| 4 | Numerical simulation of bubble plumes in overlying water of gas hydrate in the cold seepage active region显示文摘To study the seismic responses produced by gas hydrate bubble plumes in the cold seepage active region, we constructed a plume water body model based on random medium theory and acoustic velocity model of bubble medium. The plume water body model was forward simulated by finite difference. Seismic records of single shot show the scattered waves produced by the plume. The scattered wave energy is strong where the plume exists. Where the scattered wave energy is stronger, the minimum of travel time is always above the plume, which has no relationship with the shot's position. Seismic records of shot gathers were processed by prestack time migration. The migration section shows that the scattered waves produced by plumes can be imaged distinctly with higher accuracy. | 7 | Gene diagnosis and targeted breeding for blast-resistant Kongyu 131without changing regional adaptability显示文摘The fungus Magnaporthe oryzae threatens the rice production of Kongyu 131 (KY131),a leading japonica variety in Northeast China.In this study,two rice lines,KP1 and KP2-Hd1,were obtained by introgressing the blast resistance genes Pi1 and Pi2 into KY131,respectively.However,both lines headed later than KY131.RICE60K SNP array analysis showed that Hd1 closely linked to Pi2 was introgressed into KP2-Hd1,and the linkage drag of Hd1 was broken by recombination.On the other hand,no known flowering genes were introgressed into KP1.Gene diagnosis by resequencing six flowering genes showed that KP1 carried functional Hd16 and Ghd8 alleles.Due to its suppression role in heading under long-day conditions,Ghd8 was chosen as the target for gene editing to disrupt its function.Four sgRNAs targeting different sites within Ghd8 were utilized to induce large-deletion mutations,which were easy to detect via agarose gel electrophoresis.All the ghd8-mutated KP1 lines were resistant to rice blast disease and headed earlier than the control KP1,even than KY131,under natural long-day conditions,which ensures its growth in Northeast China.This study confirmed that a combination of gene diagnosis and targeted gene editing is a highly efficient way to quickly eliminate undesired traits in a breeding line. | Xiangchun Zhou Gonghao Jiang Longwei Yang Lei Qiu Ping He Chunxiao Nong Yunyue Wang Yuqing He Yongzhong Xing | 2018 | Journal of Genetics and Genomics2018,45,10: | 7 |

| 9 | Global analysis of CCT family knockout mutants identifies four genes involved in regulating heading date in rice显示文摘Many genes encoding CCT domain-containing proteins regulate flowering time. In rice(Oryza sativa), 41 such genes have been identified, but only a few have been shown to regulate heading date. Here, to test whether and how additional CCT family genes regulate heading date in rice, we classified these genes into five groups based on their diurnal expression patterns. The expression patterns of genes in the same subfamily or in close phylogenetic clades tended to be similar. We generated knockout mutants of the entire gene family via CRISPR/Cas9. The heading dates of knockout mutants of only 4 of 14 genes previously shown to regulate heading date were altered, pointing to functional redundancy of CCT family genes in regulating this trait. Analysis of mutants of four other genes showed that OsCCT22, OsCCT38, and OsCCT41 suppress heading under long-day conditions and promote heading under short-day conditions. OsCCT03 promotes heading under both conditions and upregulates the expression of Hd1 and Ehd1, a phenomenon not previously reported for other such genes. | 10 | Assessment of the MT1-MMP expression level of different cell lines by the naked eye显示文摘Membrane type 1 matrix metalloproteinase(MT1-MMP) is expressed in most tumors and is believed to play a key role in the development, invasion and metastasis of tumors. There is an urgent need to develop a simple method to detect the MT1-MMP expression level on cells. In this current study, we demonstrated a red emission Au cluster probes with the specific targeting of MT1-MMP on human dopaminergic neuroblastoma(SH-SY5 Y) cells. More importantly, utilizing the intrinsic enzyme-like activity of the Au cluster probes, the expression level of MT1-MMP on the SH-SY5 Y cells could be assessed by the naked eye without cell lysis and protein extraction process. Furthermore, SH-SY5 Y, human breast cancer(MCF-7), and human bronchial epithelial(16-HBE) cell lines with different MT1-MMP expression level could be distinguished using the Au cluster probes by the naked eye. Meanwhile, fluorescence intensity and Au count determined by inductively coupled plasma mass spectrometry(ICP-MS) were used to verify the feasibility of this simple analytical method. Creep tests of coal filled with 0 and 0.5 MPa gas were performed, and strain under different axial stress was compared.The three creep constitutive models which were analysed using the method fitting experimental data for determining which creep model can reflect the creep process of the test best. | 16 | BSA-seq-based identification of a major additive plant height QTL with an effect equivalent to that of Semi-dwarf 1 in a large rice F_(2) population显示文摘Bulked-segregant analysis is a time-and cost-saving strategy for identifying major quantitative trait loci(QTL) in a mapping population. Bulked-segregant analysis combined with whole-genome sequencing(BSA-seq) was performed to rapidly identify QTL for heading date, plant height, and panicle length in a large F_(2) population derived from two landraces: Chuan 7(C7) and Haoboka(HBK). Twenty plants with extremely low or high phenotypic values for the target traits were selected from an F_(2) population of 940 plants to construct low-and high-value bulks. Three pairs of bulks for the three traits were constructed, resulting in six DNA pools. BSA-seq revealed nine QTL: four for heading date, three for plant height, and two for panicle length. These QTL were validated in a random F_(2) population or BC_(4)F_(2) populations. The major novel plant height QTL, qPH8, acting additively with an effect equivalent to that of semi-dwarf 1(sd1), is potentially valuable for hybrid rice breeding. qPH8 controls mainly the elongation of basal internodes. | 19 | Biodegradation of polycyclic aromatic hydrocarbons in sediments from the Daliao River watershed, China显示文摘The Daliao River, as an important water system in Northeast China, was reported to be heavily polluted by polycyclic aromatic hydrocarbons (PAHs). Aerobic biodegradations of four selected PAHs (naphthalene, phenanthrene, uorene and anthracene) alone or in their mixture in river sediments from the Daliao River water systems were studied in microcosm systems. Eects of additional carbon source, inorganic nitrogen and phosphorus, temperature variation on PAHs degradation were also investigated. Results showed that the degradation of phenanthrene in water alone system was faster than that in water-sediment combined system. Degradation of phenanthrene in sediment was enhanced by adding yeast extract and ammonium, but retarded by adding sodium acetate and not significantly inuenced by adding phosphate. Although PAHs could also be biodegraded in sediment under low temperature (5°C), much lower degradation rate was observed. Sediments from the three main streams of the Daliao River water system (the Hun River, the Taizi River and the Daliao River) demonstrated dierent degradation capacities and patterns to four PAHs. Average removal rates (15 or 19 d) of naphthalene, phenanthrene, uorene and anthracene by sediment were in the range of 0.062-0.087, 0.005-0.066, 0.008- 0.016 and 0-0.059 mg/(L·d), respectively. As a result, naphthalene was most easily degraded compound, anthracene was the hardest one.
