1ELE restrains empty glumes from developing into lemmas显示文摘Although there is evident homology among reproductive organs when comparing Poaceae (grass) and eudicots, the identity of grass specific organs, such as lodicules, palea, lemma, and glumes has been the subject of a vast and largely inconclusive discussion. Here we provide some direct evidence to support the idea that the empty glumes of rice (Oryza sativa) are counterparts of lemmas. We show that the development of empty glumes is regulated by ELE (elongated empty glume), which belongs to a plant specific novel gene family. Mutations at the ELE locus cause elongated empty glumes, which mimic the lemmas and have the epidermal morphology of lemmas with four or five vascular bundles. As a nuclear-localized gene, ELE is specifically expressed at the empty glumes of immature spikelets, and its ectopic expression causes many floral development defects, including lemma-like palea, extra palea-like structures, elongated lodicules, extra stamens and stigmas. 4Contrasting elevational diversity patterns for soil bacteria between two ecosystems divided by the treeline显示文摘Above- and below-ground organisms are closely linked, but how elevational distribution pattern of soil microbes shifting across the treeline still remains unknown. Sampling of 140 plots with transect, we herein investigated soil bacterial distribution pattern from a temperate forest up to a subalpine meadow along an elevational gradient using Illumina sequencing. Our results revealed distinct elevational patterns of bacterial diversity above and below the treeline in responding to changes in soil conditions:a hollow elevational pattern in the forest(correlated with soil temperature, pH, and C:N ratio) and a significantly decreasing pattern in the meadow(correlated with soil pH, and available phosphorus). The bacterial community structure was also distinct between the forest and meadow, relating to soil pH in the forest and soil temperature in the meadow. Soil bacteria did not follow the distribution pattern of herb diversity, but bacterial community structure could be predicted by herb community composition. These results suggest that plant communities have an important influence on soil characteristics, and thus change the elevational distribution of soil bacteria. 7Effect of combined medicated thread moxibustion plus needle picking therapy of Zhuang nationality medicine on antioxidant levels in a rat model of sciatica显示文摘OBJECTIVE: To investigate the effect and underlying mechanisms of combined medicated thread moxibustion therapy plus needle picking therapy of Zhuang nationality medicine on antioxidant levels in a rat model of sciatica.METHODS: One hundred Wistar rats, of specific pathogen free level, were randomly divided into five groups: normal control group, model group,medicated thread moxibustion group, needle picking group, and combination group. Each group contained 20 rats. In the model, medicated thread moxibustion, needle picking, and combination groups,sciatica models were established through chronicconstriction injury of the sciatic nerve. After the model was established, the rats in the medicated thread moxibustion, needle picking, and combination groups were given the corresponding therapies for 21 days. The control and model groups received no treatment. Reactive oxygen species, superoxide dismutase, malondialdehyde, and total antioxidant capacity changes were determined.Nicotinamide adenine dinucleotide phosphate(NADPH) oxidase subunit NADPH oxidases 4(NOX4) m RNA expression and the morphology of cells were observed to detect apoptosis of gamma-aminobutyric acid ergic(GABAergic) neurons.RESULTS: Compared with control group, reactive oxygen species and malondialdehyde levels rose significantly in the model group(P < 0.01), while superoxide dismutase and total antioxidant capacity levels were lowered(P < 0.05). Compared with the model group, reactive oxygen species and malondialdehyde decreased in the needle picking group(P < 0.05), while superoxide dismutase levels were increased(P < 0.05); reactive oxygen species and malondialdehyde significantly decreased in the combination group(P < 0.01). In addition, the model group had higher NOX4 m RNA expression than that of the control group(P < 0.05), and the combination group had lower expression levels than that of the model group(P < 0.05). 8The geometric simulation and analysis of the blind growth fold in the overthrust belt of the southern section of the frontier Longmen Mt.during Late Triassic显示文摘The seismic profiles processed by high-resolution display that a blind growth fault-bend fold emerges in depth of seismic exploration area in Dayi in the southern section of the frontier Longmen Mt.This paper presents a detailed structural interpretation to seismic section,then describes the deformed geometry related to the pre-growth strata,the growth strata and the post-growth strata on both limbs of the fold.The computer geometric simulation,based on the model of fault-bend fold,is utilized to stimulate the geometry of the blind growth fold at different sedimentation rates in Dayi.These structural features are subsequently extracted and compared with the actual seismic reflection section.Our research suggests that a low angle overthrust formed during Late Triassic also appears in the southern section of the frontier Longmen Mt.The event of structure took place at the depositional stage of a portion of the middle and upper fourth member of Xujiahe Formation(T3x4),which corresponds to thrusting in the northern section of the frontier Longmen Mt.However,the thrusting displacement here is smaller than one of the northern section.CHEN Wei YANG KeMing LI ShuBin JIAN GaoMing TANG Jin ZHAO Yiao SU Qian 2009Chinese Science Bulletin2009,54,8:3

