| 4 | Cloning, expression and protective immunity evalua-tion of the full-length cDNA encoding succinate dehydrogenase iron-sulfur protein of Schistosoma japonicum显示文摘1071-bp fragment was obtained from the Schistosoma japonicum (Chinese strain) adult cDNA library after the 3′ and 5′ ends of the incomplete expression sequence tag (EST) of succinate dehydrogenase iron-sulfur protein of Schistosoma japonicum (SjSDISP) were amplified by the anchored PCR with 2 pairs of primers designed according to the EST of SjSDISP and the sequence of multiclone sites of the library vector. Sequence analysis indicated that the fragment was a full-length cDNA with a complete open reading frame (ORF), encoding 278 amino acid residues. The fragment was cloned into prokary- otic expression vector pQE30, and subsequently sequenced and expressed in Escherichia coli. SDS-PAGE and Western-blot analyses showed that the recombinant protein was about 32 kD and could be recognized by the polyclonal antisera from rabbits immunized with Schistosoma japonicum adult worm antigen. Compared with the FCA controls, mice vaccinated with rSjSDISP (test) or rSjGST (posi- tive control) all revealed high levels of specific antibody and significant reduction in worm burden, liver eggs per gram (LEPG), fecal eggs per gram (FEPG) and intrauterine eggs. These results suggest that SjSDISP may be a novel and partially protective vaccine candidate against schistosomiasis. | 10 | The chromosome-level reference genome of Coptis chinensis provides insights into genomic evolution and berberine biosynthesis显示文摘Coptis chinensis Franch, a perennial herb, is mainly distributed in southeastern China. The rhizome of C. chinensis hasbeen used as a traditional medicine for more than 2000 years in China and many other Asian countries. Thepharmacological activities of C. chinensis have been validated by research. Here, we present a de novo high-qualitygenome of C. chinensis with a chromosome-level genome of ~958.20 Mb, a contig N50 of 1.58 Mb, and a scaffold N50of 4.53 Mb. We found that the relatively large genome size of C. chinensis was caused by the amplification of longterminal repeat (LTR) retrotransposons. In addition, a whole-genome duplication event in ancestral Ranunculales wasdiscovered. Comparative genomic analysis revealed that the tyrosine decarboxylase (TYDC) and (S)-norcoclaurinesynthase (NCS) genes were expanded and that the aspartate aminotransferase gene (ASP5) was positively selected inthe berberine metabolic pathway. Expression level and HPLC analyses showed that the berberine content was highestin the roots of C. chinensis in the third and fourth years. | 11 | Estimating Seismic Intensity Maps of the 2021 Mw 7.3 Madoi,Qinghai and Mw 6.1 Yangbi,Yunnan,China Earthquakes显示文摘This study focuses on rapidly determining seismic intensity maps of earthquakes because it offers fundamental information for effective emergency rescue and subsequent scientific research,and remains challenging to accurately determine seismic intensity map in regions with sparse instrumental observations.Here we applied a novel method that consisted of array technology(backprojection),ground-motion prediction equations,and site corrections,to estimate the seismic intensity maps of the 2021 Mw 7.3 Madoi,Qinghai and the Mw 6.1 Yangbi,Yunnan,China earthquakes.We used seismic data recorded at European stations to back-project the source processes of the 2021 Mw7.3 Madoi,Qinghai and the Mw 6.1 Yangbi,Yunnan,China earthquakes.The back-projected energy radiations were then used as subevents or used to define the fault geometry.Summing the contributions of each subevent or estimating the shortest distances from each site to the rupture fault,we obtained the ground motion(PGA and PGV)for each site under rock site conditions.The estimated ground motions were corrected at each site for local site amplification according to the Vs30 database.Our estimated seismic intensity maps and field reports showed high similarity,which further validated the effectiveness of the novel approach,and pushed the limit of earthquake size down to~M 6.Such efforts would substantially help in the fast and accurate evaluation of earthquake damage,and precise rescue efforts. | Wenkai Chen Dun Wang Can Zhang Qiang Yao Hongjun Si | 2022 | Journal of Earth Science2022,33,4: | 2 |
