1Multimodality treatment in hepatocellular carcinoma patients with tumor thrombi in portal vein显示文摘AIM To compare the therapeutic effect andsignificances of multimodality treatment forhepatocellular carcinoma (HCC) with tumorthrombi in portal vein (PVTT).METHODS HCC patients (n = 147) with tumortrombi in the main portal vein or the first branchof portal vein were divided into four groups bythe several therapeutic methods. There wereconservative treatment group in 18 out ofpatients (group A); and hepatic artery ligation(HAL) and/or hepatic artery infusion (HAl)group in 18 patients (group B), in whompostoberative chemoembolization was doneperiodically; group of removal of HCC with PVTTin 79 (group C) and group of transcatheterhepatic arterial chemoembolization (TACE) orHAl and/or portal vein infusion (PVI) afteroperation in 32 (group D).RESULTS The median survival period was 12months in our series and the 1-, 3-, and 5-yearsurvival rates were 44.3%, 24.5% and 15.2%,respectively. The median survival times were 2,5, 12 and 16 months in group A, B, C and D,respectively. The 1-, 3- and 5-year survival rateswere 5.6%, 0% and 0% in group A; 22.2%,5.6% and 0% in group B; 53.9%, 26.9% and16.6% in group C; 79.3%, 38.9% and 26.8% ingroup D, respectively. 2Morphological study on colonic pathology in ulcerative colitis treated by moxibustion显示文摘AIM To observe the therapeutic effect ofmoxibustion on ulcerative colitis and itsinfluence on the colonic mucosal morphology.METHODS Forty-six patients with ulcerativecolitis were randomly divided into themoxibustion with herbal medicine underneathgroup and the western medicine group.Thirtypatients were treated with the abovemoxibustion and 16 patients with Salicylayefapyridine(SASP).The colonic mucosa of 13patients in the moxibustion group was observedby colonoscopy before and after the treatment.Mucin was also analyzed by H.E and AB-PASstaining.RESULTS Seventeen patients were clinicallycured,12 were improved and 1 unchanged in themoxibustion group.In the control group,5patients were clinically cured,7 improved and 4unchanged.Thirteen patients with active UCwere taken as the subjects for histopathologicanalysis in this study.The colonic mucosallesions were remarkably improved and thecharacteristic of the mucin also changed.Inmost sections,the chronic inflammation ofmucosa was geatly ameliorated(P<0.01).Theinflammatory cell infiltratation much decreased and neutrophils,disapeared in most sections(P<0.001).The goblet cells significantlyincreased(P<0.001);crypt paracrypt abscessor mucosal ulceration was seen(P<0.001).CONCLUSION The rate of cure of ulcerative colitis by moxibustion with herbal medicine beneath is superior to that by SASP. 4SHRIMP U-Pb Zircon Geochronology of the Xiaotongguanshan and Shatanjiao Intrusions and Its Petrological Implications in the Tongling Area, Anhui显示文摘This paper determines the crystallization ages of the Xiaotongguanshan quartz monzodiorite and Shatanjiao quartz monzonite porphyry from the Tongling area using the SHRIMP zircon U-Pb method. The crystallization age of the former is 142.8±1.8 Ma; that of the latter is 151.8±2.6 Ma. These data indicate that they were formed during the Late Jurassic (142.8 to 151.8 Ma). Zoned magma chamber was formed because of double diffusive convection. 6Neutrophil extracellular traps mediate the crosstalk between glioma progression and the tumor microenvironment via the HMGB1/RAGE/IL-8 axis显示文摘Objective:Neutrophil extracellular traps(NETs)produced by tumor-infiltrating neutrophils(TINs)are associated with poor prognosis in patients with several types of cancer.However,the mechanisms underlying the involvement of NETs in glioma progression remain largely unknown.This study aimed to elucidate the roles of NETs in biological processes that drive the crosstalk between glioma progression and the tumor microenvironment.Methods:Neutrophil infiltration and NETs formation were investigated in glioma tissue through immunohistochemistry,and their relationships with clinicopathological features and outcomes were statistically evaluated.The effects of NETs on glioma cell progression were studied in a co-culture system.In vivo and in vitro experiments validated the reactive oxygen species