2Anatomical variation of infra-pyloric artery origination: A prospective multicenter observational study (IPA-Origin)显示文摘Objective: Infra-pyloric artery(IPA) is an important anatomical landmark in treatment of gastric cancer and is the key vessel for pylorus-preserving gastrectomy and subgroup of infra-pyloric lymph nodes. However, its anatomical variation is not thoroughly understood. Our study aimed to clarify the origination of the IPA.Methods: We did this prospective, multicenter, open-label, observational study at gastric surgery departments of34 hospitals in China. Gastric cancer patients aged 18 years or older and scheduled to undergo elective total or distal gastrectomy were assigned. During the surgery, IPA dissecting and exposing the origination point with photographs or video clips were required. The primary outcome was the origination of the IPA. Analysis of variance, χ~2 tests and Fisher's tests were used to analyze the differences between groups. The study is registered at Clinicaltrials.gov(No. NCT03071237).Results: Between May 8 and July 31, 2017, 429 patients were assigned for the study, and 419(97.7%) patients had the IPA dissected and recorded through photograph or video and were included in the primary outcome analysis. The median age was 62 years old, and 73.7% were male. Among the patients, 78.5% received laparoscopic surgery. Single IPA origination was identified in 398(95.0%) patients, including gastroduodenal artery(GDA) in154(36.8%) patients, anterior superior pancreaticoduodenal artery(ASPDA) in 130(31.0%) patients, and right gastroepiploic artery(RGEA) in 114(27.2%) patients. Fifteen(3.6%) patients were identified with multiple IPA and 6(1.4%) patients were identified as IPA absence. The differences in the distribution of surgical approach(P=0.003) and geographic area(P=0.030) were statistically significant. 6Micron-to nano-pore characteristics in the shale of Longmaxi Formation,southeast Sichuan Basin显示文摘For shale of Lower Silurian Longmaxi Formation in Chongqing,southeast Sichuan Basin,characteristics of micro-nano pores in marine shale reservoirs were well studies by means of Field-Emission Scanning Electron Microscope and Low-temperature Low-pressure Adsorption Experiment of CO_(2)and N_(2).Results showed that six types of pore were developed in the shale of Longmaxi Formation,i.e.,organic pores,intergranular pores,intragranular pores,intercrystalline pores,dissolution pores and microfractures,among which the organic pores and intragranular pores in interlayers of clay minerals were most developed,and a plenty of dissolution pores were also well developed because of high thermal evolution degree.BET specific surface area of the shale in Longmaxi Formation ranged from 3.5 to 18.1 m^(2)/g,BJH total pore volume was from 0.00234 to 0.01338 cm^(3)/g,DA specific surface area of micropores vaired from 1.3 to 7.3 m^(2)/g,and DA pore volume ranged from 0.00052 to 0.00273 cm^(3)/g.The specific surface area of micropores in the shale accounted for 23.1%-80.2%of total specific surface area with an average of 50.3%,and the pore volume of micropores accounted for 12.1%-48.5%of total pore volume with an average of 32.3%.Micropore was the main storage space in shale reservoir for methane adsorption,that because capacities of specific surface area provided by micropores were considerably greater than those provided by mesopores and macropores.Pore size distribution of the shale was complex,and multiple different peaks occurred in the pore size curves,showing two or three peaks in the range from 0 to 100 nm and four peaks occasionally.TOC had a good linear relationship with pore structure parameters of micropores,mesopores t macropores and total pores in the shale,indicating that TOC was the most important control factor for micron-to nano-pore structure in the shale.After normalization of pore structure parameters to TOC,the pore structure parameters of total pores and mesopores t macropores,had positive linear relationships with content of clay minerals but negative linear relationships with content of brittle minerals,indicating that clay minerals and brittle minerals mainly controlled development of mesopores and macropores in the shale.Wenming Ji Yan Song Zhenxue Jiang Mianmo Meng Qingxin Liu Fenglin Gao 2017Petroleum Research2017,2,2:1

