, has been an area of intense research in recent years. A mesoporous self-healing hydrogel(DFH) with basic fibroblast growth factor(bFGF) entrapment and transforming growth factor β3(TGFβ3)-loaded chitosan microspheres(CMs) was developed. The formulation was optimized by multiple tests of self-healing, in-bottle inversion, SEM, rheological, swelling rate and in vitro degradation. In vitro tubule formation assays, cell migration assays, and osteogenic differentiation assays confirmed the ability of DFH to promote blood vessels, recruit stem cells, and promote osteogenic differentiation. The optimum DFH formula is 0.05 ml 4ArmPEG-DF(20%) added to 1 ml CsGlu(2%) containing bFGF(80 ng) and TGFβ3-microspheres(5 mg). The results of in vitro release studied by Elisa kit, indicated an 95% release of b FGF in7 d and long-term sustained release of TGFβ3. For alveolar defects rat models, the expression levels of CD29 and CD45, the bone volume fraction, trabecular number, and trabecular thickness of new bone monitored by Micro-CT in DFH treatment groups were significantly higher than others(*P < 0.05, vs Model). HE and Masson staining show the same results.In conclusion, DFH is a design of bionic alveolar remodelling microenvironment, that is in early time microvessels formed by b FGF provide nutritious to recruited endogenous stem cells, then TGFβ3 slowly released speed up the process of new bones formation to common facilitate rat alveolar defect repair. 11A Predictive Nomogram for Predicting Improved Clinical Outcome Probability in Patients with COVID-19 in Zhejiang Province,China显示文摘The aim of this research was to develop a quantitative method for clinicians to predict the probability of improved prognosis in patients with coronavirus disease 2019(COVID-19).Data on 104 patients admitted to hospital with laboratory-confirmed COVID-19 infection from 10 January 2020 to 26 February 2020 were collected.Clinical information and laboratory findings were collected and compared between the outcomes of improved patients and non-improved patients.The least absolute shrinkage and selection operator(LASSO)logistics regression model and two-way stepwise strategy in the multivariate logistics regression model were used to select prognostic factors for predicting clinical outcomes in COVID-19 patients.The concordance index(C-index)was used to assess the discrimination of the model,and internal validation was performed through bootstrap resampling.A novel predictive nomogram was constructed by incorporating these features.Of the 104 patients included in the study(median age 55 years),75(72.1%)had improved short-term outcomes,while 29(27.9%)showed no signs of improvement.There were numerous differences in clinical characteristics and laboratory findings between patients with improved outcomes and patients without improved outcomes.After a multi-step screening process,prognostic factors were selected and incorporated into the nomogram construction,including immunoglobulin A(IgA),C-reactive protein(CRP),creatine kinase(CK),acute physiology and chronic health evaluation II(APACHE II),and interaction between CK and APACHE II.The C-index of our model was 0.962(95%confidence interval(CI),0.931-0.993)and still reached a high value of 0.948 through bootstrapping validation.A predictive nomogram we further established showed close performance compared with the ideal model on the calibration plot and was clinically practical according to the decision curve and clinical impact curve.The nomogram we constructed is useful for clinicians to predict improved clinical outcome probability for each COVID-19 patient,which may facilitate personalized counselling and treatment.Jiaojiao Xie Ding Shi Mingyang Bao Xiaoyi Hu Wenrui Wu Jifang Sheng Kaijin Xu Qing Wang Jingjing Wu Kaicen Wang Daiqiong Fang Yating Li Lanjuan Li 2022Engineering2022,8,1:0