activity and cytokine production of TINs,as well as the ERK signaling pathway activation and the metastasis of gliomas.Results:Neutrophil infiltration and NETs formation were induced in high-grade glioma compared with low-grade glioma.NETs induced by TINs were determined to be an oncogenic marker of high-grade gliomas and to be involved in cell proliferation and invasion.NETs overproduction promoted glioma cell proliferation,migration,and invasion.Furthermore,HMGB1 was found to bind to RAGE and activate the NF-κB signaling pathway in vitro.In addition,NETs stimulated the NF-κB signaling pathway,thus promoting IL-8 secretion in glioblastoma.Subsequently,IL-8 recruited neutrophils which in turn mediated NETs formation via the PI3 K/AKT/ROS axis in TINs.Conclusions:Our results suggest that NETs produced by TINs mediate the crosstalk between glioma progression and the tumor microenvironment by regulating the HMGB1/RAGE/IL-8 axis.Targeting NETs formation or IL-8 secretion may be an effective approach to inhibit glioma progression.Caijun Zha Xiangqi Meng Lulu Li Shan Mi Da Qian Ziwei Li Pengfei Wu Shaoshan Hu Shihong Zhao Jinquan Cai Yanhong Liu 2020Cancer Biology & Medicine2020,17,1:19
7Post-Mesozoic Transformation of Tectonic Domain in Southeastern China and Its Geodynamic Mechanism显示文摘Since the Mesozoic and Cenozoic, a transformation from a Tethyan Himalayan tectonic domain into a circum Pacific tectonic domain from Indosinian to Yanshanian is indicated in this paper, resulting in conspicuous changes in geophysics, tectono magmatic distribution, lithofacies and paleo geography, tectonic system in southeastern China. Tectonic analysis shows that the tectonic framework resulted from the compounding, transforming and superimposing of the two tectonic domains. 8The Early Jurassic magmatism in northern Guangdong Province, southeastern China: Constraints from SHRIMP zircon U-Pb dating of Xialan complex显示文摘This paper reports SHRIMP zircon U-Pb ages of 196±2 Ma for granite, and 195±1 Ma for gabbro from the Xialan complex in the Meizhou area, northern Guangdong Province. These results shed new light on the calm stage of magmatic activity in southeastern China during 200-180 Ma, and revealed that the back-arc extension induced by the subduction of the western Pacific plate may have begun at 195 Ma at least. Field observation on the fresh outcrops allows us to recognize some features formed by magma mixing. A part of the gabbro has a fine-grained rim of 20-30 cm at the margin, and thins gradually to-ward the granite; numerous dark fine-grained to microcrystalline dioritic enclaves developed in the granite. These enclaves vary in shape and size, dark minerals concentrated at the margin of enclaves, and the contact between enclaves and host rock is either obvious or obscure, or gradational. In addi-tion, needle-shaped apatites are included in the enclaves. The REE patterns of gabbros, as well as the trace element patterns, are generally consistent with those of granitic rocks. 9Clinical significance of telomerase activity in peritoneal lavage fluid from patients with gastric cancer and its relationship with cellular proliferation显示文摘AIM: To evaluate the efficacy of telomerase activity assay and peritoneal lavage cytology (PLC) examination in peritoneal lavage fluid for the prediction of peritoneal metastasis in gastric cancer patients, and to explore the relationship between telomerase activity and proliferating cell nuclear antigen expression. METHODS: Telomeric repeated amplification protocol (TRAP)-enzyme-linked immunosorbent assay (ELISA) was performed to measure the telomerase activity in 60 patients with gastric cancer and 50 with peptic ulcer. PLC analysis of the 60 patients with gastric cancer was used for comparison. The proliferating cell nuclear antigen (PCNA) in gastric carcinoma was immunohistochemically examined. RESULTS: The telomerase activity and PLC positive rate in peritoneal lavage fluid from patients with gastric cancer was 41.7/ (25/60), and 25.0/ (15/60), respectively. The positive rate of telomerase activity was significantly higher than that of PLC in the group of pT4 (15/16 vs 9/16, P < 0.05), P1-3 (13/13 vs 9/13, P < 0.05) and diffuse type (22/42 vs 13/42, P < 0.05). The patients with positive telomerase activity, peritoneal metastasis, and serosal invasion had signifi cantly higher levels of average PCNA proliferation index (PI), (55.00 ± 6.59 vs 27.43 ± 7.72, 57.26 ± 10.18 vs 29.15 ± 8.31, and 49.82 ± 6.74 vs 24.65 ± 7.33, respectively, P < 0.05).CONCLUSION: The TRAP assay for telomerase activity is a useful adjunct for cytologic method in the diagnosis of peritoneal micrometastasis and well related to higher proliferating activity of gastric cancer. 