| 4 | Programmed Death Ligand-1 on Microglia Regulates Th1 Differentiation via Nitric Oxide in Experimental Autoimmune Encephalomyelitis显示文摘Microglia are considered to be potential antigen-presenting cells and have the ability to present antigen under pathological conditions. Nevertheless, whether and how microglia are involved in immune regulation are largely unknown. Here, we investigated the suppressive activity of microglia during experimental autoimmune encephalomyelitis(EAE) induced by myelin oligodendrocyte glycoprotein, with the goal of understanding their role in regulating the T cell reaction. Using flow cytometric analysis, we found that microglia were characterized by increased cell number and up-regulated programmed death ligand-1(PD-L1) at the peak phase of EAE. Meanwhile,both the CD^(4+)T cells and microglia that infiltrated the central nervous system expressed higher levels of PD1, the receptor for PD-L1, accompanied by a decline of Th1 cells.In an ex vivo co-culture system, microglia from EAE mice inhibited the proliferation of antigen-specific CD^(4+)T cells and the differentiation of Th1 cells, and this was significantly inhibited by PD-L1 blockade. Further,microglia suppressed Th1 cells via nitric oxide(NO), the production of which was dependent on PD-L1. | 6 | Local icariin application enhanced periodontal tissue regeneration and relieved local inflammation in a minipig model of periodontitis显示文摘Periodontitis is an inflammatory autoimmune disease. Treatment should alleviate inflammation, regulate the immune reaction and promote periodontal tissue regeneration. Icariin is the main active ingredient of Epimedii Folium, and it is a promising compound for the enhancement of mesenchymal stem cell function, promotion of bone formation, inhibition of bone resorption, alleviation of inflammation and regulation of immunity. The study investigated the effect of icariin on periodontal tissue regeneration in a minipig model of periodontitis. The minipig model of periodontitis was established. Icariin was injected locally. The periodontal clinical assessment index, a computed tomography(CT) scan, histopathology and enzyme-linked immune sorbent assay(ELISA)were used to evaluate the effects of icariin. Quantitative analysis results 12 weeks post-injection demonstrated that probing depth,gingival recession, attachment loss and alveolar bone regeneration values were(3.72 ± 1.18) mm vs.(6.56 ± 1.47) mm,(1.67 ± 0.59)mm vs.(2.38 ± 0.61) mm,(5.56 ± 1.29) mm vs.(8.61 ± 1.72) mm, and(25.65 ± 5.13) mm3 vs.(9.48 ± 1.78) mm3 in the icariin group and0.9% NaCl group, respectively. The clinical assessment, CT scan, and histopathology results demonstrated significant enhancement of periodontal tissue regeneration in the icariin group compared to the 0.9% NaCl group. The ELISA results suggested that the concentration of interleukin-1 beta(IL-1β) in the icariin group was downregulated compared to the 0.9% NaCl group, which indicates that local injection of icariin relieved local inflammation in a minipig model of periodontitis. Local injection of icariin promoted periodontal tissue regeneration and exerted anti-inflammatory and immunomodulatory function. | 9 | Deformation mechanism and microstructures on polycrystalline aluminum induced by high-current pulsed electron beam显示文摘In this paper, we present an experimental investigation of deformation twinning in polycrystalline aluminum exposed to high-current pulsed electron beam (HCPEB) irradiation. The residual tensile stress with about 10 2 MPa was introduced in the irradiated surface layer. The feature characteristic irradiated with various numbers of pulses was investigated. The formation of a large number of twin bands on the surface irradiated with multiple pulses was determined. The experimental observations indicated that the deformation twinning was indeed triggered during HCPEB irradiation. It is suggested that high value of stress and strain rate induced by rapid heating and cooling due to HCPEB irradiation may cause the shifting of whole atomic planes simultaneously. | 11 | Dark matter direct search sensitivity of the PandaX-4T experiment显示文摘The Panda X-4T experiment, a 4-ton scale dark