| 1 | Genetic analysis of 'all-fish' growth hormone gene transferred carp (Cyprinus carpio L.) and its F_1 generation显示文摘Recornbinant 'all-fish' growth hormone gene (GH) was microinjected into the fertilized eggs of carp. A comparison between the growth traits of transgenics and non-transgenics was carried out, and the transgenic individuals with significant 'fast-growing' effect were successfully gained. A comparison on the reproductivities was also given out between the transgenics and their non-transgenic siblings, and showed that the reproductive capacity of transgenics was substantially equivalent to those of the non-transgenics. On the other hand, the genetic separation and the characteristic distribution of the FI generation were genetically analyzed, which gave solid evidence for the hypothesis that 2-3 chromosomes are integrated with trans-gene. In addition, the distinct biological effects for multi-site-integrated transgenes were further discussed. | 2 | Progress in the evaluation of transgenic fish for possible ecological risk and its containment strategies显示文摘Genetically improved transgenic fish possess many beneficial economic traits; however, the commer-cial aquaculture of transgenic fish has not been performed till date. One of the major reasons for this is the possible ecological risk associated with the escape or release of the transgenic fish. Using a growth hormone transgenic fish with rapid growth characteristics as a subject, this paper analyzes the following: the essence of the potential ecological risks posed by transgenic fish; ecological risk in the current situation due to transgenic fish via one-factor phenotypic and fitness analysis, and mathe-matical model deduction. Then, it expounds new ideas and the latest findings using an artificially simulated ecosystem for the evaluation of the ecological risks posed by transgenic fish. Further, the study comments on the strategies and principles of controlling these ecological risks by using a trip-loid approach. | 3 | Rapid growth cost in “all-fish” growth hormone gene transgenic carp:Reduced critical swimming speed显示文摘Evidence has accumulated that there is a trade-off between benefits and costs associated with rapid growth. A trade-off between growth rates and critical swimming speed (Ucrit) had been also reported to be common in teleost fish. We hypothesize that growth acceleration in the F3 generation of 'all-fish' growth hormone gene (GH) transgenic common carp (Cyprinus carpio L.) would reduce the swimming abilities. Growth and swimming performance between transgenic fish and non-transgenic controls were compared. The results showed that transgenic fish had a mean body weight 1.4―1.9-fold heavier,and a mean specific growth rate (SGR) value 6%―10% higher than the controls. Transgenic fish,however,had a mean absolute Ucrit (cm/s) value 22% or mean relative Ucrit (BL/s) value 24% lower than the controls. It suggested that fast-growing 'all-fish' GH-transgenic carp were inferior swimmers. | 4 | Rapid growth and sterility of growth hormone gene transgenic triploid carp显示文摘Triploid carp(100%)with 150(3n=150)chromosomes were obtained by crossing the females of improved tetraploid hybrids(♀, 4n=200)of red crucian carp(♀)×common carp(♂)with the males of diploid yellow river carp(♂,2n=100).The crosses yielded transgenic triploid carp(positive triploid fish,44.2%of the progeny)and non-transgenic triploid carp(negative triploid fish). | 7 | Cloning and expression analysis in mature individuals of two chicken type-Ⅱ GnRH (cGnRH-Ⅱ) genes in common carp (Cyprinus carpio)显示文摘Gonadotropin-releasing hormone (GnRH) is a conservative neurodecapeptide fam-ily, which plays a crucial role in regulating the gonad development and in controlling the final sexual maturation in vertebrate. Two differing cGnRH-II cDNAs of common carp, namely cGnRH-II cDNA1 and cDNA2, were firstly cloned from the brain by rapid amplification of cDNA end (RACE) and reverse transcription- polymerase chain reaction (RT-PCR). The length of cGnRH-II cDNA1 and cDNA2 was 622 and 578 base pairs (bp), respectively. The cGnRH-II pre-cursors encoded by two cDNAs consisted of 