| 1 | Differential regulation of survivin by p53 contributes to cell cycle dependent apoptosis显示文摘Recent studies indicate that cell-cycle checkpoints are tightly correlated with the regulation of apoptosis, in which p53 plays an important role. Our present works show that the expression of E6/E7 oncogenes of human papillomavirus in HeLa cells is inhibited in the presence of anti-tumor reagent tripchlorolide (TC), which results in the up-regulation of p53 in HeLa cells. Interestingly, under the same TC-treatment, the cells at the early S-phase are more susceptible to apoptosis than those at the middle S-phase although p53 protein is stabilized to the same level in both situations. Significant difference is exhibited between the two specified expression profiles. Further analysis demonstrates that anti-apoptotic gene survivin is up-regulated by p53 in the TC-treated middle-S cells, whereas it is down-regulated by p53 in the TC-treated early-S cells. | 2 | Expression and localization of Smadl, Smad2 and Smad4 proteins in rat testis during postnatal development显示文摘Aim:To study the expression and regulation of Smad1,Smad2 and Smad4 proteins(intracellular signaling molecules of transforming growth factor-β family)in rat testis during postnatal development.Methods:The whole testes were collected from SD rats aged 3,7,14,28 and 90(adults)days.The cellular localization and developmental changes were examined by immunohistochemistry ABC method with the glucose oxidase-DAB-nickel enhancement technique.Quantitative analysis of the immunostaining was made by the image analysis system.The Smads proteins coexistence in the adult rat testis was tested by the double immune staining for CD14-Smad4 and Smad2-Smad4.The protein expression of Smad during rat testicular development was examined by means of Western blots.Results: Smadl,Smad2 and Smad4 were present throughout testicular development.The immunostaining of Smad1 and Smad2 were present in spermatogenic cells.A positive immunoreactivity was located at the cytoplasm,but the nucleus was negative,Smadl was immunolocalized at the d14,d28 and adult testes,while Smad2,at the d7,d14,d28 and adult testis.There was positive immunoreaction in the Sertoli cells and Leydig cells as well.The immunolocalization of Smad4 was exclusively at the cytoplasm of Leydig cells and the nuclei were negative throughout the testicular development.No expression was detected in the germ cells.The results of image and statistical analysis showed that generally the expression of Smad1,Smad2 and Smad4 in the testis tended to increase gradually with the growth of the rat.Conclusion;The present data provide direct evidences for the molecular mechanism of TGF-β action in rat testes during postnatal development and spermatogenesis. | JingHU Yuan-QiangZHANG Xin-PingLIU Rui-AnWANG YanJIN Ruo-JunXU | 2003 | Asian Journal of Andrology2003,5,1: | 10 |
| 3 | Preparation of nano-sized cerium and titanium pyrophosphates via solid-state reaction at room temperature显示文摘Nano-sized cerium-titanium pyrophosphates Ce1-xTixP2O7 (with x = 0, 0.2, 0.5, 0.7, 0.9, and 1.0) were obtained by grinding a mixture of Ce(SO4)2·4H2O, Ti(SO4)2, and Na4P2O7·10H2O in the presence of surfactant PEG-400 at room temperature, washing the mixture with water to remove soluble inorganic salts, and drying at 100°C. The products and their calcined samples were characterized using ultraviolet-visible spectroscopy (UV-vis), thermogravimetry and differential thermal analyses (TG/DTA), X-ray powder diffraction (XRD), and transmission electron microscopy (TEM). The results show that nano-sized Ce1-xTixP2O7 behave as an excellent UV-shielding material. Thereinto, the CeP2O7 has the most excellent UV-shielding effect, and the amorphous state of Ce0.8Ti0.2P2O7 can keep at a higher temperature than CeP2O7. Therefore, the stabilization of the amorphous state of the cerium pyrophosphates was carried out by doping titanium. | 4 | The corrosion behavior of Ti-6Al-3Nb-2Zr-1Mo alloy:Effects of HCl concentration and temperature显示文摘Investigation about the corrosion behavior of Ti alloys in different ambient environment is of great significance for their practical application.Herein,we systematically investigate the corrosion