1Advances and challenges in the treatment of esophageal cancer显示文摘Esophageal cancer(EC)is one of the most common cancers with high morbidity and mortality rates.EC includes two histological subtypes,namely esophageal squamous cell carcinoma(ESCC)and esophageal adenocarcinoma(EAC).ESCC primarily occurs in East Asia,whereas EAC occurs in Western countries.The currently available treatment strategies for EC include surgery,chemotherapy,radiation therapy,molecular targeted therapy,and combinations thereof.However,the prognosis remains poor,and the overall five-year survival rate is very low.Therefore,achieving the goal of effective treatment remains challenging.In this review,we discuss the latest developments in chemotherapy and molecular targeted therapy for EC,and comprehensively analyze the application prospects and existing problems of immunotherapy.Collectively,this review aims to provide a better understanding of the currently available drugs through in-depth analysis,promote the development of new therapeutic agents,and eventually improve the treatment outcomes of patients with EC.Shiming He Jian Xu Xiujun Liu Yongsu Zhen 2021Acta Pharmaceutica Sinica B2021,11,11:26

3CRISPR-Cas9-based genome-wide screening identified novel targets for treating sorafenib-resistant hepatocellular carcinoma:a cross-talk between FGF21 and the NRF2 pathway显示文摘The treatment of hepatocellular carcinoma(HCC)has been dominated by multikinase inhibitors for more than a decade.However,drug resistance can severely restrict the efficacy of these drugs.Using CRISPR/CAS9 genome library screening,we evaluated Kelch-like ECH-associated protein 1(KEAP1)as a key regulator of sorafenib’s susceptibility in HCC.We also investigated whether KEAP1’s knockdown can stabilize nuclear factor(erythroid-derived 2)-like 2(NRF2)protein levels that led to sorafenib’s resistance,including an NRF2 inhibitor that can synergize with sorafenib to abolish HCC’s growth in vitro and in vivo.Furthermore,we clarified that fibroblast growth factor 21(FGF21)is an important downstream regulator of NRF2 in HCC.Intriguingly,we observed that FGF21 bound to NRF2 through the C-terminus of FGF21,thereby stabilizing NRF2 by reducing its ubiquitination and generating a positive feedback loop in sorafenib-resistant HCC.These findings,therefore,propose that targeting FGF21 is a promising strategy to combat HCC sorafenib’s resistance.Jiang Chen Shi Jiang Huijiang Shao Bixia Li Tong Ji Daniel Staiculescu Jiayan He Jie Zhao Liuxin Cai Xiao Liang Junjie Xu Xiujun Cai 2022Science China(Life Sciences)2022,65,10:4

5Genetically modified CD7-targeting allogeneic CAR-T cell therapy with enhanced efficacy for relapsed/refractory CD7-positive hematological malignancies:a phase I clinical study显示文摘Chimeric antigen receptor(CAR)-T cell therapy against T cell malignancies faces major challenges including fratricide between CAR-T cells and product contamination from the blasts.Allogeneic CAR-T cells,generated from healthy donor T cells,can provide ready-to-use,blast-free therapeutic products,but their application could be complicated by graft-versus-host disease(GvHD)and host rejection.Here we developed healthy donor-derived,CD7-targeting CAR-T cells(RD13-01)with genetic modifications to resist fratricide,GvHD and allogeneic rejection,as well as to potentiate antitumor function.A phase I clinical trial(NCT04538599)was conducted with twelve patients recruited(eleven with T cell leukemia/lymphoma,and one with CD7-expressing acute myeloid leukemia).All patients achieved pre-set end points and eleven proceeded to efficacy evaluation.No dose-limiting toxicity,GvHD,immune effector cell-associated neurotoxicity or severe cytokine release syndrome(grade≥3)were observed.28 days post infusion,81.8%of patients(9/11)showed objective responses and the complete response rate was 63.6%(7/11,including the patient with AML).3 of the responding patients were bridged to allogeneic hematopoietic stem cell transplantation.With