12MXene-GaN van der Waals metal-semiconductor junctions for high performance multiple quantum well photodetectors显示文摘A MXene-GaN-MXene based multiple quantum well photodetector was prepared on patterned sapphire substrate by facile drop casting.The use of MXene electrodes improves the responsivity and reduces dark current,compared with traditional Metal-Semiconductor-Metal(MSM)photodetectors using Cr/Au electrodes.Dark current of the device using MXene-GaN van der Waals junctions is reduced by three orders of magnitude and its noise spectral intensity shows distinct improvement compared with the traditional Cr/Au–GaN–Cr/Au MSM photodetector.The improved device performance is attributed to low-defect MXene-GaN van der Waals interfaces.Thanks to the high quality MXene-GaN interfaces,it is possible to verify that the patterned substrate can locally improve both light extraction and photocurrent collection.The measured responsivity and specific detectivity reach as high as 64.6 A/W and 1.93×1012 Jones,respectively,making it a potential candidate for underwater optical detection and communication.The simple fabrication of MXene-GaN-MXene photodetectors spearheaded the way to high performance photodetection by combining the advantages of emerging 2D MXene materials with the conventional III-V materials.Lingzhi Luo Yixuan Huang Keming Cheng Abdullah Alhassan Mahdi Alqahtani Libin Tang Zhiming Wang Jiang Wu 2021Light(Science & Applications)2021,10,10:1

17Hemodynamic Shear Stress Regulates the Survival of Circulating Tumour Cells Through Piezo1-Actomyosin-Mediated YAP/TAZ Nuclear Translocation显示文摘It is critical to combat tumor metastasis by eradicating disseminated tumor cells in any step during the metastasis process.After entering the blood circulation system,tumor cells are in suspension and experience considerable levels of fluid shear stress.However,the influence of hemodynamic shear stress on the survival of CTCs and the underlying mechanotransduction mechanisms remain unclear.This study shows that fluid shear stress can eliminate the majority of CTCs and the viability of suspended tumor cells depends on the stress magnitude,indicating that tumor cells can sense and respond to fluid shear stress.Mechanistically,the expression of Piezo1 but not Piezo2 is greatly upregulated in suspended tumor cells after shear stress treatment.Inhibiting/activating Piezo1 increases/decreases the viability of suspended tumor cells in shear flow,which depends on Piezo1-mediated calcium entry.These findings suggest that Piezo1 may be the major mechanosensor by which suspended tumor cells sense fluid shear stress.As the downstream effector of Piezo1,actomyosin in tumor cells is significantly activated under increasing shear stress.Its activity influences the survival of CTCs in shear flow and rescues the effects of Piezo1 on tumor cell survival,suggesting that hemodynamic shear stress regulates the survival of CTCs through Piezo1 mediated actomyosin activity.Importantly,fluid shear stress considerably up-regulates YAP/TAZ activity of suspended tumor cells and promotes their nuclear translocation in a magnitude-dependent manner.Inhibiting YAP/TAZ enhances the viability of suspended tumor cells in shear stress,while activating their activity decreases tumor cell survival,suggesting that YAP/TAZ activation promotes the apoptosis of suspended tumor cells,which is different from the findings that YAP/TAZ facilitates the survival of adherent cells to shear flow.Further,blocking the nuclear import of YAP/TAZ inactivates the sensation of suspended tumor cells to fluid shear flow and attenuates the dependence of tumor cell survival on different magnitudes of hemodynamic shear stress.The influence of Piezo1-actomyosin pathway on suspended tumor cells can be rescued by YAP/TAZ activity,suggesting that Piezo1-mediated signaling induces tumor cell apoptosis via nuclear translocation of YAP/TAZ.In addition,fluid shear stress can also activate the expressions of LATS1/2 and MST1/2 in Hippo pathway through Piezo1,which is known to inhibit YAP/TAZ activity.Silencing/activating LATS1/2 or MST1/2 inhibits/enhances the viability of CTCs under shear stress,the effects of which can be further rescued by YAP/TAZ.These findings suggest that the responses of suspended tumor cells to hemodynamic shear stress are partially mediated by Hippo signaling.After nuclear translocation,YAP/TAZ directly bind p73/PUMA,which further promotes the transcription of pro-apoptotic genes and induces the apoptosis of suspended tumor cells.In summary,these findings show that hemodynamic shear stress considerably influences the survival of CTCs in blood circulation.We have identified the calcium channel Piezo1 as a novel mechanosensor for the response of CTCs to fluid shear stress.Hemodynamic shear stress induces the apoptosis of suspended tumor cells through Piezo1-actomyosin-YAP/TAZ-p73/PUMA signaling,which is different from the mechanotranduction mechanisms for tumor cells in adherent.Therefore,this study has unveiled the novel mechanosensor of suspended CTCs in response to fluid shear stress and the subsequent mechanisms and identified Piezo1 and YAP/TAZ as the potential therapeutic targets,through which CTCs may be effectively eradicated in the vasculature to prohibit tumor metastasis.Keming Li Ying XIN Kai Tang Youhua Tan 2019医用生物力学2019,34,A01:0
