1Activation of gibberellin 2-oxidase 6 decreases active gibberellin levels and creates a dominant semi-dwarf phenotype in rice (Oryza sativa L.)显示文摘Gibberellin (GA) 2-oxidase plays a key role in the GA catabolic pathway through 2β-hydroxylation.In the present study,we isolated a CaMV 35S-enhancer activation tagged mutant,H032.This mutant exhibited a dominant dwarf and GA-deficient phenotype,with a final stature that was less than half of its wild-type counterpart.The endogenous bioactive GAs are markedly decreased in the H032 mutant,and application of bioactive GAs (GA3 or GA4) can reverse the dwarf phenotype.The integrated T-DNA was detected 12.8 kb upstream of the OsGA2ox6 in the H032 genome by TAIL-PCR.An increased level of OsGA2ox6 mRNA was detected at a high level in the H032 mutant,which might be due to the enhancer role of the CaMV 35S promoter.RNAi and ectopic expression analysis of OsGA2ox6 indicated that the dwarf trait and the decreased levels of bioactive GAs in the H032 mutant were a result of the up-regulation of the OsGA2ox6 gene.BLASTP analysis revealed that OsGA2ox6 belongs to the class III of GA 2-oxidases,which is a novel type of GA2ox that uses C20-GAs (GA12 and/or GA53) as the substrates.Interestingly,we found that a GA biosynthesis inhibitor,paclobutrazol,positively regulated the OsGA2ox6 gene.Unlike the over-expression of OsGA2ox1,which led to a high rate of seed abortion,the H032 mutant retained normal flowering and seed production.These results indicate that OsGA2ox6 mainly affects plant stature,and the dominant dwarf trait of the H032 mutant can be used as an efficient dwarf resource in rice breeding.Jian Huang Ding Tang Yi Shen Baoxiang Qin Lilan Hong Aiqing You Ming Li Xin Wang Hengxiu Yu Minghong Gu Zhukuan Cheng 2010Journal of Genetics and Genomics2010,37,1:36
2Wx^lv、the Ancestral Allele of Rice Waxy Gene显示文摘In rice grains,the Waxy (Wx) gene is responsible for the synthesis of amylose,the most important determinant for eating and cooking quality.The effects of several Wx alleles on amylose content and the taste of cooked rice have been elucidated.However,the relationship between artificial selection and the evolution of various Wx alleles as well as their distribution remain unclear.Here we report the identification of an ancestral allele,Wx^lv,which dramatically affects the mouthfeel of rice grains by modulating the size of amylose molecules.We demonstrated that WF originated directly from wild rice,and the three major Wx alleles in cultivated rice (Wx^b,Wx^a,and Wx^in) differentiated after the substitution of one base pair at the functional sites.These data indicate that the Wx^lv allele played an important role in artificial selection and domestication.The findings also shed light on the evolution of various Wx alleles,which have greatly contributed to improving the eating and cooking quality of rice.Changquan Zhang Jihui Zhu Shengjie Chen Xiaolei Fan Qianfeng Li Yan Lu Min Wang Hengxiu Yu Chuandeng Yi Shuzhu Tang Minghong Gu Qiaoquan Liu 2019Molecular Plant2019,12,8:22

4Centromere inactivation in a dicentric rice chromosome during sexual reproduction显示文摘T0135 is a variant selected from the progeny of a rice line telotrisomic for the short arm of chromosome 11(2n+11S·).Fluorescent in situ hybridization(FISH)results indicated that T0135 contained two telocentric chromosomes,which have two centromere-specific molecular markers(5S rDNA)for chromosome 11;thus T0135 is a newly-described rice chromosome variant with two dicentric chromosomes,named 22+11L·+11L·+11S·11S·+11S·11S·(22 represents the 22 chromosomes excluding chromosome 11 in the rice genome,'·'represents the centromere).To investigate the genetic stability of the rice dicentric chromosomes during sexual reproduction,we observed the chromosome types in the progeny.Ninety-four percent of the progeny had the same chromosome type as the parental line.This result indicates that the dicentric chromosomes are mostly stable during mitosis and meiosis.Immunofluorescence analysis for centromere specific histone H3(CENH3)revealed that only one centromere is active and the other centromere is inactivated in the rice dicentric chromosomes.GONG ZhiYun XUE Chao LIU XiuXiu ZHANG MingLiang ZHOU Yong YU HengXiu GU MingHong 2013Chinese Science Bulletin2013,58,36:3

