| 3 | HrcQ is necessary for Xanthomonas oryzae pv. oryzae HR-induction in non-host tobacco and pathogenicity in host rice显示文摘Bacterial blight, caused by Xanthomonas oryzae pv. oryzae(Xoo), is one of the most destructive diseases of rice(Oryza sativa L.) worldwide. The type III secretion system(T3SS) of Xoo, encoded by the hrp(hypersensitive response and pathogenicity) genes, plays critical roles in conferring pathogenicity in host rice and triggering a hypersensitive response(HR) in non-host plants. To investigate the major genes conferring the pathogenicity and avirulence of Xoo, we previously constructed a random Tn5-insertion mutant library of Xoo strain PXO99A. We report here the isolation and characterization of a Tn5-insertion mutant PXM69. Tn5-insertion mutants were screened on indica rice JG30, which is highly susceptible to PXO99A, by leaf-cutting inoculation.Four mutants with reduced virulence were obtained after two rounds of screening. Among them, the mutant PXM69 had completely lost virulence to the rice host and ability to elicit HR in non-host tobacco. Southern blotting analysis showed a single copy of a Tn5-insertion in the genome of PXM69. PCR walking and sequencing analysis revealed that the Tn5 transposon was inserted at nucleotide position 70,192–70,201 in the genome of PXO99A, disrupting the type III hrc(hrp-conserved) gene hrcQ, the first gene in the D operon of the hrp cluster in Xoo. To confirm the relationship between the Tn5-insertion and the avirulence phenotype of PXM69, we used the marker exchange mutagenesis to create a PXO99Amutant, ΔhrcQ::KAN, in which the hrcQ was disrupted by a kanamycin-encoding gene cassette at the same site as that of the Tn5-insertion. ΔhrcQ::KAN showed the same phenotype as mutant PXM69. Reintroduction of the wild-type hrcQ gene partially complemented the pathogenic function of PXM69. RT-PCR and cellulase secretion assays showed that the Tn5-disruption of hrcQ did not affect transcription of downstream genes in the D operon and function of the type II secretion system. | 14 | RAISE:A Resilient Anonymous Information Sharing Environment显示文摘With the widespread application of cloud computing and network virtualization technologies,more and more enterprise applications are directly deployed in the cloud.However,the traditional TCP/IP network transmission model does not fully consider the information security issues caused by the uncontrollable internet environment.Network security communication solutions represented by encrypted virtual private networks(VPN)are facing multiple security threats.In fact,during the communication process,the user application needs to protect not only the content of the communication but also the behavior of the communication,such as the communication relationship,the communication protocol,and so on.Inspired by blockchain and software-defined networking technology,this paper proposes a resilient anonymous information sharing environment,RAISE.The RAISE system consists of user agents,a core switching network and a control cluster based on a consortium blockchain.User agents are responsible for segmenting,encrypting,and encapsulating user traffic.The core switching network forwards user traffic according to the rules issued by the controller,and the controller dynamically calculates the forwarding rules according to the security policy.Different from onion routing technology,RAISE adopts the controller to replace the onion routing model,which effectively overcomes the uncontrollability of nodes.The dispersed computing model is introduced to replace the TCP/IP pipeline transmission models,which overcomes the problems of anti-tracking and traffic hijacking that cannot be solved by VPNs.We propose a blockchain control plane framework,design the desired consensus algorithmand deploy a RAISE systemconsisting of 150 nodes in an internet environment.The experimental results show that the use of blockchain technology can effectively improve the reliability and security of the control plane.While maintaining high-performance network transmission,it further provides network communication security. | Ning Hu Ling Liu Xin Liu Kaijun Wu Yue Zhao | 2023 | Computer Modeling in Engineering & Sciences2023,137,12: | 0 |

Genome editing tools like transcriptional activator-like effector nucleases and clustered regularlyinterspaced short palindromic repeats (CRISPR)-associated endonuclease Cas9 (CRISPR/Cas9) haveeffectively overcome limitations of the conventional breeding methods and are being widely accepted forimprovement of crops. Among the genome editing tools, the CRISPR/Cas9 system has emerged as themost powerful tool of genome editing because of its efficiency, amicability, flexibility, low cost andadaptability. Accumulated evidences indicate that genome editing has great potential in improving thedisease resistance in crop plants. | 17 | Effects of snow absence on available N pools and enzyme activities within soil aggregates in a spruce forest on the eastern Tibetan Plateau显示文摘Winter climate change has great potential to affect the functioning of terrestrial ecosystems.In particular,increased soil frost associated with reduced insulating snow cover may impact the soil nitrogen(N)dynamics in cold ecosystems,but little is known about the variability of these effects among the soil aggregates.A snow manipulation experiment was conducted to investigate the effects of snow absence on N cycling within soil aggregates in a spruce forest on the eastern Tibetan Plateau of China.The extractable soil available N(ammonium and nitrate),net N mineralization rate,and N cycling-related enzyme activities(urease,nitrate reductase,and nitrite reductase)were measured in large macroaggregate(>2 mm),small macroaggregate(0.25–2 mm),and microaggregate(<0.25 mm)during the early thawing period in the years of 2016 and 2017.Snow absence increased soil N availabilities and nitrite reductase activity in microaggregate,but did not affect net N mineralization rate,urease or nitrate reductase activities in any of the aggregate fractions.Regardless of snow manipulations,both soil inorganic N and nitrate reductase were higher in small macroaggregate than in the other two fractions.The effect of aggregate size and sampling year was significant on soil mineral N,net N mineralization rate,and nitrite reductase activity.Our results indicated that snow cover change exerts the largest impact on soil N cycling within microaggregate,and its effect is dependent on winter conditions(e.g.,snow cover and temperature).Such findings have important implications for soil N cycling in snow-covered subalpine forests experiencing pronounced winter climate change. | ZhijieLi Rudiger Reichel Zimin Li Kaijun Yang Li Zhang Bo Tan Rui Yin Kerui Zhao Zhenfeng Xu | 2022 | Soil Ecology Letters2022,4,4: | 0 |