| 12 | Efficacy of Danggui Buxue decoction(当归补血汤)on diabetic nephropathy-induced renal fibrosis in rats and possible mechanism显示文摘OBJECTIVE:To observe the efficacy of Danggui Buxue decoction(当归补血汤,DBD) on diabetic nephropathyinduced renal fibrosis in rats,and to study the possible mechanism.METHODS:Sixty male Goto Kakizaki(GK) rats were randomly assigned to the model group,gliquidone group,astragaloside Ⅳ group,and high-,medium-and lowdoses DBD groups.After 8 weeks,changes in body weight,blood glucose,serum creatinine,serum urea nitrogen,and total cholesterol were observed.Changes in transforming growth factor-β1(TGF-β1),Smad3,and Smad5 pathways and the expression of the fibrosisrelated proteins collagen Ⅳ(col Ⅳ),α-smooth muscle actin(α-SMA),and vimentin were assessed.The degree of renal fibrosis was observed by immunohistochemistry and Mason staining.The expression of interleukin 6(IL-6),interleukin 10(IL-10),tumor necrosis factor(TNF-α),and C-reactive protein(CRP) in the kidneys was assessed using enzyme linked immunosorbent assay.RESULTS:Our experiments showed that DBD effectively reduced blood glucose,blood urea nitrogen,and creatinine levels after 8 weeks of administration,improved renal function in diabetic rats,alleviated renal fibrosis,and reduced the renal tissue levels of IL-6,IL-10,TNF-α,and CRP.Furthermore,DBD decreased the expression of TGF-β1,Smad3,col IV,α-SMA,and vimentin in renal tissues and increased the expression of Smad5.CONCLUSIONS:DBD ameliorates diabetic renal interstitial fibrosis by modulating the TGF-β1/Smads pathway. | ZHAO Ye WANG Xian GU Ling LI Zihang ZHU Jingtian WANG Wenkai ZHANG Liang XUE Mei | 2023 | Journal of Traditional Chinese Medicine2023,43,3: | 2 |

| 16 | Motivation of Parent-Child Co-residence Behavior:Evidenee from the China Health and Retirement Longitudinal Study显示文摘This paper examines the motivations of parent-child co-residence behavior in China using data from the China Health and Retirement Longitudinal Study.We test three possible motives:social norms,self-interest and altruism.We find that social norms play an important role in household co-residence behavior,showing that the belief that'sons take care of parents'is strong,and children in different birth orders take different responsibilities.Taking the one-child policy as a natural experiment,we compared co-residence behavior between only-child and multi-child families.This allowed us to test whether children in multi-child families with wealthier parents more often coreside in order to compete for a bequest.We find that parents'wealth is more appealing to children in multi-child families.The results support the life cycle theory that coresidence decisions are motivated by self-interest.We also find some evidence of altruism when parents and children make co-residence decisions.These findings provide some insights for designing future elder-support policies in China. | Yijie Wang Wenkai Sun Xianghong Wang | 2019 | China & World Economy2019,27,2: | 1 |
| 17 | Land cover classification in a mixed forest-grassland ecosystem using LResU-net and UAV imagery显示文摘Using an unmanned aerial vehicle (UAV) paired with image semantic segmentation to classify land cover within natural vegetation can promote the development of forest and grassland field. Semantic segmentation normally excels in medical and building classification, but its usefulness in mixed forest-grassland ecosystems in semi-arid to semi-humid climates is unknown. This study proposes a new semantic segmentation network of LResU-net in which residual convolution unit (RCU) and loop convolution unit (LCU) are added to the U-net framework to classify images of different land covers generated by UAV high resolution. The selected model enhanced classification accuracy by increasing gradient mapping via RCU and modifying the size of convolution layers via LCU as well as reducing convolution kernels. To achieve this objective, a group of orthophotos were taken at an altitude of 260 m for testing in a natural forest-grassland ecosystem of Keyouqianqi, Inner Mongolia, China, and compared the results with those of three other network models (U-net, ResU-net and LU-net). The results show that both the highest kappa coefficient (0.86) and the highest overall accuracy (93.7%) resulted from LResU-net, and the value of most land covers provided by the producer’s and user’s accuracy generated in LResU-net exceeded 0.85. The pixel-area ratio approach was used to calculate the real areas of 10 different land covers where grasslands were 67.3%. The analysis of the effect of RCU and LCU on the model training performance indicates that the time of each epoch was shortened from U-net (358 s) to LResU-net (282 s). In addition, in order to classify areas that are not distinguishable, unclassified areas were defined and their impact on classification.
