1Effects of a fatty acid synthase inhibitor on adipocyte differentiation of mouse 3T3-L1 cells显示文摘AIM: To investigate the influence of C75, a fatty acid synthase inhibitor, on adipocyte differentiation. METHODS: Mouse 3T3-L1 preadipocytes were induced to differentiation by insulin, isobutylmethylxanthine, and dexamethasone. Oil red O staining was performed and activity of glycerol-3-phosphate dehydrogenase (GPDH) was measured. The level of peroxisome proliferators-activated receptor γ (PPARγ) mRNA was assayed by semi-quantitative reverse transcription PCR. RESULTS: C75 blocked the adipogenic conversion in a dose-dependent manner and the inhibitory effects occurred both in the early phases (48 h) and in the latter phases (8 d) of the process. Treatment with C75 for 8 d induced more decrease in lipid content than 48 h (P<0.01). Treatment with C75 50 mg/L for 48 h or 8 d decreased GPDH activity by 52.8 % and 31.2 % of Vehicle, respectively. Treatment with C75 10-50 mg/L for 48 h or 8 d down-regulated PPARγ mRNA expression compared with control (P<0.01). 2Novel resonant pressure sensor based on piezoresistive detection and symmetrical in-plane mode vibration显示文摘In this paper,a novel resonant pressure sensor is developed based on electrostatic excitation and piezoresistive detection.The measured pressure applied to the diaphragm will cause the resonant frequency shift of the resonator.The working mode stress–frequency theory of a double-ended tuning fork with an enhanced coupling beam is proposed,which is compatible with the simulation and experiment.A unique piezoresistive detection method based on small axially deformed beams with a resonant status is proposed,and other adjacent mode outputs are easily shielded.According to the structure design,high-vacuum wafer-level packaging with different doping in the anodic bonding interface is fabricated to ensure the high quality of the resonator.The pressure sensor chip is fabricated by dry/wet etching,high-temperature silicon bonding,ion implantation,and wafer-level anodic bonding.The results show that the fabricated sensor has a measuring sensitivity of~19 Hz/kPa and a nonlinearity of 0.02%full scale in the pressure range of 0–200 kPa at a full temperature range of−40 to 80°C.The sensor also shows a good quality factor>25,000,which demonstrates the good vacuum performance.Thus,the feasibility of the design is a commendable solution for high-accuracy pressure measurements.Xiangguang Han Qi Mao Libo Zhao Xuejiao Li Li Wang Ping Yang Dejiang Lu Yonglu Wang Xin Yan Songli Wang Nan Zhu Zhuangde Jiang 2020Microsystems & Nanoengineering2020,6,1:4
3Efficacy of inverso isomer of CendR peptide on tumor tissue penetration显示文摘The dense extracellular matrix and high interstitial fluid pressure of tumor tissues prevent the ability of anti-tumor agents to penetrate deep into the tumor parenchyma for treatment effects. C-end rule(CendR) peptides can enhance the permeability of tumor blood vessels and tumor tissues via binding to neuropilin-1(NRP-1), thus aiding in drug delivery. In this study, we selected one of the CendR peptides(sequence RGERPPR) as the parent L-peptide and substituted D-amino acids for the L-amino acids to synthesize its inverso peptide D(RGERPPR). We investigated the NRP-1 binding activity and tumorpenetrating ability of D(RGERPPR). We found that the binding affinity of D(RGERPPR) with NRP-1 and the cellular uptake was significantly higher than that of RGERPPR. Evans Blue tests revealed that D(RGERPPR) exhibited improved tumor-penetrating ability in C6, U87 and A549 tumor-bearing nude mice. Using nude mice bearing A549 xenograft tumors as a model, we found that the rate of tumor growth in the group co-administered with D(RGERPPR) and gemcitabine(Gem) was significantly lower than the gemcitabine-treated group with a tumor suppression rate(TSR%) of 55.4%. 