1Challenges and strategies in high-accuracy manufacturing of the world’s largest SiC aspheric mirror显示文摘In the process of manufacturing the world’s largest silicon carbide(SiC)aspheric mirror,the primary difficulties are mirror blank preparation,asphere fabrication,and testing,as well as cladding and coating.Specifically,the challenges include the homogeneity of the complicated structure casting,accuracy and efficiency of the fabrication process,print-through effect,fidelity and precision of test procedure,stress and denseness of cladding process,the dynamic range of interferometric measurement,and air turbulence error due to the long optical path.To break through such a barrier of difficulties,we proposed the water-soluble room temperature vanishing mold and gel casting technology,homogeneous microstructure reaction-formed joint technology,nano-accuracy efficient compound fabrication,gravity unloading technology,high-denseness low-defect physical vapor deposition(PVD)Si-cladding technology,test data fusion method,and time-domain averaging method,etc.Based on the proposed technologies and methods,we have accomplished the world’s largest SiC aspheric mirror with a size of φ4.03 m.The impressive performance of the SiC aspheric mirror is validated by the characteristics of the fabricated SiC aspheric mirror.The aerial density of the SiC blank is less than 120 kg/m^(2),surface shape test accuracy is better than 6 nm RMS,thickness inhomogeneity of the cladding layer is less than 5%,and the final surface figure error and roughness are 15.2 nm RMS and 0.8 nm RMS,respectively.Xuejun Zhang Haixiang Hu Xiaokun Wang Xiao Luo Ge Zhang Wenxing Zhao Xiaoyi Wang Zhenyu Liu Ling Xiong Erhui Qi Congcong Cui Yanchao Wang Yingjie Li Xu Wang Longxiang Li Yang Bai Qiang Cheng Zhiyu Zhang Ruigang Li Wa Tang Xuefeng Zeng Weijie Deng Feng Zhang 2022Light(Science & Applications)2022,11,12:3

4A building-based data capture and data mining technique for air quality assessment显示文摘Recently,a building-based air quality model system which can predict air quality in front of individual buildings along both sides of a road has been developed.Using the Macao Peninsula as a case study,this paper shows the advantages of building-based model system in data capture and data mining.Compared with the traditional grid-based model systems with input/output spatial resolutions of 1-2 km,the building-based approach can extract the street configuration and traffic data building by building and therefore,can capture the complex spatial variation of traffic emission,urban geometry,and air pollution.The non-homogeneous distribution of air pollution in the Macao Peninsula was modeled in a highspatial resolution of 319 receptors·km^(-2).The spatial relationship among air quality,traffic flow,and urban geometry in the historic urban area is investigated.The study shows that the building-based approach may open an innovative methodology in data mining of urban spatial data for environmental assessment.The results are particularly useful to urban planners when they need to consider the influences of urban form on street environment.Ni SHENG U Wa TANG 2011Frontiers of Environmental Science & Engineering2011,5,4:1

19Targeting proteasomal deubiquitinases USP14 and UCHL5 with b-AP15 reduces 5-fluorouracil resistance in colorectal cancer cells显示文摘5-Fluorouracil(5-FU)is the first-line treatment for colorectal cancer(CRC)patients,but the development of acquired resistance to 5-FU remains a big challenge.Deubiquitinases play a key role in the protein degradation pathway,which is involved in cancer development and chemotherapy resistance.In this study,we investigated the effects of targeted inhibition of the proteasomal deubiquitinases USP14 and UCHL5 on the development of CRC and resistance to 5-FU.By analyzing GEO datasets,we found that the mRNA expression levels of USP14 and UCHL5 in CRC tissues were significantly increased,and negatively correlated with the survival of CRC patients.Knockdown of both USP14 and UCHL5 led to increased 5-FU sensitivity in 5-FU-resistant CRC cell lines(RKO-R and HCT-15R),whereas overexpression of USP14 and UCHL5 in 5-FU-sensitive CRC cells decreased 5-FU sensitivity.B-AP15,a specific inhibitor of USP14 and UCHL5,(1−5μM)dose-dependently inhibited the viability of RKO,RKO-R,HCT-15,and HCT-15R cells.Furthermore,treatment with b-AP15 reduced the malignant phenotype of CRC cells including cell proliferation and migration,and induced cell death in both 5-FU-sensitive and 5-FU-resistant CRC cells by impairing proteasome function and increasing reactive oxygen species(ROS)production.In addition,b-AP15 inhibited the activation of NF-κB pathway,suppressing cell proliferation.In 5-FU-sensitive and 5-FU-resistant CRC xenografts nude mice,administration of b-AP15(8 mg·kg^(-1)·d^(-1),intraperitoneal injection)effectively suppressed the growth of both types of tumors.These results demonstrate that USP14 and UCHL5 play an important role in the development of CRC and resistance to 5-FU.Targeting USP14 and UCHL5 with b-AP15 may represent a promising therapeutic strategy for the treatment of CRC.Wa Ding Jin-xiang Wang Jun-zheng Wu Ao-chu Liu Li-ling Jiang Hai-chuan Zhang Yi Meng Bing-yuan Liu Guan-jie Peng En-zhe Lou Qiong Mao Huan Zhou Dao-lin Tang Xin Chen Jin-bao Liu Xian-ping Shi 2023Acta Pharmacologica Sinica2023,44,12:0