86 amino acids, including a signal peptide, cGnRH-II decapeptide and a GnRH-associated peptide (GAP) linked by a Gly-Lys-Arg proteolytic site. The results of intron trapping and Southern blot showed that two differing cGnRH-II genes in common carp genome were further identified, and that two genes might exist as a single copy. The multi-gene coding of common carp cGnRH-II gene offered novel evidence for gene duplica-tion hypothesis. Using semi-quantitative RT-PCR, expression and relative expression levels of cGnRH-II genes were detected in five dissected brain regions, pituitary and gonad of common carp. With the exception of no mRNA2 in ovary, two cGnRH-II genes could be expressed in all the detected tissues. However, expression levels showed an apparent difference in different brain regions, pituitary and gonad. According to the expression characterization of cGnRH-II genes in brain areas, it was presumed that cGnRH-II might mainly work as the neurotransmitter and neuromodulator and also operate in the regulation for the GnRH releasing. | 8 | Accumulation and Output of Heavy Metals by the Invasive Plant Spartina alterniflora in a Coastal Salt Marsh显示文摘Spartina alterniflora is a foreign introduced species and has far-reaching effects on salt marsh ecosystems, particularly on the biogeochemical cycle of heavy metals. To ascertain whether the invasive plant Spartina alterniflora Loisel is a source of metals in the environment, we determined the bimonthly concentrations of heavy metals, chromium(Cr), lead(Pb), copper(Cu), zinc(Zn),and manganese(Mn), in the roots, stems, and leaves of S. alterniflora from a typical semidiurnal tidal zone in the coastal area of northern Jiangsu Province, China. Based on the measurements, we calculated annual metal primary accumulation and output. To calculate the annual output of heavy metals from S. alterniflora, a new method that calculates the annual rate of biomass loss and decomposition was developed. The annual primary accumulation of Cr, Pb, Cu, Zn, and Mn was 19.08, 84.19, 63.74, 442.58, and774.66 mg m^(-2), respectively, and the annual output from S. alterniflora to the surrounding environment was 4.01, 18.09, 14.00, 97.11,and 164.28 mg m^(-2), respectively. Spartina alterniflora only provides temporary storage, and its absorption of heavy metals could be used to remediate contaminated soil and for phytomining. The heavy metals released by S. alterniflora to the environment cannot be ignored; thus, S. alterniflora should be considered a source of metal contamination. | 10 | Transcriptome analysis reveals key differentially expressed genes involved in wheat grain development显示文摘Wheat seed development is an important physiological process of seed maturation and directly affects wheat yield and quality. In this study, we performed dynamic transcriptome microarray analysis of an elite Chinese bread wheat cultivar(Jimai 20) during grain development using the Gene Chip Wheat Genome Array. Grain morphology and scanning electron microscope observations showed that the period of 11–15 days post-anthesis(DPA) was a key stage for the synthesis and accumulation of seed starch. Genome-wide transcriptional profiling and significance analysis of microarrays revealed that the period from 11 to 15 DPA was more important than the 15–20 DPA stage for the synthesis and accumulation of nutritive reserves.Series test of cluster analysis of differential genes revealed five statistically significant gene expression profiles. Gene ontology annotation and enrichment analysis gave further information about differentially expressed genes, and Map Man analysis revealed expression changes within functional groups during seed development. Metabolic pathway network analysis showed that major and minor metabolic pathways regulate one another to ensure regular seed development and nutritive reserve accumulation. We performed gene co-expression network analysis to identify genes that play vital roles in seed development and identified several key genes involved in important metabolic pathways. The transcriptional expression of eight key genes involved in starch and protein synthesis and