4A pulsed switching peroxi-coagulation process to control hydroxyl radical production and to enhance 2,4-Dichlorophenoxyacetic acid degradation显示文摘The aim of this study was to develop a new pulsed switching peroxi-coagulation system to control hydroxyl radical(∙OH)production and to enhance 2,4-Dichlorophenoxyacetic acid(2,4-D)degradation.The system was constructed with a sacrifice iron anode,a Pt anode,and a gas diffusion cathode.Production of H_(2)O_(2) and Fe^(2+)was controlled separately by time delayers with different pulsed switching frequencies.Under current densities of 5.0 mA/cm^(2)(H_(2)O_(2))and 0.5 mA/cm^(2)(Fe^(2+)),the∙OH production was optimized with the pulsed switching frequency of 1.0 s(H_(2)O_(2)):0.3 s(Fe^(2+))and the ratio of H_(2)O_(2) to Fe^(2+)molar concentrations of 6.6.Under the optimal condition,2,4-D with an initial concentration of 500 mg/L was completely removed in the system within 240 min.The energy consumption for the 2,4-D removal in the system was much lower than that in the electro-Fenton process(686 vs.13610 kWh/kg TOC).The iron consumption in the system was~20 times as low as that in the peroxi-coagulation process(19620 vs.3940400 mg/L)within 240 min.The system should be a promising peroxi-coagulation method for organic pollutants removal in wastewater.Yaobin Lu Songli He Dantong Wang Siyuan Luo Aiping Liu Haiping Luo Guangli Liu Renduo Zhang 2018Frontiers of Environmental Science & Engineering2018,12,5:3
5A model of 3D-structure of H+, K+-ATPase catalytic subunit derived by homology modeling显示文摘AIM: To build a model of 3D-structure of H+, K+-ATPase catalytic subunit for theoretical study and anti-ulcer drug design. METHODS: The model was built on the basis of structural data from the Ca2+-ATPase. Structurally conserved regions were defined by amino acid sequence comparisons, optimum interconnecting loops were se- lected from the protein databank, and amino (N)- and carboxyl (C)-terminal ends were generated as random coil structures. Applying molecular mechanics method then minimized the model energy. Molecular dynamics tech- nique was used to do further structural optimization. RESULTS: The model of 3D-structure of H+, K+-ATPase was derived. The model is reasonable according to several validation criteria. There were ten transmembrane helices (TM1-TM10) in the model and inhibitor-binding site was identified on the TM5-8 riched negatively charged residues. 6Optimal scheduling strategy for virtual power plants based on credibility theory显示文摘The virtual power plant(VPP)is a new and efficient solution to manage the integration of distributed energy resources(DERs)into the power system.Considering the unpredictable output of stochastic DERs,conventional scheduling strategies always set plenty of reserve aside in order to guarantee the reliability of operation,which is too conservative to gain more benefits.Thus,it is significant to research the scheduling strategies of VPPs,which can coordinate the risks and benefits of VPP operation.This paper presents a fuzzy chance-constrained scheduling model which utilizes fuzzy variables to describe uncertain features of distributed generators(DGs).Based on credibility theory,the concept of the confidence level is introduced to quantify the feasibility of the conditions,which reflects the risk tolerance of VPP operation.By transforming the fuzzy chance constraints into their equivalent forms,traditional optimization algorithms can be used to solve the optimal scheduling problem.An IEEE 6-node system is employed to prove the feasibility of the proposed scheduling model.Case studies demonstrate that the fuzzy chance strategy is superior to conservative scheduling strategies in realizing the right balance between risks and benefits.Qian Ai Songli Fan Longjian Piao 2016Protection and Control of Modern Power Systems2016,1,1:3
7Effects of in-situ ZrB_2 particle on grain refinement of ZrB_2/AZ91D magnesium matrix composite显示文摘In-situ ZrB2/AZ91D magnesium matrix composite was successfully synthesized with Al/K2ZrF6+NH4BF4 by means of Direct Melt Reaction.The fabricated ZrB2/AZ91D magnesium matrix composite through direct melt mixing method was investigated.Results from X-ray diffraction (XRD) and energy dispersive X-ray spectroscopy (EDS) confirmed the existence of ZrB2 particles in the AZ91D alloy,and most ZrB2 particles were in the size range of just several microns,some even to 100 nm.The cast specimens were studied through corrosion testing and heat treatment.The average grain size of AZ91D decreased markedly from about 250μm to 50μm.In addition,the shape and size of the β-Mg17Al12 phase as well as the morphologies of primary α-Mg in the magnesium matrix composite were greatly changed.The network structure of the β-Mg17Al12 phase was broken into small blocks and the size of α-Mg decreased significantly.Wu Jiangjing Zhao Yutao Zhang Songli Chen Gang Zhang Zhenya 2012China Foundry2012,9,1:2

