1多鳞鱚phds基因家族序列特征及其在低氧胁迫后表达变化显示文摘【目的】分析多鳞鱚(Sillago sihama)脯氨酸羟化酶(prolyl hydroxylases,Phds)基因家族序列特征,探讨其在低氧响应过程的作用。[方法]通过硬骨鱼类phds基因,在多鳞鱚转录组和基因组数据中比对筛选多鳞鱚phds基因。对多鳞鱚Phds蛋白序列进行结构域预测、系统进化和共线性分析等。采用实时荧光定量PCR检测phds基因在低氧胁迫和复氧后的表达变化。【结果】在多鳞鱚基因组中共鉴定到三个phds基因,分别是phd1、phd2和phd3,开放阅读框(ORF)全长分别为1635、1092、723 bp,分别编码544、363、240个氨基酸。结构域预测表明,多鳞鱚Phd1、Phd2、Phd3蛋白均含有P4Hc保守结构域。系统进化分析发现,多鳞鱚Phds均与大黄鱼(Larimichthys crocea)亲缘关系最近。共线性分析表明,多鳞鱚phds与尼罗罗非鱼(Oreochromis niloticus)上下游基因高度一致。phd1、phd2在低氧处理4 h鳃组织中mRNA表达显著高于常氧组(P<0.05),但在心脏中无显著性差异(P>0.05);phd3在低氧处理4 h的鳃组织中显著低于常氧组(P<0.05),而在心脏中显著高于常氧组(P<0.05)。【结论】多鳞鱚存在phd1、phd2和phd3基因,并在不同组织的低氧响应中起重要作用。林星桦 叶明慧 SEATAN Wanida 潘炎杨 陈芳圆 黄洋 朱春华 李广丽 田昌绪 2020广东海洋大学学报2020,40,6:4
2Chiral nanocomposite of sulfobutyl ether-b-cyclodextrin embedded in carbon nanofibers for enantioselective electrochemical discrimination of amlodipine, metoprolol and clenbuterol enantiomers显示文摘A highly sensitive and selective electrochemical chiral sensor was developed based on the competitive supramolecular interaction of carbon nanofibers(CNFs)embedded sulfobutyl ether-b-cyclodextrin(SBEb-CD)with optically active cationic drugs at glassy carbon electrode(GCE).The difference in intermolecular hydrogen bonding/stability constant/enantioselectivity coefficient and Gibbs free energy of anionic host SBE-b-CD with enantiomers of amlodipine(R-/S-AML),clenbuterol(R-/S-CBL)and metoprolol(R-/S-MET)as a guest paved the way for efficient discrimination.The proposed sensing platform(CNFs-SBE-b-CD/GCE)could recognize the aforementioned enantiomers based on the discernible difference of peak potential(S/R-AML(△Ep=135 mV),R/S-MET(△Ep=99 mV)and R/S-CBL(△Ep=111 mV).The binding mechanisms and thermodynamic study of the enantiospecific behavior have been investigated using the host-guest chemistry approach inside the nanocavity and results suggest that S-AML,RMET,and R-CBL show stronger stability constants than their antipodes.Formation of the diastereomeric complex was taken as a measure of enantioselectivity and experimental results indicated that anionic SBE-b-CD is a better chiral ligand than neutral cyclodextrins.The fabricated sensor could be a useful lowcost electrochemical tool for molecular recognition of a variety of cationic species not only of drugs but also from other sources.Pramod K.Kalambate Sharad S.Upadhyay Yue Shen Wanida Laiwattanapaisal Yunhui Huang 2021Journal of Materiomics2021,7,2:2

13Anxiolytic and free radical scavenging potential of Chinese celery(Apium graveolens) extract in mice显示文摘Objective: To elucidate the anxiolytic and free radical scavenging effect of methanolic extract of Apium graveolens(A. graveolens) in adult C57BL/6 mice.Methods: Sixty male mice were divided into 6 groups: control, vehicle, positive control and A. graveolens(125, 250 and 500 mg/kg). Different behavioral models of elevated plus maze, open field, light/dark, hole-board and pentobarbital-induced sleep were used to assess anxiety-like behavior. Biochemical parameters including monoamine oxidase-A(MAO-A) activity, lipid peroxidation, % inhibition of superoxide anion and glutathione peroxidase activity were measured. Histologic studies were also examined.Results: Mice receiving various doses of A. graveolens(125, 250 and 500 mg/kg)showed an alleviation of anxiety-like behavior as evidenced by the battery of behavioral tests. Likewise, A. graveolens treatment was found to significantly decrease MAO-A activity, lipid peroxidation as well as cause a significant increase of % inhibition of superoxide anion and glutathione peroxidase activity in both cortex and striatum. The total number of survival neurons found in the frontal cortex and striatum was significantly higher than that of the vehicle-treated group.Conclusions: Taken together, we showed that A. graveolens improve the behavioral changes which might be related to the inhibition of free radicals and modulation of MAOA activity resulting in an increased number of survival neurons. 14Comparative Transcriptome Analysis of Heart Tissue in Response to Hypoxia in Silver Sillago (Sillago sihama)显示文摘Sillago sihama,commonly known as silver sillago,is considered as an economically important fish species in China.It is sensitive to hypoxia stress in the larval stage,and the mechanism has not been understood thoroughly.In this study,we investigated the transcriptome change in heart tissues under hypoxia stress.The fish were divided into four groups,including 1 h of hypoxia(hypoxia1h,dissolved oxygen(DO)=1.5±0.1 mg L^(−1)),4h of hypoxia(hypoxia4h,DO=1.5±0.1 mg L^(−1)),4h of reoxygen(reoxygen4h,DO=8.0±0.2 mg L^(−1))after 4h of hypoxia(DO=1.5 mg L^(−1))and normoxia or control(DO=8.0±0.2 mg L^(−1))groups.The results showed that a total of 3068 genes were identified as differentially expressed genes(DEGs)based on the criteria∣log2(Fold change)∣>1.0 and adjusted P-value<0.05.A total of 7761141 and 1151 DEGs were obtained from hypoxia1h,hypoxia4h and reoxygen4h groups,respectively.The enrichment pathway analysis showed that the DEGs were significantly enriched in ribosome biogenesis in eukaryotes,retinol metabolism,DNA replication and the oxidative phosphorylation(OXPHOS)pathways.Thirteen DEGs from the RNA-seq results were validated by quantitative real-time polymerase chain reaction(qRT-PCR).These candidate genes are considered as important regulatory factors involved in the hypoxia stress response in S.sihama.SAETAN Wanida YE Minghui LIN Xinghua LIN Xiaozhan ZHANG Yulei HUANG Yang DU Tao LI Guangli TIAN Changxu 2021Journal of Ocean University of China2021,20,4:1