a median follow-up of 10.5 months,4 patients remained in complete remission.Cytomegalovirus(CMV)and/or Epstein-Barr virus(EBV)reactivation was observed in several patients,and one died from EBV-associated diffuse large B-cell lymphoma(DLBCL).Expansion of CD7-negative normal T cells was detected post infusion.In summary,we present the first report of a Phase I clinical trial using healthy donor-derived CD7-targeting allogeneic CAR-T cells to treat CD7^(+)hematological malignancies.Our results demonstrated the encouraging safety and efficacy profiles of the RD13-01 allogeneic CAR-T cells for CD7^(+)tumors.Yongxian Hu Yali Zhou Mingming Zhang Houli Zhao Guoqing Wei Wengang Ge Qu Cui Qitian Mu Gong Chen Lu Han Tingting Guo Jiazhen Cui Xiaoyan Jiang Xiujun Zheng Shuhui Yu Xiaolong Li Xingwang Zhang Mingxi Chen Xiuju Li Ming Gao Kang Wang Cheng Zu Hao Zhang Xiaohong He Yanbin Wang Dongrui Wang Jiangtao Ren He Huang 2022Cell Research2022,32,11:2

8The impact of X-rays on the expressions of miR-130a/miR-25 and its potential role in the enhanced metastasis of A549 cell lines in vitro induced by X-rays显示文摘We aimed to investigate the impact of X-rays on miR-130a and miR-25 expressions of A549 cell lines and to understand the mechanism of miR-130a and miR-25 on the regulation of invasion and metastasis of A549 cell lines. Human adenocarcinoma cells of the line A549 were cultured and radiated by X-rays at the absorbed doses of 2 and 4, respectively by linear accelerator. Transwell invasion and migration assay were employed to exam the metastasis ability of A549 cells post X-rays irradiation. Both the miRNA and mRNA were extracted from A549 cells at different time points post radiation. The expressions of miR-130a and miR-25, as well as the expressions of VEGF and CCR-7 mRNAs, were detected in A549 cells with 2 and 4 Gy X-rays radiation, respectively by real time PCR. Results were statistically analyzed by SAS 8.2 software, which showed that the invasiveness of A549 cells post 2 and 4 Gy X-rays increased significantly compared with that of untreated A549 cells (the migration cell numbers are 20, 48 and 62 in Group 0, 2 and 4 Gy X-rays, respectively). Furthermore, the expressions of miR-130a and miR-25 also increased significantly at the time-points of 1, 2, 4, and 8 h post radiation, and began to decrease to the control level at 12 h post radiation. VEGF and CCR-7 mRNAs were detected to be up-regulated 18 h post radiation and remained at a high level till 72 h post radiation. The expression of VEGF mRNA has a close correlation with that of CCR-7 mRNA, and the expression of miR-130a also has a correlation with that of VEGF and CCR-7mRNAs. 12Effects of water and nitrogen coupling on growth,physiology and yield of rice显示文摘Water and nitrogen fertilizer are two essential factors for quality and yield formation of rice.Experimental study was carried out to investigate the effects of water and nitrogen fertilizer coupling on yield-related factors,such as growth(height),physiological indicators(chlorophyll and leaf area index(LAI))and yield composition indicators(productive panicles,thousand grain weight and total grains per panicle).Results showed that,the height difference under two irrigation regimes was not significant,and it showed no difference until the tillering stage(p>0.05).The water control method for controlled and mid-gathering irrigation(CMI)was favorable for nutrients converting to rice grain.Meanwhile the height difference for CMI and conventional irrigation(CVI)was the biggest at 80 d after rice transplantation.Variance analysis showed the effect of fertilization on height was significant(p<0.05).With organic fertilizer application,it could control plant growth and promote the nutrients converting to the panicle.The change curve of LAI was similar to chlorophyll content.Organic fertilizer application could not only promote chlorophyll content and LAI,but also delay leaf fading and promote yield.Nitrogen fertilizer