| 1 | ζ-Carotene Isomerase Suppresses Tillering in Rice through the Coordinated Biosynthesis of Strigolactone and Abscisic Acid显示文摘Rice tillering is an important agronomic trait affecting grain yield.Here,we identified a high-tillering mutant tillering20(t20),which could be restored to the wild type by treatment with the strigolactone(SL)analog rac-GR24.T20 encodes a chloroplast ζ-carotene isomerase(Z-ISO),which is involved in the biosynthesis of ca-rotenoids and their metabolites,SL and abscisic acid(ABA).The t20 mutant has reduced SL and ABA,raising the question of how SL and ABA biosynthesis is coordinated,and whether they have overlapping functions in tillering.We discovered that rac-GR24 stimulated T20 expression and enhanced all-trans-p-carotene biosynthesis.Importantly,rac-GR24 also stimulated expression of Oryza sativa 9-CIS-EPOXY-CAROTENOID DIOXYGENASE 1(OsNCED1)through induction of Oryza sativa HOMEOBOX12(0sHOX12),promoting ABA biosynthesis in shoot base.On the other hand,ABA treatment significantly repressed SL biosynthesis and the ABA biosynthetic mutants displayed elevated SL biosynthesis.ABA treatment reduced the number of basal tillers in both t20 and wild-type plants.Furthermore,while ABA-deficient mu-tants aba1 and aba2 had the same number of basal tillers as wild type,they had more unproductive upper tillers at maturity.This work demonstrates complex interactions in the biosynthesis of carotenoid,SLs and ABA,and reveals a role for ABA in the regulation of rice tillering. | Xue Liu Qingliang Hu Jijun Yan Kai Sun Yan Liang Meiru Jia Xiangbing Meng Shuang Fang Yiqin Wang Yanhui Jing Guifu Liu Dianxing Wu Chengcai Chu Steven M.Smith Jinfang Chu Yonghong Wang Jiayang Li Bing Wang | 2020 | Molecular Plant2020,13,12: | 11 |
| 2 | Expression and Chromosomal Mapping of Mouse Gpx2 Gene Encoding the Gastrointestinal Form of Glutathione Peroxidase, GPX-GI显示文摘GPX-GI is a cytosolic tetrameric Se-dependent glutathione peroxidase, similar in properties to GPX-1. Unlike the almost ubiquitous GPX-1, GPX-GI is mainly expressed in the epithelium of gastrointestinal tract. GPX-GI contributes to at least fifty percent of GPX activity in rodent small intestmal epithelium. The total GPX activity consists of at least 70% of selenium-dependent GPX activity in this compartment.By analyzing a panel of mouse mterspecies DNA from the Jackson Laboratory’s backcross resource,we mapped Gpx2 gene to mouse chromosome 12 between D12Mit4 and D12Mit5, near the Ccs1 locus which contains a colon cancer susceptibility gene. A pseudogene, Gpx2-ps is mapped to mouse chromosome 7.Comparison of Gpx2 gene expression in three pairs of C57BL/6Ha and ICR/Ha mice which are respectively resistant and sensitive to dimethylhydrazine-induced colon cancer, we found a higher Gpx2 mRNA level in C57BL/6Ha colon than ICR/Ha colon. Interestingly, a lower level of GPX activity is found in the resistant strain of mice. Because GPX-1 has three times higher specific activity than GPX GI, our data suggest that the decreased GPX activity may result from a higher level of Gpx2 gene expression in those cells co-express GPx1 | FONG-FONG CHU R. | 4 | Soil microbial activity and community structure as affected by exposure to chloride and chloride-sulfate salts显示文摘Mixed or chloride salty ions dominate in saline soils, and exert wide-ranging adversely affect on soil biological processes and soil functions. The objectives of this study were to(1) explore the impacts of mixed(0, 3, 6, 10, 20 and 40 g Cl~–/SO_4^(2–)salt/kg dry soil) and chloride(0, 1.5, 3, 5, 8 and 15 g Cl~– salt/kg dry soil) salts on soil enzyme activities, soil physiological functional(Biolog) profiles and microbial community structure by using soil enzymatic, Biolog-Eco microplates as well as denaturing gradient gel electrophoresis(DEEG) methods, and(2) determine the threshold concentration of soil electronic conductivity(EC_(1:5)) on maintaining the functional and structural diversity of soil microbial community. The addition of either Cl~– or mixed Cl~–/SO_4^(2–)salt obviously increased soil EC, but adversely affected soil biological activities including soil invertase activity, soil microbial biomass carbon(MBC) and substrate-induced respiration(SIR). Cl~– salt showed a greater deleterious influence than mixed Cl~–/SO_4^(2–)salt on soil enzymes and MBC, e.g., the higher soil MBC consistently appeared with Cl~–/SO_4^(2–)instead of Cl– treated soil. Meanwhile, we found that SIR was more reliable than soil basal respiration(SBR) on explaining the changes of soil biological activity responsive to salt disturbance. In addition, microbial community structures of the soil bacteria, fungi, and Bacillus were obviously affected by both salt types and soil EC levels, and its diversity increased with increasing of mixed Cl~–/SO_4^(2–)salt rates, and then sharply declined down after it reached critical point. Moreover, the diversity of fungal community was more sensitive to the mixed salt addition than other groups. The response of soil physiological profiles(Biolog) followed a dose-response pattern with Cl~–(R^2=0.83) or mixed Cl~–/SO_4^(2–)(R^2=0.89) salt.
