1Orexin A attenuates the sleep-promoting effect of adenosine in the lateral hypothalamus of rats显示文摘Orexin neurons within the lateral hypothalamus play a crucial role in the promotion and maintenance of arousal. Studies have strongly suggested that orexin neurons are an important target in endogenous adenosine-regulated sleep homeostasis. Orexin A induces a robust increase in the firing activity of orexin neurons, while adenosine has an inhibitory effect. Whether the excitatory action of orexins in the lateral hypothalamus actually promotes wakefulness and reverses the sleep-producing effect of adenosine in vivo is less clear. In this study,electroencephalographic and electromyographic recordings were used to investigate the effects of orexin A and adenosine on sleep and wakefulness in rats. We found that microinjection of orexin A into the lateral hypothalamus increased wakefulness with a concomitant reduction of sleep during the first 3 h of post-injection recording, and this was completely blocked by a selective antagonist for orexin receptor 1,SB 334867. The enhancement of wakefulness also occurred after application of the excitatory neurotransmitter glutamate in the first 3 h post-injection. However, in the presence of the NMDA receptor antagonist APV, orexin A did not induce any change of sleep and wakefulness in the first 3 h.Further, exogenous application of adenosine into the lateral hypothalamus induced a marked increase of sleep in the first 3-h post-injection. No significant change in sleep and wakefulness was detected after adenosine application followed by orexin A administration into the same brain area. 3Experimental and density functional theory computational evaluation of poly(N-vinyl caprolactam-co-butyl methacrylate) kinetic hydrate inhibitors显示文摘Natural gas hydrate inhibitor has been serving the oil and gas industry for many years. The development and search for new inhibitors remain the focus of research. In this study, the solution polymerization method was employed to prepare poly(N-vinyl caprolactam-co-butyl methacrylate)(P(VCap-BMA)), as a new kinetic hydrate inhibitor(KHI). The inhibition properties of P(VCap-BMA) were investigated by tetrahydrofuran(THF) hydrate testing and natural gas hydrate forming and compared with the commercial KHIs. The experiment showed that PVCap performed better than copolymer P(VCap-BMA). However,low doses of methanol or ethylene glycol are compounded with KHIs. The compounding inhibitors show a synergistic inhibitory effect. More interesting is the P(VCap-BMA)-methanol system has a better inhibitory effect than the PVCap-methanol system. 1% P(VCap-BMA) + 5% methanol presented the best inhibiting performance at subcooling 10.3 °C, the induction time of natural gas hydrate was 445 min.Finally, the interaction between water and several dimeric inhibitors compared by natural bond orbital(NBO) analyses and density functional theory(DFT) indicated that inhibitor molecules were able to form the hydrogen bond with the water molecules, which result in gas hydrate inhibition. 13Ca^(2+)-dependent TaCCD1 cooperates with TaSAUR215 to enhance plasma membrane H^(+)-ATPase activity and alkali stress tolerance by inhibiting PP2C-mediated dephosphorylation of TaHA2 in wheat显示文摘Alkali stress is a major constraint for crop production in many regions of saline-alkali land.However,little is known about the mechanisms through which wheat responds to alkali stress.In this study,we identified a calcium ion-binding protein from wheat,TaCCD1,which is critical for regulating the plasma membrane(PM)H^(+)-ATPase-mediated alkali stress response.PM H+-ATPase activity is closely related to alkali tolerance in the wheat variety Shanrong 4(SR4).We found that two D-clade type 2C protein phosphatases,TaPP2C.D1 and TaPP2C.D8(TaPP2C.D1/8),negatively modulate alkali stress tolerance by dephosphorylating the penultimate threonine residue(Thr926)of TaHA2 and thereby inhibiting PM H+-ATPase activity.Alkali stress induces the expression of TaCCD1 in SR4,and TaCCD1 interacts with TaSAUR215,an early auxin-responsive protein.These responses are both dependent on calcium signaling triggered by alkali stress.TaCCD1 enhances the inhibitory effect of TaSAUR215 on TaPP2C.D1/8 activity,thereby promoting the activity of the PM H^(+)-ATPase TaHA2 and alkali stress tolerance in wheat.Functional and genetic analyses verified the effects of these genes in response to alkali stress,indicating that TaPP2C.D1/8 function downstream of TaSAUR215 and TaCCD1.Collectively,this study uncovers a new signaling pathway that regulates wheat responses to alkali stress,in which Ca^(2+)-dependent TaCCD1 cooperates with TaSAUR215 to enhance PM H+-ATPase activity and alkali stress tolerance by inhibiting TaPP2C.D1/8-mediated dephosphorylation of PM H+-ATPase TaHA2 in wheat.Minghan Cui Yanping Li Jianhang Li Fengxiang Yin Xiangyu Chen Lumin Qin Lin Wei Guangmin Xia Shuwei Liu 2023Molecular Plant2023,16,3:1