10In situ optical backpropagation training of diffractive optical neural networks显示文摘Training an artificial neural network with backpropagation algorithms to perform advanced machine learning tasks requires an extensive computational process.This paper proposes to implement the backpropagation algorithm optically for in situ training of both linear and nonlinear diffractive optical neural networlks,which enables the acceleration of training speed and improvement in energy efficiency on core computing modules.We demonstrate that the gradient of a loss function with respect to the weights of diffractive layers can be accurately calculated by measuring the forward and backward propagated optical fields based on light reciprocity and phase conjunction principles.The diffractive modulation weights are updated by programming a high-speed spatial light modulator to minimize the error between prediction and target output and perform inference tasks at the speed of light.We numerically validate the effectiveness of our approach on simulated networks for various applications.The proposed in situ optical learning architecture achieves accuracy comparable to in silico training with an electronic computer on the tasks of object dlassification and matrix-vector multiplication,which further allows the diffractive optical neural network to adapt to system imperfections.Also,the self-adaptive property of our approach facilitates the novel application of the network for all-optical imaging through scattering media.The proposed approach paves the way for robust implementation of large-scale difractive neural networks to perform distinctive tasks all-optically.TIANKUANG ZHOU LU FANG TAO YAN JIAMIN WU YIPENG LI JINGTAO FAN HUAQIANG WU XING LIN QIONGHAI DA 2020Photonics Research2020,8,6:11
11SHRIMP zircon U-Pb dating of the intrusives in the Tongling metallogenic cluster and its dynamic setting显示文摘This paper selected five typical Mesozoic intrusives from the Tongling metallogenic cluster (Xiaotongguanshan, Fenghuangshan, Xinqiao, Dongguashan, and Shatanjiao plutons), and made a systemic SHRIMP zircon U-Pb dating for the five plutons, which produced an age range of 151.8±2.6- 142.8±1.8 Ma. This work put an accurate constraint on the formation age of the intrusives in the Tongling metallogenic cluster. These age data indicate that magmatic activity reached a peak during Late Jurassic. The intrusive sequence of magma is generally from quartz monzonite (porphyry) through monzonite to granodiorite to quartz monzodiorite to pyroxene monzodiorite to gabbro-diabase. The intrusives of different lithology differed in crystallization age, probably implying the intrusives in the Tongling area underwent an evolutional process of magma, which was closely related to geodynamical setting in the depths of the area. A dynamic model was presented for the origin of the igneous rocks in the study area as follows. The assembly between the Yangtze craton and the North China craton fini- shed at the end of T3, and then the stage of another compressional orogeny began in the Tongling area, i.e., Pacific dynamic system. Along with the subduction of the Izanagi plate underneath the Eurasian plate at J2-J3, NW-trending compression toward the East China continent was produced, and compres- sional deformation also took place, forming NE-trending fold and resulting in thickening of the crust in the Tongling area. High-density eclogite-facies rocks were produced in the low part of the crust, re- sulting in the delamination of mantle lithosphere and lower crust, and upwelling of materials in as- thenosphere. Decompression melting produced basaltic magma, and the materials in lower crust were heated by the underplating of