| 1 | Paracrine and endocrine actions of bone——the functions of secretory proteins from osteoblasts, osteocytes, and osteoclasts显示文摘The skeleton is a dynamic organ that is constantly remodeled. Proteins secreted from bone cells, namely osteoblasts, osteocytes,and osteoclasts exert regulation on osteoblastogenesis, osteclastogenesis, and angiogenesis in a paracrine manner. Osteoblasts secrete a range of different molecules including RANKL/OPG, M-CSF, SEMA3A, WNT5A, and WNT16 that regulate osteoclastogenesis. Osteoblasts also produce VEGFA that stimulates osteoblastogenesis and angiogenesis. Osteocytes produce sclerostin(SOST) that inhibits osteoblast differentiation and promotes osteoclast differentiation. Osteoclasts secrete factors including BMP6, CTHRC1, EFNB2, S1P, WNT10B, SEMA4D, and CT-1 that act on osteoblasts and osteocytes, and thereby influencea A osteogenesis. Osteoclast precursors produce the angiogenic factor PDGF-BB to promote the formation of Type H vessels, which then stimulate osteoblastogenesis. Besides, the evidences over the past decades show that at least three hormones or 'osteokines'from bone cells have endocrine functions. FGF23 is produced by osteoblasts and osteocytes and can regulate phosphate metabolism. Osteocalcin(OCN) secreted by osteoblasts regulates systemic glucose and energy metabolism, reproduction, and cognition. | 4 | Principles and technology for stepwise utilization of resources for mitigating deep mine heat hazards显示文摘As is well known, deep mines are hot. As mining depth increases, the temperature of the surrounding rock also increases. This seriously affects mine safety and production and has restricted the exploitation of deep coal resources. Therefore, reducing the working face temperature to improve working conditions by controlling these heat hazards is an urgent problem. Considering problems in cooling deep mines both domestically and abroad along with the actual conditions of the Zhangshuanglou coal mine, we propose a HEMS technology that uses heat resources from deep mines in a stepwise manner. HEMS means a high temperature ex-change machinery system. Mine inrush-water is used as a source of cooling. Twice the energy is extracted from the mine inrush water. Heat is used for building heating in the winter and cold water is used for cooling buildings in the summer. This opens a new technology for stepwise utilization of heat energy in deep mines. Energy conservation and reduced pollution, an improved environment and sustainable economic development are realized by this technique. | 5 | Metabolic engineering and flux analysis of Corynebacterium glutamicum for L-serine production显示文摘L-Serine plays a critical role as a building block for cell growth, and thus it is difficult to achieve the direct fermentation of L-serine from glucose. In this study, Corynebacterium glutamicum ATCC 13032 was engineered de novo by blocking and attenuating the conversion of L-serine to pyruvate and glycine, releasing the feedback inhibition by L-serine to 3-phosphoglycerate dehydrogenase (PGDH), in combination with the co-expression of 3-phosphoglycerate kinase (PGK) and feedback-resistant PGDH (PGDHr). The resulting strain, SER-8, exhibited a lower specific growth rate and significant differences in L-serine levels from Phase I to Phase V as determined for fed-batch fermentation. The intracellular L-serine pool reached (14.22±1.41) μmol gCDM 1, which was higher than glycine pool, contrary to fermentation with the wild-type strain. Furthermore, metabolic flux analysis demonstrated that the over-expression of PGK directed the flux of the pentose phosphate pathway (PPP) towards the glycolysis pathway (EMP), and the expression of PGDHr improved the L-serine biosynthesis pathway. In addition, the flux from L-serine to glycine dropped by 24%, indicating that the deletion of the activator GlyR resulted in down-regulation of serine hydroxymethyltransferase (SHMT) expression. | 6 | Canopy interception loss in a Pinus sylvestris var. mongolica forest of Northeast China显示文摘Pinus sylvestris var. mongolica is one of the main species to be afforested in deserts of China. But little work has been carried out on the canopy interception loss of this plant species. For researching the canopy interception loss of a natural P. sylvestris forest, we observed the gross precipitation, gross snowfall, throughfall and stemflow in a sample plot at the Forest Ecosystem Research Station of Mohe in the Great Khingan Mountains of Northeast China from July 2012 to September 2013. Considering the spatial variability of the throughfall, we increased the area rather than the number of collector and randomly relocated them once a week. The results demonstrated that the throughfall, stemflow, and derived estimates of rainfall and snowfall interception loss during the main rainy and snowy seasons were 77.12%±5.70%, 