1Telomerase Activity in Condyloma Acuminatum Tissue with Different HPV Types显示文摘Summary: The telomerse activity in condyloma acuminatum (CA) tissue with human papillomavirus (HPV) types of 6/11 and 16/18 was detected to investigate the function of telomerase in the occurrence, development and carcinogenesis of genital CA. Forty-two biopsies from patients with gennital CA and 30 control tissue samples were tested for telomerase activity, HPV presence and types. The telomerase activity was determined by modified telomerase repeat amplification protocol (TRAP) assay and HPV typing by polymerase chain reaction (PCR) with typing-specific primers. Results showed that HPV-DNA was negative and the expression rate of telomerase was 16.7 % in all normal skin samples. All CA samples were positive for HPV (6/11 type was found in 32 cases, 16/18 in 3 and mixed type in 7). Telomerase activity was detectable in all CA patients. The telomerase activity in CA of 16/18 type was apparently higher than in CA of 6/11 type. 2Extracellular histones stimulate collagen expression in vitro and promote liver fibrogenesis in a mouse model via the TLR4-MyD88 signaling pathway显示文摘BACKGROUND Liver fibrosis progressing to liver cirrhosis and hepatic carcinoma is very common and causes more than one million deaths annually.Fibrosis develops from recurrent liver injury but the molecular mechanisms are not fully understood.Recently,the TLR4-MyD88 signaling pathway has been reported to contribute to fibrosis.Extracellular histones are ligands of TLR4 but their roles in liver fibrosis have not been investigated.AIM To investigate the roles and potential mechanisms of extracellular histones in liver fibrosis.METHODS In vitro,LX2 human hepatic stellate cells(HSCs)were treated with histones in the presence or absence of non-anticoagulant heparin(NAHP)for neutralizing histones or TLR4-blocking antibody.The resultant cellular expression of collagen I was detected using western blotting and immunofluorescent staining.In vivo,the CCl4-induced liver fibrosis model was generated in male 6-week-old ICR mice and in TLR4 or MyD88 knockout and parental mice.Circulating histones were detected and the effect of NAHP was evaluated.RESULTS Extracellular histones strongly stimulated LX2 cells to produce collagen I.Histone-enhanced collagen expression was significantly reduced by NAHP and TLR4-blocking antibody.In CCl4-treated wild type mice,circulating histones were dramatically increased and maintained high levels during the duration of fibrosisinduction.Injection of NAHP not only reduced alanine aminotransferase and liver injury scores,but also significantly reduced fibrogenesis.Since the TLR4-blocking antibody reduced histone-enhanced collagen I production in HSC,the CCl4 model with TLR4 and MyD88 knockout mice was used to demonstrate the roles of the TLR4-MyD88 signaling pathway in CCl4-induced liver fibrosis.The levels of liver fibrosis were indeed significantly reduced in knockout mice compared to wild type parental mice.CONCLUSION Extracellular histones potentially enhance fibrogenesis via the TLR4–MyD88 signaling pathway and NAHP has therapeutic potential by detoxifying extracellular histones.Zhi Wang Zhen-Xing Cheng Simon T Abrams Zi-Qi Lin ED Yates Qian Yu Wei-Ping Yu Ping-Sheng Chen Cheng-Hock Toh Guo-Zheng Wang 2020World Journal of Gastroenterology2020,26,47:3

4Quantitative climatic reconstruction of the Last Glacial Maximum in China显示文摘Quantitative paleoclimatic reconstruction is crucial for understanding the operation and evolution of the global climate system.For example,a quantitative paleoclimatic reconstruction for the Last Glacial Maximum(18±2 ka14C,LGM)is fundamental to understanding the evolution of Earth’s climate during the last glacial-interglacial cycle.Previous quantitative palaeoclimate reconstructions in China are generally based on statistical comparison of modern pollen assemblages and modern climate data.These methods are based on the premise that vegetation-climate interactions remain the same through time,and implicitly assume that the interactions are independent of changes in seasonality and atmospheric CO2concentration.However,these assumptions may not always be valid,which may affect the reconstructions.Here,we present the results of a quantitative study of the LGM climate of China based on an improved inverse vegetation model which incorporates physiological processes combined with a new China Quaternary Pollen Database.The results indicate that during the LGM,mean annual temperature(ANNT),mean temperature of the coldest month(MTCO)and mean temperature of the warmest month in China were lower by^5.6±0.8,~11.0±1.6 and^2.6±0.9°C,respectively,compared to today,and that the changes in ANNT were mainly due to the decrease of MTCO.The ANNT decrease in southern China was^5.5±0.5°C.Mean annual precipitation was lower by^46.3±17.8 mm compared to today and was especially low in northern China(~51.2±21.4 mm)due to the decrease in summer rainfall.Comparison of our results with recent outputs from paleoclimatic modelling reveals that while the latter are broadly consistent with our estimated changes in mean annual climatic parameters,there are substantial differences in the seasonal climatic parameters.Our results highlight the crucial importance of developing seasonal simulation on paleoclimatic models,as well as the need to improve the quality of paleoclimatic reconstructions based on proxy records from geological archives.Haibin WU Qin LI Yanyan YU Aizhi SUN Yating LIN Wenqi JIANG Yunli LUO 2019Science China Earth Sciences2019,62,8:3

8Electrically driven single-photon sources显示文摘Single-photon sources are building blocks for photonic quantum information processes. Of the many single-photon generation schemes, electrically driven single-photon sources have the advantages of realizing monolithic integration of quantum light sources and detectors without optical filtering, thus greatly simplify the integrated quantum photonic circuits. Here, we review recent advances on electrically driven single-photon sources based on solid-state quantum emitters, such as semiconductor epitaxial quantum dots, colloidal quantum dots, carbon nanotubes, molecules, and defect states in diamond, SiC and layered semiconductors. In particular, the merits and drawbacks of each system are discussed.