matter direct detection experiment, is being planned at the China Jinping Underground Laboratory. In this paper we present a simulation study of the expected background in this experiment. In a 2.8-ton fiducial mass and the signal region between 1-10 keV electron equivalent energy, the total electron recoil background is found to be 4.9 × 10^(-5) kg^(-1) d^(-1) keV^(-1). The nuclear recoil background in the same region is 2.8 × 10^(-7) kg^(-1) d^(-1) keV^(-1). | 12 | Dynamic changes of phosphatidylinositol and phosphatidylinositol 4-phosphate levels modulate H^(+)-ATPase and Na^(+)/H^(+) antiporter activities to maintain ion homeostasis in Arabidopsis under salt stress显示文摘Plant metabolites are dynamically modified and distributed in response to environmental changes.How-ever,it is poorly understood how metabolic change functions in plant stress responses.Maintaining ion ho-meostasis under salt stress requires coordinated activation of two types of central regulators:plasma membrane(PM)H^(+)-ATPase and Na^(+)/H^(+) antiporter.In this study,we used a bioassay-guided isolation approach to identify endogenous small molecules that affect PM H^(+)-ATPase and Na^(+)/H^(+) antiporter activities and identified phosphatidylinositol(PI),which inhibits PM H^(+)-ATPase activity under non-stress conditions in Arabidopsis by directly binding to the C terminus of the PM H^(+)-ATPase AHA2.Under salt stress,the phosphatidylinositol 4-phosphate-to-phosphatidylinositol(PI4P-to-PI)ratio increased,and PI4P bound and activated the PM Na^(+)/H^(+) antiporter.PI prefers binding to the inactive form of PM H^(+)-ATPase,while PI4P tends to bind to the active form of the Na^(+)/H^(+) antiporter.Consistent with this,pis1 mutants,with reduced levels of PI,displayed increased PM H^(+)-ATPase activity and salt stress toler-ance,while the pi4kβ1 mutant,with reduced levels of PI4P,displayed reduced PM Na^(+)/H^(+) antiporter activity and salt stress tolerance.Collectively,our results reveal that the dynamic change between PI and PI4P in response to salt stress in Arabidopsis is crucial for maintaining ion homeostasis to protect plants from un-favorable environmental conditions. | Yongqing Yang Xiuli Han Liang Ma Yujiao Wu Xiao Liu Haiqi Fu Guoyong Liu Xiaoguang Lei Yan Guo | 2021 | Molecular Plant2021,14,12: | 4 |
| 13 | Co-delivery of doxorubicin and paclitaxel with linear-dendritic block copolymer for enhanced anti-cancer efficacy显示文摘A series of well-defined amphiphilic linear-dendritic block copolymers(telodendrimers, MPEG-b-PAMAM-cholesterol) with 1,2,4 or 8 cholesteryl groups(named as P1, P2, P4, P8, respectively) were synthesized. Their chemical structures were characterized with 1H NMR and mass spectrum(MALDI-TOF MS). The telodendrimers could self-assemble into micelles in aqueous solution, and encapsulate chemotherapeutic drug doxorubicin(DOX) and paclitaxel(PTX) for combination therapy. All the telodendrimers could encapsulate DOX with similar capability. However, their drug-loading capability of PTX is increased with the increasing number of cholesteryl groups. P8 exhibited much higher PTX loading efficiency than its counterparts. Thus, P8 was selected for further application of drug delivery in the paper. The drug-loading micellar nanoparticles(NPs) of P8 were spherical in shape and their diameters were less than 150 nm which were determined by dynamic light scattering measurements(DLS) and transmission electron microscope(TEM). In vitro drug release experiment demonstrated that P8 exhibited a controlled release manner for both DOX and PTX, and the two drugs were released simultaneously. In vitro cytotoxicity experiment further demonstrated that the co-delivery of DOX and PTX in P8 exhibited better anti-cancer efficiency than the delivery systems encapsulated with single drug(DOX or PTX). This indicates a synergistic effect. | 14 | Inhibitory effects of nisin-coated multi-walled carbon nanotube sheet on biofilm formation from Bacillus anthracis spores显示文摘Multi-walled carbon nanotube(MWCNT)sheet was fabricated from a drawable MWCNT forest and then deposited on poly(methyl methacrylate)film.The film was further coated with a natural antimicrobial peptide