stress defense was further validated by q RT-PCR. | 11 | Oleic Acid and Eicosapentaenoic Acid Reverse Palmitic Acid-induced Insulin Resistance in Human HepG2 Cells via the Reactive Oxygen Species/JUN Pathway显示文摘Oleic acid(OA),a monounsaturated fatty acid(MUFA),has previously been shown to reverse saturated fatty acid palmitic acid(PA)-induced hepatic insulin resistance(IR).However,its underlying molecular mechanism is unclear.In addition,previous studies have shown that eicosapentaenoic acid(EPA),aω-3 polyunsaturated fatty acid(PUFA),reverses PA-induced muscle IR,but whether EPA plays the same role in hepatic IR and its possible mechanism involved need to be further clarified.Here,we confirmed that EPA reversed PA-induced IR in HepG2 cells and compared the proteomic changes in HepG2 cells after treatment with different free fatty acids(FFAs).A total of 234 proteins were determined to be differentially expressed after PA+OA treatment.Their functions were mainly related to responses to stress and endogenous stimuli,lipid metabolic process,and protein binding.For PA+EPA treatment,the PA-induced expression changes of 1326 proteins could be reversed by EPA,415 of which were mitochondrial proteins,with most of the functional proteins involved in oxidative phosphorylation(OXPHOS)and tricarboxylic acid(TCA)cycle.Mechanistic studies revealed that the protein encoded by JUN and reactive oxygen species(ROS)play a role in OA-and EPA-reversed PA-induced IR,respectively.EPA and OA alleviated PA-induced abnormal adenosine triphosphate(ATP)production,ROS generation,and calcium(Ca^(2+))content.Importantly,H_(2)O_(2)-activated production of ROS increased the protein expression of JUN,further resulting in IR in HepG2 cells.Taken together,we demonstrate that ROS/JUN is a common response pathway employed by HepG2 cells toward FFA-regulated IR. | Yaping Sun Jifeng Wang Xiaojing Guo Nali Zhu Lili Niu Xiang Ding Zhensheng Xie Xiulan Chen Fuquan Yang | 2021 | Genomics, Proteomics & Bioinformatics2021,19,5: | 6 |
| 12 | Highly efficient Zr doped-TiO_2/glass fiber photocatalyst and its performance in formaldehyde removal under visible light显示文摘Zr-doped-TiO_2 loaded glass fiber(ZT/GF) composite photocatalysts with different Zr/Ti ratios were prepared with a sol–gel process. Zr4+can replace Ti4+in the TiO_2 lattice, which is conducive to forming the anatase phase and reducing the calcination temperature. The glass fiber carrier was responsible for better dispersion and loading of Zr-doped-TiO_2 particles, improving the applicability of the Zr-doped-TiO_2. The ZT/GF photocatalysts were characterized by X-ray diffraction(XRD), scanning electron microscope(SEM), Fourier transform infrared spectroscopy(FT-IR), ultraviolet–visible spectroscopy(UV–vis) and Barrett–Joyner–Halenda(BJH). The performance of photocatalysts with different loading was evaluated in formaldehyde degradation under visible light at room temperature. ZT/GF0.2exhibited the highest activity, with a formaldehyde removal rate as high as 95.14% being observed, which is better than that of the photocatalyst particles alone. | 14 | Neural Stem Cells Transfected with Leukemia Inhibitory Factor Promote Neuroprotection in a Rat Model of Cerebral Ischemia显示文摘Leukemia inhibitory factor(LIF) contributes to the neuroprotection by neural stem cells(NSCs) after ischemic stroke. Our aim was to explore whether LIFtransfected NSCs(LIF-NSCs) can ameliorate brain injury and promote neuroprotection in a rat model of cerebral ischemia. To accomplish this goal, we transfected NSCs with a lentivirus carrying the LIF gene to stably overexpress LIF. The LIF-NSCs reduced caspase 3 activation under conditions of oxygen-glucose deprivation in vitro.Transient cerebral ischemia was induced in rats by 2 h of middle cerebral artery occlusion(MCAo), and LIF-NSCs were intravenously injected at 6 h post-ischemia. LIF-NSC treatment reduced the infarction volume and improved neurological recovery. Moreover, LIF-NSCs improved glial cell regeneration and ameliorated white matter injuryin the MCAo rats. | 15 | Time course of foreign gene integration and expression in transgenic fish embryos显示文摘Using