12Construction and evaluation of in-house methylation-sensitive SNaPshot system and three classification prediction models for identifying the tissue origin of body fluid显示文摘The identification of tissue origin of body fluid can provide clues and evidence for criminal case investigations.To establish an efficient method for identifying body fluid in forensic cases,eight novel body fluid-specific DNA methylation markers were selected in this study,and a multiplex single base extension reaction(SNaPshot)system for these markers was constructed for the identification of five common body fluids(venous blood,saliva,menstrual blood,vaginal fluid,and semen).The results indicated that the in-house system showed good species specificity,sensitivity,and ability to identify mixed biological samples.At the same time,an artificial body fluid prediction model and two machine learning prediction models based on the support vector machine(SVM)and random forest(RF)algorithms were constructed using previous research data,and these models were validated using the detection data obtained in this study(n=95).The accuracy of the prediction model based on experience was 95.79%;the prediction accuracy of the SVM prediction model was 100.00%for four kinds of body fluids except saliva(96.84%);and the prediction accuracy of the RF prediction model was 100.00%for all five kinds of body fluids.In conclusion,the in-house SNaPshot system and RF prediction model could achieve accurate tissue origin identification of body fluids.Yating FANG Man CHEN Bofeng ZHU 2023Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2023,24,9:0
13CircUCP2 promotes the tumor progression of non-small cell lungcancer through the miR-149/UCP2 pathway显示文摘Non-small cell lung cancer(NSCLC)is a highly lethal cancer,and better treatments are urgently needed.Many studies have implicated circular RNAs(circRNAs)in the progression of multiple malignant tumors.Nonetheless,the functions of circRNAs in NSCLC remain unclear.To study new targets for the treatment of NSCLC,circRNA expression profiling was performed on NSCLC tissues and para-carcinoma nonmalignant tissues.RNA was isolated and used for circRNA sequencing.Biological studies were performed in vitro and in vivo to determine the functions of circRNAs in NSCLC,including their functions in cell proliferation and migration.How circRNAs function in NSCLC was explored to clarify the underlying regulatory mechanisms.We found that circUCP2 was upregulated in NSCLC tissues compared with neighboring nonmalignant tissues.circUCP2 promoted the proliferation and metastasis of NSCLC cells.circUCP2 promoted NSCLC progression by sponging miR-149 and upregulating UCP2.The circUCP2/miR-149/UCP2 axis accelerates the progression of NSCLC,and circUCP2 may therefore be a novel diagnostic biomarker for the progression of NSCLC.WEI DU FANG YIN YATING ZHONG MINJIE LUO ZHEN WANG PENG LIN QING LIU HAN YANG 2023Oncology Research2023,31,6:0
14An endophytic fungus interacts with the defensinlike protein OsCAL1 to regulate cadmium allocation in rice显示文摘Defensin-like proteins are conserved in multicellular organisms and contribute to innate immune re-sponses against fungal pathogens.In rice,defensins play a novel role in regulating cadmium(Cd)efflux from the cytosol.However,whether the antifungal activity of defensins correlates with Cd-efflux function remains unknown.In this study,we isolated an endophytic Fusarium,designed Fo1o,by a comparative mi-crobiome analysis of rice plants grown in a paddy contaminated with Cd.Fo10 is tolerant to high levels of Cd,but is sensitive to the defensin-like protein OsCAL1,which mediates Cd efflux to the apoplast.We found that Fo10 symbiosis in rice is regulated by OsCAL1 dynamics,and Fo10 coordinates multiple plant pro-cesses,including Cd uptake,vacuolar sequestration,efflux to the environment,and formation of Fe pla-ques in the rhizosphere.These processes are dependent on the salicylic acid signaling pathway to keep Cd levels low in the cytosol of rice cells and to decrease Cd levels in rice grains without any yield penalty.Fo10 also plays a role in Cd tolerance in the poaceous crop maize and wheat,but has no observed effects in the eudicot plants Arabidopsis and tomato.Taken together,these findings provide insights into the mech-anistic basis underlying how afungal endophyte and host plant interact to control Cd accumulation in host plants by adapting defense responses to promotethe establishment ofa symbiosis that permits adaptation to high-Cd environments.Tianyu Gu Ziai Qi Yating Wang Siying Chen Jing Yan Huapeng Qiu Yanxuan Yu Zjun Fang Junmin Wang Jiming Gong 2024Molecular Plant2024,17,2:0