12Comparative analysis of symbiotic phenotypes of soybean ’’Tianlong 1’’ with two different rhizobia and co-inoculation at different developmental stages显示文摘Symbiotic nitrogen fixation between soybean and rhizobia, which directly catalyze reduction of atmospheric nitrogen into ammonia, has critical importance in agricultural economy and environment. However, soybean symbiotic nitrogen efficiency, which changes with growth stage, largely relies on symbiotic host-specificity and nodule development. In this report, symbiotic phenotypes of soybean ‘Tianlong 1’ with different inoculation (inoculated with Bradyrhizobium japonicum strain 113-2, Sinorhizobium fredii USDA205 and co-inoculated with 113-2 and USDA205) were comparatively studied at different developmental stages. Without additional nitrogen, chlorophyll content, above ground fresh weight,plant height, nodule number and nodule dry weight changed with different rhizobia at various developmental stages. 113-2 had more advantages than USDA205 in promoting plant growth and nodulation. Nodulation morphology of co-inoculation was more similar to 113-2 than USDA205. Competitive nodulation between two rhizobia promoted plant growth, which had significant agricultural application prospect. 16Engineered platelets-based drug delivery platform for targeted thrombolysis显示文摘Thrombolytic agents have thus far yielded limited therapeutic benefits in the treatment of thrombotic disease due to their short half-life,low targeting ability,and association with serious adverse reactions,such as bleeding complications.Inspired by the natural roles of platelets during thrombus formation,we fabricated a platelet-based delivery system(NO@uPA/PLTs)comprising urokinase(uPA)and arginine(Arg)for targeted thrombolysis and inhibition of re-embolism.The anchoring of uPA to the platelet surface by lipid insertion increased the thrombotic targeting and in vivo circulation duration of uPA without disturbing platelet functions.Nitric oxide(NO)generated by the loaded Arg inhibited platelet aggregation and activation at the damaged blood vessel,thereby inhibiting re-embolism.NO@uPA/PLTs effectively accumulated at the thrombi in pulmonary embolism and carotid artery thrombosis model mice and exerted superior thrombolytic efficacy.In addition,the platelet delivery system showed excellent thrombus recurrence prevention ability in a mouse model of secondary carotid artery injury.The coagulation indicators in vivo showed that the platelet-based uPA and NO co-delivery system possessed a low hemorrhagic risk,providing a promising tool for rapid thrombolysis and efficient inhibition of posttreatment re-embolism.Songli Wang Ruifeng Wang Nana Meng Linwei Lu Jun Wang Jianfen Zhou Jiasheng Lu Qianzhu Xu Cao Xie Changyou Zhan Yao Li Yang Yu Weiyue Lu Min Liu 2022Acta Pharmaceutica Sinica B2022,12,4:1

20Herbivore defense responses and associated herbivore defense mechanism as revealed by comparing a resistant wild soybean with a susceptible cultivar显示文摘Plants have evolved sophisticated defense mechanisms against herbivores to help them adapt to the environment. Understanding the defense mechanisms in plants can help us control insects in a more effective manner. In this study, we found that compared with Tianlong 2(a cultivated soybean with insect susceptibility), ED059(a wild soybean line with insect resistance)contains sharper pubescence tips, as well as lower transcript levels of wound-induced protein kinase(WIPK) and salicylic acid-induced protein kinase(SIPK), which are important mitogen-activated protein kinases involved in early defense response to herbivores. The observed lower transcript levels of WIPK and SIPK induced higher levels of jasmonic acid(JA), JA biosynthesis enzymes(AOC3) and some secondary metabolites in ED059. Functional analysis of the KTI1 gene via Agrobacterium-mediated transformation in Arabidopsis thaliana indicated that it plays an important role in herbivore defense in ED059. We further investigated the molecular response of third-instar Helicoverpa armigera(Hübner) larvae to Tianlong 2 and ED059. We found apoptotic cells only in the midguts of larvae that fed on ED059. Compared with larvae reared on the susceptible cultivar Tianlong 2, transcript levels of catalase(CAT) and glutathione S-transferase(GST) were up-regulated, whereas those of CAR, CHSB, and TRY were down-regulated in larvae that fed on the highly resistant variety ED059.