17Maternal butyrate supplementation affects the lipid metabolism and fatty acid composition in the skeletal muscle of offspring piglets显示文摘Maternal sodium butyrate(SB)intake has important effects on offspring growth and development.This study aimed to investigate the impacts of maternal SB supplementation during gestation and lactation on fatty acid composition and lipid metabolism in the offspring skeletal muscle of pigs.Twenty sows(Yorkshire,parity 2 to 3)were assigned to the control group(diets without SB,n=10)and SB group(diets with 0.1%SB,n=10).The results showed maternal SB supplementation throughout gestation and lactation increased(P<0.05)body weight of offspring piglets at weaning.The concentrations of triglyceride in plasma and milk were enhanced(P<0.05).Maternal SB induced(P<0.05)lipid accumulation with increased expression of peroxisome proliferator activated receptorγ(PPARγ)by enrichment of the acetylation of H3 acetylation K27(H3 K27)in offspring skeletal muscle.Meanwhile,the concentrations of C18:2 n-6,C18:3 n-3,total polyunsaturated fatty acid(PUFA),n-6 PUFA and n-3 PUFA decreased(P<0.05)in skeletal muscle of weaning piglets derived from SB sows.Together,these results showed that maternal SB supplementation could influence offspring growth performance,lipid metabolism and fatty acid composition of the skeletal muscle.Yongsen Zhao Danping Wang Yanping Huang Dangdang Wu Xiaoming Ji Xiaobing Zhou Dong Xia Xiaojing Yang 2021Animal Nutrition2021,,4:1

19Jumonji domain-containing 6 (JMJD6) identified as a potential therapeutic target in ovarian cancer显示文摘Jumonji domain-containing 6(JMJD6)is a candidate gene associated with tumorigenesis,and JMJD6 overexpression predicts poor differentiation and unfavorable survival in some cancers.However,there are no studies reporting the expression of JMJD6 in ovarian cancer,and no JMJD6 inhibitors have been developed and applied to targeted cancer therapy research.In the present study,we found that the high expression of JMJD6 in ovarian cancer was correlated with poor prognosis in ovarian cancer.A potential inhibitor(SKLB325)was designed based on the crystal structure of the jmjC domain of JMJD6.This molecule significantly suppressed proliferation and induced apoptosis in a dose-dependent manner in SKOV3 cell lines as detected by CCK-8 cell proliferation assays and flow cytometry.A Matrigel endothelial tube formation assay showed that SKLB325 inhibited capillary tube organization and migration in HUVECs in vitro.We also observed that JMJD6 colocalized with p53 protein in the nucleus,with mRNA and protein expression of p53 as well as its downstream effectors significantly increasing both in vitro and in intraperitoneal tumor tissues treated with SKLB325.In addition,SKLB325 significantly reduced the intraperitoneal tumor weight and markedly prolonged the survival of tumor-bearing mice.Taken together,our findings suggest that JMJD6 may be a marker of poor prognosis in ovarian cancer and that SKLB325 may be a potential candidate drug for the treatment of ovarian cancer.Heng Zheng Yan Tie Zhen Fang Xiaoai Wu Tao Yi Shuang Huang Xiao Liang Yanping Qian Xi Wang Ruyu Pi Siyuan Chen Yong Peng Shengyong Yang Xia Zhao Xiawei Wei 2019Signal Transduction and Targeted Therapy2019,4,1:1
20Incremental QR-based tensor-train decomposition for industrial big data显示文摘Industrial big data was usually multi-source, heterogeneous, and deeply intertwined. It had a wide range of data sources, high data dimensions, and strong data correlation. In order to effectively analyze and process streaming industrial big data generated by edge computing, it was very important to provide an effective real-time incremental data method. However, in the process of incremental processing, industrial big data incremental computing faced the challenges of dimensional disaster, repeated calculations, and the explosion of intermediate results. Therefore, in order to solve the above problems effectively, a QR-based tensor-train(TT) decomposition(TTD) method and a QR-based incremental TTD(QRITTD) method were proposed. This algorithm combined the incremental QR-based decomposition algorithm with an approximate singular value decomposition(SVD) algorithm and had good scalability. In addition, the computational complexity, space complexity, and approximation error analysis were analyzed in detail. The effectiveness of the three algorithms of QRITTD, non-incremental TTD(NITTD), and TT rank-1(TTr1) SVD(TTr1 SVD)were verified by comparison.