the basaltic magma. Thus, melting of lower crust yielded granitic magma, which intruded along deep and large faults through various geological processes (J3-K1). The SHRIMP U-Pb zircon age of 151.8±2.6-142.8±1.8 Ma for intrusives in the Tongling area suggests that the de- lamination of lithosphere mantle and lower crust at least began at middle-late stage of Late Jurassic, resulting in sharp thinning of lithosphere and intense extension of middle-upper crust. Thus, a lot of decollements were produced between cover and cover, basement and cover, and middle and lower crust. This was structural layering or detachment of lithosphere in the Tongling area. 13A feasible,economical,and accurate analytical method for simultaneous determination of six alkaloid markers in Aconiti Lateralis Radix Praeparata from different manufacturing sources and processing ways显示文摘Aconiti Lateralis Radix Praeparata(Fuzi) is a commonly used traditional Chinese medicine in clinic for its potency in restoring yang and rescuing from collapse. Aconiti alkaloids, mainly including monoester-diterpenoidaconitines(MDAs) and diester-diterpenoidaconitines(DDAs), are considered to act as both bioactive and toxic constituents. In the present study, a feasible, economical, and accurate HPLC method for simultaneous determination of six alkaloid markers using the Single Standard for Determination of Multi-Components(SSDMC) method was developed and fully validated. Benzoylmesaconine was used as the unique reference standard. This method was proven as accurate(recovery varying between 97.5%-101.8%, RSD < 3%), precise(RSD 0.63%-2.05%), and linear(R > 0.999 9) over the concentration ranges, and subsequently applied to quantitative evaluation of 62 batches of samples, among which 45 batches were from good manufacturing practice(GMP) facilities and 17 batches from the drug market. The contents were then analyzed by principal component analysis(PCA) and homogeneity test. The present study provided valuable information for improving the quality standard of Aconiti Lateralis Radix Praeparata. 14A two-step lineage reprogramming strategy to generate functionally competent human hepatocytes from fibroblasts显示文摘Terminally differentiated cells can be generated by lineage reprogramming,which is,however,hindered by incomplete conversion with residual initial cell identity and partial functionality. Here, we demonstrate a new reprogramming strategy by mimicking the natural regeneration route, which permits generating expandable hepatic progenitor cells and functionally competent human hepatocytes. Fibroblasts were first induced into human hepatic progenitor-like cells (hHPLCs), which could robustly expand in vitro and efficiently engraft in vivo. Moreover, hHPLCs could be efficiently induced into mature human hepatocytes (hiHeps) in vitro, whose molecular identity highly resembles primary human hepatocytes (PHHs). Most importantly, hiHeps could be generated in large quantity and were functionally competent to replace PHHs for drug-metabolism estimation, toxicity prediction and hepatitis B virus infection modeling. Our results highlight the advantages of the progenitor stage for successful lineage reprogramming. 15Effects of early oral feeding after radical total gastrectomy in gastric cancer patients显示文摘BACKGROUND Gastric cancer(GC)is a heavy burden in China.Nutritional support for GC patients is closely related to postoperative rehabilitation.However,the role of early oral feeding after laparoscopic radical total gastrectomy in GC patients is unclear and high-quality research evidence is scarce.AIM To prospectively explore the safety,feasibility and short-term clinical outcomes of early oral feeding after laparoscopic radical total gastrectomy for GC patients.METHODS This study was a prospective cohort study conducted between January 2018 and December 2019 based in a high-volume tertiary hospital in China.A total of 206 patients who underwent laparoscopic radical total gastrectomy for GC were enrolled.Of which,105 patients were given early oral feeding(EOF group)after surgery,and the other 101 patients were given the traditional feeding strategy(control group)after surgery.Perioperative clinical data were recorded and