6Isolation and identification of the three rice monotelosomics显示文摘In order to obtain rice monotelosomic, the progeny of 24 telotrisomics, derived from an indica rice vari-ety, Zhongxian 3037, were screened. The variants that dif-fered morphologically from the diploids and the original primary trisomics as well as the telotrisomics were collected for cytological identification. The variants with 24 chromo-somes were selected according to the prometaphase chromo-somes. From these variants, three monotelosomics with one chromosome arm deletion in each were verified by fluores-cence in situ hybridization (FISH) using a rice centromeric BAC clone of 17p22 as a marker probe. The three monote-losomics were derived from telotrisomic 1S, 4L and 11L, re-spectively. Further identification was conducted on the pro-metaphase or pachytene chromosomes of the three variants, which were probed with the same centromeric BAC clone together with the corresponding chromosome arm specific makers, a0059H02 (on the short arm of chromosome 1), a0034E24 (on the long arm of chromosome 4), and a0071H11 (on the long arm of chromosome 11). The results indicated that the telocentric chromosomes in the three monotelosom-ics were derived from their respective corresponding telotrisomics. 7Generation of selectable marker-free soft transgenic rice with transparent kernels by downregulation of SSSⅡ-2显示文摘The amylose content(AC)of rice endosperm starch varies from 0 to 35%,and is associated with rice cooking and eating quality.Soft rice has low AC,generally between 6%and 15%,and its eating quality is high whether it is consumed hot or cold.However,the appearance quality of current soft rice cultivars needs to be improved,especially opaque endosperm.Conventional genetic engineering has improved some agronomic traits of soft rice varieties,but not endosperm appearance.In the present study,a RNAi construct of the soluble starch synthase II-2(SSSII-2)and the hygromycin phosphotransferase(HPT)gene were introduced into an elite japonica rice variety,Kangtiaowuyunjing(KWY8)by co-transformation.Several selectable marker-free(SMF)transgenic lines were obtained,and SSSII-2 expression was significantly downregulated in selected transgenic lines,resulting in lower AC of the endosperm.The physicochemical properties of the transgenic rice kernels,including gel consistency(GC)and rapid visco analyzer(RVA)profile,differed significantly from those of wild-type rice and were similar to those of a soft rice variety,Nanjing 46(NJ46).These findings indicate that the cooking,eating,and processing qualities of transgenic rice are comparable to those of NJ46.However,the transgenic rice endosperm retained a transparent appearance under low-moisture conditions.Thus,SMF SSSII-2 RNAi rice provides a resource for breeding soft rice with transparent endosperm.Zhihao Xu Meimei Yu Youwei Yin Cenwen Zhu Wen Ji Changquan Zhang Qianfeng Li Honggen Zhang Shuzu Tang Hengxiu Yu Qiaoquan Liu 2020The Crop Journal2020,8,1:2

14Generating of rice OsCENH3-GFP transgenic plants and their genetic applications显示文摘In order to investigate rice functional centromeres, OsCENH3-GFP chimeric gene was constructed and transformed into the indica rice variety, Zhongxian 3037, mediated by Agrobacturium. The integration of the exogenous genes in the transgenic plants was confirmed by PCR and Southern blotting. The transgenic plants grow normally during their whole life time, just like Zhongxian 3037. No significant defects were detected in either mitosis or meiosis of the transgenic plants. The overlapping of GFP signals and anti-CENH3 foci in both mitotic and meiotic cells from T0 and T1 generation plants indicated that GFP had been successfully fused with CENH3, so the GFP signals can well represent the CENH3 locations on each chromosome. To evaluate the applicability of the transgenic plants to other genetic studies, fluorescence in situ hybridization (FISH) using rice centromeric tandem repetitive sequence CentO as the probe was conducted on the zygotene chromosomes of pollen mother cells (PMCs). It has been revealed that the GFP signals are overlapping with CentO FISH signals, showing that CentO is one of the key elements constituting rice functional centromeres. Immunofluorescent staining using anti-α-tublin antibody and anti-PAIR2 antibody on the chromosomes during mitosis and meiosis stages of the transgenic plants further reveals that OsCENH3-GFP transgenic plants can be widely used for studying rice molecular biology, especially for tagging functional centromeres in both living cells and tissues.YU HengXiu WANG Xin GONG ZhiYun TANG Ding GU MingHong CHENG ZhuKuan 2008Chinese Science Bulletin2008,53,19:0