8Adsorption of Naphthenic Acids from Dewaxed Vacuum Gas Oil by Activated Clay: Kinetics, Equilibrium and Thermodynamic Studies显示文摘This work was mainly concentrated on the removal of naphthenic acids(NAs) from dewaxed vacuum gas oil(VGO) by adsorption using a commercial grade activated clay(AC) adsorption during lube base oil refining. The NAs in dewaxed VGO cut-4 were identified by negative-ion electrospray ionization Fourier transform ion cyclotron resonance mass spectrometry(ESI FT-ICR MS). The AC sample from a refinery was characterized by XRD, BET, TG/DTA, and SEM. A series of experiments were carried out to investigate the performance of NAs adsorption by AC using a batch adsorption technique, in which some key experimental parameters such as temperature, contact time, initial concentration of NA in oil sample as well as the dosage of adsorbent were investigated. Equilibrium isotherms were analyzed using the Langmuir, Freundlich, Tempkin and Dubinin-Radushkevich(D-R) adsorption models. The pseudo-first order, the pseudo-second order, and intraparticle diffusion models were employed to describe the kinetics data. The results revealed that the D-R isotherm provided a better fit to the experimental data than other isotherms, and the adsorption kinetics followed the pseudo-first order kinetic equation. The thermodynamic data indicated that the adsorption process was feasible and spontaneous as an endothermic process. 11Improving the protective effects of a FGF for peripheral nerve injury repair using sulfated chitooligosaccharides显示文摘Injury to the peripheral nerves can result in temporary or life-long neuronal dysfunction and subsequent economic or social disability. Acidic fibroblast growth factor(a FGF) promotes the growth and survival of neurons and is a possible treatment for peripheral nerve injury. Yet,the actual therapeutic utility of a FGF is limited by its short half-life and instability in vivo. In the present study,we prepared sulfated chitooligosaccharides(SCOS),which have heparinlike properties,to improve the bioactivity of a FGF. We investigated the protective effects of SCOS with or without a FGF on RSC96 cells exposed to Na2 S2 O4 hypoxia/reoxygenation injury. Cell viability was measured by MTT assay and cytotoxicity induced by Na2 S2 O4 was assessed by lactate dehydrogenase(LDH) release into the culture medium. Pretreatment with a FGF and SCOS dramatically decreased LDH release after injury compared to pretreatment with a FGF or SCOS alone. We subsequently prepared an a FGF/SCOS thermo-sensitive hydrogel with poloxamer and examined its effects in vivo. Paw withdrawal thresholds and thermal withdrawal latencies were measured in rats with sciatic nerve injury. Local injection of the a FGF/SCOS hydrogels(a FGF: 40,80 μg/kg) increased the efficiency of sciatic nerve repair compared to a FGF(80 μg/kg) hydrogel alone. Especially a FGF/SCOS thermo-sensitive hydrogel decreased paw withdrawal thresholds from 117.75 ± 8.38(g,4 d) to 65.74 ± 3.39(g,10 d),but a FGF alone group were 140.58 ± 27.54(g,4 d) to 89.12 ± 5.60(g,10 d)(a FGF dose was 80 μg/kg,P < 0.05,n = 8). The thermal withdrawal latencies decreased from 11.61 ± 2.26(s,4 d) to 2.37 ±0.67(s,10 d). However,a FGF alone group were from 17.69 ± 1.47(s,4 d) to4.65 ± 1.73(s,10 d)(P < 0.05,n = 8). Furthermore,the a FGF/SCOS hydrogels also exhibited good biocompatibility in mice. In summary,SCOS improved the protective effects of a FGF in RSC96 cells injured with Na2S2O4 and increased the efficiency of nerve repair and recovery of function in rats with sciatic nerve injury. 12Metabolic engineering of yeasts for green and sustainable production of bioactive ginsenosides F2 and 3β,20S-Di-O-Glc-DM显示文摘Both natural ginsenoside F2 and unnatural ginsenoside 3β,20S-Di-O-Glc-DM were reported to exhibit anti-tumor activity.Traditional approaches for producing them rely on direct extraction from Panax ginseng,enzymatic catalysis or chemical synthesis,all of which result in low yield and high cost.Metabolic engineering of microbes has been recognized as a green and sustainable biotechnology to produce natural and unnatural products.Hence we engineered the complete biosynthetic pathways of F2 and 3β,20S-Di-OGlc-DM in Saccharomyces cerevisiae via the CRISPR/Cas9 system.The titers of F2 and 3β,20S-Di-O-GlcDM were increased from 1.2 to 21.0 mg/L and from 82.0 to 346.1 mg/L at shake flask level,respectively,by multistep metabolic engineering strategies.Additionally,pharmacological evaluation showed that both F2and 3β,20S-Di-O-Glc-DM exhibited anti-pancreatic cancer activity and the activity of 3β,20S-Di-O-GlcDM was even better.Furthermore,the titer of 3β,20S-Di-O-Glc-DM reached 2.6 g/L by fed-batch fermentation in a 3 L bioreactor.To our knowledge,this is the first report on demonstrating the anti-pancreatic cancer activity of F2 and 3β,20S-Di-O-Glc-DM,and achieving their de novo biosynthesis by the engineered yeasts.Our work presents an alternative approach to produce F2 and 3β,20S-Di-O-Glc-DM from renewable biomass,which lays a foundation for drug research and development.Fenglin Jiang Chen Zhou Yan Li Haidong Deng Ting Gong Jingjing Chen Tianjiao Chen Jinling Yang Ping Zhu 2022Acta Pharmaceutica Sinica B2022,12,7:0