analyzed.The primary endpoints were gastrointestinal function recovery time and postoperative complications,and the secondary endpoints were postoperative nutritional status,length of hospital stay and expenses,etc.RESULTS Compared with the control group,patients in the EOF group had a significantly shorter postoperative first exhaust time(2.48±1.17 d vs 3.37±1.42 d,P=0.001)and first defecation time(3.83±2.41 d vs 5.32±2.70 d,P=0.004).In addition,the EOF group had a significant shorter postoperative hospitalization duration(5.85±1.53 d vs 7.71±1.56 d,P<0.001)and lower postoperative hospitalization expenses(16.60±5.10 K¥vs 21.00±7.50 K¥,P=0.014).On the 5th day after surgery,serum prealbumin level(214.52±22.47 mg/L vs 204.17±20.62 mg/L,P=0.018),serum gastrin level(246.30±57.10 ng/L vs 223.60±55.70 ng/L,P=0.001)and serum motilin level(424.60±68.30 ng/L vs 409.30±61.70 ng/L,P=0.002)were higher in the EOF group.However,there was no significant difference in the incidence of total postoperative complications between the two groups(P=0.507).CONCLUSION Early oral feeding after laparoscopic radical total gastrectomy can promote the recovery of gastrointestinal function,improve postoperative nutritional status,reduce length of hospital stay and expenses while not increasing the incidence of related complications,which indicates its safety,feasibility and potential benefits for gastric cancer patients.Yi-Xun Lu Yan-Jun Wang Tian-Yu Xie Shuo Li Di Wu Xiong-Guang Li Qi-Ying Song Li-Peng Wang Da Guan Xin-Xin Wang 2020World Journal of Gastroenterology2020,26,36:9
16Research on Flexural Behavior of Coral Aggregate Reinforced Concrete Beams显示文摘Through the flexural behavior test of coral aggregate reinforced concrete beams(CARCB) and ordinary Portland reinforced concrete beams(OPRCB), and based on the parameters of concrete types, concrete strength grades and reinforcement ratios, the crack development, failure mode, midspan deflection and flexural capacity were studied, the relationships of bending moment-midspan deflection, load-longitudinal tensile reinforcement strain, load-maximum crack width were established, and a calculation model for the flexural capacity of CARCB was suggested. The results showed that with the increase in the reinforcement ratio and concrete strength grade, the crack bending moment(Mcr)and ultimate bending moment(Mu) of CARCB gradually increased. The characteristics of CARCB and OPRCB are basically the same. Furthermore, through increasing the concrete strength grade and reinforcement ratio, Mcr/Mu could be increased to delay the cracking of CARCB. As the load increased, crack width(w) would also increase. At the beginning of the loading, w increased slowly. And then it increased rapidly when the load reached to the ultimate load, which then led to beam failure. 17Interaction between cyclooxygenase-2,Snail,and E-cadherin in gastric cancer cells显示文摘AIM:To investigate the mechanisms of how cyclooxygenase-2(COX-2)regulates E-cadherin in gastric cancer cells.METHODS:COX-2 expression in human gastric cancer cell lines SGC-7901,BGC-823,MGC-803 and AGS were measured at the mRNA and protein level.COX-2 rich cell line SGC-7901 was chosen for subsequent experiments.siRNA mediated gene knockdown was used to investigate the impact of COX-2 on nuclear factor-κB (NF-κB),Snail,and E-cadherin in gastric cancer cells.Gene expression was determined by Western blot and real-time polymerase chain reaction.To analyze whether NF-κB inhibition could interrupt the modulatory effect of COX-2 or prostaglandin E2(PGE2)on E-cadherin,gastric cancer cells were treated with celecoxib or PGE2,in the presence of NF-κB specific siRNA.RESULTS:Highest expression level of COX-2 was found in SGC-7901 cells,both at mRNA and protein levels.siRNA mediated down-regulation of COX-2 led to a reduced expression of NF-κB and Snail,but an increased expression of E-cadherin in SGC-7901 cells.siRNA mediated down-regulation of NF-κB also led to a reduced expression of E-cadherin and Snail in SGC-7901 cells.However,COX-2 expression did not alter after cells were treated with NF-κB specific siRNA in SGC-7901 cells.Treatment of SGC-7901 cells with celecoxib led to a reduced expression of Snail but an increased expression of E-cadherin.In contrast,treatment of SGC-7901 cells with PGE2 led to an increased Snail and a decreased E-cadherin.However,siRNAmediated knockdown of NF-κB partially abolished the effect of celecoxib and PGE2 on the regulation of E-cadherin and Snail in SGC-7901 cells.CONCLUSION:COX-2 likely functions upstream of NF-κB and regulates the expression of E-cadherin via NF-κB/Snail signaling pathway in gastric cancer cells.Xiao-Jun Liu Zhao-Feng Chen Hai-Long Li Ze-Nan Hu Min Liu Ai-Ping Tian Da Zhao Jing Wu Yong-Ning Zhou Liang Qiao 2013World Journal of Gastroenterology2013,19,37:8