0.80%, 22.08%±5.51% and 21.39%±1.21% of the incident rainfall or snowfall, respectively. The stemflow didn't occur unless the accumulated rainfall reached up to 4.8 mm. And when the gross precipitation became rich enough, the stemflow increased with increasing tree diameters. | 11 | A transgene-free method for rapid and efficient generation of precisely edited pigs without monoclonal selection显示文摘Gene-edited pigs for agricultural and biomedical applications are typically generated using somatic cell nuclear transfer(SCNT).However, SCNT requires the use of monoclonal cells as donors, and the time-consuming and laborious monoclonal selection process limits the production of large populations of gene-edited animals. Here, we developed a rapid and efficient method named RE-DSRNP(reporter RNA enriched dual-sg RNA/CRISPR-Cas9 ribonucleoproteins) for generating gene-edited donor cells. RE-DSRNP takes advantage of the precise and efficient editing features of dual-sg RNA and the high editing efficiency, low off-target effects, transgene-free nature, and low cytotoxic characteristics of reporter RNA enriched RNPs(CRISPR-Cas9ribonucleoproteins), thus eliminating the need for the selection of monoclonal cells and thereby greatly reducing the generation time of donor cells from 3–4 weeks to 1 week, while also reducing the extent of apoptosis and chromosomal aneuploidy of donor cells. We applied RE-DSRNP to produce cloned pigs bearing a deletion edit of the wild-type p53-induced phosphatase 1(WIP1)gene: among 32 weaned cloned pigs, 31(97%) carried WIP1 edits, and 15(47%) were homozygous for the designed fragment deletion, and no off-target event was detected. The WIP1 knockout(KO) pigs exhibited male reproductive disorders, illustrating the utility of RE-DSRNP for rapidly generating precisely edited animals for functional genomics and disease research. Development of cultivars with improved P-deficiency tolerance is an efficient strategy for sustainable agriculture. Plant roots play an important role in crop growth and development, especially in nutrient uptake and improvement of P-efficiency. Mapping quantitative trait loci (QTLs) for root traits and their response to low P stress at seedling stage will facilitate the development of P-efficient wheat cultivars. In this study, 30 QTLs (LOD>2.0) were mapped for the three root traits, such as root length, root number and root dry matter under different P supply conditions and their response to P-stress. These QTLs were distributed on 14 chromosomes, with each of the 5 QTLs explaining more than 10% phenotype variance. Analyses showed that root traits and their response to P-deficiency were controlled by different QTLs. | 14 | Natural continuous influent nitrifier immigration effects on nitrification and the microbial community of activated sludge systems显示文摘Two sequencing batch reactors(SBRs) were operated for 100 days under aerobic conditions,with one being fed with unsterilized municipal wastewater(USBR), and the other fed with sterilized municipal wastewater(SSBR). Respirometric assays and fluorescence in situ hybridization(FISH) results show that active nitrifiers were present in the unsterilized influent municipal wastewater. The maximum ammonia utilization rate(AUR) and nitrite utilization rate(NUR) of the unsterilized influent were 0.32 ± 0.12 mg NH4+-N/(L·hr) and0.71 ± 0.18 mg NO2--N/(L·hr). Based on the maximum utilization rates, the estimated seeding intensity for the ammonia oxidizing bacteria(AOB) and nitrite oxidizing bacteria(NOB) of the USBR was 0.08 g AOB/(g AOB·day) and 0.20 g NOB/(g NOB·day) respectively. The fraction of nitrifiers/total bacteria in the influent was 5.35% ± 2.1%, the dominant AOB was Nitrosomonas spp., Nitrosococcus mobilis hybridizated with Nsm156, and the dominant NOB was Nitrospira hybridizated with Ntspa662. The influent nitrifiers potentially seeded the activated sludge of the bioreactor and hence demonstrated a mitigation of the acclimatization times and instability during start-up and early operation. The AUR and NUR in the USBR was 15% and 13% higher than the SSBR respectively during the stable stage, FISH results showed that nitrifiers population especially the Nitrospira in the USBR was higher than that in the SSBR. | 15 | Spatial and Temporal Variability of Thermal Stress to China's Coral Reefs in South China Sea显示文摘Coral bleaching, caused by elevated sea surface temperature(SST), is occurring more frequently and seriously worldwide. Due to the lack of field observations, we understand little about the large-scale variability of thermal stress in the South China Sea(SCS) and its effect on China's coral reefs. This paper used 4-km high resolution gap-filled SST(Filled SST) data and thermal stress data related to coral bleaching derived from Coral Reef Temperature Anomaly Database(Co RTAD) to quantify the spatial and temporal