behavior of a newfound Ti-6 Al-3 Nb-2 Zr-1 Mo(Ti80)alloy in hydrochloric acid(HCl)ranging from 1.37 to 7 M,and temperature ranging from 25 to 55℃,by means of electrochemical measurements,static immersion tests and surface analysis.Results manifest that increasing either HCl concentration or temperature can accelerate the corrosion of Ti80 alloy via promoting the breakdown of native protective oxide film and then further facilitating the active dissolution of Ti80 matrix.According to potentiodynamic polarization curves,Ti80 alloy displays a spontaneous passive behavior in 1.37 M HCl at 25℃,compared to a typical active-passive behavior under the other conditions.As indicated by cathodic Tafel slope,the rate determining step for cathodic hydrogen evolution reaction is likely the discharge reaction step.The apparent activation energies obtained from corrosion current density and maximum anodic current density for Ti80 alloy in 5 M HCl solution are 62.4 and 55.6 kJ mol-1,respectively,which signifies that the rate determining step in the corrosion process of Ti80 alloy is mainly determined by surface-chemical reaction rather than diffusion.Besides,the electrochemical impedance spectroscopy tests demonstrate that a stable and compact oxide film exists in 1.37 M HCl at 25℃,whereas a porous corrosion product film forms under the other conditions.Overall,the critical HCl concentration at which Ti80 alloy can maintain passivation at 25℃can be determined as a value between 1.37 and 3 M.Furthermore,the corroded surface morphology characterization reveals that equiaxedαphase is more susceptible to corrosion compared to intergranularβphase due to a lower content of Nb,Mo,and Zr in the former. | Baoxian Su Binbin Wang Liangshun Luo Liang Wang Yanqing Su Fuxin Wang Yanjin Xu Baoshuai Han Haiguang Huang Jingjie Guo Hengzhi Fu | 2021 | Journal of Materials Science & Technology2021,,15: | 5 |
| 5 | Preparation of rabbit anti-rat LRRN3 polyclonal antibody and study of its expression显示文摘BACKGROUND:Studies have shown that the LRRN3,a member of the Neuron Leucine-Rich Repeat superfamily,could be related to neural development,differentiation,information transmission,and other functions,but most studies have focused on nucleic acid levels and few have reported on LRRN3 protein levels.OBJECTIVE:To prepare rabbit anti-rat LRRN3 polyclonal antibody and to observe protein tissue expression profiles.DESIGN,TIME AND SETTING:In vitro,molecular,biological experiments were performed from October 2007 to April 2009 in Laboratory of Neurobiology at Xiangya School of Medicine,Central South University.MATERIALS:Immunization antigen,namely rat Mal-LRRN3C-His recombinant protein,was provided by the Laboratory of Neurobiology at Xiangya School of Medicine,Central South University.METHODS:Rat Mal-LRRN3C-His recombinant protein was used to immunize male,New Zealand rabbits,and rabbit anti-rat LRRN3 polyclonal antibody was prepared.MAIN OUTCOME MEASURES:Antibody purification was conducted using Protein A affinity chromatography,and the LRRN3 anti-serum titer was identified using enzyme-linked immunosorbent assay.Immunohistochemical techniques and Western blot preliminary tests were used to determine LRRN3 protein expression profiles in adult rats.RESULTS:A highly purified rabbit anti-rat LRRN3 polyclonal antibody was obtained.Western Blot results from rat brain total protein revealed a band at 79 kD,which was consistent with the size of LRRN3.Immunohistochemistry results showed that protein was mainly expressed in the central nervous system,and no significant positive signals were observed in other tissues.Positive cells included neurons of cerebral cortex and hippocampal dentate gyrus granule cell layer,and cerebellar Purkinje cells.There was no positive expression in glial cells.CONCLUSION:Rabbit anti-rat LRRN3 polyclonal antibody was successfully prepared at a high purity from the prokaryotic-expressed Mal-LRRN3C-His recombinant protein,which served as an antigen.Rat LRRN3 protein was primarily expressed in cerebral cortex neurons,hippocampal dentate gyrus granule cell layer neurons,and cerebellar Purkinje cells. | Jing Yang Yanjin Wang Yan Cai Zhiying Yang Ganqiu Wu Jianguo Wen Xuegang Luo He Huang | 2010 | Neural Regeneration Research2010,5,13: | 3 |

