1Flow cytometric detection of hepatitis C virus antigens in infected peripheral blood leukocytes: Binding and entry显示文摘AIM: We designed two synthetic-core-specific peptides core 1 (C1) and core 2 (C2), and an E1-specific peptide (E1). We produced specific polyclonal antibodies againstthese peptides and used the antibodies for detection of HCV antigens on surface and within infected peripheral blood leukocytes.METHODS: Peripheral blood from a healthy individual who tested negative for HCV RNA was incubated with HCV type 4 infected serum for 1 h and 24 h at 37 ℃. Cells were stained by direct and indirect immunofluorescence and measured by flow cytometry.RESULTS: After 1 h of incubation, antibodies against C1,C2, and E1 detected HCV antigens on the surface of 27%,26% and 73% of monocytes respectively, while 10%, 5% and 9% of lymphocytes were positive with anti-C1, anti-C2 and anti-E1 respectively. Only 1-3% of granulocytes showed positive staining with anti-C1, anti-C2 and anti E1 antibodies. After 24 h of incubation, we found no surface staining with anti-C1, anti-C2 or anti-E1. Direct immunostaining using anti-C2 could not detect intracellular HCV antigens, after 1 h of incubation with the virus, while after 24 h of incubation, 28% of infected cells showed positive staining. Only plus strand RNA was detectable intracellularly as early as 1 h after incubation, and remained detectable throughout 48 h post-infection.Interestingly, minus RNA strand could not be detected after 1 h, but became strongly detectable intracellularly after 24 h post-infection.CONCLUSION: Monocytes and lymphocytes are the preferred target cells for HCV infection in peripheral blood leukocytes. METHODS: Formalin-fixed paraffin-embedded gas-trointestinal biopsy blocks from patients (n = 48) with non-inflammatory bowel disease (irritable bowel syndrome and functional abdominal pain) and inflammatory bowel disease were sectioned and stained for IL-6, mast cells, enterochromaffin cells, 5-hydroxytryptamine, and substance P. All children had chronic abdominal pain as part of their presenting symptoms. Biopsy phenotype was confirmed by a pathologist, blinded to patient information. Descriptive statistics, chi-square, and independent sample t tests were used to compare differences between the inflammatory and non-inflammatory groups. RESULTS: The cohort (n = 48), mean age 11.9 years (SD = 2.9), 54.2% females, 90% Caucasian, was comprised of a non-inflammatory (n = 26) and an inflammatory (n = 22) phenotype. There was a significant negative correlation between substance P expression and mast cell count (P = 0.05, r = -0.373). Substance P was found to be expressed more often in female patient biopsies and more intensely in the upper gastrointestinal mucosa as compared to the lower mucosa. There were significantly increased gastrointestinal mucosal immunoreactivity to IL-6 (P = 0.004) in the inflammatory phenotype compared to non-inflammatory. Additionally, we found significantly increased mast cells (P = 0.049) in the mucosa of the non-inflammatory phenotype compared to the inflammatory group. This difference was particularly noted in the lower colon biopsies. 5Early detection of hepatocellular carcinoma co-occurring with hepatitis C virus infection: A mathematical model显示文摘AIM: To develop a mathematical model for the early detection of hepatocellular carcinoma(HCC) with a panel of serum proteins in combination with α-fetoprotein(AFP).METHODS: Serum levels of interleukin(IL)-8, soluble intercellular adhesion molecule-1(s ICAM-1), soluble tumor necrosis factor receptor Ⅱ(s TNF-RⅡ), proteasome, and β-catenin were measured in 479 subjects categorized into four groups:(1) HCC concurrent with hepatitis C virus(HCV) infection(n = 192);(2) HCV related liver cirrhosis(LC)(n = 96);(3) Chronic hepatitis C(CHC)(n = 96); and(4) Healthy controls(n = 95). The R package and different modules for binary and multi-class classifiers based on generalized linear models were used to model the data. Predictive power was used to evaluate the performance of the model. Receiver operating characteristic curve analysis over pairs of groups was used to identify the best cutoffs differentiating the different groups. RESULTS: We revealed mathematical models, based on a binary classifier, made up of a unique panel of serum proteins that improved the individual performance of AFP in discriminating HCC patients from patients with chronic liver disease either with or without cirrhosis. We discriminated the HCC group from the cirrhotic liver group using a mathematical model(-11.3 + 7.38 × Prot + 0.00108 × s ICAM + 0.2574 ×β-catenin + 0.01597 × AFP) with a cutoff of 0.6552, which achieved 98.8% specificity and 89.1% sensitivity. For the discrimination of the HCC group from the CHC group, we used a mathematical model [-10.40 + 1.416 × proteasome + 0.002024 × IL + 0.004096 × s ICAM-1 +(4.251 × 10-4) × s TNF + 0.02567 ×β-catenin + 0.02442 × AFP] with a cutoff 0.744 and achieved 96.8% specificity and 89.7% sensitivity. Additionally, we derived an algorithm, based on a binary classifier, for resolving the multi-class classification problem by using three successive mathematical model predictions of liver disease status. 6Radiolarian and Planktic Foraminifera Biostratigraphy of the Early Albian Organic Rich Beds of Fahdene Formation,Northern Tunisia显示文摘The lower part of Fahdene Formation outcropping in northeastern Tunisia is represented by pelagic sequences and characterized by an important siliciclastic fraction that includes radiolarian and organic-rich beds of Allam Member.?Litho-biostratigraphic analysis based on planktic foraminifera and radiolaria shows that deposition of organic-rich beds of Allam Member is confined to the Microhedbergella rischi Zone through the lower part of Ticinella primula Zone.?Age diagnostic radiolaria recovered from these beds has been identified to constrain a direct age of black shale deposition.The assemblages can be correlated with the radiolarian biochronozone U.A.10–11 indicating an Early Albian to early Middle Albian in age.Radiolarian assemblages composed of species characteristic of U.A.10 biochronozone(A.montisserei,D.gracilis)are associated with radiolarian taxa belonging to Costata Zone(U.A.9 Pseudoeucyrtis hanni and Thanarla pseudodecora),which can be correlated with Dercourt Member in the Ionian Zone and Paquier level of the Vocontian Basin(southeast of France).Biostratigraphic and abundance curves analysis has demonstrated significant diversification of cryptocephalic Nassellaria and Archaeodictyomitrae,probably adapted to eutrophic conditions that characterized the Aptian-Albian transition.Ecological conditions may have governed the stratigraphic range of many cosmopolitan taxa(i.e.,Pseudodictyomitrae lodogaensis)compared with stratigraphic distributions schemes reported from other domains.The timing of black shales deposition was diachronic due to local geodynamic conditions and upwelling currents distribution.The Allam black shales are correlative with the oceanic anoxic event OAE1b which is characterized by the widespread of supraregionnally organic-rich beds in the Mediterranean Tethys basins across the Aptian-Albian transition.M Ben Fadhel T Zouaghi A Amri M Ben Youssef 2014Journal of Earth Science2014,25,1:2