a nuclear transplantation approach, the integration and expression of the green fluorescent protein (GFP) gene in the embryogenesis of transgenic loach (Misgurnus anguillicaudatus Cantor) have been studied. The GFP gene expression is first observed at the gastrula stage, which is consistent with the initiation of cell differentiation of fish embryos. The time course of the foreign gene expression is correlated with the regulatory sequences. The expression efficiency also depends on the gene configuration: the expression of pre-integrating circular plasmid at early embryos is higher than that of the linear plasmid. The integration of the GFP gene is first detected at the blastula stage and lasts for quite a long period. | 19 | High thermal stability Cu_(2)O@OZrP micro-nano hybrids for melt-spun excellent antibacterial activity polyester fibers显示文摘Public safety incidents caused by bacterial infections have attracted widespread attention towards antibacterial textiles(fibers,fabrics,etc.).Nevertheless,it is still challenging to efficiently load inorganic nano-antibacterial materials in polymer fibers.In this work,zirconium phosphate(ZrP,layered micro-nano materials)was utilized as a micro-nano carrier.The octadecyl triphenyl phosphonium bromide(OTP)was intercalated between the ZrP sheets by the ion exchange method to improve the carrier-polymer compatibility and the antibacterial performance.Through in-situ chemical reduction,the ultra-small nano-sized cuprous oxide(Cu_(2)O<5 nm)was loaded on the outer surface of ZrP to realize the uniform and stable dispersion of the Cu_(2)O on the carrier and improve the antibacterial performance.The ZrP nanosheets loaded with Cu_(2)O and OTP(Cu_(2)O@OZrP)had excellent antibacterial properties,and the antibacterial rate against E.coli,S.aureus and C.albicans was more than 99%.The intercalation amount of OTP in Cu_(2)O@OZrP can reach 16%,and the thermal stability was excellent and a significant increase in the Zeta potential.Indeed,the decomposition temperature was greater than 350℃,which was suitable for high-temperature melt processing of polymers.Consequently,we prepared PET/Cu_(2)O@OZrP fibers using polyethylene glycol terephthalate(PET),which accounts for 70%of the total chemical fibers,as the fiber matrix.PET/Cu_(2)O@OZrP fibers exhibited excellent mechanical property and antibacterial performance when the content of Cu_(2)O@OZrP was only 0.2%.The antibacterial rate against five types of bacteria including super bacteria(MRSA,VRE)was more than 99%. | Jialiang Zhou Yaping Wang Weinan Pan Hengxue Xiang Peng Li Zhe Zhou Meifang Zhu | 2021 | Journal of Materials Science & Technology2021,,22: | 4 |
| 20 | Acute and chronic un-ionized ammonia toxicity to ‘all-fish’ growth hormone transgenic common carp(Cyprinus carpio L.)显示文摘Ammonia is toxic to fish in natural and artificial waters.We evaluated the acute(96 h) and chronic(21 d) toxicity of un-ionized ammonia to GH transgenic common carp(Cyprinus carpio L.) and non-transgenic common carp using a static-renewal bioassay.The 24,48,72 and 96 h median lethal concentrations(LC50) of un-ionized ammonia were slightly lower in transgenic carp(2.64,2.44,2.28 and 2.16 mg N/L,respectively) than in non-transgenic carp(2.70,2.64,2.52 and 2.33 mg N/L,respectively).Similarly,the median lethal time(LT50) was significantly shorter for transgenic carp(1.41,7.91 and 117.42 h) than for non-transgenic common carp(2.53,14.06 and 150.44 h) following exposure to 3.86,3.29,or 2.09 mg N/L,respectively.Moreover,the mortality of transgenic carp was significantly higher than that of non-transgenic carp at all un-ionized ammonia concentrations((0.91 ± 0.12),(0.48 ± 0.06) and(0.12 ± 0.01) mg N/L) during the 21 d chronic toxicity test.Our results suggest that GH transgenic carp are less tolerant of unionized ammonia than non-transgenic carp.Our data are useful for evaluating potential environmental risk,optimizing stocking density in intensive aquaculture and establishing water quality criteria for ammonia in aquaculture. | GUAN Bo HU Wei ZHANG TangLin DUAN Ming LI DeLiang WANG YaPing ZHU ZuoYan | 2010 | Chinese Science Bulletin2010,55,35: | 4 |