nisin.We studied the effects of nisin coating on the attachment of Bacillus anthracis spores,the germination of attached spores,and the subsequent biofilm formation from attached spores.It was found that the strong adsorptivity and the super hydrophobicity of MWCNTs provided an ideal platform for nisin coating.Nisin coating on MWCNT sheets decreased surface hydrophobicity,reduced spore attachment,and reduced the germination of attached spores by 3.5 fold,and further inhibited the subsequent biofilm formation by 94.6%compared to that on uncoated MWCNT sheet.Nisin also changed the morphology of vegetative cells in the formed biofilm.The results of this study demonstrated that the anti-adhesion and antimicrobial effect of nisin in combination with the physical properties of carbon nanotubes had the potential in producing effective anti-biofilm formation surfaces. | Xiuli Dong Eric McCoy Mei Zhang Liju Yang | 2014 | Journal of Environmental Sciences2014,26,12: | 4 |
| 15 | Prussian blue nanoparticle-loaded microbubbles for photothermally enhanced gene delivery through ultrasound-targeted microbubble destruction显示文摘By adsorbing chitosan(CS)-functionalized Prussian blue(PB) nanoparticles(CS/PB NPs) complexing DNA onto the surface of gas encapsulated microbubbles(MBs), a multifunctional gene delivery system of MBs@CS/PB/DNA was fabricated for photothermally enhanced gene transfection through ultrasound-targeted microbubble destruction. CS/PB NPs of(2.69 ± 0.49) nm could complex DNA effectively when the mass ratio was2:1. It was found that MBs@CS/PB/DNA could enhance ultrasound imaging greatly both in vitro and in vivo. In addition, MBs@CS/PB/DNA could be disrupted by applying a higher-intensity ultrasound irradiation to release CS/PB/DNA, which could effectively transform the nearinfrared(NIR) light into heat to assist the uptake of CS/PB/DNA by cells. With the aid of ultrasound irradiation and NIR light irradiation, the gene transfection efficiency was significantly enhanced to(43.08 ± 1.13) %, much higher than polyethylenimine. | 17 | Effectiveness of tonifying-kidney and regulating-liver therapy on diminished ovarian reserve: a systematic review and Meta-analysis of randomized controlled trials显示文摘OBJECTIVE: To evaluate the effectiveness of the Traditional Chinese Medicine tonifying-kidney and regulating-liver therapy on diminished ovarian reserve(DOR).METHODS: The literature was comprehensively searched up to August 2019 using four Chinese and three English electronic databases to extract randomized clinical trials(RCTs) comparing Traditional Chinese Medicine tonifying-kidney and regulating-liver prescriptions(combined with hormone therapy or not) with Western Medicine. Data quality evaluation was conducted using the Cochrane risk of bias tool. Meta-analysis was conducted using Revman 5.3 software with effect estimates presented as mean difference(MD), risk ratio(RR), and95% confidence interval(CI).RESULTS: A total of nine RCTs with 512 participants were extracted and eligible for Meta-analysis.There were no significant differences between Chinese medicine and Western Medicine on basal serum follicle-stimulating hormone(FSH) level(MD0.11, 95% CI-0.52 to 0.74, 392 participants, seven trials), anti-Müllerian hormone level(MD 0.48, 95%CI-0.62 to 1.58, 95 participants, two trials), and the FSH and luteinizing hormone ratio(MD 0.01,95% CI-0.95 to 0.96, 115 participants, two trials).Chinese medicine was more effective at improving Traditional Chinese Medicine symptom scores(TCMSS)(MD-2.39, 95% CI-3.83 to-0.94, 160 participants, three trials), effective rate of TCMSS(RR1.18, 95% CI 1.02 to 1.36, 160 participants, three trials), antral follicle count(AFC)(MD 0.55, 95% CI 0.05 to 1.04, 155 participants, three trials), and FSH levels at 3 months post-treatment(MD-4.77, 95% CI-6.09 to-3.45, 137 participants, two trials).CONCLUSION: Compared with Western Medicine,tonifying-kidney and regulating-liver therapy is more effective at relieving symptoms and improving AFC and FSH at 3 months post-treatment. | Liu Liuqing Liu Yanfeng Yang Ming Xu Guiqin Li Ruiqi Xu Xiuli Pan Xue Liang Jialing | 2020 | Journal of Traditional Chinese Medicine2020,40,3: | 3 |