factors showed significant difference on rice yield,compared to irrigation regimes showing no significance.Considering the irrigation and fertilizer factors together,the interaction was significant.The descending orders for the effects of water and nitrogen on rice yield were fertilizer,water and fertilizer,water.Regression analysis showed that the productive panicles and total grains per panicle of rice were extremely significant on rice yield,and the direct effect of total grains per panicle on yield was greater than that of productive panicle.This study results could provide theoretical basis for water and nitrogen management to improve rice production.Yuanyuan Li Xiaohou Shao Daoxi Li Menghua Xiao Xiujun Hu Jing He 2019International Journal of Agricultural and Biological Engineering2019,12,3:0
13Selective coordination of polyazin sapanisertib with halide vacancy for printable mesoscopicperovskite solar cells显示文摘The mitigation of under-coordinated Pb^(2+)(halide vacancy)defect remains an imperative challenge in the perovskite solar cells,especially printable mesoscopic perovskite solar cells(FP-PsCs).Here we re port a commercial-available polyazin anticancer drug Sapanisertib as coordination passivator of halide vacancies in FP-PSCs,thereby achieving the photoelectric conversion efficiency(PCE)to 18.46%,along with a record certified PCE of 18.27%.In polazin Sapanisertib(Sap),there exists two kinds of nitrogen atoms:in-aromatic ring(in purine and oxazole rings,IAR-Ns)and out-aromatic ring(substituted amino groups,OAR-Ns).Through multiple characterizations,and DFT calculations show that substituted amino groups OAR-Ns hardly get interaction with the halide vacancy due to the distribution of charge density in Sapanisertib.Our work suggests that the selective coordination is of great significance for the design of high-performance passivators for printable mesoscopic perovskite solar cells.Dun Ma Xiujun He Jingshan He Huanxin Guo Yongzhen Wu Wenjun Wu Weihong Zhu 2023Chinese Chemical Letters2023,34,12:0
14Targeting LncRNA LLNLR-299G3.1 with antisense oligonucleotide inhibits malignancy of esophageal squamous cell carcinoma cells in vitro and in vivo显示文摘Accumulating evidence has indicated that long non-coding RNAs(lncRNAs)play critical roles in the development and progression of cancers,including esophageal squamous cell carcinoma(ESCC).However,the mechanisms of lncRNAs in ESCC are still incompletely understood and therapeutic attempts for in vivo targeting cancer-associated lncRNA remain a challenge.By RNA-sequencing analysis,we identified that LLNLR-299G3.1 was a novel ESCC-associated lncRNA.LLNLR-299G3.1 was up-regulated in ESCC tissues and cells and promoted ESCC cell proliferation and invasion.Silencing of LLNLR-299G3.1 with ASO(antisense oligonucleotide)resulted in opposite effects.Mechanistically,LLNLR-299G3.1 bound to cancerassociated RNA binding proteins and regulated the expression of cancer-related genes,including OSM,TNFRSF4,HRH3,and SSTR3.ChIRP-seq(chromatin isolation by RNA purification and sequencing)revealed that these genes contained enriched chromatin binding sites for LLNLR-299G3.1.Rescue experiments confirmed that the effects of LLNLR-299G3.1 on ESCC cell proliferation were dependent on interaction with HRH3 and TNFRSF4.Therapeutically,intravenous delivery of placental chondroitin sulfate A binding peptide-coated nanoparticles containing antisense oligonucleotide(pICSA-BP-ANPs)strongly inhibited ESCC tumor growth and significantly improved animal survival in vivo.Overall,our results suggest that LLNLR-299G3.1 promotes ESCC malignancy through regulating gene-chromatin interactions and targeting ESCC by pICSA-BP-ANPs may be an effective strategy for the treatment of lncRNA-associated ESCC.LI TIAN YONGYI HUANG BAOZHEN ZHANG YI SONG LIN YANG QIANQIAN CHEN ZHENG WANG YILING WANG QIHAN HE WENHAN YANG SHUYONG YU TIANYU LU ZICHEN LIU KAIPING GAO XIUJUN FAN JIAN SONG RIHONG ZHAI 2023Oncology Research2023,31,4:0