| 9 | Sorl1 knockout inhibits expression of brain-derived neurotrophic factor:involvement in the development of late-onset Alzheimer's disease显示文摘Sortilin-related receptor 1(SORL1)is a critical gene associated with late-onset Alzheimer’s disease.SORL1 contributes to the development and progression of this neurodegenerative condition by affecting the transport and metabolism of intracellularβ-amyloid precursor protein.To better understand the underlying mechanisms of SORL1 in the pathogenesis of late-onset Alzheimer s disease,in this study,we established a mouse model of SorI1 gene knockout using cluste red regularly inters paced short palindro mic repeats-associated protein 9 technology.We found that Sorl1-knocko ut mice displayed deficits in learning and memory.Furthermore,the expression of brain-derived neurotrophic factor was significantly downregulated in the hippocampus and co rtex,and amyloidβ-protein deposits were observed in the brains of 5orl1-knockout mice.In vitro,hippocampal neuronal cell synapses from homozygous Sorl1-knockout mice were impaired.The expression of synaptic proteins,including Drebrin and NR2B,was significantly reduced,and also their colocalization.Additionally,by knocking out the Sorl1 gene in N2a cells,we found that expression of the N-methyl-D-aspartate receptor,NR2B,and cyclic adenosine monophosphate-response element binding protein was also inhibited.These findings suggest that SORL1 participates in the pathogenesis of late-onset Alzheimer s disease by regulating the N-methyl-D-aspartate receptor NR2B/cyclic adenosine monophosphate-response element binding protein signaling axis. | Mingri Zhao Xun Chen Jiangfeng Liu Yanjin Feng Chen Wang Ting Xu Wanxi Liu Xionghao Liu Mujun Liu Deren Hou | 2024 | Neural Regeneration Research2024,19,7: | 2 |

| 11 | Ginger-indirect moxibustion plus acupuncture versus acupuncture alone for chronic fatigue syndrome:a randomized controlled trial显示文摘OBJECTIVE:To assess the efficacy and safety of gingerindirect moxibustion for chronic fatigue syndrome(CFS).METHODS:In this central randomized,controlled trial,290 CFS participants were recruited and randomly allocated to group A(ginger-indirect moxibustion plus acupuncture)or group B(acupuncture alone).The study consisted of a treatment period of 8 weeks with a total of 24 treatments(3 sessions per week,every other day),and a follow-up period of 12 weeks.The outcome was measured by Fatigue Severity Scale(FSS),Psychological Health Report(SPHERE),the Self-rating depression scale(SDS)and the Hamilton anxiety scale(HAMA)at baseline,2,4,6,8,12 and 20 weeks.RESULTS:With the treatment undergoing,the changes of FSS,SPHERE,SDS and HAMA scores in both groups increased gradually,and the effect maintained at the 12th week.Between groups,significantly higher score changes were seen in group A in FSS after 4 weeks treatment(11.94 vs 9.12,95%CI:0.94,4.7)and in SPHERE after 2 weeks treatment(3.7 vs 2.27,95%CI:0.56,2.31).But for SDS and HAMA,the improvement did not differ significantly between groups.No severe adverse events were reported.CONCLUSION:Ginger-indirect moxibustion is a safe and effective intervention to relieve fatigue and accompanying physical symptoms of CFS. | MA Tingting WU Jie YANG Lijie FENG Fen YANG Huilin ZHANG Jinhua ZHONG Yanjin NING Qing HUANG Lirong LIN Youbing YAN Jue CHEN Guiquan HOU Tianshu WANG Li REN Yuanfang TAN Jing | 2022 | Journal of Traditional Chinese Medicine2022,42,2: | 2 |

| 20 | Multi-Plan Integration: A Pilot Study of Mianzhu, Sichuan Province显示文摘The current planning system in China lacks coordination, which is a crucial issue to be explored to fully implement the multi-plan integration. Mianzhu is an example that is officially named as one of the pilot cities for multi-plan integration on the county level. The paper firstly analyzes the problems associated with multi-plan integration in Mianzhu, including the indeterminate hierarchy of the planning system, the disunity of technical standards, problems regarding spatial order, coordination and division of powers and functions, and the intensive utilization of spatial resources, the lack of a coordination mechanism, and low participation of experts and the public.