characteristics of chronic thermal stress and acute thermal stress to China's coral reefs in SCS from 1982 to 2009. We analyzed the trend of SST in summer and the thermal stress frequency, intensity and duration during this period. The results indicate that, as a chronic thermal stress, summer mean SST in SCS shows an average upward trend of 0.2℃/decade and the spatial pattern is heterogeneous. Waters of Xisha Islands and Dongsha Islands of the northern SCS are warming faster through time compared to Zhongsha Islands and Nansha Islands sea areas of the southern SCS. High frequency bleaching related thermal stress events for these reefs are seen in the area to the northwest of Luzon Island. Severe anomaly thermal stress events are more likely to occur during the subsequent year of the El Nino year for these coral reefs. | 17 | Research on sintering process of YSZ electrolyte显示文摘Yttria stabilized zirconia (YSZ) has widely been used as electrolyte in solid oxide fuel cell (SOFC). The microstructure of YSZ related to the fabrication process was discussed in the paper. With YSZ nano-powders about 40-100 nm as raw material, the YSZ adobe was manufactured by tape calendering process. The named three-step sintering process was performed at 1000 ℃ for 2 h, then raised the temperature with normal rate and as soon as up to 1400 ℃, the furnace was controlled at 1250-1300 ℃ for 10-20 h. The high dense YSZs with the relative density of 96%-99% were obtained; the grain size of YSZ could be reduced to 0.5-3 μm. | 18 | Designing near-natural planting patterns for plantation forests in China显示文摘Background: China has a long tradition of managing planted forests. Different species of Populus, Eucalyptus, Larix, Cunninghamia and Pinus are planted to satisfy the local demand for wood products and provide ecological services at the same time. Evidence of the greater resilience of natural forests provides the motivation to develop asymmetric planting patterns, which is the focus of this study. We present a new method for designing plantation patterns that follow those observed in natural ecosystems and to maintain some regularity for operational convenience. Methods: Based on the uniform angle index, we analyzed the spatial structure of six natural forests in different regions of China. The uniform angle index describes the degree of spatial uniformity of the n nearest neighbors of a given reference tree. Accordingly, we identified all possible patterns of a neighborhood group within a regular planting pattern and developed a method to optimize planting point arrangements that contain some randomness as well as a minimum degree of regularity. Results:(1) There are 13 types of structural units in a regular planting, including seven random units, five even units and one cluster unit;(2) Five near-natural arrangements are presented with a minimum proportion of 50% of random units. These five arrangements represent a combination of regularity for operational convenience and asymmetry. Conclusions: The new planting patterns developed in this study are expected to increase the asymmetric competition and resilience of these important ecosystems. These extracts contain pseudo- homologous families of 5,5-diethylalkanes (5,5- DEAs), 6,6-diethylalkanes, 5-butyl-5-ethylalkanes and 6-butyl-6-ethylalkanes, which are first detected in modern soils. All these pseudohomologous series display exclusively odd or exclusively even carbon numbers, clearly indicating a biosynthetic origin, and their carbon number distributions generally covary. The distributions of 5,5-DEAs exhibit strong regional variations, correlated with the spatial distributions of climate and vegetation. The lower molecular weight 5,5-DEAs, maximizing at C21 or C23, dominate in southeast and southwest China samples, while the higher molecular weight 5,5-DEAs with a mode at C29 dominate in cold, arid northwest China and Mongolia. | 20 | Mechanical regulation of bone remodeling显示文摘Bone remodeling is a lifelong process that gives rise to a mature, dynamic bone structure via a balance between bone formation by osteoblasts and resorption by osteoclasts. These opposite processes allow the accommodation of bones to dynamic mechanical forces, altering bone mass in response to changing conditions. Mechanical forces are indispensable for bone homeostasis;skeletal formation, resorption, and adaptation are dependent on mechanical signals, and loss of mechanical stimulation can therefore significantly weaken the bone structure, causing disuse osteoporosis and increasing the risk of fracture. The exact mechanisms by which the body senses and transduces mechanical forces to regulate bone remodeling have long been an active area of study among researchers and clinicians. Such research will lead to a deeper understanding of bone disorders and identify new strategies for skeletal rejuvenation.