14The interphasial degradation of 4.2 V-class poly(ethylene oxide)-based solid batteries beyond electrochemical voltage limit显示文摘Solid-state polymer electrolytes(SPEs)have attracted increasing attention due to good interfacial contact,light weight,and easy manufacturing.However,the practical application of SPEs such as the most widely studied poly(ethylene oxide)(PEO)in high-energy solid polymer batteries is still challenging,and the reasons are yet elusive.Here,it is found that the mismatch between PEO and 4.2 V-class cathodes is beyond the limited electrochemical window of PEO in the solid Li Ni_(1/3)Mn_(1/3)Co_(1/3)O_(2)(NMC)-PEO batteries.The initial oxidation of PEO initiates remarkable surface reconstruction of NMC grains in solid batteries that are different from the situation in liquid electrolytes.Well-aligned nanovoids are observed in NMC grains during the diffusion of surface reconstruction layers towards the bulk in solid batteries.The substantial interphasial degradation,therefore,blocks smooth Li+transport across the NMC-PEO interface and causes performance degradation.A thin yet effective Li F-containing protection layer on NMC can effectively stabilize the NMC-PEO interface with a greatly improved lifespan of NMC|PEO|Li batteries.This work deepens the understanding of degradations in high-voltage solid-state polymer batteries.Renzhi Huang Yang Ding Fenglin Zhang Wei Jiang Canfu Zhang Pengfei Yan Min Ling Huilin Pan 2022Journal of Energy Chemistry2022,,12:0

16Inflammation as a Mediator of Microbiome Dysbiosis-Associated DNA Methylation Changes in Gastric Premalignant Lesions显示文摘Evidence for the influence of chronic inflammation induced by microbial dysbiosis on aberrant DNA methylation supports a plausible connexion between disordered microbiota and precancerous lesions of gastric cancer(PLGC).Here,a comprehensive study including multi-omics data was performed to estimate the relationships amongst the gastric microbiome,inflammatory proteins and DNA methylation alterations and their roles in PLGC development.The results demonstrated that gastric dysbacteriosis increased the risk of PLGC and DNA methylation alterations in related tumour suppressor genes.Seven inflammatory biomarkers were identified for antrum and corpus tissues,respectively,amongst which the expression levels of several biomarkers were significantly correlated with the microbial dysbiosis index(MDI)and methylation status of specific tumour suppressor genes.Notably,mediation analysis revealed that microbial dysbiosis partially contributed to DNA methylation changes in the stomach via the inflammatory cytokines C-C motif chemokine 20(CCL20)and tumour necrosis factor receptor superfamily member 9(TNFRSF9).Overall,these results may provide new insights into the mechanisms that might link the gastric microbiome to PLGC.Lingjun Yan Wanxin Li Fenglin Chen Junzhuo Wang Jianshun Chen Ying Chen Weimin Ye 2023Phenomics2023,3,5:0
17Abivertinib inhibits megakaryocyte differentiation and platelet biogenesis显示文摘Abivertinib,a third-generation tyrosine kinase inhibitor,is originally designed to target epidermal growth factor receptor(EGFR)-activating mutations.Previous studies have shown that abivertinib has promising antitumor activity and a well-tolerated safety profile in patients with non-small-cell lung cancer.However,abivertinib also exhibited high inhibitory activity against Bruton’s tyrosine kinase and Janus kinase 3.Given that these kinases play some roles in the progression of megakaryopoiesis,we speculate that abivertinib can affect megakaryocyte(MK)differentiation and platelet biogenesis.We treated cord blood CD34+hematopoietic stem cells,Meg-01 cells,and C57BL/6 mice with abivertinib and observed megakaryopoiesis to determine the biological effect of abivertinib on MK differentiation and platelet biogenesis.Our in vitro results showed that abivertinib impaired the CFU-MK formation,proliferation of CD34+HSC-derived MK progenitor cells,and differentiation and functions of MKs and inhibited Meg-01-derived MK differentiation.These results suggested that megakaryopoiesis was inhibited by abivertinib.We also demonstrated in vivo that abivertinib decreased the number of MKs in bone marrow and platelet counts in mice,which suggested that thrombopoiesis was also inhibited.Thus,these preclinical data collectively suggested that abivertinib could inhibit MK differentiation and platelet biogenesis and might be an agent for thrombocythemia.Jiansong Huang Xin Huang Yang Li Xia Li Jinghan Wang Fenglin Li Xiao Yan Huanping Wang Yungui Wang Xiangjie Lin Jifang Tu Daqiang He Wenle Ye Min Yang Jie Jin 2022Frontiers of Medicine2022,16,3:0