18Cloned pigs derived from somatic cell nuclear transferembryos cultured in vitro atlow oxygen tension显示文摘Pig cloning has great potential to human xenotransplantation. The present study was designed to establish a more efficient system for producing cloned pigs by somatic cell nuclear transfer (SCNT). Our approach was as follows: SCNT embryos were reconstructed by using fetal fibroblasts of Chinese miniature pig as donors and in vitro matured oocytes of prepubertal gilts as recipients. Reconstructed em- bryos were induced by electrical fusion/activation and cultured in BSA-containing North Carolina State University 23 medium (NCSU-23) or Porcine Zygote Medium (PZM-3) at the gas condition of 5% CO2, 7% O2, 88% N2. A total of 230 cloned embryos were transferred to three surrogate sows, producing three piglets. One of them is apparently healthy. The clonal provenance of the piglet was indicated by its coat color and confirmed by DNA microsatellite analysis. 19Effects of yaw-roll coupling ratio on the lateral-directional departure prediction and restraint显示文摘The experimental data obtained from yaw-roll coupled wind tunnel tests are used for lateral-directional departure prediction,by linearizing the_b model to extract nominal dynamic derivatives at each coupling ratio.The prediction results are compared with those of the existing engineering methods which are based on the conventional aerodynamic derivatives.The comparison shows that the yaw-roll coupling ratio has a great influence on the departure susceptibility.The departure resistance will loss in partial region of the coupling ratio when the angle of attack is higher than a critical value.According to the stable and unstable regions of coupling ratio,a two-segment stability augmentation system with two different feedback gain matrices is obtained by pole-placement method.The two-segment stability augmentation system is used in the simulations of straight and level flight,steady turn,spin recovery and Herbst maneuver.The simulation results are also compared with the applications of a fixed-gain stability augmentation system designed by the conventional aerodynamic derivatives.When the yaw-roll coupling effects are fully considered,the two-segment stability augmentation system is more effective for departure restraint and can provide a better flying quality with less control energy.Lin SHEN Da HUANG Genxing WU 2019Chinese Journal of Aeronautics2019,32,10:8
20The origin of Wx^(la) provides new insights into the improvement of grain quality in rice显示文摘Appearance and taste are important factors in rice(Oryza sativa) grain quality. Here, we investigated the taste scores and related eating-quality traits of533 diverse cultivars to assess the relationships between—and genetic basis of—rice taste and eating-quality. A genome-wide association study highlighted the Wx gene as the major factor underlying variation in taste and eating quality. Notably, a novel waxy(Wx) allele, Wx^(la), which combined two mutations from Wx^(b) and Wx^(in), exhibited a unique phenotype. Reduced GBSSI activity conferred Wx^(la) rice with both a transparent appearance and good eating quality. Haplotype analysis revealed that Wx^(la) was derived from intragenic recombination. In fact,the recombination rate at the Wx locus was estimated to be 3.34 kb/c M, which was about 75-fold higher than the genome-wide mean, indicating that intragenic recombination is a major force driving diversity at the Wx locus. Based on our results, we propose a new network for Wx evolution, noting that new Wx alleles could easily be generated by crossing genotypes with different Wx alleles.
